Abstract

Calcitriol supplementation in ovariectomized rats decreased bone resorption and increased bone formation, however, it depend on dose. The objective of the research was to study the response of femur bone in osteoporosis rats consuming calcitriol. Thirty female Wistar rats at 8 weeks of age were randomly divided into six groups (sham operated rats as normal control rats/group N and NK, ovariectomized control rats/group Ov and OvK, ovariectomized rats/group OvDand OvE) of five each. All rats were fed standard diet for 8 weeks. At 16 weeks  of age, group N and Ov  were euthanized, the right femur were taken for histopathological  examination. Group NK and OvK were fed a standard diet, group OvD was fed a standard diet +40ng calcitriol;  and group OvE was fed a standard diet+25µg ethynil ethyl estradiol. Treatments were done for six weeks. At the end of study, blood samples were taken from plexus orbitalis medialis for estrogen analysis. All rats were euthanized using ketamine10% and xylazine 2%. Right femur was taken for histopathological examination  using hematoxylin and eosin stain, and immunohistochemistry using monoclonal antibody anti TRAP5b which was detected with streptavidin-biotin.         The results showed that estradiol level of the rats in group OvD was not significantly different compared with the rats in OvK group, however, it was significanly lower compared to the rats in group OvE. Histopathologic figure of right distal femur metaphysis in group OvD was shown lesser adipocyte in the bone marrow and more trabecular bone speculum compared to group  OvK, however, there was more adipocyte in the bone marrow and lesser trabecular bone speculum compared to group OvE. Immunohistochemistry of distal femur metaphysis in group OvD and OvE were revealed tartrate resistant alkaline phosphatase 5b (TRAP5b) expression in trabecular bone, which was located in bone marrow space and trabecular speculum surface as well. Based on the results, it can be concluded that calcitriol 40ng/day supplementation in osteoporosis rats for 6 weeks decreased bone resorption and increased bone formation distal femur metaphysis.

Abstract

Antimicrobial resistance is amongst the primary concern in the field of veterinary medicine worldwide. The research was conducted to evaluate the resistance of Escherichia coli due to inappropriate antibiotic treatment. Escherichia coli in this research was cultured in vitro using 5-ml Tryptone Soya Broth (TSB), which was mixed with an antibiotic, then was incubated at 37 0C for four days (stage I). Subsequently, a part of the culture was transferred  into another 5-ml TSB and incubated at 37 0C for four days (stage II). This procedure was undertaken continuously for stage III, IV, V,and VI (until day 24). Antibiotics in this research was oxytetracycline with three various doses, which are lower dose (1 mg/ml), normal dose (2 mg/ml), and higher dose (8 mg/ml). The development of the antrimocobial resistance was evaluated every four days, using disk diffusion method and the data were analysed descriptively. The results showed that the normal and higher doses of oxytetracycline has the same rate (day 16) in causing E. coli resistant to oxytetracycline. Therefore, the treatment of oxytetracycline with a normal and higher doses continuously could accelerate the emergence of antimicrobial resistance in Escherichia coli.

Abstract

This research has purposes to identify parasites as well as to reward determining the prevalence of parasites on water monitor lizard (Varanus salvator) was captured in Banda Aceh. As many as 50 water monitor lizard were used this research and examined at the Laboratory of Parasitology, Faculty of Veterinary Medicine University of Syiah Kuala. Examination of ectoparasites on the surface of the body was done by direct examination. Endoparasit examination carried out by observation in blood using the blood smear and centrifugation techniques microhematocrit (mHCT), while fecal examination using flotation method, Borray sedimentation and digestive tract surgery digestive tract surgery if obtained at least 3 types of helminth eggs (1 cestodes, 1 nematodes, and 1 trematodes). Results of observation on blood is no parasites were found. Observations on the skin was obtained ectoparasites Amblyomma sp. with a prevalence rate of 100% category always. Observations of feces obtained Ascaris sp. with a prevalence rate of 74% category usualy and Stronyloides spp. with a prevalence rate of 8% category sometimes. Based on these results it can be concluded that the parasites that can infest the water monitor lizard (Varanus salvator) is Amblyomma sp, Ascaris sp, and Stronyloides spp. with variant  prevalence rates.

Abstract

Aeromonas hydrophila causes a disease that often infects fish and is known as Motile Aeromonas Septicaemia (MAS), Hemorrhagi Septisemia, Ulcer disease or Red-Sore disease. The   aims of this study were to develop polyclonal antibody of  Aeromonas hydrophila in the rabbits to   confirm the diagnosis of Aeromonas hydrophila  in the fish by immunohistochemistry staining method. Preparation of polyclonal antibodies was performed on the rabbits used to Aeromonas hydrophila bacteria that have been tested biochemically by intravenous and intraperitoneal injection. Doses of Aeromonas hydrophila  bacteria were 109 CPU/ml  of 0.5 ml at first injection, 1 ml at second injection, 2 ml at thirth injection and 3 ml at fourth injection. Blood serum collection was performed at week 5 after injection from  an  ear and intracardial vein. The result of antibody titer was 28 = 1024 which measured by   tube test. Furthermore, polyclonal   antibody was used to immunohistochemistry  staining with 400x dilution. The results of the staining showed that an immunopositive reaction in the liver, skin,lien,  gill, kidney, and heart of fish to Aeromonas hydrophila antibody. The research conclution was polyclonal antibody from rabbit can be used to accurately confirm the diagnosis of Aeromonas hydrophila  based on antigen and antibody reaction.

Abstract

Leptospirosis is a zoonotic disease of global concern, and is caused by pathogenic serovar Leptospira interrogans. Canine Leptospirososis is widespread worldwide, dogs can act as incidental hosts or maintenance hosts for various serovars. The purpose of this research was to identify leptospire serovars that infect healthy and suspected leptospirosis dogs in Yogyakarta. A total of 56 dogs (36 healthy dogs and 20 suspect leptospirosis dogs) sera were taken from cephalica vein as much as 3 ml. Sera were examined for leptospirosis with Microscopic Aglutination Test (MAT) which conducted at the Research Center for Veterinary Science, Bogor. Microscopic Aglutination Test carried out on various Leptospire serovar, namely: Ichterohaemorrhagiae, Javanica, Celledoni, Ballum, Pyogenes, Cynopeteri, Rachmati, Australis, Pomona, Canicola, Grippotyphosa, Bataviae, Hardjo, and Tarrasovi. The results showed that Celledoni serovars infected 25% of healthy dogs and 5% of suspect leptospirosis dogs, Javanica serovar infected 19% of healthy dogs, Bataviae serovars infected 15% of suspect leptospirosis dogs, Grippotyphosa serovar infected 11% of healthy dogs, Tarrasovi serovar infected 10% of suspect leptospirosis dogs, serovars Cynopteri infects 5% of healthy dogs and 5% of suspect leptospirosis dogs, serovar Pyrogenes infects 5% of healthy dogs and 5% of suspect leptospirosis dogs, and serovar Rachmati infects 5% of suspect leptospirosis dogs. Seven healthy dogs (19%) and 2 suspect leptospirosis dogs (10%) were infected with more than 2 leptospire serovars. From the results of this study it can be concluded that Celledoni serovar of Leptospira interrogans infection causes subclinical leptospirosis, while Bataviae serovar infection causes clinical leptospirosis in dogs in Yogyakarta.
