Abstract

The Effect of Culture Container and Carbon Source Concentration on In Vitro Shoot Propagation of Atlantic PotatoABSTRACTPotato is a food commodity that has the potential to support food diversification in Indonesia. There is an increasing demand for Atlantic potatoes as the raw material for processed potato products. The demand, which has not been met by the increased production, has been the cause of the ongoing potato import activities in Indonesia. The limitation of producing quality Atlantic potato seeds economically is one of the obstacles to increasing the production of Atlantic potatoes in Indonesia. The aim of this research was to study the effect of various table sugar concentrations as the carbon source and the type of the culture containers used for Atlantic potato shoot multiplication in vitro. The propagation was carried out in bioreactors and culture bottles with MS liquid medium + coconut water at a concentration of 150 mL/L medium, and 3 concentration levels of table sugar, namely 0; 7.5; and 15 g/L medium. The use of bioreactor significantly increased the height of the Atlantic potato plantlets. The use of bioreactor combined with table sugar addition decreased hyperhydricity level. The highest number of shoots, leaves, and roots were found at the table sugar concentration of 15 g/L medium in both containers.Keywords: bioreactor, micropropagation, shoot culture, Solanum tuberosum, sucrose ABSTRAKKentang merupakan komoditas pangan yang berpotensi mendukung program diversifikasi pangan di Indonesia. Peningkatan permintaan terhadap kentang Atlantik sebagai bahan baku kentang olahan yang tak diimbangi dengan peningkatan produksi kentang Atlantik menjadi penyebab masih berlangsungnya impor kentang Atlantik di Indonesia. Keterbatasan menghasilkan benih kentang Atlantik berkualitas yang ekonomis merupakan salah satu hambatan dalam meningkatkan produksi kentang Atlantik di Indonesia. Penelitian ini bertujuan mempelajari pengaruh variasi konsentrasi sukrosa teknis sebagai sumber karbon dan penggunaan jenis wadah terhadap perbanyakkan tunas kentang Atlantik secara in vitro. Perbanyakkan tunas kentang Atlantik menggunakan media MS cair + 150 mL/L air kelapa dalam wadah bioreaktor dan botol kultur dengan 3 taraf konsentrasi sukrosa, yaitu 0; 7,5; dan 15 g/L media. Penggunaan bioreaktor secara signifikan meningkatkan tinggi planlet kentang Atlantik yang dihasilkan. Penggunaan bioreaktor yang dikombinasikan dengan penambahan sukrosa teknis menurunkan tingkat hiperhidrisitas. Tunas, daun, dan akar terbanyak dihasilkan oleh perlakuan sukrosa teknis 15 g/L media dalam kedua jenis wadah.Kata Kunci: bioreaktor, kultur tunas, mikropropagasi, Solanum tuberosum, sukrosa

Abstract

Genetic Transformation and Expression of Sucrose Phosphate Synthase Mutant in Tomato Plant ABSTRACTSucrose phosphate synthase (SPS) is a key enzyme responsible for sucrose biosynthesis. In its regulation, SPS activity is modulated by an allosteric effector glucose-6-phosphate (G6P) suggested to have an ability to bind SPS N-terminus domain. To understand the role of N-terminus in regulating SPS, the SPS gene was mutated with the deletion of N-terminus domain (∆N-SPS). The ∆N-SPS gen was transformed into tomato plants with 5% transformation efficiency. Three transgenic tomato plant 4.20, 5.5.1, and 5.10 were obtained and confirmed by PCR analysis. Transgenic tomato expression was characterized by enzymatic analysis. Result showed that the G6P allosteric regulation in transgenic ∆N-SPS had lost and the SPS activity increased by 2-fold compared to non-transgenic plant. This showed that N-terminus domain-deleted SPS could be actively expressed in plant. Keywords: enzyme, genetic transformation, N-terminus domain deletion, sucrose phosphate synthase, tomato ABSTRAKSucrose phosphate synthase (SPS) merupakan enzim kunci yang bertanggung jawab dalam sintesis sukrosa. Dalam regulasinya, aktifitas SPS dipengaruhi oleh alosterik efektor glukosa-6-fosfat (G6P) yang diduga dapat berikatan pada domain N-terminus SPS. Untuk mengetahui peran N-terminus pada regulasi SPS, dilakukan mutasi SPS dengan penghilangan domain N-terminus (∆N-SPS). Gen ∆N-SPS diinsersi pada tanaman tomat melalui transformasi genetik dengan efisiensi transformasi 5%. Tiga tanaman transgenik tomat (event4.20; 5.5.1; dan 5.10) didapatkan dan positif terkonfirmasi melalui analisis PCR. Ekspresi mutan dikarakterisasi melalui analisis enzimatik. Hasil menunjukkan bahwa tanaman tomat transgenik ∆N-SPS tidak dipengaruhi regulasi alosterik G6P dan aktifitas SPS 2 kali lipat lebih tinggi daripada tanaman bukan transgenik. Ini menunjukkan bahwa SPS dengan delesi domain N-terminus dapat terekspresi aktif pada tanaman.  Kata Kunci: delesi domain N-terminus, enzim, sucrose phosphate synthase, tomat, transformasi genetik

