Abstract

Lola (Trochus niloticus) is one of the largest sea snails that lives in the shallow water in Eastern Indonesia and the Pacific. In Saparua Island (Central Maluku), lola population tends to decline due to its exploitation for industrial need. The aim of this study is to determine the reproductive potential of lola snail in Saparua Island, Central Maluku. Lola snails were collected from six locations i.e Booi, Haria, Ullath, Ouw, Itawaka, and Nolloth (03.290-03.800 LT dan 128.320-128.430 BT). In each location a strip transect 100 x 2 m was made perpendicular to the coastline started from the lower tide level. Lola snail populations were grouped into different age classes based on the shell diameter using Bhattacharya method. The reproductive potential of lola snail was analyzed by the value of the net reproductive rate. Gonad histology analyses were made by using paraffin methods and stained by Ehrlich’s Haematoxylin-Eosin. The results showed that the net reproductive rate of lola population is 226 individuals and its generation time is 2.88 years. The histological observations showed that the oocyte consist of proliferation, early developing and mature stages. Key words: Bhattacharya, oocyt, reproductive potential, Trochus

Abstract

A research was undergone to study the effect of single and double infection of Cucumber Mosaic Virus (CMV) and Chili Veinal Mottle Virus (ChiVMV) on the growth and yield of five chilli cultivars, i.e. Prabu, Taro, Jatilaba, Laris, and Keriting Bogor. Mechanical inoculation was conducted to transmit the virus. Infection of the virus was then confirmed with DAS-ELISA. Severe symptom was observed on plant given double infection compared to those given single infection. The rate of plant growth and the amount and weight of fruits were reduced. The type of interaction between CMV and ChiVMV on most chilli cultivar can be considered as interference and additive. Synergism interaction was only observed on cultivar Laris. Based on symptom expression and reduction on yield, it can be concluded that all chilli cultivars used in this study could not hold up the virus infection. Abstract

Pathogenic bacteria produced protease that involved in molecular mechanism of foodborne disease. Produced protease involved in molecular mechanisms of foodborne diseases. The purpose of this research was to screen, identify and characterize the potential microorganisms associated with sponge as producer of protease inhibitor. Among 96 isolates examined, four isolates i.e 10A6, 6A3, 9A51, and 1A12 yielded protease inhibitors which were potential to inhibit protease substrates (40-90%). One of the most potential protease inhibitor producer, the bacteria isolate 6A3, was identified as Chromohalobacter sp. Chromohalobacter sp.6A3 produced protease inhibitor with optimum temperature and pH 300 C and 5, respectively. The inhibitor activity was stable when incubated at 400 C for ten minutes or at 300 C for 8 hours. Key words: Bacteria, Chromohalobacter sp., protease inhibitor, screen, sponge

Abstract

Four oligoresveratrols ranging from dimer to tetramer, isolated from stem bark of Hopea mengarawan (Dipterocarpaceae) plants were tested for their activity as hydroxyl radical scavenger. The activity of these compounds was evaluated against the 2-deoxyribose degradation induced by the hydroxyl radical generated via a Fenton-type reaction. Result showed that balanocarpol, heimiol A, vaticanol G, and vaticanol B had IC50 3.83; 15.44; 2.01; and 4.71 uM, respectively. These results suggest that oligoresveratrols from stem bark of H. mengarawan maybe useful as potential sources of natural antioxidants. Key words: Oligoresveratrol, Hopea mengarawan

Abstract

The aims of this research were to determine diurnal patterns of testosterone and cortisol metabolites to predict the testis functional status. In this study, fecal testosterone and cortisol were quantified in 77 samples from three male Hylobates moloch during a course of three months period. These data showed that the highest concentration of fecal testosterone occured at 18.00-06.00 (23.61 ng/g dried feces), then declined gradually. The lowest concentration was in the evening (5.54 ng/g dried feces). Our tests showed that there was a decrease in the mean testosterone concentration from 06.00-10.00 to 10.00-14.00 to 14.00-18.00. For cortisol, the highest concentration occured at 06.00-10.00 (597.84 ng/g dried feces), then decline gradually in the evening (225.73 ng/g dried feces). Abstract

The objectives of this experiment were to analyze the endo- or exo-chitinase activities of healthy and Sclerotium rolfsii infected peanuts. The experiment analyzed 24 different peanut genotypes. Results of the experiment showed chromogenic dimer was the most suitable substrate for analysing chitinase activities. Both endo- and exo-chitinases activities were detected in leaf, stem, and crown tissues. Increased in chitinase activities were detected in S. rolfsii infected peanut tissues than in healthy plant. Regression analysis showed negative slope between disease intensity and chitinase activity in S. rolfsii infected peanut tissue (R2= 0.45). Abstract

The term of allelopathy refers to chemical interactions (inhibitory or stimulatory) between plants, between plants and microorganisms, and between microorganisms. The wealth of information on the processes, procedures, and practices of allelopathy has contributed to understanding this field of science. Recently, researches of allelopathy have been conducted in laboratory, greenhouse, and field with multifaceted standpoint in some concerning area: (i) allelochemicals identifications and screening test; (ii) ecological and physiological aspects of allelopathy; (iii) genetic studies and the possibilities of using plant breeding or genetic manipulation to enhance allelopathic varieties; (iv) the use of allelopathic potential in the biological control, including as natural pesticide, of weeds and plant diseases as eco-friendly approach for sustainable agriculture scheme. Abstract

This study was conducted to observe intracellular antioxidant cooper,zinc-superoxide dismutase (Cu,Zn-SOD) in liver tissue of rats under hypercholesterolemic condition by using immunohistochemical technique. A total of twenty male Wistar rats were used for this study. Those rats were divided into two groups; (i) control group and (ii) hypercholesterolemic group, which were fed died containing 1% cholesterol for eight weeks. Rat livers were taken at the end of treatment, and processed by using paraffin embedding standard method. The tissues were stained immunohistochemically to Cu,Zn-SOD. Observation of Cu,Zn-SOD content in the tissue was performed qualitatively in the cytoplasm and quantitatively in the nucleus of hepatocytes based on colour intensity of enzyme reaction product. The profile of antioxidant-Cu,Zn-SOD decreased (P < 0.05) in the hypercholesterolemic group compared to the control group. Key words: Superoxide dismutase (SOD), hypercholesterolemia, liver, rat

Abstract

The research aim was to examine the enhancement of catharanthine content in Catharanthus roseus callus culture added with different concentration of Naphtalene Acetic Acid (NAA). NAA treatment produced callus that formed hairy roots. Fresh and dry weight of callus increased as the increasing of NAA concentration. The catharanthine content of C. roseus callus culture was increased by adding NAA as well. The highest catharanthine content was found in 2.5 ppm NAA added callus. Abstract

Both Lumbricus rubellus and Pseudomonas putida decompose soil organic matters. The population of P. putida Pf-20 increased if L. rubellus was introduced to the cucumber growth medium. The process of organic decomposition was much better if the medium was introduced with both L. rubellus and P. putida Pf-20, compared to the medium contained only either one of those organisms. The activity of L. rubellus may serve to provide nutrients for both the cucumber and P. putida. The role of P. putida to reduce disease severity was increased if L. rubellus was introduced to the growth medium. The synergism of these two organisms, reduced either the level of disease severity to CMV-48 and C/N ratio of medium, but increased the content of available phosphor and potassium.
