Abstract

The Effect of Culture Container and Carbon Source Concentration on In Vitro Shoot Propagation of Atlantic PotatoABSTRACTPotato is a food commodity that has the potential to support food diversification in Indonesia. There is an increasing demand for Atlantic potatoes as the raw material for processed potato products. The demand, which has not been met by the increased production, has been the cause of the ongoing potato import activities in Indonesia. The limitation of producing quality Atlantic potato seeds economically is one of the obstacles to increasing the production of Atlantic potatoes in Indonesia. The aim of this research was to study the effect of various table sugar concentrations as the carbon source and the type of the culture containers used for Atlantic potato shoot multiplication in vitro. The propagation was carried out in bioreactors and culture bottles with MS liquid medium + coconut water at a concentration of 150 mL/L medium, and 3 concentration levels of table sugar, namely 0; 7.5; and 15 g/L medium. The use of bioreactor significantly increased the height of the Atlantic potato plantlets. The use of bioreactor combined with table sugar addition decreased hyperhydricity level. The highest number of shoots, leaves, and roots were found at the table sugar concentration of 15 g/L medium in both containers.Keywords: bioreactor, micropropagation, shoot culture, Solanum tuberosum, sucrose ABSTRAKKentang merupakan komoditas pangan yang berpotensi mendukung program diversifikasi pangan di Indonesia. Peningkatan permintaan terhadap kentang Atlantik sebagai bahan baku kentang olahan yang tak diimbangi dengan peningkatan produksi kentang Atlantik menjadi penyebab masih berlangsungnya impor kentang Atlantik di Indonesia. Keterbatasan menghasilkan benih kentang Atlantik berkualitas yang ekonomis merupakan salah satu hambatan dalam meningkatkan produksi kentang Atlantik di Indonesia. Penelitian ini bertujuan mempelajari pengaruh variasi konsentrasi sukrosa teknis sebagai sumber karbon dan penggunaan jenis wadah terhadap perbanyakkan tunas kentang Atlantik secara in vitro. Perbanyakkan tunas kentang Atlantik menggunakan media MS cair + 150 mL/L air kelapa dalam wadah bioreaktor dan botol kultur dengan 3 taraf konsentrasi sukrosa, yaitu 0; 7,5; dan 15 g/L media. Penggunaan bioreaktor secara signifikan meningkatkan tinggi planlet kentang Atlantik yang dihasilkan. Penggunaan bioreaktor yang dikombinasikan dengan penambahan sukrosa teknis menurunkan tingkat hiperhidrisitas. Tunas, daun, dan akar terbanyak dihasilkan oleh perlakuan sukrosa teknis 15 g/L media dalam kedua jenis wadah.Kata Kunci: bioreaktor, kultur tunas, mikropropagasi, Solanum tuberosum, sukrosa

Abstract

Aglaonema sp. is well known as an ornamental plant in Indonesian society. It has leaf with various beautiful patterns. Aglaonema pink katrina is one of the imported hybrids Aglaonema. A method to increase plant yield quickly is tissue culture. The crucial step in plant tissue culture is sterilization. This study aimed to find the best sterilization method by looking at the effect of concentration and immersion time of sodium hypochlorite (NaOCl) as disinfectant to reduce contaminants explant Aglaonema Pink Katrina leaves. In this study, there were 11 groups consisting of positive control, negative control, and immersion with 2,5; 5%; and 10% NaOCl for 1 minute and 3 minutes. Parameters observed included the time of first contamination, percentage of bacterial and fungal contamination, and appearance of explants for 28 days. The DMRT test showed significant differences in all treatments of disinfectant concentration and soaking time.The results show contamination first time occurred in code S0a (negative control), the highest percentage of sterile explants was in code A5, and explants that were still green/fresh were found in codes A5 and A8 after observation.

Abstract

A taxonomic status of Pucuk Seminyak plant is still unclear. This plant has a fruit like Melinjo (Gnetum gnemon) fruit and is often classified as a member of Gnetaceae family. This study aims to determine the taxonomic status of Pucuk Seminyak from Riau using trnL-trnL-trnF intergenic spacer (IGS) and matK sequences. Methods included leaf samples from three individuals collected from the Kampar Regency, Riau Province, total DNA extraction, electrophoresis, polymerase chain reaction (PCR), and data analysis. The results showed that the trnL-trnL-trnF IGS DNA sequences from the three samples of Pucuk Seminyak were obtained 937 bp length, while the matK had a size of 775 bp. These sequences have been registered in GenBank with registration numbers OQ174512, OQ174513, and OQ174514 for trnL-trnL-trnF IGS, and OQ174525, OQ174526, and OQ174527 for matK. The highest similarity was found between Pucuk Seminyak and Champereia manillana with an identity value of 99.67% based on trnL-trnL-trnF IGS and 99.74% based on matK. BLASTn analysis by default settings and organism Champereia (taxid: 50116) resulted in hits on the Champereia genus of the Opiliaceae. Further phylogenetic analysis placed the study objects in the Champereia clade (bootstrap support 49 for trnL-trnL-trnF, 68 for matK, and 71 for concatenated sequences). Lastly, identification by morphological characters also confirmed that the specimen agreed with the description and identification key for C. manillana. The species is classified into two varieties: var. longistaminea and var. manillana. The former was distributed in Southwest China, and the latter was distributed in Malesia to India. Furthermore, the phylogenetic analysis separated the two varieties into different clades. Therefore, it can be concluded that our specimen of Pucuk Seminyak is identified as C. manillana var. manillana Merr. (Opiliaceae).

