Abstract

DNA isolation is a fundamental step in various molecular biology applications, with its success highly dependent on the purity and integrity of the isolated DNA. This review article aims to highlight the vital role of alcohol, particularly ethanol and isopropanol, in the DNA isolation process from various biological sources. This comprehensive review explores the biophysical principles underlying DNA precipitation by alcohol, where the reduction of the solution's dielectric constant and disruption of the hydration layer lead to DNA precipitation. The article also evaluates the application of alcohol in different DNA isolation protocols, including the classic phenol-chloroform extraction method, commercial silica column-based kits, and salting-out techniques. Recent advancements in optimizing alcohol precipitation parameters indicate that process efficiency can be enhanced by adjusting the type of alcohol, incubation time, and salt concentration. Current research trends also point toward the development of environmentally friendly approaches that minimize alcohol usage, as well as alternative alcohol-free methods such as magnetic-based technology and paper-based DNA isolation systems.The future prospects of DNA isolation are projected to integrate traditional alcohol-based methods with advanced technologies such as automation, nanomaterials, and microfluidic systems. A deeper understanding of alcohol’s role in DNA isolation is expected to optimize existing protocols and drive innovation in DNA isolation techniques for applications ranging from diagnostics to genomic research.

Abstract

Protein-energy malnutrition (PEM) is a nutritional disorder that arises from low energy and protein consumption in the daily diet, and is influenced by certain health conditions. According to the 2023 Indonesian Health Survey (SKI) data, 29.7% of children in East Nusa Tenggara Province are malnourished. This study aims to evaluate the impact of adding red bean flour on the acceptability level and macro-nutrient content of cakalang fish basreng products. The study used a Completely Randomized Design (CRD) with red bean flour additions of 10%, 20%, and 30% in sequence, involving 30 panelists. The organoleptic test results showed that the average liking level (color, aroma, texture, and taste) fell into the “somewhat liked” to “liked” category. The normality test results for color, aroma, texture, and taste indicated that all the data presented were not normally distributed. Statistical test results using the Kruskal-Wallis test showed that there was no significant effect on color, aroma, texture, and taste in each treatment. Nutritional value calculations showed that the addition of red bean flour significantly contributed to an increase in the macro-nutrient content of skipjack tuna basreng. It is hoped that future researchers will use a drying oven in the red bean drying process to minimize cross-contamination from the surrounding environment.

Abstract

The period between childhood and adulthood, known as adolescence, is marked by a number of social, mental, and physical changes. Rapid increases in height and weight are indicators of adolescent physical development. Due to changes in body composition and nutrient excess or deficiency, nutritional needs increase during this phase. To determine the relationship between parental characteristics and nutritional knowledge with students' consumption of vegetables and fruits at Mercusuar Kupang Christian High School, the research instruments were questionnaires and the study was quantitative research using the observational method with a cross-sectional design. To see the relationship between parental characteristics and nutritional knowledge with students' vegetable and fruit consumption, the data was analyzed using the Chi-Square Test. The results of the univariate analysis showed that 53 students (43.8%) had good knowledge, 55 students (45.5%) had sufficient knowledge, and 13 students (10.7%) had poor knowledge. The most common educational background of the fathers was high school graduates, with 60 people (49.6%), while the most common educational background of the mothers was high school graduates, with 58 people (47.9%). Regarding the parents' occupations, the most common occupation for fathers was self-employed, with 53 people (43.8%). For mothers, the most common occupation was homemaker, with 53 people (43.8%). Based on bivariate analysis, there was no significant correlation between the mother's education level and vegetable consumption (p-value 0.937), no significant correlation between the father's education level and vegetable consumption (p-value 0.127), and no significant correlation between nutrition knowledge and vegetable consumption (p-value 0.137) and fruit consumption (p-value 0.137). However, for fruit consumption (p-value 0.885), there was a relationship between the father's occupation and vegetable consumption (p-value 0.005 ≤0.05), but there was no relationship with fruit consumption (p-value 0.682). There was no significant relationship between the mother's occupation and vegetable consumption (p-value 0.997), while for fruit consumption (p-value 0.434). Suggestion: For Mercusuar Kupang Christian High School, continue to provide education and motivation to adolescents so they can apply existing nutrition knowledge more effectively. The results of this research are expected to be used as a consideration for making policies on school children's nutrition services, such as establishing a canteen at school that offers affordable vegetables and fruits.Keywords: Student knowledge, Education, Parental occupation