Abstract

Growth Ability of Jatropha Curcas L. Explants on the In Vitro Media Containing IBA and BAProbiotic product is one of the applications of biotechnology that utilize lactic acid bacteria, especially lactobacilli. Some important requirements for microbes that can be used as probiotic include resistance to low pH, ability to grow on bile salts and colonize, and having antimicrobial activity. Each species of the genus Lactobacillus has different characteristics. These characteristics are strongly influenced by the environment in which the bacteria live. This study was carried out in order to characterize Lactobacillus casei which was isolated from dadih. The result of the experiment showed that the isolated L. casei was able to grow on the bile salt at the concentration of 15%, resistant to acid media until pH 2, had antimicrobial activity (significantly inhibited the growth of Escherichia coli, Staphylococcus aureus, and Enterococcus faecalis). The local L. casei isolate has a potential application for use as probiotic microbe.Keywords: Lactobacillus casei, probiotic, lactic acid bacteria, characterization,dadih ABSTRAKProduk probiotik merupakan salah satu aplikasi bioteknologi yang memanfaatkan bakteri asam laktat terutama jenis Lactobacillus. Beberapa syarat utama mikroba yang dapat difungsikan sebagai mikroba probiotik antara lain tahan terhadap pH rendah, mampu tumbuh pada garam empedu, mampu berkoloni, memiliki aktivitas antimikroba. Masing-masing spesies dari Genus Lactobacillus memiliki karakteristik yang berbeda-beda. Karakteristik ini sangat dipengaruhi oleh lingkungan dimana bakteri tersebut hidup. Pada penelitian ini telah dilakukan karakterisasi Lactobacillus casei yang merupakan hasil isolasi dari susu kerbau fermentasi. Dari hasil percobaan menunjukkan bahwa L.casei hasil isolasi mampu hidup sampai dengan konsentrasi garam empedu 15%, tahan terhadap media asam sampai dengan pH 2, memiliki aktivitas antimikroba (positif menghambat Escherichia coli, Staphylococcus aureus, dan Enterococcus faecalis). Abstract

