Abstract

One of the steps in making histological preparations is staining. Staining is a stage that aims to simplify the process of observing tissue structure and also makes it easier to distinguish the parts of the tissue that will be observed. Before staining, mast cells are difficult to identify visually under a microscope. Hematoxylin eosin is a routine stain in histotechnics and alcian blue safranin is a special dye that can stain mast cells. Mast cells can be found at the boundaries of body tissue and the external environment, one of which is the skin. Mast cells found in the skin are located in the dermis of the skin. The aim of this study was to determine the histology of skin connective tissue mast cells in Wistar rat (Rattus norvegicus) preparations stained with hematoxylin eosin and Alcian blue safranin. This research uses a experimental laboratory research type with a qualitative descriptive research design. The results obtained from observations of the histology of mast cells in the connective tissue of the skin of Wistar rats (Rattus norvegicus) with hematoxylin eosin staining from 16 preparations were found to be 62.5% good and 37.5% unfavorable. Staining with alcian blue safranin of 16 preparations was found to be 81.25% good and 18.75% unfavorable. The research results from preparations stained with alcian blue safranin showed better results compared to preparations stained with hematoxylin eosin.

Abstract

Elderly people experience a decline in physical function that can affect their quality of life, one of which is a decline in digestive function, namely a reduction in stomach acid production of up to 30%. In Indonesia, many elderly people have a habit of drinking coffee. Coffee contains caffeine and tannin polyphenols, which are known to inhibit iron absorption. These physiological changes and consumption habits have the potential to affect iron metabolism, including serum iron and ferritin levels. This study aims to determine the difference in serum iron and ferritin levels in elderly coffee consumers and non-consumers in RW 8, Simomulyo Baru Village, Sukomanunggal District, Surabaya City. Serum iron levels were examined using the colorimetric method, while ferritin levels were examined using the Hipro A1 tool using the Nephelometry Immunoassay method. Data were analyzed using the Mann-Whitney test. The results showed a significant difference in serum iron levels between coffee consumers and non-consumers (p = 0.029). However, no significant difference was found in ferritin levels between the two groups (p = 0.468). This suggests that coffee consumption may influence fluctuating serum iron levels, but does not significantly impact long-term iron stores as reflected in ferritin levels.

Abstract

Malondialdehyde (MDA) can reveal the oxidation processes occurring within cell membranes and is a reliable and accurate biomarker of oxidative stress. Increased levels of MDA are linked to pathological situations damage to blood vessel walls, heart disease, and cancer. Garcinia tetrandra Pierre, a Garcinia species that is rich in secondary metabolite compounds but has not been widely studied. This study aims to evaluate MDA levels in liver tissue homogenates of rats in a carrageenan-induced peritonitis model. This study began with a qualitative test of secondary metabolites to analyze the content of tannins, saponins, flavonoids, terpenoids and phenolics. In vivo tests using 30 male Wistar The rats were split up into six groups: the standard drugs group, the G. tetrandra methanolic extract treatment group dose of 200 mg/kg BW (P1), dose of 400 mg/kg BW (P2), and dose of 600 mg/kg BW (P3), the positive control group, and the negative control group. Liver tissue was made into homogenate to measure MDA levels using the UV-Vis spectrophotometer method. The results showed that the MDA levels in the liver of the treatment group were significantly different from the positive control group (P<0.05). This is supported by the presence of secondary metabolite compounds that are radical scavenging. It can be concluded that the methanol extract of G. tetrandra fruit skin is able to reduce liver MDA levels in peritonitis model mice so that it can be said to have the potential as a candidate for anti-inflammatory drugs.

