Abstract

The aim of the study was to determine the diversity of fruit fly species (Bactrocera spp.) found in fruit trees in the lowlands Bontomarannu and highlands Tinggimoncong of Gowa Regency, South Sulawesi. The research took place for five months, starting from February to July 2020 which was carried out in two stages. The first stage was to fruit attacked by fruit flies and then put them in a plastic jar filled with sterile fine sand. The jar is covered with gauze and left until a cocoon is formed and an adult insect is born. Adult insects were transferred to petri dishes containing cork and had been smeared with honey as food for fruit flies. The population of adult insects was counted and then identified using a microscope. The second stage is setting traps containing attractant Methyl Eugonol for seven days on fruit plants. Observations were made daily on trapped fruit flies. The attractants in the traps were updated daily for subsequent observations for seven days. Fruit flies were brought to the laboratory for identification using a microscope. The identification results showed that there were five species of fruit flies namely B. papajae, B. carambolae, B. umbrosa, B. occipitalis and B. Cucurbitae.

Abstract

This research is a continuation of previous research on the diversity of pitcher plants (Nepenthes sp) at the University of Palangka Raya. This study aims to determine the habitat characteristics of the pitcher plant (Nepenthes sp) at the University of Palangka Raya. We carried out this research in June-September 2022. The habitat characteristics of pitcher plants were collected by purposive sampling to analyze habitat characteristics data using a quadratic sampling method consisting of two transects with 15 plots based on the findings of Nepenthes sp. at the research site. The first transect is in a secondary forest, and the second transect is in the scrub with slightly open conditions after the fire. The research area was dominated by tree species Cratoxylum glaucum, Acacia mangium, Melaleuca cajuputi, and understorey species dominated by Stenochlaena palustris. A diversity index value is classified as a medium for understorey and saplings and low for poles and trees.

Abstract

The Barcode of Life Consortium (CBOL) recommended a standard method for the identification of plant species using matK and rbcL barcoding gene. This study was aimed to evaluate the efficiency of matK DNA barcoding for identification of local durian (Durio sp.) from Ternate Island. Total 15 local durian has been used in this study. Whole genom DNA was isolated by Geneid plant DNA kit and then successfully amplified by the Polymerase Chain Reaction (PCR) technique using specific primer. MatK successfully amplified with 245 bp in length. MatK has sequencing success (71.3%) and relative high of Quality value 20+ (86%). BLAST analysis of the sequence showed that local durian in Ternate are identifies as Durio zibethinus and Neesia malayana with query cover 97%-99%. It could be concluded that matK with short sequence is not efficience for durian identification. The recommended in this studies for using of molecular markers with sequence lengths above 500 bp would be more effective for the identification of cryptic species Durio sp. and other.

Abstract

Herbal medicine's development as an antifertility agent is the condensed tannins of the leaves of beluntas (Pluchea indica). This study was to determine the safety of P. indica condensed tannin extract based on the behaviour of male Rattus norvegicus. This research design was qualitative descriptive with two weeks direct observation of animal models and scoring based on rats' behaviour parameters. Comparative research involving 24 white male rats based on negative control group (aquades), dose groups : I (0.4 ml/KgBW), II (2 ml/KgBW), and III (2 ml/KgBW and 3 ml/KgBW). Parameters observed were body posture, motoric activity, ataxia, righting reflex, gauze test, analgesia, ptosis, and respiratory death or cardiac arrest. The result showed a significant correlation between three of the eight parameters, namely body posture, motor activity, and ptosis based on the dose that had been given. The safe dossage as an antifertility candidate for humans was range I to dose II (0.4ml/KgBW-2 ml/KgBW) because it was included in the practically non-toxic category. Therefore, this research was important to make condensed tannins safe in their application as an antifertility drug for humans in the future.

Abstract

This study at to determine the changes in liver histology of male rats after induction of condensed tannin extract of Beluntas leaves with control doses, 0.4 ml/kgBW, 2 ml/kgBW, and 3 ml/kgBW. This study is an experimental study for 14 days involving 24 samples of male rats. Parameters of histological structure changes were assessed by the scoring system for normal hepatocyte cell condition, parenchymatous degeneration, hydropic degeneration, and necrosis. One Way Anova test obtained a p value< 0.001 which means there was a significant difference in changes in the histopathological structure of liver cells significantly. Based on the Post Hoc test, it was obtained. between the control and treatment groups showed a significant difference between the control dose of 0.4 ml/kgBW, 2 ml/KgBW dose, and 3ml/KgBW dose. Meanwhile, the changes in liver histopathology between 2 ml/KgBW, and a dose of 3ml/KgBW were significantly the same.

