Abstract

Ameliorant is a substance that can increase soil fertility by improving the soil's physical, chemical, and biological conditions, and it can be organic or inorganic. Some ameliorants often used are manure, lime, or a combination of all these fertilizers. This study aimed to determine the effect of ameliorants on the growth and production of sweet corn. The study was conducted at the Experimental Station of the Faculty of Agriculture, University of Muhammadiyah Jakarta, from July to October 2022. The study used a Randomized Completely Block Design with five ameliorant treatments and five replications, so there were 25 experimental units. Each experimental unit consisted of three plants. The treatments given were P0 (without ameliorant as control)), P1 (calcite 5 g/plant), P2 (dolomite 5 g/plant), P3 (green mussel shell waste 5 g/plant, and P4 (zeolite 5 g/plant). The results showed that ameliorants gave similar results as controls and did not increase the growth and production of sweet corn plants. Keywords: ameliorant, green mussel shell, soil pH

Abstract

Soft rot disease in orchid plants is a factor in decreasing orchid production. One way of biological control that has the potential to spur growth is to utilize endophytic bacteria. This study aims to describe the diversity of orchid endophytic bacteria that have the potential as growth promoters and biocontrol agents. Observation parameters include indole acetic acid (IAA) production, inorganic phosphate dissolution, ammonia production of orchid endophytic bacteria, and antagonism of endophytic bacteria to pathogenic bacteria that cause soft rot in vitro. The diversity of orchid endophytic bacteria was analyzed using rep-PCR with BOX-A1R primer. The results showed that 10 isolates of orchid endophytic bacteria were able to produce IAA with a concentration of 79–321 μg/mL, able to dissolve phosphate, and produce ammonia. In antagonism testing against soft rot-causing bacteria, all isolates were able to form an inhibitory zone with a diameter of 3–7.2 cm. Furthermore, diversity analysis using rep-PCR showed that isolates of endophytic bacteria DnPh5, BgCt2, BgVt10, DnLp7, DnBl1, and DnAr4 had a uniform DNA band pattern so that they were grouped together. Meanwhile, isolates of TbPh7, IbtPhm1, DnDr2, and AkOc1 showed different DNA band patterns, indicating that the four isolates were different species or subspecies of bacteria. In dendrogram analysis, DnPh5, BgCt2, BgVt10, DnLp7, DnBl1, and DnAr4 isolates have a similarity of 100%. The similarity of isolates IbtPhm1, DnDr2, and AkOc1 was 90%, 79%, and 68%, respectively, against the other six isolates. TbPh7 isolate is an isolate with a low level of similarity to other isolates, which is about 51%. Keywords: orchid, endophytic bacteria, growth promoter, biocontrol agent, rep-PCR

Abstract

Sugar cane (Saccharum officinarum L.) is the main ingredient in sugar production. Sugarcane is widely cultivated in warm and tropical areas and is widely used as a sweetener because it contains a lot of fructose and glucose. National sugar production shows a decline, the majority of which is caused by pathogens, especially the fungus Bipolaris sp. capable of producing up to 85% damage per Ha if no control is applied. Biological control using Trichoderma spp. can control this pathogen and is able to promote sugarcane growth. The purpose of this study was to determine the effectiveness of Trichoderma spp. in controlling Bipolaris sp. in vitro. The results showed isolates TD1, TD2, TD3 were able to inhibit Bipolaris sp. antagonistically and volatilely, the antagonist test showed a result of 86% (TD1) while the volatile test of 65% (TD2). Conclusion Trichoderma spp. can inhibit the growth of Bipolaris sp. on an in vitro scale. Keywords: BCA, Bipolaris sp. Tebu banyak dibudidayakan didaerah hangat dan tropikal dan banyak digunakan sebagai pemanis dikarenakan mengandung banyak fruktosa dan glukosa. Produksi gula nasional menunjukan adanya penurunan yang mayoritas disebabkan oleh patogen terutama jamur Bipolaris sp. yang mampu menghasilkan kerusakan hingga 85% per Ha jika tidak diberlakukan pengendalian. Pengendalian biologis menggunakan Trichoderma spp. dapat mengendalikan patogen ini dan mampu mendorong pertumbuhan tebu. Tujuan penelitian ini ialah untuk mengetahui efektivitas Trichoderma spp. dalam mengendalikan Bipolaris sp. secara In vitro. Hasil menunjukkan isolat TD1, TD2, TD3 mampu menghambat Bipolaris sp. secara antagonis dan volatile, uji antagonis memperlihatkan TD1 memiliki hasil 86% sementara uji volatile TD2 memiliki hasil 65%. Kesimpulan Trichoderma spp. dapat menghambat pertumbuhan Bipolaris sp. dalam skala In vitro. Kata kunci: APH, Bipolaris sp. Trichoderma spp.

