Abstract

Papaya (Carica papaya L.) is one of the most popular fruits in the community and is generally only used as an ingredient for consumption in a fresh state. Papaya fruit production tends to be dynamic where sometimes the abundant amount of fruit may not necessarily be marketed directly. This causes the stock of papaya fruit during storage to rot and cannot be reused. This study aims to explore the potential of ripe papaya fruit to be processed as a substrate for the manufacture of liquid fertilizer using local microorganisms or effective microorganisms (EM). The quality of EM-based liquid fertilizer was determined based on the bacterial population density (CFU/mL) using the standard plate count (SPC) method on different growth media, then the levels of IAA and NPK in the applied fermented papaya fruit. The results showed the occurrence of nitrogen fixing bacteria, phosphate solubilizing bacteria and chitinolytic bacteria besides the cosmopolite bacteria. The optimum level of IAA in the fermented papaya fruit was EM + molasses (100%) while the NPK profile was still higher in the EM treatment without molasses.

Abstract

Mango is a climacteric fruit that can increase in maturity after harvest. As a result, mangoes will rot more easily because they are infected with pathogens such as fungi. The objectives of the study were to identify, examine the pathogenicity, and virulence of the fungus that causes postharvest disease in mangoes. Sampling of mangoes was carried out at the Pal market, Cimanggis, Depok by selecting fruits that were indicated to be infected with post-harvest diseases. Symptoms that appear are then described visually. Isolation of the fungus was carried out from the part of the mango fruit on Potato Dextrose Agar (PDA) media, then it was identified morphologically both macroscopically and microscopically. The virulence test was carried out by inoculating the fungus at 4 inoculation points of mango fruit. The results showed that the symptoms that appeared were symptoms of anthracnose in the form of blackish brown, sunken, and widespread spots on the mango skin. The results of morphological identification showed that the associated fungus was Colletotrichum gloeosporiodes which was characterized by white fungal colonies and fast growth, while the condia were cylindrical in shape with rounded ends. Confirmation results with Koch's postulates showed that postharvest anthracnose in mangoes was caused by C. gloeosporiodes. Meanwhile, virulence testing showed that C. gloeosporiodes had moderate to high virulence levels with symptomatic lesions ranging from 2,50 to 5,85 cm.

Abstract

The primary need for cooking oil or edible oil, the demand for cooking oil raw materials increases from year to year, this increase forces producers of crude palm oil (CPO) to compete to produce quality CPO products. PT. Sinar Pandawa, which is located in the Sennah plantation village, Batang downstream district, Labuhan Batu district, is one of the companies that produce CPO. Therefore, this study aims to determine the quality of CPO PT. Sinar Pandawa has good quality by using an analysis process of free fatty acid levels, namely the amount of free fatty acids resulting from hydrolysis and fat oxidation reactions, water content, namely the amount of water content in CPO and levels of impurities are foreign substances that do not dissolve. The method used in this research is titration using 0.1 NaOH solution for analysis of free fatty acid levels. The oven is open for moisture analysis i.e. heating the sample until the remaining water evaporates. Gravimetry for analysis of impurities content by filtering the sample using Whatman filter paper. The results show that the quality of CPO at PT. Sinar Pandawa which has the highest levels of fatty acids at 5.08%, water content of 0.35% and dirt content of 0.034%. The results of the analysis that has been carried out stated that the quality of CPO PT. Sinar Pandawa has not met the quality standards of SNI. The high value of each parameter, namely free fatty acid content, water content and dirt content with test values in accordance with SNI-2901-2006 maximum free fatty acid content of 5%, 0.25% water content and 0.25% dirt content indicates a lack of quality. CPO produced by PT. Sinar Pandawa due to lack of quality control

Abstract

Apis Dorsata Binghami honey bee is a honey bee endemic to Indonesia, living naturally in the forests of Sulawesi. This study aimed to obtain isolates and characteristics of endogenous fungi antibiotic activity from Apis dorsata Binghami nest. The research consisted of isolation of fungi from beehives using potato dextro agar, pure culture of fungi, antagonist test and antibiotic test using disc diffusion method. Antibiotic test was performed on oral bacteria and Staphylococcus aureus. The results obtained six isolates of fungi from the nest of Apis dorsata Binghami, namely isolates FAB1, FAB2, FAB3, FAB4, FAB5 and FAB6. The results of the antagonist test showed that the isolates FAB2 and FAB3 had the best antagonist properties, while the FAB6 isolates had the weakest antagonist properties. The FAB2 isolate showed the best bacterial growth inhibition zone average for the isolates of oral bacteria and Staphylococcus aureus. The inhibition zone lasts up to 3 x 24 hours so that the activity of the bacteria is bactericidal. From the results of this study, it can be concluded that the endogenous fungus Apis dorsata Binghami is a potential source of antibacterial bioactives.
