Abstract

Spiny Lobster, Panulirus homarus, farming is very potential to be developed in Lombok Island, but physiological aspects such as nutrient needs and growth hormone, especially ecdysone titer, were little known. This research aimed to determine the ecdysone titer of Panulirus homarus during the last pre-molting phase. This study was conducted as a quasi-experiment with one group post-test design. The lobsters used in this experiment have the length of carapacs and the body weight of 1.5–2.0 cm and 4.33–5.77 g respectively. The lobster fed for 42 days with gel pellets three times a day before ecdysone titer measurement. The ecdysone hormone was then isolated from the second segment of the abdomen on the basal of the fifth pereiopod. Ecdyson titer was measured by using High-Performance Liquid Chromatography (HPLC). The result showed that the titer of ecdysone was 5.27 µg/ml hemolymph. The titer of ecdysone was found higher compared to previous research on Homarus americanus; this result suggested that the spiny lobster Panulirus homarus needs a higher amount of ecdysone hormone to initiate the molting process.

Abstract

Induced Mutations with EMS (Ethyl Methane Sulfonate) has been widely reported to increase genetic diversity and improve quality of the crop. This study aimed to find out the effect of EMS immersion on short pod winged bean plant (Psophocarpus tetragonolobus (L.) DC) resistance from Rhizoctonia solani pathogens attack and to determine the optimum concentration to obtain mutan plants which resistant to R. solani pathogen attack. The method was experimental with Completely Randomized Design (CRD). Variables used were 2, i.e. independent and dependent variables. The independent variable was 4 levels of EMS concentrations (E0 = no EMS, E1 = 0,1% EMS, E2 = 0,3% EMS and E3 = EMS 0,5 %) and the dependent variable was the resistance of plants against disease intensity caused by R. solani. Observed parameters were dumping-off disease intensity caused by R. solani. The experiment was replicated 6 times. Data were analyzed using F test with confident levels of 5% and 1%. Furthermore, LSD (Least Significant Difference) test were carried out to determine the differences of each treatment. The results indicated that EMS caused the short pods winged bean plants more vulnerable against the attack of R. solani. Mutants categories obtained in the three treatments of EMS concentrations. Mutants were found in E1, E2 and E3 treatments in the intensity of dumping-off disease of R. solani pathogens in categories 1 and 4; 4; 3 and 4, respectively.

Abstract

Spiders are crucial in controlling insect pest population. The various cultivation managements such as fertilizer and pesticide application, weeding, pruning, harvesting, and cropping system affect their diversity. In the plantation, vegetation diversification has applied various practices, including monoculture, and intercropping, which influence the spider community. Thus, this study was intended to determine the spider abundance and diversity of the tea plantation, and the intercropping field (tea and strawberry) at Serang village, Karangreja Sub-District, District of Purbalingga. A survey and purposive sampling techniques were conducted, then the spiders were hand collected. Shannon-Wiener diversity (H’), Evenness (E), Simpson’s dominance (D), and Sorensen’s similarity (IS) indices were used to measure the spider diversity. The results revealed a total number of 575 individual spiders from 10 families, i.e., Araneae, Araneidae, Clubionidae, Linyphiidae, Lycosidae, Nephilidae, Oxyopidae, Salticidae, Tetragnathidae, Theridiidae, and Thomisidae. Araneidae was the most abundant in both fields. The total abundance of spiders in tea plantation (379 individuals), however, was greater than that in the intercropping field (196 individuals). Shannon-Wiener diversity reached H’= 1.873 in the plantation, and H’= 1.975 in the intercropping field.

Abstract

The research objectives were to investigate nematode community response to decomposing plant litter of varying proportions, and to measure the litter decomposition rate. This greenhouse experiments included three treatments of decomposing plant litter with proportions of 30:70, 50:50, and 70:30 (leaf litter to stem-paddy litter, weight to weight), and two controls (commercial organic fertilizers and no-organic matter). We applied the prepared plant litter and organic fertilizers on the surface of the agricultural sandy soil. Soil samples were taken after two, seven, 14, 28, 49, and 77 days to follow nematode temporal changes. Nematodes were assigned to functional groups and family level of a taxon. We used Anova to test the significance of nematode abundance among the treatments, exponential decay model to measure decomposition rates of the plant litter, and canonical correspondence analysis to investigate the correlation of varying plant proportion and nematode community composition. The results showed that nematode functional groups and family did not differ among soils of various plant litter proportions. Bacterivorous Rhabditidae and Cephalobidae, fungivorous Aphelenchidae, and fungivorous-root feeder Tylenchidae inhabited plant litter amended soils. Their abundance, however, differed significantly (p<0.001) and revealed two temporal patterns, i.e., linear (50:50) and unimodal (30:70 and 70:30). No nematodes were observed in organic fertilizer amended soil and no-organic matter soil. Nematode community composition changed over time according to the litter proportion, in which by the end of experiments, community of 50:50 and 30:70 were the least similar (r: 0.57 of axis-1 and 0.54 of axis-2, CPV: 89.06). We concluded that all litter proportions retained similar quality and supported the growth of similar nematode groups. The litter proportion, however, appeared to influence nematode abundance, their temporal patterns, and community compositions.

