Abstract

The Present study was designet to investigate the influence of vanadyl sulphate towards GLUT-4 protein activities in skeletal muscle tissue in streptozotocin induced diabetic mice. Twenty five mice divided into five groups i.e. placebo group, diabetic group, and three treatment groups based on vanadyl's doses (5,30 or 100 mg/kg BW, respectively). Diabetic mice model was induced by twice intraperitoneal administration of streptozotocin. The first dose of streptozotocin is 100 mg/kg that were inject in the first day and then dose 50 mg/kg, in the day 14th. Diabetes occured on 21st day after streptozotocin injection and it was shown by increasing blood glucose level from 151.4 ± 25.1 mg/dL to 237.1 ± 33.0 mg/dL. Administration of vanadyl sulphate at the dose of 5,30 or 100 mg/kg BW was significantly reduces blood glucose concentration (p<0,001). The muscular tissue were harvested on the day 28th and stained using routine histology staining, hematoxylin-eosin for morphological qualitative analysis and immunohistochemical examination to observe the activities of GLUT-4 protein in skeletal muscle. The result showed that vanadyl sulphate restore atrophic condition of muscular cells and inhibit cell necrosis in muscular tissue. On immunohistochemical examination, vanadyl sulphate might increased the GLUT-4 protein activities in skeletal muscle in streptozotocin-induced diabetic mice Keywords : vanadyl sulphate, streptozotocin, diabetes mellitus, GLUT-4 Abstrak Penelitian ini bertujuan untuk mengetahui pengaruh vanadil sulfat terhadap aktivitas protein GLUT-4 pada otot skelet mencit yang menderita diabetes mellitus akibat induksi streptozotocin. Sebanyak 25 ekor mencit dibagi menjadi 5 kelompok, yaitu kelompok kontrol, diabtes dan perlakuan yang diberi vanadil sulfat dengan dosis 5,30 dan 100 mg/kgBB. Keadaan diabetes diinduksi dengan pemberian streptozotocin dosis 100 mg/kgBB pada hari pertama dan 50 mg/kgBB pada hari ke -14. Pada hari ke-21 terjadi peningkatan kadar glukosa dari 151,4 ± 25,1 mg/dL menjadi 237,1 ± 33,0 mg/dL. Pemberian vanadil sulfat selama 7 hari akan menurunkan kadar glukosa darah secara signifikan (p<0,001). Otot skelet diambil pada hari ke-28 dan dipreparasi dengan hematoksilin-eosin untuk diamati histologinya dan secara imunohistokimia untuk pengamatan aktivitas protein GLUT-4. Hasil pengamatan menunjukkan pemberian vanadil sulfat akan memperbaiki keadaan atropi dan menghambat nekrosis pada sel otot skelet. Pengamatan secara imonohistokimia menunjukkan bahwa vanadil sulfat dapat meningkatkan aktivitas GLUT-4. Kata Kunci: vanadil sulfat, streptozotocin, diabetes mellitus, GLUT-4

Abstract

The aims of this research is to observe the inhibition activity of sulfonamides or sulfacetamides substituted of 3-Phenyl-2-styril-4(3H)-quinazolinones with COX-2. The study of COX-2, binding inhibition and dynamics interaction was done with in silico method by molecular docking with Auto Dock 4.0 and molecular dynamics in 2 nanoseconds with Amber 11. Those compound could be divided into 3 groups, based on Î?G scores of docking result: very selective group (-10.92 to -11.33 kcal/mol) compared to SC-558 (-10.90 kcal/mol); selective group compound (-9.22 to -10.68 kcal/mol) compared to celecoxib (-10.63 kcal/mol); non selective group, compound (-6.48 to -6.98 kcal/mol) compared to aspirin (-4.82 kcal/mol). Molecular dynamics simulation of 6COX complex with several quinazolinon derivates showed number and stability of hydrogen bond.Keywords : COX-2, anti-inflammatory, molecular docking, molecular dynamics.

