Abstract

Agriculture is an important sector in the Indonesian economy, with vegetables such as mustard greens (Brassica juncea) that have high economic value and are important for the healthy diet of the community. One of the main challenges in mustard cultivation is the optimal management of soil moisture, as unstable moisture levels can affect the quality and quantity of the harvest. This research aims to design and implement an automatic irrigation system based on Internet of Things (IoT) technology using the ESP32 microcontroller and soil moisture sensors, as well as applying Mamdani Fuzzy logic to manage irrigation adaptively. This system is capable of increasing water usage efficiency by up to 50% by maintaining soil moisture at optimal levels, which positively impacts the growth of mustard plants. In addition, this system allows for remote monitoring through an IoT application, making it easier for farmers to manage irrigation more efficiently. The research results show that the implementation of this automated system not only reduces dependence on manual labor but also ensures more efficient water usage, thereby providing an innovative solution for irrigation efficiency in the agricultural sector.  Keywords - ESP32, Mamdani Fuzzy, Smart Irrigation System, Soil Moisture.

Abstract

Most infectious diseases occur in the digestive tract caused by Salmonella typhimurium, while in the respiratory tract caused by Staphylococcus aureus. Continuous use of antibiotics as a treatment for bacterial diseases can result in resistance, therefore it needs to be balanced with the discovery of herbal medicines. Jatropha curcas is thought to have the ability to inhibit the growth of gram-positive and gram-negative bacteria. The purpose of this study was to obtain data and information on the antibacterial activity of Jatropha curcas leaf extract. This study used the paper disc diffusion method with 6 treatments and 3 repetitions. The treatments used were positive control (chloramphenicol), negative control (DMSO), and concentration of Jatropha curcas leaf extract of 10%; 20%; 40% and 80%. The results showed that Jatropha curcas leaf extract at concentrations of 10%, 20%, 40% and 80% had an effect on the growth of Salmonella typhimurium with the average inhibition zone obtained being 9.78; 9.19; 11.95; and 16.88 mm. At concentrations of 10%, 20%, 40% and 80%, it has an effect on the growth of Staphylococcus aureus with the average inhibition zone results obtained being 13.23; 14.83; 16.01; and 18.03 mm.Keywords – Jatropha Leaf Extract, Salmonella Typhimurium, Staphylococcus Aureus.

Abstract

High quality extracted RNA from plant tissues is used in downstream transcriptomic application that needs high quantity for multiple analysis. Attaining high quality and high yield for banana plant tissue is a challenge with its high number of secondary metabolites becoming contaminants, effecting the purity of RNA extracted. A procedure to extract banana plant RNA with high quality and quantity in this study is developed to be simple, robust, affordable and accessible using basic lab equipment and materials. This study optimizes RNA extraction from the leaf of juvenile Pisang Susu (Musa spp.) using a combination of Li buffer (modified CTAB) and GENEzol reagent. Treatment differs in the use of β-mercaptoethanol and the duration of sample precipitation with isopropanol. NucleoSpin RNA Plant extraction kit was made as comparison. Measurement of RNA quantity used quantus fluorometer, and quality measured by spectrophotometer. Results showed that the addition of β-mercaptoethanol in Li buffer is vital with samples left to precipitate overnight providing best results. The total RNA obtained had a higher yield compared to the commercial kit with 108 – 211 ng/µl and 0.35 – 0.37 ng/µl respectively. Light absorbance A260/280 indicating sample purity between method has similar RNA quality of 0.917 – 1.084 and 0.843 – 1.026. This study proved that modification of CTAB buffer using Li buffer for RNA extraction resulted in better quality and quantity of RNA compared to the commercial RNA extraction kit.Keywords – Banana, Optimization, RNA Extraction.

Abstract

Urban forests hold potential as carbon sinks while also providing vital ecosystem services. This study aims to analyze the carbon sequestration potential of Cipayung and Munjul Urban Forests in East Jakarta and to offer policy recommendations for the Jakarta Provincial Government. Field measurements were conducted using Diameter at Breast Height (DBH) data, which were collected through stratified random plot sampling. Biomass was estimated using the IPCC (2006) allometric equation, combined with species-specific wood density values. The results show that Cipayung Urban Forest sequesters approximately 111.52 tons CO₂ eq, while Munjul Urban Forest sequesters 42.10 tons CO₂ eq. These differences are influenced by vegetation structure and species composition. Ecologically, urban forests contribute to local greenhouse gas reduction while supporting adaptation to extreme weather, including reducing urban temperature and improving water absorption capacity. Policy implications include the need for the Jakarta Provincial Government to strengthen urban forest protection regulations, integrate urban forests into spatial planning, and enhance participatory community-based management. The limitation of this study lies in its coverage, which was restricted to only two urban forests; therefore, future studies should expand to other urban green areas across Jakarta.Keywords - Carbon Sequestration, Jakarta, Urban Forest.

Abstract

PT XYZ is a manufacturing company that provides hot metal transportation handling services and supports Blast Furnace (BF) operations at the Krakatau Posco Plant using heavy equipment, namely Multi Mover M600U. To ensure smooth production processes, effective and efficient maintenance management of the heavy equipment are essential to enhance reliability. This study evaluates potential failure modes, their impacts and appropriate handling measures for the Multi Mover M600U using the Failure Mode and Effect Analysis (FMEA) method. The result of the study shows the highest Risk Priority Number (RPN) value of 350 in the Electrical PLC component, with a total RPN of 777 for the Electrical and Brake systems. These findings indicate that the Electrical and Brake systems have the highest failure rates due to downtime compared to other systems. Through the Reliability Centred Maintenance (RCM) approach, this study proposes a maintenance scheduling plan to minimize the risk of sudden downtimes, and their impacts on time and maintenance costs.Keywords - Downtime, Failure Mode and Effect Analysis, Heavy Equipment Maintenance, Reliability Centred Maintenance, Risk Priority Number.