Abstract

Penelitian ini bertujuan untuk menganalisis dan membuktikan adanya pengaruh: 1) kepemimpinan kepala sekolah terhadap motivasi kerja, 2) budaya organisasi sekolah terhadap motivasi kerja, 3) kepemimpinan kepala sekolah terhadap profesionalisme guru, 4) budaya organisasi sekolah terhadap profesionalisme guru, dan 5) motivasi kerja terhadap profesionalisme guru. Penelitian ini menggunakan pendekatan kuantitatif dengan populasi penelitian ini adalah seluruh guru SMP yang ada di kecamatan Jeunib sebanyak 147 orang yang kemudian menjadi sampel sebanyak 106 orang setelah dihitung dengan rumus Slovin. Hasil penelitian didapatkan bahwa 1) adanya pengaruh yang positif dan signifkan antara variabel kepemimpinan kepala sekolah terhadap motivasi kerja, 2) adanya pengaruh yang positif dan signifikan antara variabel budaya organisasi sekolah terhadap motivasi kerja, 3) adanya pengaruh positif dan signifikan antara variabel kepemimpinan kepala sekolah terhadap profesionalisme guru, 4) adanya pengaruh antara variabel budaya organisasi sekolah terhadap profesionalisme guru, dan 5) adanya pengaruh langsung antara variabel motivasi kerja terhadap profesionalisme guru.

Abstract

Pterocarpus indicus Willd. also known as redwood is a native Philippine species belonging to the Fabaceae family that has many uses and high economic value and highly exploited. However, it is not accompanied by good regeneration which causes this species to be categorized as endangered (IUCN v. 2021-1). The in vitro technique is an alternative to overcome this problem. This study aims to determine the response of P. indicus Willd. shoot growth to the addition of BAP and TDZ in vitro. The research was conducted from August to December 2023, at the Biotechnology Laboratory, KST BJ. Habibie Puspiptek Serpong, South Tangerang, Banten. This study used a completely randomized design (CRD) consisting of 9 treatments and 14 replications. The treatments consisted of: R0 = Control, R1 = BAP 0,25 ppm, R2 = BAP 0,5 ppm, R3 = BAP 0,75 ppm, R4 = BAP 1 ppm, R5 = TDZ 0,25 ppm, R6 = TDZ 0,5 ppm, R7 = TDZ 0,75 ppm, R8 = TDZ 1 ppm. The results showed that the concentration of BAP 0,5 ppm gave the best results in height of shoot and TDZ 0,25 ppm gave optimum results in the number of shoots compared to the control and other treatment concentrations.

Abstract

ABSTRACTThe recent scarcity of fuel has stimulated many efforts to remove dependence on petroleum oil by finding alternative sources of new and renewable energy. Jatropha curcas L. has been chosen as a source of biofuel that has an important role as a petroleum oil substitute, especially for producing biodiesel. Possible efforts for the development of Jatropha in Indonesia include exploration of Jatropha plants from various regions of Indonesia, introducing the plants from abroad and plant breeding. The purpose of this study was to study the genetic parameters of morpho-agronomic traits and selection of potential genotype for genetic improvement of Jatropha curcas. The experiment was conducted at the Biotech Center, BPPT from November 2011 to July 2012. The experiment was performed using a complete randomized block design with sixteen genotypes of J. curcas. Five plants were used for each genotype, with three replications. Results of this study showed that genetic diversity of the genotypes based on morpho-agronomic characters was relatively narrow, with genetic diversity coefficient ranged from 2.73% to 9.02%. The broad-sense heritability was high for all characters, ranged from 32.26% to 85.89%. None of morpho-agronomic characters observed in the 16 genotypes was directly correlated to productivity and seed oil content. Number of fruit and total seed number per bunch were the best characters for selection criteria to obtain high dry seed weight. Based on the bi-plot analysis on seed oil content and dry weight characters among the observed genotypes, LMP and CRP x PDI could be recommended as potential F1 parents to be used in constructing F2 population.Keywords: biplot analysis, broad-sense heritability, genetic diversity coefficient
