Abstract

The aim of this study was to evaluate the effect of different IgY concentrations on the production, reproduction and immunity of Sentul chicken. This research used 20 month-old Sentul chickens which consisting of 10 chickens with high IgY concentration and 10 chickens with low IgY concentration Parameters observed included prelayer performance (feed consumption, weight gain, feed conversion, morbidity and mortality), layer performance (feed consumption, egg production, egg weight, feed conversion, hatchability, morbidity and mortality), and immunity at chicks (IgY concentration of eggs and chicks serum). The experiment was conducted as a completely randomized design. The treatments were high and low IgY concentration. All treatments were repeated 10 times. The data obtained were analyzed using the SAS program t-test. Morbidity and mortality were analyzed descriptively. The results of this research indicated that the prelayer period, chickens with high IgY concentration had a higher performance than chicken with low IgY concentration. This is caused by about 30% chickens with low IgY were in sick condition for approximately seven days. Further, the layer period showed that the chickens with high IgY concentration had lower production performance than the chicken with low IgY concentration. The study concluded that chickens at low IgY concentrations are susceptible to exposure to antigens that lead to decreased productivity. In normal conditions hens, Sentul with high IgY concentration resulted in low production. The observation of chicken immunity showed that concentration of IgY from chicks and eggs did not differ between hens with high IgY concentrations and hens with low IgY concentrations.

Abstract

The aim of this study was to determine genetic diversity of Calpastatin gene (CAST) in Pasundan cattle. Forty-four fresh blood samples were collected from UPTD BPPT Beef Cattle Ciamis West Java. Genomic DNA samples were extracted using High Salt method. A 523 bp fragment of Calpastatin gene was successfully amplified using PCR method. Genotyping of CAST gene was conducted by PCR-RFLP method using RsaI restriction enzyme (GT^AC). Genotypes and alleles were analyzed using software Cervus 3.0.7. Parameters were observed i.e genotypes and alleles frequencies, heterozygosity observed (Ho) and expected (He), Hardy Weinberg Equilibrium (HWE), and Polymorphic Information Content (PIC). Result showed that three variant genotypes of GG, GC and CC were found and two variant alleles of G and C. Allele G was found higher (0.77) than allele C (0.23). Population of Pasundan cattle was found polymorphism and in the Hardy Weinberg Equilibrium. Polymorphic Information Content (PIC) value showed in a moderate (0.290) condition. Values of Heterozygosity observed and expected were 0.409 and 0.355 respectively. This research concludes that there is polymorphism of CAST gene in Pasundan cattle population and has genetic diversity. This result could be used as early genetic information in exploration of Pasundan cattle.

Abstract

The current study was aimed to evaluate the ability of phenol content in cashew nutshell liquid (CNSL) in binding protein and its effect on in vitro nutrient digestibility. This research divided into three steps, there were determination of total phenol and tannins content in CNSL, optimalization binding of CNSL tannin to bovine serum albumin with the determination of protein-percipitable phenolics compound and protein content using Lowry method, after that amount of optimalization supplementation of CNSL was used for measuring on in vitro nutrient digestibility by Tilley and Terry method. The results showed that phenol and tannin content of CNSL were 148.69 g/100 ml and 28.3 g/100 ml. One gram tannin in CNSL could be bound by 65.83 g protein. Optimum tannin were used in this study was 5.85 g tannin or equivalent to 163.91 mg of CNSL. Amount of CNSL were used in various levels, there are 0, 50, 100, 150 dan 200 mg. CNSL supplementation reduced dry matter, organic matter crude protein digestibility in rumen, and didn’t reduce dry matter dan crude protein in vitro post rumen digestibility. Supplementation of CNSL increased feed efficiency on in vitro.

Abstract

This study aimed to analyzing the Strategies for fulfillment animal nutritionist competency at feedloter in Indonesia. Methods in this study using Analytical Hierarchy Process (AHP). The data collection technique was conducted by judgement sampling to seven competent resource persons in the field of animal feed they are feed director in Ministry of Animal Health and Husbandry Republic of Indonesia, Chairman AINI and Director of the competency development and certification AINI period 2015-2019, Academics of lecturer in ransum formulation, operational manager and animal nutritionist in industry feedlot. The questions posed to the resource persons include factors, actors, goals and alternatives that most decisive in the fulfillment animal nutritionist competency at feedloter in Indonesia. The results of the AHP indicate that individual competence factors are critical in the strategy of the needs of livestock animal nutritionist. The Indonesian Association of Nutrition and Feed Technology (AINI) is actor the most influential in fulfilling the needs of animal nutritionist. AINI can conduct competency-based training, skills and knowledge development, latest technological innovations in ration making, and certification of competence of animal nutritionist to enhance individual competence. The alternative that a feedloter should do is to adapt the competency standards policy that exists in our country. It is concluded that the Strategies for fulfillment animal nutritionist competency at feedloter in Indonesiacan be fulfilled through the individuals as animal nutritionist, role of AINI increasing the competence of animal nutritionist, and the application of customized Indonesian welfare in Indonesia.

