Abstract

A known compound caffeine from the water extract of parasitic tea plants, Scurrula junghunii (Loranthaceae) have been isolated and identified. Identification the compound based on spectra of ultraviolet, infra red, nuclear magnetic resonance ('H- & 13C-NMR) and mass spectra.

Abstract

The purpose of this study was to provide information on the antibacterial activity of ethyl acetate culture broth extract of endophytic fungi associated with Rhizophora mucronata. An isolated fungi was identifi ed as Nigrospora oryzae using microscopic and molecular analysis. Theantibacterial activity was carried out using micro dilution method to get MIC value. The ethyl acetate extract showed antibacterial activity with MIC value of 250 μg/mL against and Escherichia coli and 500 μg/mL against Staphylococcus aureus.

Abstract

Solid lipid nanoparticle (SLN) has been proposed as colloidal carriers using solid lipid as matrix material. In this study, gliclazide-loaded solid lipid nanoparticle has been prepared with high shear homogenization and freeze drying method using stearic acid as lipid material, Tween 80 and PEG 400 as surfactant, ethanol as`co-solvent and lactose as cryoprotectant. Characterization was performed on SLN dosage from before and after freeze drying, which includes the analysis of particle size, zeta potential value, morphology analysis and entrapment efficiency of gliclazide. Results indicated gliclazidecan be formulated in SLN dosage form using high shear homogenization and freeze drying method. The morphology study revealed that the prepared SLN were irregular in shape with mean particle size of 878.0 ± 246.3 nm and 745.8 ± 204.0 nm. Abstract

Ethanolic extract of mango’s leaves variety of bapang (Mangifera indica L. ‘Bapang’) has been known to contains mangiferin which has antidiabetic activity in patients with diabetes mellitus. This study aimed to obtain a formulation of tablets of ethanolic extract mango leaves with variety of bapang with good physical characteristic and know the effect of variations concentration of guar gum on the properties of mucoadhesive. Tablets were prepared using wet granulation method varying levels of guar gum as a bioadhesive matrix. Tablets were tested physical characteristics and mucoadhesive strength in vitro by using a rotating cylinder and observed within 8 hours. The results showed that the tablets of ethanolic extract mango leaves with variety of bapang have good physical characteristics. The result of mucoadhesive in vitro test showed that all formulas have good mucoadhesive strength. Mucoadhesive strength increased with the increase in the concentration of guar gum. The result of this research showed that the tablets of formula 4 with the highest concentration of guar gum showed the best mucoadhesive strength where they could be attach of 7 ± 1 hour.

Abstract

Recrystallization with acetone of antalgyne, phenylbutazone, and the mixture of binary system has been characterized. The aim of this research was looking for thermodynamic and x-ray diffraction characteristic of re-crystallite to compare with raw material characteristics. The result used to confirm validity of cool contact methods to identify physical interaction of antalgine-phenylbutazone that has been developed in the preliminary research. Recrystallites characterized by DSC and powder-XRD shows the polymorphism cases, but the co-recrystallite of binary system show the peritecticum physical interaction, similar with the physical interaction of the raw material binary system before recrystallization. Principally, the results of this investigation prove that cool contact methods by acetone could be used for identify physical interaction of antalgine-phenylbutazone.

Abstract

Rambutan (Nephelium lappaceum L.) is a plant which can be used to treat various diseases, including diabetes mellitus. This research was conducted to investigate the effect of rambutan seed infusion on the mice blood glucose levels and body weight. In this research, 30 mice (Mus musculus) were divided into 6 groups, each group consists of 5 mice. The 6 groups consists of a normal control group (distilled water), negative control group (alloxan tetrahydrate 250 mg/kg bw + distilled water), three experimental groups (induced with alloxan 250 mg/kg bw) were given rambutan seed infusion of 1.56 gram/kg bw, 2.34 gram/kg bw and 3.12 gram/kg bw and a positive control group (alloxan tetrahydrate 250 mg/kg bw + treated with glibenclamide 0.65 mg/kg bw). After given treatment for 16 days, it was observed that high dose of rambutan seed infusion has significant effects in reducing the blood glucose and body weight of mice. Histology of the pancreas were checked and the ammount of live pancreatic beta cells found in high dose of rambutan seed infusion were almost the same as the positive control group. It can be concluded that the effect of high dose of rambutan seed infusion (3.12 gram/kg bw) was not significantly different from glibenclamide (0,65 mg/kg bw).

