Abstract

Kumis kucing leaves (Orthosiphon stamineus Benth.) are one type of plants that is able to inhibit the activity of α-glucosidase enzyme. This study aims to formulate Solid Lipid Nanoparticle from ethanol 70% standardized extract of kumis kucing leaves and which give α-glucosidase enzyme inhibitory activity. The result of SLN characteristic for particle size of formula I, II and III shows the result of 51,60 nm; 12.81 nm; 11.87 nm and zeta potential of formula I, II and III show the results -27.67; -10,7; -15.0, respectively. The results of the α-glucosidase enzyme inhibitory activity of the standardized extract, SLN formula I, II and III respectively showed IC50 of 132.9 ppm; 130.3 ppm; 131.4 ppm; 132.2 ppm and Acarbose as comparison showed IC50 of 50.0 ppm. Data processing using t-test statistically at α = 0,05 showed that extract and SLN of formula I had significant difference, thus the best SLN formula was formula I with concentration of tween 80 and propylenglycol of 6% and 10%.

Abstract

Cyclodextrin glycosyltransferase (CGTase) is an extracellular enzyme which is produced by some bacteria and archae that converts starch into cyclodextrin (CD). This enzyme has a high commercial value because of its product (CD) is widely used in several industries, such as pharmaceutical, chemistry, cosmetic, food, textile and also for environment protection. In Indonesia, there are still a few bacteria as CGTase producer were discovered and observed. The aim of this research was to find bacteria as CGTase producer from soil of corn farm land in Sumedang, West Java, and further characterized the isolate in produce CGTase. Horikoshi agar media was used as screening media. An alkalophylic bacteria as CGTase producer, JS-1 was successfully gained from this process. This isolate was characterized in the term of Gram staining, oxygen demand, motility and catalase test. The characterization results showed that JS-1 is a bacillus, Gram positive, facultative anaerob, motil and catalase positive. JS-1 isolate was also further characterized in order to determine optimum incubation period and temperature for CGTase production. The results were showed that JS-1 isolate was produced CGTase in maximum level if incubated for 48h at 37 °C.

Abstract

A study has been carried out to find out the effect of temperature, solvent flow rate and ethanol-water solvent composition on curcuminoid content in the extract produced from the extraction process (percolation) of Curcuma xanthorriza Roxb. Experiment was designed and analysed using complete random analysis with 3 factors and 2 duplications. Observed variables were: (1) varied extraction temperatures of 35°C, 45° C and 55° C; (2) varied solvent flow rates of 40 ml/minute, 60 ml/minute and 80 ml/minute; and (3) various solvent (ethanol 96%-water) compositions of 100:0,75:25 and 50:50, respectively. The measurement of curcuminoid content in the extract was done using spectrophotometer UV-visible light with wavelength setting of 420 nm. The results of analysis statistically explained that temperature and solvent flowrate did not affect significantly on curcuminoid content in the extract, but ethanol-water (solvent) composition did significantly influence on extracted curcuminoid content each shown in F table 0.01. Therefore, the optimum result of the study showed that the higher the ethanol concentration in the solvent was, the higher the curcuminoid content in the extract became. The highest curcuminoid content of 10.7% in extract was resulted at temperature of 35° C, solvent flow rate of 40 ml/minute and ethanol: water (solvent) composition of 100:0.

Abstract

Ekstrak daun sambung nyawa (Gynura procumbens (Lour.) Merr.) telah terbukti secara in vitro berkhasiat sebagai antidiabetes karena mampu menghambat enzim α-glukosidase. Penelitian ini bertujuan untuk memformulasikan ekstrak etanol 70% daun sambung nyawa menjadi sediaan tablet yang memenuhi parameter mutu fisik dan uji in vivo. Ekstrak kental daun sambung nyawa diformulasikan menjadi 3 formula dengan variasi pengisi: kalsium fosfat dibasa (formula I), laktosa (formula II) dan mikrokristalin selulosa (formula III) dengan metode granulasi basah. Selanjutnya, dilakukan uji penghambatan α-glukosidase, formula yg mempunyai aktivitas teraktif dilakukan uji aktivitas secara in vitro dan in vivo. Hasil evaluasi mutu fisik dari ketiga formula tablet yaitu organoleptik: warna hijau, rasa pahit dan berbau khas; keseragaman bobot: 510,26; 506,61 dan 508,08 mg; kekerasan tablet: 6,2; 5,6 dan 5,2%; friabilitas: 0,14; 0,13 dan 0,15%; waktu hancur: 6 menit 50 detik, 7 menit 49 detik dan 7 menit 39 detik dan hasil uji aktivitas penghambatan enzim α-glukosidase: 76,35; 67,13 dan 78,14%. Pada pengujian ini digunakan akarbose sebagai kontrol positif dengan penghambatan sebesar 88,49%. Dapat disimpulkan bahwa ketiga formula memenuhi syarat mutu fisik tablet dan mampu menghambat enzim α-glukosidase. Formula III merupakan sediaan teraktif. Hasil uji in vivo pada hari ke-14 dengan dosis penggunaan 3x2 tablet per hari dapat menurunkan kadar glukosa dalam darah.