Abstract

This experiment aimed to determine the nutrient digestibility of Bligon and Kejobong goats fed by king grass and peanut straw. The sample used was 6 to 8 months of five male Bligon goats, and five male Kejobong goats with the initial body weight of 15 to 20 kg and placed in separated metabolism cages. The diet and water were supplied ad libitum. This experiment was done in 14 days of adaptation period and seven days of the collection period. In the collection period, samples of feed, refusal feed, and feces were collected to get the chemical content includes dry matter (DM), organic matter (OM), ether extract (EE), crude fiber (CF), crude protein (CP), nitrogen-free extract (NFE), and total digestible nutrient (TDN). The data obtained were subjected to a T-test analysis. Results showed that nutrients intake, digested nutrient as well as nutrient digestibility of Bligon and Kejobong goats were not significantly different. However, Kejobong goats had EE and CP intake of 15.42% and 14.02%, digested DM, OM, and NFE of 16.29%, 15.71%, and 13.93% respectively, as well as NFE digestibility 4.37% higher than those Bligon goats (P< 0.05). Therefore, there was no difference in nutrient intake, digested, and digestibility of nutrients in Bligon and Kejobong goats fed by king grass and peanut straw. However, Kejobong goats appeared to have higher EE and CP intake, as well as digested and digestibility of DM, OM, and NFE than Bligon goats.

Abstract

The aim of this research was to understand the effect of Azolla microphylla flour supplementation in the basal feed on carcass production and meat tenderness characteristics of culled quail. The materials used in this research were 100 culled quails at 48-week-old age and commercial quail feed. The research was conducted as an experimental research and used Completely Randomized Design. Four treatments were done in this research, which was basal feed without Azolla microphylla flour supplementation; basal feed supplemented with 1% Azolla microphylla flour; basal feed supplemented with 2% Azolla microphylla flour, and basal feed supplemented with 3% Azolla microphylla flour. Each treatment was done 4 times with each fed into 5 quails. The observed variables include carcass production (carcass percentage, breast meat percentage, thigh meat percentage, back meat percentage, and wing meat percentage) and meat tenderness characteristics (meat fat content, collagen content, pH and meat tenderness). The obtained data were then analyzed by analysis of variance and if the result showed a significant effect, further analysis will be done by honestly significant difference test. The analysis of variance showed that Azolla microphylla flour supplementation showed significant effect (P<0.05) on the carcass percentage, breast meat percentage, meat fat content, collagen content, and meat tenderness but showed no significant effect (P>0.05) on the thigh meat percentage, back meat percentage, wings meat percentage and meat pH. The research concluded that Azolla microphylla flour supplementation in the basal feed will increase carcass production and meat tenderness of culled quail meat at maximum 2% of supplementation level.

Abstract

The purpose of this research was to analysis the value of repeatability and correlation among the traits affecting the production of frozen semen from Holstein’s bull in Indonesia. Repeatability and correlation were calculated based on the data of frozen semen production of 15.699 records from 44 Holstein bulls at Singosari Artificial Insemination Center (SAIC) and 8.935 records from 39 Holstein bulls at Lembang Artificial Insemination Center (LAIC). Repeatability for volume, motility, fresh semen concentration and frozen semen production was evaluated by intraclass correlation method. The repeatability values of LAIC for volume, motility, fresh semen concentration and frozen semen production were 0.60; 0.54; 0.37 and 0.47. The repeatability values of SAIC for volume, motility, fresh semen concentration and frozen semen production were 0.54; 0.30; 0.43 and 0.29. The linear correlation value between volume, motility and fresh semen concentration with the amount of semen produced per collections were 0.41, 0.36, and 0.58. Concentration was the most factors influencing the number of frozen semen produced. The effectiveness of the selection of Holstein's frozen semen producing could be determined by the value of repeatability and the phenotypic correlation among semen quality traits such as volume, motility, concentration and frozen semen production.

