Abstract

Ovarian cancer is a serious disease that affects the ovaries, and its early detection is challenging due to vague symptoms often dismissed as minor ailments. Currently, natural sources have gained attention for their potential role in anticancer treatment. This study aimed to utilize network pharmacology to explore the potential targets and mechanisms of Zanthoxylum acanthopodium in the treatment of ovarian cancer. This study utilized the KNApSAcK and Swiss Target Prediction to identify active compounds and target genes. Additionally, ovarian cancer-specific target genes were sourced from the GEO database. To identify possible key target genes, the network interaction between protein-protein using the STRING database and visualized them in Cytoscape. Subsequent analysis using Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways enabled us to focus on primary therapeutic targets. Our investigation into Zanthoxylum acanthopodium revealed 10 active compounds that pass Lipinski rule of five and oral bioavailability with acceptable pharmacokinetic profiles, 88 therapeutic targets, and identified 5 hub genes: SRC, CCNB2, MMP9, PTGS2, and PTPRC, which are strongly associated with ovarian cancer progression. Pathway enrichment analysis highlighted several pathways significantly related to the pathogenesis of ovarian cancer. This study elucidates the therapeutic potential and mechanisms of action of Z. acanthopodium as a promising candidate for ovarian cancer treatment. However, further research, including both in vitro and in vivo studies, is necessary to understand its molecular mechanisms comprehensively.

Abstract

Background: High yield carbohydrate diet is common in Indonesia mainly in North Sumatra province. In the long run consuming primary carbohydrate form rice may increase the risk for diabetic and its complication. Not only the amount of rice but certain beverages also such as tamarillo juice, though it’s rich amount of nutritious benefit, some product may yield more sugar to improve from its sour or bitter flavor. Objectives: studies aim to find the diet correlation between amount of carbohydrate total and Vitamin C intake in fresh and store brought tamarillo juice affect animal model with high sucrose induction. Material and Methods: Fresh tamarillo fruit grew in north Sumatra blended versus store brought juice are given daily towards obese female rats induced with 2 ml of lard and 5% sucrose orally for five weeks. Beforehand sample had its total carbohydrate and Vitamin C content with iodometric method. The last 2 weeks then rats treated with fresh blend and store brought product juice. Results: Total content value obtained from fresh compared with store brought tamarillo juice in vitamin C, (18.63 ± 7.16 mg/ml); (11.57 ± 6.8 mg/ml) and total carbohydrate (15.02 ± 2.35 mg/ml); (25.37 ± 28.17 mg/ml) respectively. During treatment increase in body weight significantly in store brought product. Histology remarks in pancreas do not show any significance damage in cell especially for fresh blend treatment group. Conclusion: Fresh blend tamarillo juice may improve health and can be considered safe for subject that like to consume high carbs in their diet.

Abstract

The degenerative disease of memory that most attacks the frontal lobe is Alzheimer's. Therapy for Alzheimer's is mostly done by targeting an increase in the amount of acetylcholinesterase in the synapse cleft by inhibiting the enzyme acetylcholinesterase. The purpose of this study is to conduct a comparative study of vitexifolin E compounds from the vitexium plant family with donepezil to obtain more precise and targeted drug candidates in Alzheimer's therapy. The results showed that donepezil compounds have a more active probability (0.53) of being carcinogenic compared to vitexifolin E with the web-based application Protox II. Tests with in silico studies found that the hydrogen bonding results of vitexifolin E had values close to those of donepezil drugs, as well as RMSD results with receptors between donepezil and vitexifolin E (1,781; 1,281) and free energy obtained (-6.9; -7.8). Based on these results, it can be concluded that vitexifolin E compounds can have good enough potential to be developed in implementing the treatment of Alzheimer's disease therapy.

Abstract

Stres oksidatif akibat radikal bebas beperan dalam patogenesis penyakit degeneratif, mendorong pemanfaatan antioksidan alami seperti kunyit putih yang kayak fenolik sebagai alternatif yang lebih aman dari sintetis. Penelitian ini mengevaluassi hubungan antara kandungan fenolik dan kapasitas antioksidan kunyit putih menggunakan metode DPPH dan FRAP. Aktivitas antioksidan dan kadar total fenol ekstrak etanol kunyit putih dianalisis menggunakan metode DPPH, FRAP, dan Folin-Ciocalteu. Ekstrak etanol kunyit putih mengandung total fenol 189,21 mg GAE/g dan menunjukkan aktivitas antioksidan sangat kuat dalam menangkal radikal bebas dengan nilai IC50 5,30 ppm (DPPH) dan 24,02 ppm (FRAP).

Abstract

Glibenclamide is an antidiabetic drug that is not permitted for use in traditional herbal medicines because it can cause serious side effects such as hypoglycaemia and organ damage. This study aims to identify the presence of glibenclamide as a Chemicals Drug (CD) in antidiabetic herbal medicines found in Medan Petisah. Two herbal medicine samples, namely Unlabelled Powdered Herbal Medicine and JSR Diabetes Herbal Medicine (Jamu), were analysed using Fourier Transform Infrared Spectroscopy (FTIR). The infrared spectra of the samples were compared with the standard Glibenclamide spectrum to identify characteristic wavenumber patterns. The analysis results showed similarities in the patterns at 3280 cm⁻¹ (N–H bond), 2922 cm⁻¹ (C–H bond), and 1237–1282 cm⁻¹ (S=O bond), confirming the presence of Glibenclamide in both samples. These findings indicate a violation of Indonesian Minister of Health Regulation No. 007 of 2012, which prohibits the use of Chemicals Drug (CD) in traditional preparations. Therefore, this study recommends strict supervision of the distribution of antidiabetic herbal medicines and public education about the dangers of consuming herbal medicines containing CD to prevent health risks.