Abstract

Malnutrition (wasting) is a condition caused by starvation over a certain period of time and the presence of chronic diseases. Based on the Indonesian health survey (SKI 2023) The prevalence of malnutrition (wasting) based on BB/TB or BB/PB indicators in Indonesia in 2023 was 6.4%. The prevalence of malnutrition status (wasting) based on BB/PB or BB/TB indicators in toddlers in East Nusa Tenggara was 9.4% and the Kupang City area was 12.1%. Referring to information from the Service (2024), there were 909 cases of malnutrition recorded in the Oepoi Health Center working area during the period of January to December 2024. To find out the results of the Evaluation of the Nutrition Management Program for Malnutrition (wasting) Toddlers at the Oepoi Health Center in 2024. The type of research used was qualitative research using the CIPP method (Context, Input, Process and Product) with a descriptive design. This research was conducted in May 2025. The sampling technique used in this study was the Snowball sampling technique, which consisted of 14 respondents. The data collected included the planning, implementation, and results of the six programs that had been implemented at the Oepoi Community Health Center in 2024. The results showed that the nutrition education program, exclusive breastfeeding, complementary foods (MP-ASI), supplementary feeding (PMT), vitamin A, and deworming had been planned and implemented, but with various obstacles. The nutrition education program and exclusive breastfeeding had not reached optimal coverage, while the MP-ASI and PMT programs showed better results even though there were still problems with malnutrition among toddlers. Evaluation of the Nutrition Management Program for Malnourished Toddlers at the Oepoi Community Health Center in Kupang City in 2024 had been implemented in accordance with applicable procedures, but of the six programs evaluated, three of the six programs had not achieved maximum results

Abstract

To achieve optimal growth and development, it is important to consider the golden age, often referred to as the vital development phase for children. According to the 2023 Indonesian Health Survey (SKI), 9.4% of children in East Nusa Tenggara (NTT) province are underweight. Therefore, researchers decided to investigate the impact of replacing catfish flour with green bean flour on biscuit acceptance. The objective of this study was to determine the nutritional components and biscuit acceptance test of biscuits made using green bean flour as a substitute for catfish flour. This study is an experimental study, with food formulations using proportions of catfish flour (10%, 20%, 30%) and green bean flour (30%, 20%, 10%). Acceptance test data were analyzed descriptively, using the Normality Test, ANOVA, and Tukey's Test. It can be concluded that the preference level for each treatment received a score of 4, indicating preference. Based on statistical analysis, there was a significant difference in taste between treatments P0 and P3. The macronutrient values of the biscuits, with varying proportions of catfish flour to green bean flour, were affected (energy, protein, fat, and carbohydrates). To prevent cross-contamination from the environment, researchers are advised to use an oven when drying the green beans and catfish, and to use uniform molds to ensure consistent product weight.

Abstract

Chronic energy deficiency (CED) is a disorder that affects adolescent girls and can last for years, characterized by inadequate food intake, especially in terms of energy and protein. Adolescents who have good knowledge of nutrition will be able and willing to regulate their diet so that it is balanced, neither deficient nor excessive. This study aims to assess the relationship between nutritional knowledge, energy intake, and protein intake with the incidence of chronic energy deficiency (CED) among female students at SMA Negeri 1 Kupang Timur. This study involved 210 tenth-grade female students at SMA Negeri 1 Kupang Timur as the population. Of these, 68 female students were selected as samples using stratified random sampling. Data analysis was performed using the Chi-square test. The results of the study at SMA Negeri 1 Kupang Timur showed that nutritional knowledge was not significantly related to CEE status (p = 0.734). Similarly, energy intake (p = 1.000) and protein intake (p = 0.540) also did not show a significant relationship with KEK status. There was no significant relationship between nutritional knowledge and energy and protein intake with KEK status.

Abstract

The prevalence of anemia is 32%, meaning that 3 to 4 out of every 10 adolescents have anemia. One way to address anemia is through the consumption of foods rich in iron and vitamin C. Jackfruit seeds and pumpkin are local food ingredients that contain iron, protein, and vitamin C, and can be processed into functional food products such as cookies. This study aimed to determine the effect of substituting jackfruit seed flour and pumpkin flour on the organoleptic properties and nutritional value of cookies as an effort to diversify nutritious local foods to prevent anemia in adolescent girls. This research was an experimental study using a Completely Randomized Design (CRD) with three treatments of jackfruit seed flour and pumpkin flour ratios: P1 (20%:25%), P2 (25%:20%), and P3 (30%:15%). Organoleptic tests were conducted by 30 student panelists from the Nutrition Department of Poltekkes Kemenkes Kupang. Data were analyzed using ANOVA to examine differences between treatments. Results: Conversion results showed that 1,000 grams of jackfruit seeds produced 650 grams of flour (conversion ratio 1.53:1), while 1,500 grams of pumpkin produced 400 grams of flour (conversion ratio 3.75:1). The organoleptic test results indicated that all treatments received average scores close to 5, categorized as “liked very much.” Treatment P3 (30%:15%) received the highest scores for color (4.90), aroma (4.73), texture (4.80), and taste (4.73). ANOVA results showed no significant differences (p>0.05) in all organoleptic aspects. Based on nutritional analysis, cookies from P1 (20%:25%) had the highest nutritional content, with protein 0.66 g, iron 0.17 mg, and vitamin C 0.36 IU per piece. To meet 10% of the daily iron requirement for adolescent girls, it is recommended to consume 11 pieces of P1 cookies. Conclusion: Substitution of jackfruit seed flour and pumpkin flour up to 45% (P3) produced cookies with a high level of preference without reducing organoleptic quality. P1 cookies are recommended because they have higher nutritional content, especially iron, protein, and vitamin C, which are beneficial in helping prevent anemia in adolescent girls.