Abstract

Lactic Acid Bacteria (LAB) has long been used in fermented milk because its stability, unique taste and texture and also for its health benents that are provided by the probiotic. There are many fermented dairy products in the market nowadays that declared containing living culture or as probiotics. The aim of this study was to identify LAB from fermented milk product by Visual inspection on plate culture for their morphology, as well by molecular technique performing PCR and DNA sequencing methods. Morphology and growth incubation time of each isolate was observed. A pair of primer that is highly speciiic for LAB targeting the gene encoding for 168 rRNA, which represents sequences of region 677 to 693 of E. coli numbering has been used for PCR identiiication in this study. A total of nineteen isolates were obtained from 10 fermented milk samples that were grown on de Mann Rogosa Sharpe Agar (MRSA) media. Nine out of 19 isolates which represent each of the samples were selected. Genomic DNA extracted from those 9 isolates were used as template for PCR. The results showed that 8 amplicons were positively identitied as LAB, but only three identities indicated similar species as declared, while 5 others did not.

Abstract

Inorganic mercury compound have been used for long time in cosmetics preparations for its skin lightening effect. The used of mercury in cosmetics has been prohibited as stipulated in the decision of the BPOM RI (Head of the Food and Drug Supervisory Agency) No.HK.00.01.432.6147. Based on health department research from various countries, there are a number of skin lightening products containing mercury between 660-57.000 μg/kg. People needs a quick and easy way to detect mercury in a lightening cream, so that people can test the safety of cream that used. One alternative that can be used to detect mercury in skin lightening cosmetics is using detecting reagents. The aim of this study was preparation of specific and sensitive mercury detecting reagents. The detecting reagents consisted mixtures of diphenylcarbazone and acetic acid in 96% of ethanol. The specificity test is performed by adding diphenylcarbazone solution into a lightening cream containing whitening agents such as mercury, hydroquinone, bangkuang extract and mixture of these substances. Assay was also performed on simulated cream that contained mercury, zinc oxide, bismuthsubnitrate, and mixture of mercury, zinc oxide and bismuthsubnitrate. The mercury detecting reagents gives a distinctive result that shown by the formation of purple mercury complex in less than 3 minutes.

Abstract

Biji pepaya mengandung vitamin E dan flavonoid yang diketahui memiliki aktivitas sebagai antioksidan. Tujuan penelitian ini untuk mengetahui efek antioksidan ekstrak etanol biji pepaya (Carica papaya L.) terhadap aktivitas SOD sel darah merah dengan metode adenokrom dan kadar MDA plasma dengan metode TBARs pada mencit yang diberi aktivitas fisik perenangan. Mencit diberi ekstrak etanol biji pepaya selama 7 hari dan perenangan selama 65 menit pada hari ke-7. Pengujian ini menggunakan 36 ekor mencit yang dibagi 6 kelompok, yaitu kelompok I (normal), kelompok II (kontrol negatif), kelompok III (kontrol positif vitamin E), kelompok IV (dosis 0,105 g/kg bb), kelompok V (dosis 0,210 g/kg bb) dan kelompok VI (dosis 0,420 g/kg bb). Hasil penelitian kelompok I, II, III, IV, V, VI berturut-turut untuk aktivitas SOD sel darah merah adalah 89±6,4329; 47,37±3,8225; 85,56±3,9210; 46,35±10,0929; 58,89±3,8271 dan 81,59±5,7700 U/mL, kadar MDA plasma berturut-turut adalah 1,6831±0,0942; 6,3567±0,1046; 1,3697±0,2254; 4,7914±0,2139; 3,8231±0,3574 dan 1,7560±0,1215 nmol/mL. Ekstrak etanol biji pepaya dosis 0,420 g/kg bb memberikan efek antioksidan paling tinggi.

Abstract

Benalu batu (Begonia sp.) is used as traditional medicine by Wana tribe in Central Sulawesi to treat various diseases, including cancer. In an effort to search potential anticancer drugs from natural sources, investigation was conducted on the cytotoxic activity of Begonia sp. methanol extract against cervic cancer cells (HeLa) and breast cancer cells (T47D) and identification of the chemical compound groups in methanol extract that is responsible for the anticancer activity. Investigations including the extraction of Begonia sp. dried herb by soxhletation using methanol, followed by evaporation on rotary vapor until viscous extract gained. The cytotoxic test were carried out by tetrazolium bromide (MTT) method. The concentration series of Begonia sp. methanol extract were 250, 125, 62.5 and 31.25 μg /mL. Results showed that the Begonia sp. methanol extract inhibited the growth of cervic cancer cells (HeLa) (IC50 = 70.97 μg/mL) stronger than inhibition on breast cancer cells (T47D) (IC50 = 122.21 μg/mL). Analysis of chemical compound groups in the methanol extract using thin-layer chromatography (TLC) method indicated that polyphenolic flavonoid might be the main compound responsible for the anticancer activity

Abstract

Factors attributing to the behavior of non-prescription used of antibiotics in Indonesia setting have not been identified yet. The availability of effective and efficient instrument or method to identify the attributing factors might be the cause of non-optimal identifying process. The objective of present study was to develop instrument, i.e questionnaire, to identify factors contributing to the behavior of non-prescription used of antibiotics. The questionnaire was developed based on factors identified on published scientific literatures. Face and content validity were conducted by having the experts’ judgement, while the construct validity was conducted by using the Spearman Correlation Test. Reliability was tested by using Cronbach’s alpha test. There were 14 rating and 8 multiple answer questions in the questionnaire that could be classified unto 5 domains. After conducting the validity and reliability test, the final questionnaire had a R value 0.276-0.628 and Cronbach’s alpha 0.833. This valid and reliable questionnaire could be used to portray the attributing factors of non-prescription used of antibiotics in larger area, therefore, the most appropriate intervention could be planeed based on the identified factors.