Abstract

Dengue Hemorrhagic Fever (DHF) remains a major health problem in endemic regions, requiring accurate laboratory examinations to support diagnosis. Serological testing of immunoglobulin G (IgG) and immunoglobulin M (IgM) is widely used to differentiate between primary and secondary dengue infections. This study aimed to analyze the serological profile of IgG and IgM as indicators of primary and secondary dengue infection in patients with DHF. A descriptive cross-sectional design was employed using secondary data from 170 patients who underwent IgG and IgM testing at RSUD Haji Surabaya. Data were analyzed descriptively by calculating frequency distributions and percentages. The results showed that the distribution was dominated by IgG positive and IgM negative findings (62%), followed by both IgG and IgM positive (17%), IgM positive only (10%), and both negative results (11%). These findings indicate that most patients experienced secondary dengue infection. Therefore, IgG and IgM serological testing plays an important role in determining the phase of dengue infection; however, it should be combined with other diagnostic methods to improve diagnostic accuracy.

Abstract

Type 2 Diabetes Mellitus is often accompanied by lipid metabolism disorders that may increase the risk of cardiovascular complications. This study aimed to determine the relationship between glucose levels and High-Density Lipoprotein (HDL) and Low-Density Lipoprotein (LDL) levels in patients with Type 2 Diabetes Mellitus. This study used a cross-sectional design with a descriptive-analytic approach. The study was conducted at the Clinical Pathology Laboratory of RSDI Banjarbaru in July 2025 using secondary data from the medical records of 45 patients with Type 2 Diabetes Mellitus selected through purposive sampling. Data were analyzed using the Chi-Square test and Fisher’s Exact Test. The results showed that most respondents had high glucose levels with normal HDL levels in 33 patients (73.3%) and high LDL levels in 41 patients (91.1%). Statistical analysis showed no significant relationship between glucose levels and either HDL or LDL levels (p > 0.05). It can be concluded that glucose levels were not significantly associated with HDL and LDL levels in patients with Type 2 Diabetes Mellitus.

Abstract

Typhoid fever is a systemic infectious disease caused by Salmonella typhi and remains a health problem in developing countries, including Indonesia. The Widal test is still widely used as a supporting diagnostic examination for typhoid fever because it is simple and rapid. Typhoid infection may also cause changes in hematological parameters such as leukocyte and platelet counts. This study aimed to determine the relationship between Widal titers and leukocyte and platelet counts in typhoid fever patients. This study used a quantitative design with a cross-sectional approach. A total of 21 typhoid fever patients were selected using purposive sampling technique. Leukocyte and platelet examinations were performed using the Mindray BC 5300 Hematology Analyzer. Data analysis was conducted using the Shapiro-Wilk normality test followed by Spearman correlation test. The results showed that the correlation between Widal titer and leukocyte count obtained p=0.987 and r=0.004, while the correlation between Widal titer and platelet count obtained p=0.935 and r=0.019. These findings indicate that there was no significant relationship between Widal titer and leukocyte or platelet counts in typhoid fever patients. However, there was a significant relationship between leukocyte and platelet counts with p=0.000 and correlation coefficient r=0.753. In conclusion, Widal titer had no significant relationship with leukocyte and platelet counts in typhoid fever patients, while leukocyte and platelet counts showed a strong positive correlation.

Abstract

Blood glucose examination is an important parameter for screening and monitoring diabetes mellitus. The accuracy of glucose testing is influenced by pre-analytical factors, particularly delayed examination and specimen storage conditions. Delayed examination may cause a decrease in glucose levels due to ongoing glycolysis after blood collection. This study aimed to determine the effect of examination delay time and storage conditions on plasma glucose levels. This laboratory experimental study used a factorial design with repeated measures. The delay times consisted of 0, 2, and 4 hours, while the storage conditions included EDTA plasma stored at room temperature, EDTA plasma stored at 4ºC, and NaF plasma. Plasma glucose levels were measured using the GOD-PAP method. Data were analyzed using the Shapiro-Wilk test, Levene’s test, Two-Way ANOVA, and Tukey Post Hoc test. The results showed that plasma glucose levels decreased as the examination delay time increased. After 4 hours of delay inEDTA plasma at room temperature, glucose levels were reduced by 55.8%, EDTA plasma stored at 4ºC by 52.3%, and NaF plasma by 30.7%. There was a significant effect of variations in examination delay time and storage conditions on plasma glucose levels (p<0.005). It can be concluded that NaF plasma was more effective in maintaining glucose stability compared with EDTA plasma stored at room temperature or under refrigeration conditions.