Abstract

Penelitian ini dilakukan untuk mengisolasi dan menguji aktivitas antibakteri dari isolat bakteri X2 yang berasosiasi dengan spons Xestospongia testudinaria dari Pantai Pasir Putih Situbondo. Isolasi bakteri dilakukan dengan metoda pour plate kemudian koloni bakteri yang diperoleh dimurnikan dengan metode streak plate. Isolat bakteri X2 dihasilkan 12 isolat bakteri yang akan diuji aktivitas antibakteri terhadap bakteri uji Klebsiella pneumonia, Pseudomonas aeruginosa dan Salmonella typhii menggunakan metode overlay. Hasil uji mendapatkan isolat X2-10 yang aktif terhadap P. aeruginosa ATCC27853 dan MDR dengan menggunakan metode difusi agar. Karakterisasi isolat X2-10 dilakukan dengan uji pewarnaan Gram, uji biokimia dan identifikasi dengan 16s-rRNA. Hasil penelitian menunjukkan isolat bakteri X2-10 yang berasosiasi dengan spons X. testudinaria memiliki aktivitas antimikroba terhadap bakteri P. aeruginosae ATCC27853 dan MDR dan hasil karakterisasi dari isolat bakteri X2-10 merupakan B. licheniformis.

Abstract

Klebsiella pneumoniae is one of coliform bacteria which causes human and mammalian diseases. The bacteria dominate in dairy cow milk which has been infected by mastitis and has resistent on antibiotic. Vaccination is one of aims to prevent the diseases. Nuclear technique could be used to have a vaccine candidate.This research was conducted to get inactivated K. pneumoniae by gamma irradiation and heat inactivated as vaccine candidate. The experiments were done by determination of inactivated doses, protein content, and protein profile. analysis. K. pneumoniae could be inactivated using gamma rays by doses higher than 600 Gy. Neither irradiation nor heat (65 oC) influenced the K. pneumoniae total protein content The intensity of protein profile in gamma ray inactivated was higher than heat inactivated. There were 35, 36, and 60 kDa protein which were diagnosed as antigen protein. It could be concluded that inactivated by gamma irradiation K. pneumoniae could be chosen as a vaccine candidate and as a model for other bacterial vaccine.

Abstract

Endophytic microbe is a definition which is given to all microorganisms that during a more or less long period of their life, colonize symptomnlessly the living tissues of their hosts. The endophytes can produce useful substances such as oligosaccharide degrading enzyme, antimicrobial substances and plant growth regulator. Quisqualis indica is one of tropical herb that is widely known for traditional medicine. There is no information about endophytic microbes of this plant. The aim of this study are to isolate the endophytic microbes from Q. indica, and screen on their ability in producing antimicrobial substances. Isolation is conducted by direct seed plating method using PDA and NA medium. Liquid fermentation method is taken place using supernatant F4 and PDY medium, then followed by bioassay of the supernatant on three test microbes: Staphylococcus aureus, Escherichia coli and Candida albicans. Result of isolation is 25 bacterial isolates and 12 fungal isolates. Result of bioassay showed that there are some isolates that have ability in producing antimicrobial substances. There are 1 bacterial isolate and 2 fungal isolates that showed wide spectrum activity of antimicrobial substance.

Abstract

Sonchus arvensis L. leaves have been used traditionally to treat various disease conditions. This study is designed to evaluate cardioprotective potential of ethanol extract of S. arvensis leaves on isoproterenol-induced myocardial infarction in Wistar rat. Male Wistar albino rats were divided into three main groups: negative control (saline only), positive control (isoproterenol only), and S. arvensis extract treated groups. S. arvensis extract was administered in three doses; 50, 100, and 200 mg/kg b.w. p.o for 14 days. On day 13 and 14, isoproterenol (85 mg/kg bw) was given intraperitoneally to positive control and extract treated groups. The parameters studied were cardiac biomarker enzymes which were Creatine Kinase (CK), Creatine Kinase-MB (CK-MB), Aspartate Transaminase (AST), Alanine Transaminase (ALT) and Lactate Dehydrogenase (LDH). The results showed that S. arvensis at dose of 50 mg/kg b.w. could significantly (P<0.05) reduce the level of CK, CK-MB, AST, ALT, and LDH in myocardial infarcted rats compared to positive control. The increase of the dose of S. arvensis extract was not followed by an increase of its cardioprotective activity. In conclusion, Sonchus arvensis L. leaves extract at dose of 50 mg/kg b.w. has potential to be developed as cardioprotective drug.
