Abstract

Deteksi dan Pembedaan Praktis Fitoplasma dari Beberapa Tanaman Inang Menggunakan PCR-RFLP Fitoplasma adalah prokariota fitopatogenik dengan kisaran inang yang luas dan merupakan patogen yang perlu mendapatkan perhatian lebih banyak di Indonesia. Patogen ini relatif sulit untuk dideteksi dan diidentifikasi mengingat sifat biologinya yang rumit. Penelitian ini meliputi deteksi fitoplasma menggunakan teknik polymerase chain reaction (PCR) dengan primer P1/P7 dari tujuh tanaman bergejala, yaitu daun putih rumput Bermuda, kuning bambu, sapu kacang tanah, kedelai, kacang panjang dan kaktus, serta daun kecil ubi jalar. DNA gen 16S rRNA fitoplasma hasil amplifikasi PCR selanjutnya direaksikan dengan masing-masing enzim endonuklease restriksi AluI, RSaI dan MSeI untuk menghasilkan pola restriction fragment length polymorphism (RFLP). Tujuh tanaman sakit tersebut terkonfirmasi berasosiasi dengan fitoplasma yang ditunjukkan oleh produk PCR sebesar 1800 pb. Berdasarkan profil RFLP tiga enzim, fitoplasma dibedakan ke dalam dua kelompok, yaitu kelompok I (rumput Bermuda dan bambu) dan kelompok II (kacang tanah, kedelai, kacang panjang, kaktus dan ubi jalar). Fitoplasma kaktus merupakan sub-kelompok (galur) karena memiliki sedikit perbedaan fragmen dari profil RFLP dengan enzim MSeI.

Abstract

Penyakit antraknosa dapat terjadi pada seluruh fase pertumbuhan tanaman bawang merah (Allium cepa var. aggregatum) dan belum diketahui proses perkembangannya serta karakteristik penyebabnya. Penelitian dilakukan untuk mempelajari etiologi penyakit antraknosa yang meliputi perkembangan gejala, periode inkubasi dan karakteristik patogen melalui pengujian secara in vivo dan in planta. Awal gejala ialah munculnya bercak putih yang kemudian membesar hingga menyebabkan jaringan mati. Gejala khas yang ditimbulkan ialah adanya pola konsentris berwarna jingga kecokelatan pada bercak dan memiliki periode inkubasi 2-4 hari. Karakteristik morfologi koloni patogen berwarna putih, konidium berbentuk silindris, hifa bersekat. Karakter lain ditandai dengan keberadaan aservulus dan setae. Patogen yang diidentifikasi secara molekuler menunjukkan 99.51% homologi dengan Colletotrichum gloeosporioides aksesi KT390192.1 dari China yang berasal dari tanaman tembakau.

Abstract

Application of Low Temperature and Antiviral for Elimination of Chrysanthemum stunt viroid and Its Effect on the Growth of Shoot Tip Culture of Chrysanthemum Chrysanthemum stunt viroid has been widely reported to be a limiting factor in the production of chrysanthemum cut flowers. The aim of this study was to determine the effect of low temperature treatment (5 ℃) and antiviral ribavirin on the growth and elimination of Chrysanthemum stunt viroid (CSVd) in chrysanthemum plantlets cultured from shoot tips. CSVd infected plants were obtained from a government experimental garden in Cianjur, West Java. The research was carried out in several stages, namely 1) plant sampling, 2) viroid detection using RT-PCR method, 3) initiation of explants and propagation of plant material in vitro, 4) low temperature treatment 5 ℃ with three levels of incubation time at 1, 3, and 5 months, 5) ribavirin antiviral treatment with three levels of concentration (25, 100, and 125 ppm), and 6) confirmation of viroid-free plant materials by RT-PCR. The results showed that low temperature storage and ribavirin treatment reduced the rate of growth of shoot height and number of leaves. Ribavirin did not cause phytotoxity up to concentration level of 125 ppm, although it caused inhibition on shoot growth and leaf number. However, the combination of low temperature treatment (1, 3, and 5 months) with ribavirin (25, 100, and 125 ppm) was not able to eliminate CSVd from tissues at all treatment levels.

Abstract

Tanaman mentimun varietas lokal Padang merupakan tanaman sayuran unggulan bagi petani perkotaan karena umur panen yang pendek. Hasil survei menemukan adanya gejala bercak kuning dan klorosis dengan tulang daun hijau. Gejala ini mirip dengan infeksi Papaya ring spot virus (PRSV) yang menginfeksi mentimun di Jawa dan Zucchini yellow mosaic virus (ZYMV) yang menginfeksi mentimun pada umumnya. Deteksi virus dilakukan dengan teknik reverse transcription polymerase chain reaction (RT-PCR) menggunakan primer spesifik PRSV dan primer universal Potyvirus, perunutan dan analisis DNA. RT-PCR menggunakan primer spesifik PRSV DNA target tidak menghasilkan pita DNA, namun pita DNA dengan ukuran ±320 pb berhasil teramplifikasi dengan primer universal Potyvirus. Analisis sikuen nukleotida menunjukkan bahwa similaritas virus tertinggi sebesar 93.8% dan 93.5%, berturut-turut dengan ZYMV isolat Singapura dan India. Infeksi ZYMV merupakan kejadian pertama pada tanaman mentimun di Padang, Sumatera Barat.

Abstract

Composition of Functional Bacteria on Suppressive and Conducive Soil for Fusarium Wilt on Chilli The phenomenon of suppressive soil has been widely studied to control plant diseases in the field. Suppresive soil is characterized by its contain of functional microbes that can suppress pathogen populations. This study aims to determine and compare the composition of functional bacteria on suppressive and conducive soil of fusarium wilt. The research method consists of soil sampling in the field, isolation of functional bacteria from suppressive soil (SS) and conducive soil (CS) samples, calculation of bacterial population and type, biosafety screening, and characterization of functional bacteria in suppressing of Fusarium oxysporum. The results showed that the bacterial population in SS was higher than those in CS, especially for heat-tolerant bacteria and the fluorescent Pseudomonads. The high population of bacteria is supported by the high value of functional bacterial diversity in SS (H’ 3.70 > 3) compared to CS, which is classified as medium (1 < H’ 2.07 < 3), with the distribution of functional bacteria classified as almost evenly distributed and no dominance of certain types in SS and CS. The percentage of nonpathogenic bacteria in SS is higher than in CS, i.e. 51% and 23%, respectively. These bacteria have potential as plant growth promoting bacteria (PGPB) by producing IAA or dissolving phosphate alone of 24% and 10%, respectively in suppressive soils; and of 14% and 29%, respectively in conducive soils. Only bacteria derived from suppressive soils were able to produce IAA while dissolving phosphate with a percentage of as much as 48%.
