Abstract

Marine biota has many benefits for human life. Sponges are a species of marine biota that can be used as a producer of antimicrobial compounds. The bacteria found in sponges have an important role in the continuity of life in the sea. The symbiotic lifestyle that occurs in bacteria and sponges has the opportunity to form substitutions for the content of secondary metabolites and enzymes, especially extracellular enzymes (amylase, protease, cellulose and lipase). This study aims to determine how to isolate sponge symbiotic bacteria and identify spongy symbiotic bacteria. The method used is purposive to take sponges. Characterization of bacteria was carried out based on morphology and gram staining. Enzymatic bacterial selection was carried out by testing the activity of the amylase enzyme (soluble starch), lipase enzyme (Teen 80), protease enzyme (skim milk) and cellulose enzyme (carboxyl methyl cellulose). Data analysis was performed in a qualitative descriptive manner by measuring the clear zone in the extracellular enzyme test results. Meanwhile, other research parameters measured in this study were temperature, salinity and pH. The results showed that out of 47 isolated bacteria and 33 of them had extracellular enzymes with 10 bacteria had amylase enzymes, 27 bacteria had lipase enzymes, 2 bacteria had protease enzymes and 1 isolate had cellulose enzymes.

Abstract

One potential plant species for restoration in peat swamp forests is Cratoxylon arborescens. A well-managed nursery includes the application of chicken manure. This study aims to determine the effect and best dose of chicken manure fertilizer on the growth of Cratoxylon arborescens seedlings on peat planting media. This research used RAL (Completely Randomized Design) experimental method, which consisted of 4 treatments (P0 = control, P1 = 10% chicken manure + 90% peat, P2 = 30% chicken manure + 70% peat and P3 = 50% chicken manure + 50% peat) with 5 replications. The data obtained were analyzed statistically using variance analysis, followed by Duncan's New Multiple Range Test (DNMRT) at the 5% level. The results showed that chicken manure had a significant effect on the growth of Cratoxylon arborescens seedlings, because it was able to increase fertility and nutrient availability, improve the physical, chemical and biological properties of peat. The most effective and best treatment for four-month-old Cratoxylon arborescens seedlings was P2 (30% chicken manure + 70% peat) with 100% survival percentage, average height increase of 14.39 cm, average diameter increase of 1.56 mm, dry weight of 7.41 g and seedling robustness of 8.35

Abstract

Browning of explants is a common problem in Kepel (Stelechocarpus burahol (Blume) Hook.f. & Thomson) in vitro culture, resulting in low explant in vitro regeneration of Kepel. Using nodal explant, the effect of explant immersion in ascorbic acid, the addition of ascorbic acid to the media, and dark and light incubation conditions were investigated. The browning prevention method was selected based on the delayed browning appearance time, the lowest broning intensity, and the highest percentage of callus. This present study used a completely randomized design with treatment variations: immersion explant in ascorbic acid 100 mg/L, addition of ascorbic acid 100 and 200 mg/L to the media, addition of ascorbic acid 100 and 200 mg/L and activated charcoal 1 g/L to the media, and incubation in dark and light conditions for 28 days. The results showed that the combination of the addition of ascorbic acid 200 mg/L to MS media and incubation in dark conditions were effective browning prevention methods in inhibiting browning appearance time (5 DAP), significantly reducing browning intensity (0.3), and increasing callus growth (100%) of kepel node explants during 28 days of culture. The results of this study are useful in establishing protocols for in vitro culture of Kepel plants, especially at the initiation stage, and can be expected to support the successful conservation of Kepel plants through in vitro propagation. Keywords: Ascorbic acid, Activated Charcoal, Browning, In Vitro Culture, Stelechocarpus burahol