Abstract

Rasbora is a genus of freshwater fish of the family Cyprinidae. Three species belonging to this genus were found in Java Island; R. aprotaenia, R. lateristriata, and R. argyrotaenia. Rasbora inhabits clear running water with sandy and gravel bottom. This study aimed to determine the longitudinal distribution of Rasbora spp. based on their species richness and abundance at Banjaran River; to determine the population structure of Rasbora spp based on their body weight, standard and total length; and to identify interactions between physical-chemical factors with species richness and abundance of Rasbora spp. This research was a survey, with purposive random sampling technique applied to collect samples. The research divided the study sites into five stations based on their altitude and topography. The Cluster Analysis was calculated based on species richness and relative abundance of Rasbora spp., while the longitudinal distribution and the population structure were presented descriptively. The interaction between the physical-chemical factors and the species richness and abundance of Rasbora were analyzed using Principal Component Analysis (PCA). Thi study found two species of Rasbora, namely Rasbora argyrotaenia and Rasbora lateristriata with 80 and 2 individuals respectively. R. argyrotaenia was distributed longitudinally along Banjaran River. Meanwhile, R. latestriata was only found at the upstream. Based on the body weight, the population of R. argyrotaenia at the station three were found highest by the average body weight of 1–6 gr of a total 31 individuals. The maximum measurements of the standard length were observed at the station 3 with a range of diameter 4.0–5.9 cm of 30 individuals. The total length were also found at the station 3 with a range of measuremtnt 5.0–7.9 cm of 33 individuals. R. latestriata was found only two individual with body weights measured were 9 gr and 10 gr, the standard lengths were 7.6 and 8.5 cm, and the total lengths were 9.5 and 10.5 cm. The distribution of R. argyrotaenia was influenced mostly by pH, the speed of the water, and DO. The presence of R. lateristriata was influenced primarily by BOD and depth.

Abstract

Bawang merah merupakan salah satu tanaman hortikultura yang memiliki nilai ekonomi tinggi sebagai bumbu masakan. Ketersediaan bawang merah seringkali bersifat fluktuatif akibat pengaruh cuaca serta serangan hama dan penyakit yang berakibat terjadinya lonjakan harga di pasaran. Busuk pangkal umbi atau moler yang disebabkan oleh cendawan Fusarium oxysporum f. sp. cepae merupakan salah satu penyakit penting pada bawang merah yang secara signifikan dapat menyebabkan kehilangan hasil. Oleh karena itu, perakitan varietas unggul baru (VUB) bawang merah yang memiliki karakter ketahanan terhadap penyakit tersebut perlu terus diupayakan. Penelitian ini bertujuan untuk menganalisis kekerabatan empat genotipe bawang merah menggunakan 40 pasang marka mikrosatelit dan menganalisis polimorfisme dari marka-marka tersebut. Penelitian ini dilaksanakan pada bulan Maret hingga Agustus 2018 di Laboratorium Biologi Molekuler BB Biogen, Bogor. Hasil amplifikasi diskor sebagai data biner dan dianalisis menggunakan perangkat lunak NTSYS dan PowerMarker. Hasil penelitian menunjukkan bahwa keempat varietas bawang merah mengelompok menjadi dua klaster utama pada koefisien kemiripan genetik 0,58. Berdasarkan hasil estimasi nilai jarak genetik, sebanyak lima kombinasi tetua persilangan, yaitu antara varietas Mentes dan Sembrani (jarak genetik sebesar 45%), Mentes dan Tiron (jarak genetik sebesar 46%), Sembrani dan Kuning (jarak genetik sebesar 41%), Sembrani dan Tiron (jarak genetik sebesar 29%), serta Kuning dan Tiron (jarak genetik sebesar 37%) direkomendasikan sebagai kombinasi persilangan yang ideal untuk perakitan VUB tahan terhadap penyakit busuk umbi Fusarium. Di antara 40 marka yang digunakan, sebanyak 25 marka bersifat polimorfik dan akan menjadi bagian set marka molekuler yang akan digunakan dalam pemetaan gen terkait karakter ketahanan Fusarium ke depannya. Abstract