Abstract

The aim of this study was to determine the antibacterial activity and the ccombined effect of methanol extract of the ketepeng cina leaves (Cassia alata L.) with six selected plant extracts against Staphylococcus epidermidis using the diffusion and the paper strip methods at a concentration of 10% w/v. The results showed the average antibacterial activity of each extract showed the inhibitory growth of Staphylococcus epidermidis. The average value of the inhibition zones of the single extract samples of ketepeng cina, sambang darah, ruku-ruku, avocado seed, bungli, pacar air, and sungkai resulted in inhibition zones of 6.28±0.20 mm, 7.90±0.31 mm, 2.30±0.37 mm, 3,53±0.50 mm, 4.35±0.52 mm, and 4.69±0.17 mm, respectively. Furthermore, the combination of methanol extract of ketepeng cina leaves (C. alata) with the six selected plants extract in an average diameter of inhibition zone greater than the average diameter of the inhibition zone of each extract. However, the average diameter of the inhibition zone of the combined extract was not more significant than the sum of the diameter of the inhibition zone of each extract. The conclusion of this study is the combination of the methanol extract of ketepeng cina leaves (C. alata) with six selected plant extracts has an additive/indifferent effect in inhibiting the growth of S. epidermidis.

Abstract

This study aimed to determine the phytochemical content and potency of the ethanol extract of lemongrass (Cymbopogon citratus) stem and red galangal rhizome (Alpinia purpurata) in inhibiting the growth of Shigella sonnei in vitro. Phytochemical testing of ethanol extract using Thin Layer Chromatography (TLC). Antibacterial test was carried out by applying the Kirby Baeur disk diffusion method with swab technique with a series treatment of extract concentrations of 6.25%, 12.5%, 25%, 50%, positive control (tetracycline 20%), and negative control (DMSO 10%) with 5 replicates. From the results of phytochemical tests, showed that secondary metabolites of flavonoids and terpenoids were found in red galangal extract. While the lemongrass extract showed positive results for alkaloids, flavonoids, terpenoids and phenolic compounds. The ethanol extract of lemongrass stem and galangal rhizome potential as an antibacterial with the highest inhibitory activity obtained at a concentration of 50% in both lemongrass and galangal extracts, respectively 8.4 mm and 7 mm. The results of the antibacterial test showed that red galangal extract and lemongrass extract had phytochemical contents that could inhibited the growth of S. sonnei.

Abstract

Dragonflies are insects that mostly live as nymphs and are related to aquatic habitats and act as bioindicators of environmental quality. The purpose of the study was to identify the diversity of dragonflies in various habitat types in Cipeuteuy Village. This study uses the line transect method measuring 100 meters with a width of 20 meters right and left and the duration of observation is 15 minutes per each observation line. The study was conducted in nite habitat types from 52 locations. The total dragonfly found were 13 species from four families. The highest dragonfly richness was found in the rice field habitat as many as 12 species and the lowest species was found in the citrus garden habitat as many as three species. The dominant species of dragonfly found in all habitat types is Rhodothemis rufa. The overall dragonfly diversity index was 2.56 and ranged from 1.10 to 2.48, and the evenness index ranged from 0.57 to 0.90.

Abstract

The highest plant diversity in the tropical rainforest area in Matalawa National Park is the Angiospermae plant. There are still many people in Praingkareha Village who are not familiar with Angiospermae plants or closed seed plants. This study aims to describe the diversity index of Angiosperm plants found in the Laputi Forest block, Matalawa National Park. This research was limited to the Laputi Forest block, so it did not cover the entire forest in Praingkareha Village, Matalawa National Park, East Sumba Regency. This study used a double plot method with a purposive sampling technique. The results showed that there were 30 species of Angiosperms from 17 families with a total of 499 individuals. The diversity of Angiosperm plants in the Laputi Forest block of the Matalawa National Park area is moderate with (Ĥ)=2.90.

Abstract

Sanana Honey is a local brand, a type of honey produced by forest bees which is well known in the Sula Islands, North Maluku. The forest bees of the Sula Islands have the potential to be developed as a producer of quality honey. At this time there has been no scientific report regarding the types and characteristics of the forest bee species from the Sula Islands. This study aims to identify the types of honey-producing forest bees by applying the LCO gene DNA barcode. The methods used in this study used: 1) DNA extraction from bee body tissues using the DNA Miniprep Kit procedure (Zymo Research, D6016); 2). DNA amplification by PCR amplification procedure with TOYOBO KOD FX Neo (KFX-201); 3) bidirectional DNA sequencing process (Bi-directional sequencing). The results showed that the specimen was successfully amplified with an amplicon size of 677 bp. Furthermore, the results of BLASTN analysis revealed that the sequence has a 97.78% similarity with the Apis cerana mitochondria complete genome gene country of Russia, but is phylogenetically different between the two.
