Abstract

Insertion and deletion are information that reflect evolutional potency of species or population, that nowadays, are central topics in conservation genetics. This research aims to reveal the proportion of insertion and deletion on RDP1of species Megapodes in Indonesia. Whole blood DNA was extracted according to the protocol of Qiamp DNA Blood Mini Kit (Qiagen), and used as template for PCR (Polymerase Chain Reaction) which was conducted according the protocol of Takara Ex TaqTM PCR Kit in a Perkin-Elmer Thermal Cycler 9600. PCR products were purified using a Qiaquick PCR Purification Kit (Qiagen, USA). The sequencing reaction for RDP1 was performed using BigDye v.3.1. Cycle Sequencing Kit (Applied Biosystems,USA) followed the protocol of ABI Prism 3100-Avant Genetic Analyzer (Applied Biosystems, USA). Alignment of RDP1 nucleotide sequences of megapodes and that are obtained from international DNA data base (DDBJ-DNA Data Bank of Japan) with accession number AF 394649 (RDP1) were conducted by using Clustal-X. Characteristics of RDP1 were analyzed by employing the DNA Sequence Polymorphism (DnaSP) version 3.51 and Program MEGA version 4. The analysis result demonstrated there were insertion and deletion on RDP1 of species of Megapodes in Indonesia, which resulting in a gap of 1-10 bp and was concentrated in nucleotide 407-568. Alignments between species of Megapodes  and two species of Galliformes showed more gaps, which were concentrated on nucleotides 480-708.

Abstract

The study used quatitative approach with comparative study. The aims were to determine the student achievement and learning response as well as the different of student achievemnt  tought by inquiry method with concept maps. The population were students of Class X SMA Negeri 2 Sigli consisting of 227 pupils and  the samples were 35 students (experimental-group class) and 35 students (control-group class). Data of teaching learning type was collected throught observation. interview, and field data whereas student achievement collected by design and presentation of concept map, student worksheets, and final test. Data of student response in using the map was collected by questionnaire. The result showed that the value of t-count t-table for student achievement which was 3,45 1,67 indicating  the method has affected the student achievement. The mean values of student responses were 3,73 (agree category) for positive qustions and 2,27 (uncentain category) for negative questions. It can be concluded that learning learning process on concept of environment throught inquiry method with concept map has increased student achievement and response of students of SMA Negeri 2 Sigli.

Abstract

Bacterial leaf blight (BLB) disease caused by plant pathogenic bacteria Xanthomonas oryzae pv. oryzae (Xoo) could lead to death of rice plants. Control of plant pathogenic bacteria can be performed using biological control agents. The aim of this research was to study the inhibitory ability of eight isolates of biocontrol bacteria against Xanthomonas oryzae pv. oryzae (Xoo) under greenhouse conditions. Isolates of biological agents used in the study were obtained from IPB Culture Collection (IPBCC), Department of Biology, IPB and microbial gene bank collection of Indonesian Center for Agriculture Biotechnology and Genetic Resources (BB Biogen), consisting of Pseudomonas aeruginosa C32a and C32b, P. fluorescens ATCC 13525 Pf , Serratia marcescens E31, Bacillus sp. I.5, B. cereus I.21 and II.14, and B. firmus E65 isolates. The methods used in the research i.e. hypersensitivity test, antagonistic test of biocontrol bacteria to Xoo, and in vivo application of biological agents in the greenhouse condition. Hypersensitivity test on tobacco plants using C32a and C32b inoculums showed characteristics of slightly leaf yellowing but did not cause necrosis. Injection using Xoo inoculum showed necrosis on tobacco leaves. Antagonist isolates i.e. C32a, C32b, Pf, I.21, and I.5 showed inhibitory activity against Xoo, whereas others isolates did not show inhibitory activity. In greenhouse experiments IR 64 rice plants were sprayed with biological control agents (107cfu/ml) at 7 days, 14 days, 28 days, and 42 days after planting. The results showed that C32a isolate could suppress better the lesion length of BLB than that of chemical control (copper sulphate).

Abstract

Paradigm of education requires a better change in students’ behavior. To challenge these problems, the improvement way of learning is needed. Inquiry method is associated to improve student learning outcomes and activities. The purpose of this study was to determine the differences of student learning outcomes and activities that is learned between the inquiry method and conventional learning. The population of this study was all students from five classes in grade VII MTsN Bakti Pidie City academic year of 2010/2011. The samples were randomly selected (random sampling) with VII2 grade students as the experimental group and VII4 grade students as a control group. The data was collected through paper and pencil test: pretest and posttest and non-test through observation sheet. The data is analyzed using t-test, moreover student activities were analyzed by percentage formula. The analysis of data shows t count= 3.80 and ttable = 2.64, it is indicated that the student learning outcomes by methods of inquiry is significantly different from student learning outcomes with conventional methods. Students teaching by Inquiry learning are more active than students teaching by conventional learning.

Abstract

The simple and efficient method for genomic DNA isolation protochol from clove, its woody crops containing high polysaccharide and phenol levels has been described here. In the present study, using modified CTAB for plant DNA isolation method were studied for removing the highly concentrated polysaccharides from genomic DNA of woody crops.This method involves the modified CTAB   at the pvp concentration, time incubate  and precipitate procedure employing DNA purification  step to remove polysaccharides and phenol residu.  Compared with the two studied DNA isolation methode of clove using standart CTAB and modified CTAB the everage yield high quality DNA whole genom is 1.90 purity and DNA was suitable for PCR and RAPD analyses.