Cephalosporin is a β-lactam antibiotic produced by Acremonium chrysogenum using submerged fermentation. Carbon and nitrogen are the most influential medium ingredients for cephalosporin formation. The purpose of this study was to obtain the best composition of media for cephalosporin C production. Response surface methodology was used for production optimization. The results showed that molasses of 70 g/Lwas the best carbon source, while the best nitrogen source was the combination of corn steep liquor, urea and ammonium sulphate. DL-methionine, carbon, and nitrogen source significantly affected  the production of cephalosporin C. The mathematically modelled optimization showed that the highest production of cephalosporin C (3876 mg/L) was obtained using medium composition of 68.28 g/L molasses, 71.61 g/L nitrogen, and 0.4 g/L DL-methionine. Laboratory verification using the same medium composition produced 3696 mg/L of cephalosporin C, being 4.65% different from the mathematically optimized results. Medium optimization increased the cephalosprin C production which was 1.48 times higher than that using the previous medium, where the maximum production was only 2487 mg / L.Keywords: Carbon,  cephalosporin C, cultivation medium, nitrogen, A. chrysogenum ABSTRAKSefalosporin C adalah golongan antibiotik β-lactam yang dihasilkan Acremonium chrysogenum melalui fermentasi cair. Komponen yang sangat berpengaruh terhadap produksi sefalosporin C adalah sumber karbon dan nitrogen. Penelitian ini bertujuan mendapatkan komposisi media terbaik untuk produksi sefalosporin C. Optimasi dilakukan menggunakan metode respon permukaan. Hasil menunjukkan bahwa molases 70 g/L adalah sumber karbon terbaik dan kombinasi corn steep liquor, urea dan ammonium sulfat adalah sumber nitrogen terbaik. DL-methionin, sumber karbon, dan nitrogen berpengaruh nyata terhadap produksi sefalosporin C. Optimasi menggunakan model matematika menunjukkan produksi sefalosporin C tertinggi (3876 mg/L) yang diperoleh dengan komposisi media 68,28 g/L molases, 71,61 g/L nitrogen, dan  0,4 g/L DL-methionin. Verfikasi di laboratorium menggunakan komposisi media yang sama menghasilkan sefalosporin C sebesar 3696 mg/L, berbeda 4,65% dibanding dengan hasil optimasi matematis. Optimasi media mampu meningkatkan produksi sefalosprin C sebesar 1,48 kali dibanding media yang digunakan sebelumnya, dimana maksimal hanya menghasilkan 2487 mg/L.Kata kunci: Karbon, sefalosporin C, media kultivasi, nitrogen, A. chrysogenum

Abstract

Review on Rapid Diagnosis Method and Treatment of Salmonella typhi Infection ABSTRACTSalmonella is a genus of gram-negative bacilli which are pathogenic for human. Recently over 2,500 serotypes of Salmonella have been reported. Of these, the most common serotype causing typhoid fever which is acute infectious disease in small intestine due to S. typhi entering the body through contaminated food or drink. S. typhi infection remains a major public health concern worldwide because of the subsequent economic burden for the cost of surveillance, prevention, and treatment. In Indonesia, typhoid fever is an endemic disease that threatens public health and becomes a complex problem because it increases career cases and drug resistance, so its diagnosis is needed. Although there is already a diagnosis method of typhoid fever conventionally, a fast, easy and reliable diagnosis method is needed to diagnose typhoid fever by medical personnel in endemic countries. Typhoid fever is treated by antibiotics and prevention efforts are carried out through vaccination.Keywords: antibiotics, pathogen, rapid detection, Salmonella typhi, typhoid fever ABSTRAKSalmonella adalah bakteri basil gram negatif yang bersifat patogen terhadap manusia dan saat ini telah dilaporkan lebih dari 2.500 serotipe. Salah satu serotype Salmonella diketahui menyebabkan penyakit demam tifoid yaitu infeksi akut pada usus halus akibat S. typhi yang masuk ke dalam tubuh melalui makanan dan minuman yang tercemar. Infeksi S. typhi menjadi masalah utama dalam kesehatan masyarakat di seluruh dunia karena bebani ekonomi yang ditimbulkannya untuk biaya pengawasan, pencegahan, dan pengobatan. Di Indonesia, demam tifoid merupakan penyakit endemis yang mengancam kesehatan masyarakat dan menjadi masalah kompleks karena demam tifoid meningkatkan kasus-kasus karier dan resistensi obat sehingga diperlukan diagnosisnya. Meskipun sudah ada diagnosis demam tifoid secara konvensional, tetapi diperlukan metode diagnosis yang cepat, mudah dan andal untuk mendeteksi demam tifoid oleh tenaga medis yang bekerja di negara-negara endemik. Abstract