Abstract

The world population is proliferating, and the proportion of people living in urban areas is also increasing. It is challenging to achieve the SDGs, especially SDG 11, which aims to make cities and communities more inclusive, safe, resilient, and sustainable. Meeting the needs of urban residents, such as housing, public infrastructure, a clean environment, security, jobs, and other basic needs to live comfortably in a city with limited resources, is a problem. Sustainable cities are becoming increasingly important today to facilitate the growth of sustainable cities that integrate economic, sociocultural, environmental, ICT/transverse, and urban dimensions. Effective evaluation of urban sustainability is essential for sustainable development. Banten Province is one of the regions that has committed to achieving the SDGs. This commitment is contained in Banten Governor Regulation Number 47 of 2019 concerning the Regional Action Plan for the Sustainable Development Goals of Banten Province for 2020‒2022. Therefore, this study aims to assess and map the sustainability status of cities in Banten Province using Multi-Criteria Decision Analysis (MCDA) analysis and processed with PROMETHEE (Preference Ranking Organization METHods for Enrichment Evaluation). The number of criteria for a sustainable city in 2012 was 82 criteria and 109 criteria in 2022, where the highest number of criteria is on the sociocultural and economic dimensions. The PROMETHEE results show that the cities in Banten Province with the best sustainability levels are Tangerang City and South Tangerang City. City governments with low sustainability status or experiencing a decrease in sustainability status need to improve the performance of sustainable city indicators so that development can be sustainable. Abstract

This study aims to describe the status of soil fertility and corn yield on different slopes of agricultural land in UPT SP3 Saritani Village, Boalemo Regency. The study used survey methods at 3 land slope locations (20--30%, 30–40%, and 50–60%), determined purposively. Soil samples and maize yields were analyzed quantitatively and descriptively. Corn yields were collected and compared over 3 harvesting seasons based on the parameters of agronomic components. Soil fertility was analyzed at BSIP PALMA Manado laboratory, with parameters including macronutrient components. An essential finding of this study is that there is a difference in corn yield and nutrients on the sloping lands. The fertility status of farmers' land on the average sloped land of the three stations is very low levels of nitrogen and carbon nutrients and high levels of phosphate and potassium. The agronomic components of Stations 1 and 2 were relatively similar compared to the yield of components at Station 3. In the 3 harvest seasons, Station 1's yield decreased every season; from Station 2 it also decreased, but only at the time of the third harvest, although it was insignificant. Abstract

Greenbelt and green infrastructure are still very limited in Jakarta. Transjakarta Corridor 8 Harmoni - Lebak Bulus covers Central Jakarta, West Jakarta, and South Jakarta which are government centers and densely populated settlements. The high activity in the area causes an increase in transportation flows and greenhouse gas emissions. The transportation sector is the highest contributor to CO2e emissions in the capital city, thus reducing environmental quality, especially the greenbelt in Transjakarta Corridor 8. The assessment of ecological functions in this study consists of 7 variables, namely noise reduction, pollution reduction, air humidity, view control, glare control, direction and accident prevention. The research location is divided into 5 segments with the analysis method used, namely the assessment of ecological function aspects and the formulation of recommendations based on Hierarchical Process Analysis. The results of the assessment on ecological functions with noise-canceling variables were (Polyalthia longifolia) Glodongan Tiang, (Casuarina sp.) Spruce, (Acacia auriculiformis) Acacia, and (Archontophoenix alexandrae) Alexander Palm. The results of the pollution reduction assessment of each segment are dominated by vegetation types (Mangifera indica) Mango, (Polyalthia longifolia) Glodongan Tiang, (Carbera odollam) Bintaro. Based on the results of the questionnaire, the AHP assessment weight was obtained. The combined AHP results show the highest priority scale for the evaluation recommendations of this research is Pollution Reduction as the most important variable with a weight of 0.348, the second rank is Accident Reducer with a weight of 0.300, the third rank is Glare Control with a weight of 0.150. The combined results of 3 experts produce a consistency ratio value of 0.05, the consistency ratio value of 0.05 is smaller than 0.10 so it can be said to have met the consistent requirements. Abstract