Abstract

This research was done in wild life conservation area of Sibela mountain, South Halmahera Regency, North Maluku Province from November 2009 to February 2010. The purpose of the research is to give evidence about how is the people’s behavior towards the wild life conservation area of Sibelia Mountain, the. The population of the research was all of patriarchs in three villages: Sawadai village, Tuokona and Gandasuli village. Furthermore, 46-47 people are chosen randomly from each village to get 140 patriarchs of total sample. Questionnaire instrument was used to collect the data. Questionnaire instrument and closed response interview guideline were used to collect the data of people’s behavior toward the wild life area conservation. Prior to distribute the instrument to the respondents, a test is carried out to the people who live in east Halmahera wild life area excluding from the research sample to get its validity and reliability. The result of analysis on people’s behavior in three different villages living in wild life area shows that they still have a low level of land utilization (about 20% - 25%). This result indicates that respondents who answered the questions of utilize of the conservation area tend to be in small number. This result also shows that the people of the three villages are having a tendency to do activities that caused environmental damage (contra conservation). The analysis result on people’s behavior in maintaining wild life conservation area of Sibela Mountain is also at a low level. From the three villages, the percentages are 8% - 13%. It indicates that people’s behavior tendency of maintaining or preserve the conserved area is at a very low level.

Abstract

Telah dilakukan penelitian tentang  penerapan model concept attainment terhadap hasil belajar   siswa pada materi metabolisme. Tujuan penelitian ini  adalah untuk mengetahui  pengaruh penerapan model concept attainment terhadap peningkatan pemahaman konsep, keterampilan proses sains, dan kemampuan berpikir kritis siswa pada materi metabolisme. Penelitian ini dilakukan sejak bulan September sampai dengan November 2013. Metode yang digunakan adalah metode ekperimen dengan desain  pretest postest control group design. Instrumen yang  digunakan adalah Rencana Program pembelajaran (RPP), soal pemahaman konsep, soal keterampilan proses`sains, soal kemampuan berpikir kritis, pedoman observasi, dan angket untuk mengetahui tanggapan siswa terhadap model pembelajaran yang diterapkan. Pengumpulan data dilakukan melalui pretest dan posttest. Data peningkatan hasil belajar berupa gain ternormalisasi (N-Gain). Perbedaan rata-rata N-gain kelas eksperimen dan kelas kontrol dianalisis dengan independent sample t-test. Hasil penilitian ini  menunjukkan bahwa penerapan model concept attainment dapat meningkatkan pemahaman konsep, keterampilan proses sains, dan keterampilan berpikir kritis siswa pada materi metabolisme

Abstract

Green mussel is a mollusk class that has been developing to culture system.  The perna viridis culture using bagan with rope as settling media.  Settlement of perna on rope that strongly will support the survival rate of mussel.  This research done to assessing bysus on rope media in water column.   Sampel take from culturing area in Muara Kamal station.  The Measurement of bysus strength use Shimadzu Autograp AGS-D Series.  Data collected analyzed are descriptive and  principal component  analyses.  Water quality that measure are temperature (30,1 oC), current (0,115 m/dt), Salinity (28,89 promil).  Bysus elongasi strength between 4,598-7,892 with bysus line reach 20-60 piece.  Break point  yarn much happen at plaque site from bysus.

Abstract

Kajian penerapan pembelajaran berbasis masalah terhadap peningkatan keterampilan metakognitif siswa pada konsep sistem reproduksi manusia telah dilakukan di SMA Negeri 2 Sigli dengan menggunakan metode kuasi eksperimen dengan desain penelitian the one group pre-test and pos-test. Populasi pada penelitian ini adalah seluruh siswa kelas XI semester ganjil pada Tahun Ajaran 2012/2013 yang berjumlah 120 orang. Sampel diambil siswa kelas XI-B yang berjumlah 30 orang. Analisis data dilakukan dengan uji t menggunakan program SPSS 16.0 for windows. Hasil penelitian menunjukkan bahwa: terdapat perbedaan skor keterampilan metakognitif siswa yang signifikan (P0,05) dengan nilai thiting= 16,76 ttabel =2,045  antara sebelum dan sesudah pembelajaran berbasis masalah.  Dapat disimpulkan bahwa penerapan pembelajaran berbasis masalah dapat meningkatkan keterampilan metakognitif siswa pada konsep sistem reproduksi manusia.

Abstract

The research about food preference of Sumatran elephant have conducted in Jantho Natural Reserve Aceh Besar District inAcehProvincein April to June 2006. The aim of this research was to identified elephant food species vegetation in elephant habitat. This research used direct observation method in 20 transect. We found 22 species eaten Sumatran elephant in Jantho Natural Reserve Aceh Besar District. The food preference consist of Moraceae, 11 species (18,74%), Poaceae, 15 species (15,64%), Arecaceae, 9 species (14,52%), Euphorbiaceae, 6 spesies (9,95%), Mimosaceae, 5 spesies (6,25%), Caesalpiniaceae, 2 spesies (3,30%), Cyperaceae,4 spesies, (3,27%), Bombacaceae, 2 species (3.23 %), Gleicheniaceae, 4 species (3.22%), Zingiberaceae 4 species (2.77%),Fabaceae, 2 species (2.58%), Musaceae, 1 species (2,10%), Apocynaceae, 2 species (1.94 %), Ebonaceae, 1 species (1,70%), Sapindaceae, 1 species (1.41%).
