Abstract

People generally use larvicide to kill mosquito larvae. Synthetic chemical larvicides leave residues that have a negative impact on the environment. Therefore, research on natural larvicides has been developed to suppress the negative effects of chemical larvicides. Microparticle preparations can speed up the compound delivery system so that it will be effective in its use as a larvicide. The important composition in making microparticle preparations is polymer. The polymer used in this research is chitosan. The formulation of papaya seed extract and alum microparticles using chitosan polymer with the spray drying method was made into three formulas with different concentrations of extract and alum, namely formula 1 (0.1% : 0.05%), formula 2 (0.15% : 0.075%). %), and formula 3 (0.2% : 0.1%). The results obtained show that formula 3 with a concentration of 0.2% papaya seed extract and 0.1% alum has the best quality characteristics based on the polydispersity index parameters, namely 0.181, particle size 1233.67 nm, zeta potential value 72.6 mV. The number of samples was 500 test larvae consisting of 5 treatment groups. Each group contained 25 larvae and 4 replicates. The test larvae were observed after 24 hours and then analyzed. The test analysis carried out was the Probit test. The LC50 value obtained in this study was 95.5 ppm. These results indicate that the microparticle powder preparation has larvicidal efficacy against third/IV instar Aedes aegypti larvae.

Abstract

Curcumin, found in turmeric (Curcuma longa L.), is a pharmacologically active component with hepatoprotective properties. In addition to alleviating pain, the synergistic effects of paracetamol protect the liver from harm. Oral solid dose formulations of curcumin have low bioavailability due to its low solubility. This study aims to characterize the physical interactions that occur in the binary mixture of curcumin and paracetamol and to determine its impact on the solubility of curcumin. The curcumin-paracetamol binary mixture with a 1:1 stoichiometric ratio was prepared by a wet grinding method with the addition of a small amount of ethanol. The characterization of the physical interactions in the wet milling result was carried out using a powder X-ray diffractometer (PXRD) and a differential scanning calorimeter (DSC). Evaluation of physicochemical properties was carried out by testing its solubility in water and its dissolution rate in 40% v/v ethanol. The PXRD pattern of the curcumin-paracetamol milling result did not show any new peaks that were different from the typical peaks of the two components (curcumin form I and paracetamol form 1). The thermogram DSC of the binary mixture curcumin-paracetamol (1:1) only showed one wide endothermic transition at 151.2°C which is below the melting point of curcumin and paracetamol which is thought to be the melting point of the eutectic mixture of curcumin-paracetamol (1:1). The solubility of curcumin from the curcumin-paracetamol (1:1) milled binary mixture was 8.3-folds higher than that of pure curcumin. The dissolution rate of curcumin from the wet milling of the curcumin-paracetamol binary mixture (1:1) was also faster than that of pure curcumin. The research results can be concluded that wet milling of the binary mixture of curcumin-paracetamol (1:1) with a little ethanol shows a physical interaction with the formation of a simple eutectic mixture between the two substances which has an impact on increasing the solubility and dissolution rate of curcumin.

Abstract

The liver is the organ most susceptible to the adverse effects of alcohol use. Decreased superoxide dismutase can be used as an indicator of liver cell damage due to excessive alcohol consumption. Red ginger (Zingiber officinale var. Rubrum) is known to have a hepatoprotective effect through antioxidant and anti-inflammatory mechanisms. This study was to prove the effect of red ginger extract from Batu Malang, Indonesia to restore normal levels of superoxide dismutase in the liver of Wistar rats induced by ethanol. Gas Chromatography-Mass Spectroscopy (GC-MS) metabolite profiling analysis of the red ginger ethanol extract was also carried out to ensure the presence of antioxidant compounds. Male Wistar rats were divided into 5 groups with 6 rats each, including negative control (C-) which was only given standard feed, positive control (C+) which was only given 1.8 mL/200g body weight of 40% ethanol orally, and treatment group which were given 40% ethanol as much as 1.8 mL/200 g body weight and red ginger extract at a dose of 250 (T1), 500 (T2), and 750 (T3) mg/kg body weight. Liver organs were taken to examine superoxide dismutase levels after 14 days of treatment in all groups. This research shows that ethanol with a concentration of 40% can reduce superoxide dismutase levels in the liver of rats significantly (p<0.05). All treatment groups that were given red ginger extract had higher superoxide dismutase levels than the positive control (C+) group (p <0.05), but the optimal dose to increase superoxide dismutase levels in Wistar rats induced by oral administration of 40% ethanol is 250 mg/kg body weight. Based on the results of metabolite profiling using GC-MS, found three dominant compounds thought to act as antioxidants, including Zingiberene, Gingerol, and 6-shogaol. Furthermore, further research is needed to evaluate the safety of using red ginger extract.

Abstract

Bones in squid (Loligo sp.) can be used as a source of medicinal ingredients, one of which is chitosan. The polymer in chitosan contains amino and hydroxyl groups per residue, giving chitosan many biological activities, such as anti-inflammatory, antimicrobial, hypoglycemic, hypocholesterolemic, and immunostimulatory effects. This study aims to determine the antioxidant activity and the effect of chitosan squid bone powder on the total cholesterol levels of rats (Rattus norvegicus). This type of research used an experimental method with a one-group pre-test and post-test design, in which 12 male white rats, body weight 150 g, were divided into four groups. Group 1 was a negative control of 1% Na-CMC, group II chitosan 50 mg/kg, group III chitosan 100 mg/kg, and group IV positive control of simvastatin 10 mg, which each group was given orally; the treatment was carried out for two weeks. The results showed a decrease in the mean after treatment in the treatment group with chitosan powder, but there was no significant difference compared to the control group (P>0.005). This shows that squid bone chitosan powder reduced total cholesterol levels in rats.

Abstract

Cantigi (Vaccinium varingiaefolium (Blume) Miq.) contains anthocyanins, chlorophyll, phenols, saponins, steroids, tannins, triterpenoids, flavonoids, and antioxidant activity. This study aimed to obtain the concentration of cremophor RH-40 as an optimal and stable surfactant during storage in Cantigi leaf extract nanoemulsion formulation process. Cantigi leaf extract was made by kinetic maceration in stages with n-hexane, ethyl acetate, and 96% ethanol as solvent, then evaporated with an evaporator. The extract was tested for solubility using various solvents before making the nanoemulsion. Nanoemulsions were prepared by varying the amount of cremophor RH-40 in three formulas (5%, 7.5%, and 10%) with stirring at 400 rpm at 40 °C. The nanoemulsions were characterized by particle size, zeta potential, and storage for 5 days. The evaluation results of Formula 1, 2, and 3 have an average particle size of 224.7, 226.5, and 198.3 nm, with a polydispersity index of 0.565, 0.237, and 0.223. The zeta potential values are - 43.7, - 39.2, and - 42.3 mV, respectively. The results of the storage of nanoemulsions for 5 days showed that in formulas 1 and 2 a precipitate formed, while in formula 3, there was no precipitate. Based on the optimization results obtained, cremophor RH-40 10% obtained a stable nanoemulsion that does not occur with no observed sedimentation/precipitation during storage.