Dedak padi dan ampas kelapa dicampur dengan perbandingan tertentu dan kemudian difermentasi menggunakan Rhizopus oryzae untuk pakan ikan. Uji pemberian pakan lalu dilakukan untuk mengetahui pengaruh pakan terhadap pertumbuhan ikan mas (Cyprinus carpio L.). Dalam penelitian ini digunakan 5 perlakuan: satu perlakuan pakan tanpa fermentasi (pakan komersial 100%), dan empat perlakuan pakan fermentasi substrat campuran bekatul dan ampas kelapa dengan empat perbandingan yang berbeda, yakni 75%:25%, 50%:50%, 25%:75%, dan 0%:100%. Hasil penelitian menunjukkan bahwa pemberian pakan komersial 100% (protein sejati 15,25% dan serat kasar 6,27%) memperlihatkan hasil terbaik terhadap pertumbuhan ikan mas dengan pertambahan bobot badan 2,56 g dan rasio konversi pakan 1,95. Sementara itu pemberian pakan fermentasi (protein sejati berkisar 4,89-9,97% dan serat kasar 22,87-25,70%) hanya menghasilkan pertambahan bobot badan ikan pada kisaran 0,47-0,64 g dengan rasio konversi pakan 2,50-2,64. Dengan demikian pakan fermentasi tersebut mampu mendorong pertumbuhan ikan mas meskipun masih kurang optimal dibandingkan dengan pakan komersial.Kata Kunci: Rhizopus oryzae, Cyprinus carpio, rice bran, coconut bagasse, fermentation  ABSTRACTRice bran and coconut bagasse were mixed and then fermented using Rhizopus oryzae for preparing aquafeed. Subsequent feeding test was carried out to determine the effect on the growth of carps (Cyprinus carpio L). Five feeding treatments were employed, one unfermented feed (commercial feed 100%), and the other four feeds produced by fermentation using substrate mixture of rice bran and coconut pulp in four different ratios, namely 75%:25%, 50%:50%, 25%:75%, and 0%:100%. The results showed that feeding 100% commercial feed (true protein 15.25% and crude fibre 6.27%) showed the best results on the fish growth with body weight gain of 2.56 g and feed conversion ratio of 1.95. Meanwhile, feeding fermented feeds (true protein 4.89-9.97% and crude fiber 22.87-25.70%) only resulted in body weight gain in the range of 0.47 to 0.64 g with feed conversion ratio of 2.50 to 2.64. Thus, the fermented feeds promoted growth in tested carps albeit less optimally than commercial feed.Keywords: Rhizopus oryzae, Cyprinus carpio, dedak, ampas kelapa, fermentasi

Abstract

In detergent industry, enzymes are used enormously in terms of quantity and economic value. Lipase catalyzes the hydrolysis of triglycerides into diglycerides and monoglycerides by releasing fatty acids. Lipase is produced by bacteria, fungi, and yeasts. This study aims to determine the potential of Alcaligenes faecalis lipase  for its application  as biodetergen, through stability testing of its lipase activity against detergent components by exposing the enzyme to the commercial detergents, as well as performance testing through washing. Alcaligenes faecalis lipase was produced using Luria Bertani (LB) culture medium supplemented with 1% olive oil inducer. Production is carried out for 24 hours, and the enzyme was harvested at the 18th hour. The harvested enzyme was tested for their stability after being exposed to commercial detergents at a concentration of 1-5%. Results showed that the exposure to the detergents decreased the enzyme activity to 22, 38, 48, 68 and 90%. Performance test showed that the olive oil impurity removal from the fabric was 29%.Keywords: Alcaligenes faecalis Lipase, biodetergent, lipase activities, washing test AbstrakPada industri detergen penggunaan enzim sangatlah besar baik secara jumlah maupun nilai ekonomi. Lipase mengkatalis hidrolisis trigliserida menjadi digliserida dan monogliserida dengan membebaskan asam lemak. Lipase dihasilkan oleh bakteri, jamur, dan yeast. Penelitian ini bertujuan untuk mengetahui potensi lipase Alcaligenes faecalis untuk aplikasi biodeterjen, melalui uji stabilitas aktivitas lipase terhadap komponen deterjen dengan memaparkan terhadap deterjen komersial serta uji kinerja melalui washing test. Lipase Alcaligenes faecalis diproduksi menggunakan media Luria Bertani (LB) dengan penambahan induser minyak zaitun 1%. Produksi dilakukan selama 24 jam dengan waktu pemanenan enzim pada jam ke-18. Enzim yang sudah dipanen diuji stabilitasnya setelah dipapar dengan deterjen komersial pada konsentrasi 1-5%. Berdasarkan hasil pengujian aktivitas setelah dipapar terlihat penurunan aktivitas berturut-turut sebesar 22, 38, 48, 68 dan 90%. Hasil uji kinerja menunjukkan bahwa noda minyak zaitun yang hilang dari kain sebesar 29%.Kata kunci: Lipase Alcaligenes faecalis, biodeterjen, aktivitas lipase, washing test
