Abstract

Welding was one of the industry in Purwokerto City. One of the exhaust particulate material from the welding process, this will be the potential occurrence of exposure to workers and caused renal dysfunction. Several risk factors have contributed in caused kidney difunction of workers. This study aims to determine the levels of cadmium metal (Cd), chromium (Cr), lead (Pb) with creatinine and Glomerulus Filtration Rate (GFR). The correlation of risk factors for renal dysfunction with creatinine and GFR. The risk factors that play a major role in caused kidney dysfunction. The method used is cross sectional. Sampling using purposive sampling with giving inform consent to the respondent. Provision of questionnaires and blood sampling as much as 3 ml according to ethical clearens. Measurement of metal content of Cd, Cr and Pb; serum creatinine and LFG levels. Number of respondents 30 people with 95% confidence level. The results showed that levels of Cr, Cd and Pb were 0.049 + 0.12 ppm; 1,029 + 2.38 ppm; 4,933+ 11,66 ppm. Cr levels are still within normal limits with levels but Cd and Pb exceeding the normal threshold. Creatinine levels of 1.867 + 0.446 mg / dl showed higher than normal. Minimum of GFR decrease in welding workers was 44 mL / min / 1.73 m2. Risk factors significantly correlated with creatinine and GFR was levels of Pb (p <0.05). Risk factors that play a major role in caused kidney dysfunction in welding workers in Purwokerto city is Pb content with 12.9% percentage effect on serum creatinine level and 9% effect on Glomerulus Filtration Rate.

Abstract

This research aimed to determine the various diseases caused by fungi, to identify the fungi causing the disease, and to determine the most dominant disease in red chili crops (Capsicum annum L.) from smallholder agriculture located in Kaligiri Sirampog Brebes. The research was a survey applying purposive random sampling technique. This study collected samples from five locations, within five plots placed diagonally at each site. This study picked ten infected plants from each plot, identified the disease; isolated and identified the fungi causing the disease. This research also measured the environmental factors such as temperature, humidity, and soil pH. The results showed diseases in red chili plants (Capsicum annum L.) were the leaf spot caused by Cercospora sp., the anthracnose caused by Colletotrichum sp., and the fusarium wilt caused by Fusarium sp., and the most dominant were leaf spot disease.

Abstract

The productivity of strawberry (Fragaria sp.) depends on the presence of pollinator insects that need nectar or pollen as their food. The diversity and the population size of pollinator insects can be increased by the companion planting of Borreria laevicaulis among the strawberry. This research aimed to determine the diversity of pollinator insects of strawberry plantation companion by B. laevicaulis and to determine the relationship between the diversity of pollinator insects with the strawberry fruit produced. This research was conducted in the strawberry plantation of Serang Village, of the District Karangreja, Purbalingga, from June to August 2012. The research was designed to determine the effect of companion planting of B. laevicaulis at the 0%, 5%, 10%, and 15% of the total individuals of strawberry. The results showed that the companion planting of B. laevicaulis in the strawberry field did not affect the species diversity of pollinator insects. But the B. laevicaulis companion planting affected the amount of strawberry yielded. The maximum yield was obtained by planting B. laevicaulis at the 10% of the strawberry individuals; it was increased by 76.12% of the control without companion planting.

Abstract

Surra merupakan kasus Infeksi Trypanosoma evansi pada ternak, yang menyebabkan banyak kerugian ekonomi yang dihadapi oleh peternak. Pemerintah  mengupayakan pengobatan penyakit ini dengan mengunakan obat-obatan jenis trypanosidal. Penggunaan obat secara terus menerus dapat menyebabkan terjadinya mekanisme resistensi. Di Indonesia, obat ISMM belum pernah dilakukan evaluasi terhadap penggunaannya dalam mengobati surra. Sehingga dibutuhkan penelitian untuk dapat menggambarkan kondisi T. evansi setelah dilakukan pengobatan dengan ISMM dan respon kesembuhan hospesnya. Metode infeksi T. evansi dilakukan secara intra peritoneum dan derajat parasitemia di amati setiap hari dibawah mikroskop, selanjutnya dibedakan menjadi 4 kelompok. Kelompok P1 diinjeksi dengan obat ISM dosis 0.05 mg/kg, P2 dosis 0.10mg/kg, P3 dosis 0.30 mg/kg dan kontrol yang diinjeksi dengan aquades. Selanjutnya mencit dengan parasitemia + 4 (108) di ambil darahnya melalui intracardial, kemudian darah yang diambil di amati gambaran darahnya. Hasilnya hampir keeluruhan mencit menunjukkan adanya parasit T. evansi setelah di obati dengan ISMM baik kelompok kontrol dan kelompok perlakuan,  meskipun kelompok perlakuan dapat bertahan lebih lama dibandingkan kontrol. Hasil pengamatan gambaran darah menunjukkan penurunan jumlah eritrosit hampir disemua mencit kontrol dan perlakuan (kisaran 5x106/mm3) jika dibandingkan dengan kondisi normal harusnya 7-12 x106/mm3 yang menandakan terjadi anemia, sedangkan jumlah leukositnya menunjukkan kelompok Kontrol dan P1 masih menunjukan dalam kisaran 6-12.60x106/mm3 dan kelompok P2, P3 menunjukkan 13,7x106/mm3 dan 17,4x106/mm3. Kesimpulannya T. evansi isolate ngawi diduga mengalami resistensi terhadap obat Isometamidiuklorida (ISMM) dengan menunjukan tetap adanya perkembangan T. evansi pasca pemberian ISMM, serta dosis ISMM yang diberikan tidak begitu efektif dalam mendukung hospes untuk proses penyembuhan dengan tidak adanya perbedaan gambaran darah dari mencit kontrol dan mencit yang di obati dengan ISMM.Key Words: Trypanosoma evansi, isometamidium klorida, gambaran darah,  Mus musculus, resistensi