Abstract

The stability of commercial control sera is affected by storage techniques and duration, directly impacting laboratory accuracy. This study aimed to analyze the differences in creatinine stability in control sera stored using aliquot and non-aliquot methods based on storage time. A quasi-experimental approach (one-group pretest-posttest) was applied to control sera stored at 2-4°C and observed on days 0, 5, 7, and 9. Data from 48 tests were evaluated using the Independent Sample T-test. Results indicated an escalation in creatinine levels corresponding to storage duration. The aliquot group increased from an average of 1.36 g/dL to 1.52 g/dL, while the non-aliquot group spiked more sharply from 1.39 g/dL to 1.63 g/dL. Significant instability emerged on the ninth day (p<0.05). In conclusion, the non-aliquot technique triggers more severe deterioration or pseudo-level increases in creatinine compared to the aliquot technique over extended storage periods. Therefore, the use of the aliquot method is highly recommended to maintain the stability of control sera during storage.

Abstract

Fungal infections may occur in the lungs due to Aspergillus infection or in subcutaneous tissues as a result of chromoblastomycosis. Histopathological staining methods commonly used for fungal detection include Periodic Acid–Schiff (PAS) and Gomori's Methenamine Silver (GMS). This study aimed to compare the staining quality of conventional PAS and GMS methods with their modified counterparts, PAS-Tartrazine (PAS-T) and GMS-Phloxine Tartrazine (GMS-PT), in tissues infected with Aspergillus and chromoblastomycosis. This study employed a descriptive exploratory design and was conducted at Sudarma Medical Laboratory, Surabaya, in May 2024. The study samples consisted of paraffin-embedded tissue blocks stained using PAS, GMS, PAS-T, and GMS-PT methods. A total of six samples were examined, with each sample processed in duplicate. Staining quality was evaluated using a four-point scoring system (scores 1–4) based on the clarity of fungal structures, cell nuclei, and cytoplasm. The results were statistically analyzed using the Mann–Whitney test. The comparison between PAS and PAS-Tartrazine staining demonstrated significant differences (p < 0.05) in the visualization of fungal structures and cell nuclei. Likewise, comparison between GMS and GMS-Phloxine Tartrazine staining revealed significant differences (p < 0.05) in the visualization of cell nuclei and cytoplasm. PAS-Tartrazine staining provided clearer visualization of fungal structures than conventional PAS staining, whereas both GMS and GMS-Phloxine Tartrazine produced equally clear visualization of fungal structures.

Abstract

Hyperlipidemia is a lipid metabolism disorder characterized by elevated levels of cholesterol, triglycerides, and low-density lipoprotein (LDL), which can lead to increased creatinine and uric acid levels due to impaired kidney function and oxidative stress. Moringa leaves (Moringa oleifera) are known to contain flavonoids, phenolics, and antioxidants that have the potential to improve these conditions. This study aims to determine the effect of moringa leaf extract on creatinine and uric acid levels in male Wistar strain white rats with hyperlipidemia. The study was conducted from May to June 2026, using a pretest-posttest control group design with five treatment groups of hyperlipidemic rats: a negative control, a positive control (simvastatin at a dose of 0.18 mg/200 g body weight), and moringa leaf extract administered at doses of 100 mg/200 g body weight, 150 mg/200 g body weight, and 200 mg/200 g body weight. The results of the Two-Way ANOVA test showed that creatinine and uric acid levels decreased in all groups after treatment. The time factor (before and after treatment) showed a significant difference (p<0.001), but between treatment groups did not show a significant difference in creatinine levels p=0.113 or uric acid levels p=0.096 (p>0.05). In conclusion, the administration of Moringa oleifera leaf extract has an effect on reducing creatinine and uric acid levels in male white rats of the Wistar strain with hyperlipidemia, but there was no significant difference in effectiveness between treatment groups.