Abstract

Feed is a main factor influencing fish growth. However, feed formulation using traditional methods such as the Pearson Square is often considered complex and time-consuming for fish farmers. This study aims to design and development an Android-based application named “Hitung Pakan”, which can calculate fish feed formulations quickly and accurately. The research employed the waterfall method, including requirement analysis, system design, implementation, and testing. The application was developed using Java programming language and tested by 10 respondents from a fish farming group. The results showed no differences between the application’s calculations and manual calculations using Pearson Square method equations. This indicates that the developed application provides accurately results, simplifies the formulation process, and reduces human error. Therefore, the application is feasible to be used as a formulation tool in fish farming group.Keywords - Android Application, Calculation Efficiency, Fish Feed Formulation, Pearson Square.

Abstract

Corn production produces waste in the form of corn cobs that have not been optimally utilized, even though they contain complex carbohydrates that can be converted into Bioethanol. The problem faced is the cessation of the use of agricultural waste as an alternative energy source, especially in the context of direct conversion into electrical energy. This study aims to analyze the potential for converting corn cob waste into Bioethanol and then calculate the potential electrical energy that can be generated from a mixture of Bioethanol and gasoline (E0, E10, E15, E20) through simulation using Aspen Plus V10 software. The results show that from 825 kg of corn cob waste per ton of plant, 58.5684 L/hour of Bioethanol can be produced, with the highest electrical energy potential of 542.42 kWh in the E20 mixture. Utilization of corn cob waste as Bioethanol offers an environmentally friendly and sustainable renewable energy solution.Keywords – Bioethanol, Corn Humps, Energy, Fermentation.

Abstract

Coffee is one of the world's most popular beverages and is renowned for its health benefits derived from its bioactive compounds, such as caffeine, chlorogenic acid, melanoidins, cafestol and kahweol. This research aims to assess the potential functional properties of coffee based on the latest scientific literature. Seventy-three scientific articles were systematically reviewed to identify coffee’s diverse biological activities its diverse biological activities, including antioxidant, anti-inflammatory, antimicrobial, anticancer, anti-obesity, anti-diabetic, hepatoprotective, and cognitive-enhancing effects. Furthermore, compounds in coffee have shown potential as prebiotics and anti-aging agents. However, coffee consumption carries toxic risks associated with excessive caffeine intake, ochratoxin A, and 5-hydroxymethylfurfural (5-HMF). Therefore, understanding the correct consumption dosage and safe processing methods is crucial. This review concludes that coffee possesses significant potential as a functional food that supports overall public health and warrants further development in the food and health sectors.Keywords – Antioxidants, Coffee, Functional Foods, Bioactive Compounds, Toxicity.

Abstract

Effective waste management is a key prerequisite for achieving sustainable tourism. Sustainable tourism provides important benefits such as environmental conservation, improved community well-being, and economic contributions to destinations. This study aims to analyze the existing waste management system in the Ancol tourism area, identify the challenges faced, and formulate recommendations for its optimization. The analysis focuses on the waste management system at the Integrated Waste Processing Facility (TPST) Ancol, Jakarta, with reference to national regulations (Law No. 18/2008, Jakarta Governor Regulations No. 102/2021 and No. 142/2019) and the Global Sustainable Tourism Council (GSTC) standards. A descriptive qualitative approach was applied through observations, interviews, and document analysis. The results show that TPST Ancol has implemented four-stream waste segregation and circular economy-based processing, such as composting for parks and sorting recyclable waste for waste banks. However, challenges remain in terms of limited visitor participation in source segregation, constraints in human resources, and inadequate technological infrastructure. The Soft Systems Methodology (SSM) analysis highlights the importance of visitor education, infrastructure improvement, and multi-stakeholder collaboration to strengthen the triple bottom line (people, planet, profit). This study recommends optimizing monitoring systems, enforcing stricter environmental policies, and integrating educational programs to position Ancol as a world-class sustainable tourism destination.Keywords - Ancol, Soft Systems Methodology, Sustainability, Tourism, Waste.

Abstract

Hypertension is a major risk factor for cardiovascular diseases, leading to serious health complications. Genetic polymorphisms in the COX-2 gene, especially the rs689466 variant, are associated with increased cardiovascular risk, but its link to hypertension in the Indonesian population remains limited. This study investigates the relationship between COX-2 rs689466 polymorphism and hypertension in Indonesia, using a cross-sectional design with 200 samples (100 hypertensive patients and 100 healthy individuals). DNA was isolated from whole blood using the salting-out method, and the COX-2 rs689466 polymorphism was analyzed by PCR-RFLP. Statistical analysis was performed using the Chi-square and Mann-Whitney tests. The results showed a significant association between the COX-2 rs689466 polymorphism and hypertension (p<0.001), with the AA genotype carrying a higher risk (p<0.001, OR=8.609) compared to individuals with the GG or AG genotypes. Smoking exacerbated this association, with smokers being more likely to carry the AA genotype and have an increased risk of hypertension (p<0.001, OR=6.091). This study highlights the role of COX-2 rs689466, particularly the AA genotype, in increasing hypertension risk in Indonesia, with smoking further worsening vascular inflammation and resistance, contributing to hypertension development.Keywords - Cyclooxygenase-2 (COX-2), Hypertension, Polymorphism, Smoking, Rs689466.