Abstract

Contamination that decreased chicken meat quality could be prevented using natural preservatives. Bay leaves (Syzygium polyanthum) contain volatile fatty acids, tannin, and flavonoid that possess bacteriological and fungicidal activity as well as preventing bacterial spore growth. The purpose of this study was to determine the effect of fresh bay leaf infusion on microbiological, chemical, and physical qualities of chicken meat. This study used bay leaves, water, chicken meat, eight strain of bacteria, chemicals and materials for the analysis of chicken meat. The experiment consisted of two steps, the first was the antibacterial properties of bay leaves and the second was the application of bay leaf infusion for chicken meat. Eight bacteria was used for the bacterial inhibition of bay leaf at the concentration of 0, 5, 10, 15 and 20%. The experiment on antibacterial properties of bay leaf (Syzygium polyanthum) used one way randomized design with five concentration treatments, while the application of bay leaf infusions on chicken meat using factorial completely randomized design 2x5 (2 types of soaking and 5 observation time). At the second step, chicken meat was divided into 2 groups, the first group was soaked in water only and the second group was soaked in 15% bay leaf infusion. They were then stored for 0 (control), 2, 4, 6, and 8 hours at the room temperature. Each treatment was repeated four times. The microbiological, chemical, physical qualities of chicken meat were observed. The results showed that bay leaf infusion had the ability to inhibit the growth of bacteria (P<0.05). The highest growth inhibition was found on Leuconostoc mesenteroides, whereas the lowest growth inhibition was on Pseudomonas putida. Bay leaf infusion influenced the number of bacteria in chicken meat. The concentration of 15% bay leaf infusion could decrease bacterial number amounting to 1 log CFU/mL compared to control. The number of bacteria of chicken meat stored significantly increased during the storage (P<0.05). However, the number of bacteria of chicken meat soaked in bay leaf infusion was significantly lower than the control. Bay leaf infusion has no significant effect on the chemical quality of chicken meat but it did on the physical quality of chicken meat. The conclusion of the study was bay leaf infusion could inhibit the bacterial growth and reduced the amount of bacteria in chicken meat, whereas storage time influenced the microbiological and physical qualities of chicken meat.

Abstract

The objective of this study was to determine the activity and the stability of bacteriocin from lactic acid bacteria (BAL) isolated from rumen fluid of thin-tail sheep under the temperature (80, 100, and 121°C), pH (3, 7, and 10), and the length of storage (for 2 weeks under the temperature -8, 11, and 29°C). Lactic acid bacteria obtained by isolation, selection, and identification of thin-tailed sheep rumen fluid were used for bacteriocin production. The crude bacteriocin was partially purified using 70% ammonium sulfate, then was dialysis for 12 hours. The obtained bacteriocin then tested its inhibitory activity against E.coli (representing Gram-negative) and S. aureus (representing Gram-positive) under temperature (80, 100, and 121°C), pH (3, 7, and 10), and the length of storage (for 2 weeks under the temperature -8, 11, and 29°C). The data of bacteriocin activity based on pH, temperature, and the length of storage were analyzed with factorial, then when there was a significant difference of variable because treatment was continued with Duncan's Multiple Range Test (DMRT) test. The results showed that the bacteriocin activity of the three types of BAL against S.aureus is greater than E.coli. The highest activity was shown in pH 3, while the lowest activity was shown at pH 10 (P<0.01). The highest activity was shown at a heating temperature of 100°C, while the lowest activity was shown at a heating temperature of 80°C (P<0.01). The activity of bacteriocin produced by BAL 0 A, BAL 1 A, and BAL 4 C tended to be stable to the heating temperature of 80, 100, and 121°C but decreased with increasing pH value (pH 3, 7, and 10). The best of bacteriocin activity was found at pH 3 (acid), heating at 100°C, and stored at -8°C for 14 days.

Abstract

The aim of this study was to evaluate the effect of different IgY concentrations on the quality of semen (macroscopic and microscopic) and testicular morphometry (weight, length and width of the testes) of Sentul cock testis and its ability to fertilize hens. This study used 4 months of 20 Sentul cocks consisting 10 high IgY concentration cocks and low IgY concentration cocks. This study used a complete randomized design (RAL) with 2 treatments namely low and high IgY concentration and10 times repetitions. Data were analyzed using t-test minitab program versions 16. Based on the result, different IgY concentrations did not affect the quality of semen and testicular morphometrics, but high IgY concentration cocks were able to fertilize more hens than the low IgY concentrations one by using dilution methods. This proves that high IgY concentration cocks are more effective and efficient cocks or stud.