Abstract

Peanut is one of food crops commonly consumed in Indonesia. This species comprises several cultivars such as Kancil, Bison, Jerapah, Talam, and Tuban, each of which has its individual advantages and disadvantages. The vast variation among peanut cultivars leads to the need of study on genetic diversity and relationship among them using particular molecular marker. This study aims to see whether variation on DNA sequences among some peanut cultivars amplified with atpB-rbcL primers exists or not and to know the relationship among the cultivars based on the amplicon sequences. The method involves some sequential steps, i.e. genomic DNA isolation using CTAB protocol, amplification of DNA sequence using atpB-rbcL primers and sequencing of the amplification products. Data on sequences were edited manually using Bioedit version 7.0.4.1. Sequence alignment was performed using ClustalW, which is also implemented in Bioedit version 7.0.4.1. Arlequin 2.0 was used to calculate nucleotide diversity p. Phylogenetic analysis was performed using Maximum Parsimony in MEGA 5.0. The results showed that considerably high variation in DNA sequences of some peanut cultivars amplified with atpB-rbcL primers are observed. On the other hands, very close genetic relationship among cultivars is found.

Abstract

The fish auction site at Ocean Fishing Port of Cilacap is one of the major fish landings sites in Indonesia. Fishes landed in this port mainly of large pelagic fish, bycatch, crustaceans including kemprit fish (Ilisha megaloptera Swainson,1839). Kemprit fish is a member of the family Pristigasteridae, which has an elongated body shape-sprawl. Fishers use this fish as the main ingredient to produce fish crackers and dried salted fish. Overfishing is the greatest threat to kemprit fish, and without conservation efforts the future population is uncertain. Fish farming is one right answer in kemprit fish conservation. However, information on this fish biology is insufficient. One of the biological aspects is sexual dimorphism, the morphological properties that can be used to distinguish between male and female fish. Truss morphometrics can measure the morphological traits differences of male and female. This technique in which the measurement of the distance Truss morphometrics in certain parts, by the benchmark points (dots Truss morphometrics), then compared with standard size. The result of the preliminary survey indicated that kemprit fish did not show sexual dimorphism, therefore truss morphometric technique applies to distinguish sexes of this fish. This research was a survey, and purposive sampling techniques were used to collect data. Data were then analyzed using the “t” test. The results showed that distinct truss distance between male and female fish was the distance between the anterior base of the dorsal fin to the anterior base of the ventral fins and the distance between the posterior base of the dorsal fin to the anterior base of the anal fin.

Abstract

Mutation induction in soybeans was carried out to develop high genetic diversity as a basis for plant breeding to get superior varieties.  Mutation induction can be done using physically mutagen gamma rays and chemically using EMS mutagens.  This study aims to determine the effectiveness of chemical mutagen (EMS) and physical mutagen (gamma rays) in the induction of genetic diversity in soybeans. Mutation detection can be done with molecular markers to characterize plant genetic diversity. In this study, ISSR molecular markers were used consisting of 4 primers. The method of mutation induction used EMS and gamma rays treatment. The results of the primary PCR amplification of ISSR2, ISSR3 and UBC888 showed 100% polymorphism. The treatment of mutation induction using EMS chemical agents showed a higher degree of polymorphism compared to gamma rays treatment

Abstract

Probiotics MEP+ can increase fowl weight and weft efficiency, therefore it is important to know probiotics MEP+ effect at different dosage toward reproduction aspect. This research aimed to examine duck reproduction organ size suplemented with probiotics MEP+ with different dosage within 30 days. This research used Completely Randomized Design (CRD) with 4 treatments with different dosages within 30 days which was without probiotic’s application or control (K), 0,75 ml/kg wefts (P1) dose, 1,5 ml/kg wefts (P2) dose, a n d 3 ml/kg wefts (P3) dose. Each treatment repeated 8 times. Total 40 ducks raised in floor dry cage system. At 31st day of treatment duck reproduction system organ was measured. Whole results show increase average data (±SD) for weight of both right and left testis, and liver weight with highly probiotics dosage it, however the analysis result statistic not significant (P>0,05) except weight of right left testis with duck weight or gonadosomatic indeks (GSI) were very significant (P<0,01) among all treatment at different dosages was compared control. The results is confirmed that probiotic's MEP+ treatment with different dosages within 30 days gave no effect towards duck reproduction system organ size except to gonadosomatic indeks (GSI) male duck.