Abstract

ABSTRACT: The use of herbal preparations to improve fertility were empirically beneficial for reproductive health, however its use needs to be supported by scientific evidence. In this study we observed the effects of herbal preparations on haematological parameters, liver function and kidney function of female rats. The estrus cycle of the experimental female rats were previously synchronized by injection with prostaglandin twice in two days interval. The rats were randomly assigned into eight groups i.e., two negative control groups, two positive control groups (injected with pregnant mare serum gonadotropin with effective and double of effective doses), two groups were given herbal preparation A (with effective and double of effective doses), and two groups were given herbal preparation B (with effective and double of effective doses). The treatments were given for 3 consecutive estrous cycles. Parameters measured were hematological parameters (erythrocyte, leukocyte, and hemoglobin), liver functions (SGOT and SGPT concentrations), and renal function (serum ureum concentration). The results showed that the administration of herbal preparation for fertility improved erythrocyte, leukocyte, and hemoglobin parameters, improved liver functions, and optimized renal functions. It was concluded that the administration of herbal preparation for fertility for 3 consecutive estrous cycles did not disturb physiological functions, instead its improved hematological parameters as well as liver and renal functions.Keywords: hematological, herbal, liver function, renal function, female rats  ABSTRAK: Penggunaan jamu kesuburan secara empiris bermanfaat untuk kesehatan reproduksi, namun penggunaannya perlu didukung oleh data ilmiah. Dalam penelitian ini dilakukan pengamatan efek pemberian jamu kesuburan terhadap parameter hematologis, fungsi hati dan fungsi ginjal dari tikus betina. Penelitian ini diawali dengan sikronisasi siklus estrus tikus percobaan dengan penyuntikan prostaglandin dua kali dalam selang waktu dua hari. Tikus-tikus percobaan secara acak dibagi ke dalam delapan kelompok, yaitu dua kelompok kontrol negatif yang hanya diberi pelarut persiapan jamu kesuburan, dua kelompok kontrol positif yang disuntik pregnant mare serum gonadotropin untuk dosis efektif dan 2x dosis efektif, dua kelompok diberi jamu kesuburan A dengan dosis efektif dan 2x dosis efektif, dan dua kelompok yang diberi jamu kesuburan B untuk dosis efektif dan 2x dosis efektif. Perlakuan diberikan selama 3 siklus estrus berturut-turut. Parameter yang diukur adalah status hematologi (eritrosit, hemoglobin, dan leukosit), fungsi hati (kadar SGOT dan SGPT), dan fungsi ginjal (kadar ureum). Hasil penelitian menunjukkan bahwa pemberian jamu kesuburan memperbaiki parameter hematologis, memperbaiki fungsi hati, dan mengoptimalkan fungsi ginjal. Dari hasil penelitian disimpulkan bahwa pemberian jamu kesuburan selama tiga siklus estrus berturut-turut tidak mengganggu fungsi fisiologis tubuh bahkan terbukti memperbaiki parameter hematologis, fungsi hati, dan fungsi ginjal. Abstract

ABSTRAK : Lendir bekicot (Achatina fulica) dalam bentuk lotion telah diuji secara ilmiah aktivitasnya terhadap bakteri penyebab infeksi jerawat. Penelitian bertujuan menghasilkan formula lotion lendir bekicot yang bisa mengobati jerawat. Metode pengujian antibakteri menggunakan turbidimetri. Evaluasi sediaan yang dilakukan meliputi pengamatan organoleptik dan pH, ujidaya sebar dan uji daya lekat. Berdasarkan hasil pengujian yang telah dilakukan, lendir bekicot dan lotion lendir bekicot memiliki aktivitas antibakteri terhadap Propionibacterium acnes. Lotion lendir bekicot yang menghasilkan efek anti jerawat paling baik adalah yang mengandung konsentrasi lendir 16%. Sediaan lotion memiliki karakteristik, daya sebar, dan daya rekat yang baik. Bentuk sediaan stabil dalam penyimpanan suhu kamar selama lima minggu.

Abstract

One targeting point of cancer treatment, especially liver cancer is the suppression of N-Ras expression, inhibition of c-Src activity and CYP1A2 in the liver cells. The aim of the research in to explore the anticarcinogenesis effect of the ethanolic extract of C. reticulata peel through suppression of N-ras expression and the binding afinity and interaction of polimetoksiflavon compound found in the peel of Citrus Reticulata, namely tangeretin and nobiletin to the target protein using molecular docking. Geometry structure optimization tangeretin and nobiletin were done with the software Molecular Operating Environment (MOE) for Windows. The optimum structure conformation of tangeretin and nobiletin were approached using semiempirik AMBER99 method. The process of docking of the test compound to the bindingsite c-Src (PDB ID: 1FMK) and CYP1A2 (PDB ID: 1AE4) were done using the software Molecular Operating        Environment (MOE) for Windows in the conditions without water. From the docking process found that the lowest scoring value is tangeretin in conditions without water.  Docking Results of tangeretin compared with the experimental ligan in the target CYP1A2, shows the interaction of tangeretin stronger than the interaction of ligan an α-naphtoflavon. Meanwhile, the target protein, c-Src, interaction endogenous ligan (ATP) is much stronger than the interaction with the test compound and ligan comparison Imatinib. Therefore, it is estimated that the mechanism of liver cancer hepar inhibition in molecular docking is through CYP1A2 inhibition. ABSTRAK Salah satu titik tangkap pengobatan kanker khususnya kanker hepar adalah penekanan ekspresi N-Ras, penghambatan protein c-Src dan aktivitas CYP1A2 di hepar.  Penelitian ini bertujuan untuk mengetahui afinitas dan interaksi senyawa berkerangka polimetoksiflavon pada kulit jeruk keprok (Citrus reticulata) yaitu tangeretin dan nobiletin terhadap protein target tersebut menggunakan molecular docking. Optimasi geometri struktur tangeretin dan nobiletin dilakukan dengan piranti lunak Molecular Operating Environment for Windows.  Konformasi optimum tangeretin dan nobiletin dihasilkan menggunakan metode semiempirik AMBER99. Kemudian dilakukan proses docking senyawa uji dengan bindingsite c-Src (PDB ID : 1FMK) dan CYP1A2 (PDB ID : 1AE4) menggunakan piranti lunak Molecular Operating Environment for Windows dalam kondisi tanpa air. Dari hasil yang diperoleh dapat diperkirakan bahwa mekanisme penghambatan kanker hepar secara docking molekuler adalah melalui penghambatan CYP1A2