Processed food SMEs in Gorontalo Province are one of the sectors contributing to regional economic development. The government implements policies and programs to develop competitive processed food SMEs. This study aims to analyze the formulation of policies for the development of processed food SMEs, with the hope of optimizing the production and added value of these SMEs through increasing production volume and market access. This study used a quantitative approach using primary data and secondary data in the form of time series. A dynamical system method with six scenarios is used and simulated until 2045. The results showed that the best scenario for achieving the research objectives was scenario six. Scenario six is carried out by simulating business capital assistance programs as well as increasing partnerships, business legality, and digitalization simultaneously. The production volume of processed food SMEs will reach 764,352 pcs in 2045, with a market access value of 41.67%. Policies need to focus on business financing and increasing the business capacity of processed food SMEs in Gorontalo province. The results of the policy simulation can be considered in realizing the achievement of accelerating the development of competitive SMEs, as stated in the Long-Term Development Plan of Gorontalo Province. Abstract

Seaweed plays an essential role in the people's economy in Pari Island because its cultivation has been the main livelihood for a long time. Currently, seaweed cultivation activities are no longer found, possibly due to tourism activities that directly degrade the cultivation activities. The purpose of this study is to evaluate the status of sustainability and determine the strategy of cultivation management in 5 aspects: ecological, social, economic, institutional, and infrastructure. Data were collected from primary and secondary sources related to socio-ecology in Pari Island. The assessment of the level of sustainability in each dimension was carried out using multidimensional scaling analysis, and a fairly sustainable average suitability value was obtained, namely the infrastructure (73.37), institutional (71.24), ecological (62.48), and economics (62.16). Meanwhile, the dimension with a very suitable sustainability value is the social dimension, with a value of 76.33, meaning that it is very suitable. Overall, the sustainability of seaweed cultivation, if viewed from several dimensions, shows the level of suitability of cultivation, both in terms of infrastructure, institutions, ecology, and economics. However, several dimensions, such as economic and ecological dimensions, must still be considered and continue to be encouraged so that sustainability can be achieved in all dimensions. Abstract

Fungi, including Rhizopus sp., are food sources that commonly contain high nucleic acid levels. Therefore, the nucleic acid content must be reduced to achieve health standard requirements. This study aimed to isolate, identify, and produce Rhizopus sp. mycelium containing low nucleic acid. The Rhizopus spp. were isolated from tempeh collected from 12 different areas in Indonesia. Fungal identification was conducted based on morphological characteristics. The fungal isolates were selected based on mycelial growth and spore production on PDA. Biomass production of mycelium was carried out in potato extract and soybean extract media obtained from 200 g/L and 333.3 g/L, respectively. In each medium, 6 sugar levels were added, namely 0, 2, 3, 4, 5, and 6 g/L. Mycelium nucleic acid content reduction was achieved by heat treatment at 50°C and 60°C for 15 minutes and measured by a spectrophotometer at 260 nm. Fifty-eight isolates that were identified into 3 species were obtained in this experiment: R. oryzae, R. stolonifera, and R. microsporus. R. Microsporus had higher mycelium biomass and lower spore number than the other species. R. Microsporus produced a higher mycelium biomass in the soybean extract medium with 5 g/L additional sugar. The nucleic acid content of the 50°C heat-treated mycelium was 1.82% and 1.73% at 65°C. These values fulfilled the standard of mycelial nucleic acid content permitted in food by the USDA. Abstract

The antifungal properties of clove basil (Ocimum gratissimum L.) extract on Fusarium oxysporum f. sp. Cepae and Alternaria porri were evaluated in vitro and in vivo. Thirty grams of the clove basil sterilized leaves were mashed in a sterile mortar, soaked for 24 hr in 100 mL of 96% ethanol as an extractant. The extract was filtered through sterile Whatman No. 1 filter paper and stored at 4°C for subsequent use. Three different concentrations (5, 10, and 15%) were applied using the poisoned food technique. A 4-mm diameter mycelial disc of each tested fungus was inoculated on each amended agar plate. Daily radial growth was recorded for 7 days. For the in vivo test, two different concentrations (15% and 20%) were used. No spray and mancozeb spraying were used as controls. Seed bulbs of shallots were planted in a 25 cm x 25 cm polybag and allowed to be naturally infected by Alternaria porri. The clove basil leaf extract and mancozeb were sprayed onto the plants once a week, starting when the first symptom of the purple blotch disease appeared. The results showed that clove basil leaf extract had good efficacy in inhibiting F. Oxysporum f. sp. Cepae and A. Porri in vitro, with a growth inhibition up to 55%. The extract can also reduce the disease intensity of purple blotch with an infection rate of 0.029, indicating that it has the potential to be developed as a botanical fungicide.