The aim of this research was to know the effect of supplemented Spirulina platensis in diet on leukocytes differential of gourami (Osphronemous gouramy Lac.) and to obtain the best dose. This research was carried out experimentally using a completely randomized design (RAL), which consists of 4 treatments and 3 repetitions. The fish fed with diets which contain different levels of (0, 2.4, and 6g/kg) Spirulina platensis were investigated. Sampling was done 4 times at intervals of 2 weeks of the feeding experiment, leukocyte differential (monocyte, lymphocyte, neutrophil, eosinophyl) of blood taken from fish were determined. The results showed that the highest percentage of lymphocytes was found in level of supplementation of 4 g/kg of diets, whereas the highest percentage of monocytes, neutrophils and eosinophils were found at the level suplemetation 2 g/kg of diets. Level supplementation of S. platensis 4 g/kg of diets is an efficient and effective level in stimulating immune system cells of gourami, particularly lymphocytes cell.

Abstract

Plants (Tetracera indica) commonly used by communities Musi North Rawas District for a variety of treatments. This study aims to see the effect of the use of leaf memelas to motor movement male mice. This research is true experimental with the design of Post-test only with control group design. 25 male mice divided into 5 groups. P0 was given 0 g leaf mampelas, P1 was given 2 g leaf mampelas, P2 was given 5 g leaf mampelas, P3 10 g leaves mampelas and P4 extra joss 10 mg. Treatment activities for 7 days. The calculation of motor motion is done by performing a series of tests with each of 3 repetitions, which consist of negative geotaxis test, swing test and swimming test. From the result of statistical analysis using Kruskal Wallis test, it is known that the boiled leaf water does not affect the negative geotaxis test (0,33>0,05), does not affect the hanging test (0,57>0,05) and swimming endurance  Based on the above data it can be concluded that boiling water does not affect the influence of motor activity of mice.

Abstract

Cyprinidae is a family of freshwater fish that has quite a lot of valuable members and spread almost all over the world. The purpose of this study was to determine the number of fish of the family Cyprinidae in Serayu River and their relationships. This study used survey method with purposive random sampling. Research materials were members of Cyprinidae taken from the River Serayu Banyumas, which was the collection of Mrs. Dian Bhagawati. The sequence of this study included sample collection, sample preservation, sample observation in the laboratory, i.e., identification, determination of morphological characters and species name of fish and their relationships. Observed variables were morphological characters that included body shape, colors, fins specification, the total and standard length of the body, the shape of the lateral line, the shape of the caudal fin, the position of the mouth, body length, the height and width of the caudal peduncle. The data were then computed to get the coefficient of association among species (S), clustered using UPGMA Cluster Analysis as implemented in Numerical Taxonomy System (NTSYS) version 2.02i. The observed data were also descriptively analyzed based on phenogram obtained. The fish diversity recorded were eight species i.e., Puntius orphoides, Barbodes gonionotus, Osteochilus sp., Osteochilus vittatus, Osteochilus hasselti., Labiobarbus leptocheilus, Cyprinus carpio, and Rasbora sp.. The results showed that the closest relationship based on morphological characteristics was between Puntius orphoides and Rasbora sp. which had the greatest association coefficient of 0.8292. The smallest value of the association was followed by P. orphoides and Osteochilus sp. with the value of 0.3000. This study was expected to provide information of Cyprinid fish in Serayu River and their phenetic relationship.

Abstract

The pepsin-like and lipase activities of eel with different maturity stage has been studied by an experimental method using complete randomized design (RAL) with 3 treatments and four replications. A total of 51 juvenile stage of eels (mean weight 41.25 ± 0.898 g), 14 yellow stage of eels (mean weight 319.8 ± 4.66g) and 6 silver stage of eels (mean weight 569.5 ± 9.150g) have been used for this study. Measurement of pepsin-like and lipase activities were using spectrophotometer method. The results showed that pepsin-like and lipase activities of eel did not differ significantly (P> 0.05) between different maturity stage, but pepsin-like activity was significantly different between incubation pH (P <0.05), and pH 3,4 is optimal for pepsin-like activity. Eel have equal capacity for protein and lipid digestion between different maturity stage, and has true gastric with pepsin active at acid condition (pH 3.4).