Abstract

Extract of god leaves [Gynura pseudochina (L.) DC] has been characterized by using the high performance liquid chromatography method (HPLC). Dried god leaves were extracted by 70% ethanol and followed by fractionation successively with hexane, ethyl acetate, butanol and water. Fractions of ethyl acetate, butanol and water were dissolved in methanol respectively and injected into HPLC system which is optimized previously. Profiles of respective fractions showed a number of specific peaks that can be used to identify and control the quality of god leaves extract.   ABSTRAK Karakterisasi ekstrak daun dewa [Gynura pseudochina (L.) DC] telah dilakukan dengan menggunakan metode kromatografi cair kinerja tinggi (KCKT). Daun dewa kering diekstraksi dengan etanol 70% dan kemudian difraksinasi berturut-turut dengan heksana, etil asetat, butanol dan air. Fraksi-fraksi etil asetat, butanol dan air dilarutkan dalam metanol dan diinjeksikan ke sistem KCKT yang telah dioptimasi sebelumnya. Pola KCKT masing-masing fraksi menunjukkan puncak-puncak khas yang dapat dipakai untuk identifikasi dan pengendalian mutu ekstrak daun dewa.

Abstract

The slow release of drug from tablets dosage form obtained by adding cellulose derivative as matrix which could gel form. The aims of the study were to investigate the profile and the rate of diclofenac sodium release from sustained released tablets using Methocel K15M as matrix intragranular in medium with intestine pH. Diclofenac sodium sustained release were made with wet granulation method in five formulas based on the different concentration of Methocel K15M that added of 0mg (F Control), 25mg (F I), 50mg (F II), 75 mg (F III) dan 100mg (F IV). Dissolution testing used paddle stirrer with 100 rpm in a buffer phosphate medium of pH 7,6 at 37±0,5oC for 5 hours. Sampel analyzed by spectrophotometer at 274 nm. The result showed that the Methocel K15M matrix decreased the diclofenac sodium release from the tablets and it had a linear relationship with time. The more Methocel K15M added in the matrix the slower the rate of diclofenak sodium released from it. Drug release from the matrix followed zero order kinetics and mechanisms was a combination of erosion and diffusion, the diffusion mechanism was the dominant one. The results of t test with 95% confidence level the significant differences between four formulas. ABSTRAK Pelepasan obat secara perlahan dari sediaan lepas lambat dapat diperoleh dengan penambahan turunan selulosa sebagai matriks yang dapat membentuk gel. Penelitian ini bertujuan untuk mengetahui profil pelepasan natrium diklofenak dari tablet lepas lambat dengan penambahan Methocel K15M sebagai matriks intragranular dalam medium pH usus. Sediaan lepas lambat natrium diklofenak dibuat dengan cara granulasi basah dalam 5 formula yang didasarkan pada perbedaan kadar Methocel K15M yang ditambahkan, yaitu 0 mg (F Kontrol), 25mg (F I), 50mg (F II), 75 mg (F III) dan 100mg (F IV). Uji disolusi menggunakan pengaduk dayung dengan kecepatan pengadukan 100 rpm dalam medium dapar fosfat pH 7,6 pada suhu 37±0,5oC selama 5 jam. Sampel dianalisa secara spektrofotometri UV pada panjang gelombang 274 nm. Hasil uji disolusi menunjukkan kecepatan pelepasan obat dari matriks Methocel K15M berlangsung secara perlahan dan mempunyai hubungan yang linear dengan waktu. Semakin banyak Methocel K15M yang ditambahkan dalam matriks semakin lambat natrium diklofenak dilepaskan. Pelepasan natrium diklofenak dari matriks mengikuti kinetika orde nol dengan mekanisme kombinasi difusi dan erosi, mekanisme difusi lebih dominan.

Abstract

In this study a High Performance Liquid Chromatography (HPLC) has been used for determination of rubraxanton on bark extracts Garcinia spp (Garcinia mangostana, Garcinia Cowa, Garcinia griffitii, Garcinia dioica and Garcinia forbesii). HPLC system consisted of C-18 reversed phase column with a length of 250 mm, diameter 4.6 mm, 20 mL injection volume, mobile phase methanol: water (gradient system with polarity) and flow rate of 1ml/min. Rubraxanton levels obtained in this study; 9.161% for G.mangostana, 6.942% for G.cowa, 6.762% for G.dioica, 0.499% for G.forbesii and 0.229% for G.griffitii. The method has been validated for specificity, linearity, accuracy, precision, limits of detection (LOD) and limits of quantitation (LOQ). The linearity of the method can be seen from the regression coefficient r=0.9996 with a linearity range from 1.72 to 55 ug/ml. Recovery of rubraxanton in the extract of G.mangostana was between 99.61 to 101.08%. Intra and inter day precision showed relatively small level of standard deviation (lower than 2%). Limit Of Detection (LOD) and Limit Of Quantitation (LOQ) are 0.55 ug/ml and 1.82 ug/ml respectively.Keywords : rubraxanton, bark extracts, Garcinia spp.

Abstract

Iron supplement containing ferrous sulfate is commonly used for anaemia. Unfortunately, it has bad taste, can cause nausea, and made adverse effects if taken in large doses for long periods. It is necessary to 􏰁ind an alternative source of raw materials, including those from plants. Moringa pterigospera Gaertn leaves was selected because it contains iron and other nutritions. The purpose of this work was to make instant powder formula for anti anaemia using Moringa leaves extract as an alternative for ferous suphate iron suplementation. The extraction was performed by Microwave Assisted Extraction method. Optimization of extraction condition was performed by creating some variations in solvent composition (0- 70% ethanol), microwave power (450 to 900 watts) and extraction time (3 to10 min). Iron content was determined by Atomic Absorption Spectrophotometer at wave length of 248 nm. Instant powder formula was made in 3 concentrations of sodium CMC as suspending agent. Results of the study showed that the most optimal extraction condition which resulted the highest iron content (2.4 mg/g extracts) achieved with 900 watts microwave power, 10 min extraction time and aqua demineralisata. According to 30 panelists of hedonic test, formula which used 5% w/w of sodium CMC got the highest scores. Its powder form had 2.31% of loss on drying and 7.74 g/sec of 􏰁low rate for powder form and pH of 5.78, viscosity of 15.98 cps for reconstituted form.

Abstract

Dry extract of sarang semut (Myrmecodia pendens Merr & Perry) water/n-butanol fraction is native plant from Papua is proven to be active against cancer cells. This research aims to improve the quality of extracts of sarang semut become the raw material that meets the standards of medicinal plant extracts in the fulfillment of the quality of the extracts into anticancer Standardized Herbal Medicine / Obat Herbal Terstandar (OHT). Extraction with ultrasonic methods (1:10 with methanol) and fractionation with water:butanol (1:1) as much as 1:20. Standardization and data analysis based on parameters of the Medicinal Plant Extracts (Kepmenkes RI No: 55/Menkes/SK/I/2000) and BPOM RI Regulation No. 12, Year 2014 about quality requirements of traditional medicine. Dry extract of sarang semut (water/n-butanol fraction) met the standards of raw material preparation for OHT with organoleptics shaped dried extracts, dark brown color, aromatic smell, and has a sour-bitter taste. Moisture content was 9,259 ± 0.001 (� 10%). Total Plate Count was 5 x 103 colonies/g (< 104 colonies/g). Yeast and Mold Count was 0,5 x 101 colonies/g (< 103 colonies/g). Pathogenic bacteria test shows that MPN Coliform value was 0 colonies/g and negatively to the presence of Escherichia coli, Salmonella sp, Staphylococcus aureus, Pseudomonas aeruginosa, and Shiggella sp. The extract was not detected the presence of aflatoxin total (aflatoxin B1, B2, G1 and G2). The content of heavy metal Cd was 0,0052 ± 0.0005 ppm (� 0.3 ppm) and heavy metals Pb was 0,9429 ± 0,0087 ppm (� 10 ppm).