Abstract

The nutritional status of women before marriage significantly impacts their physical readiness and reproductive health. An unbalanced diet—whether due to deficiency or excess—can affect fertility and fetal health. Women with undernutrition may experience menstrual irregularities, difficulty conceiving, or give birth to low-birth-weight babies. Meanwhile, overnutrition can lead to obesity and pregnancy complications. Therefore, maintaining a balanced diet before marriage is crucial for supporting pregnancy and the health of both the mother and the baby. According to the results of the 2023 Indonesia Health Survey, the prevalence of BMI-based nutritional status in NTT Province was 14.5% wasting, 56.3% normal, 12.1% overweight, and 17.1% obese, while the prevalence of KEK was 28.0% among pregnant women and 35.7% among non-pregnant women. This study aims to determine the nutritional status (BMI, mid-upper arm circumference, and waist circumference) of premarital women. The study design used was a quantitative descriptive study with a cross-sectional approach. The study results show that the majority of respondents had a BMI-based nutritional status in the normal category (65.3%), while 27.8% were overweight and 6.9% were underweight. Based on LILA measurements, the majority of respondents fell into the normal category (88.9%), while 13.9% were at risk of Chronic Energy Deficiency (CED). Meanwhile, waist circumference measurements indicated that 37.5% of respondents were in the obese category. The nutritional status of premarital women in the Alak Community Health Center’s service area is mostly normal. However, some still suffer from obesity; therefore, regular nutrition education and monitoring are necessary.

Abstract

Type 2 diabetes mellitus occurs when the body can no longer produce sufficient insulin or is unable to use insulin effectively. According to SKI 2023, the prevalence of type 2 diabetes among individuals aged ≥15 years reached 50.2%, while East Nusa Tenggara Province reported a prevalence of 45.9% in the same year. Medical record data from Prof. Dr. W. Z. Johannes Kupang Regional General Hospital indicated that there were 797 cases of type 2 diabetes mellitus from January to December. This study was conducted to assess the relationship between magnesium and vitamin C intake and random blood glucose levels in patients with type 2 diabetes mellitus. The study employed a descriptive analytical approach with a cross-sectional design, conducted from June 17 to July 17, 2025. Samples were selected using purposive sampling, resulting in 40 respondents who met the inclusion criteria. The variables examined included magnesium intake, vitamin C intake, and random blood glucose levels, all analyzed using the Chi-Square test. The findings revealed that 42.5% of respondents (17 individuals) had deficient magnesium intake. The Chi-Square test produced a p-value of 0.002, indicating a significant association between magnesium intake and random blood glucose levels. Meanwhile, 20 respondents (50%) had deficient vitamin C intake, but the statistical test showed a p-value of 0.184, demonstrating no significant relationship. Additionally, 39 respondents (97.5%) had high random blood glucose levels, with only one respondent (2.5%) falling within the normal category. Overall, the study concludes that magnesium intake is significantly associated with random blood glucose levels, whereas vitamin C intake shows no such association among patients with type 2 diabetes mellitus.

Abstract

Stunting remains a prominent nutritional issue in Indonesia, including in the city of Kupang, where the prevalence reaches 31.9%. Identifying local risk factors is crucial for determining appropriate intervention strategies. This study aims to analyze the relationship between direct breastfeeding, a history of low birth weight (LBW), maternal knowledge level, and family income with the incidence of stunting in toddlers aged 52–104 weeks. This study employed a cross-sectional design. A sample of 76 toddlers was selected from a population of 301 toddlers in the service area of the Oesapa Community Health Center, Kupang City. Data analysis showed no significant association between stunting and direct breastfeeding (p=0.204), a history of LBW (p=0.204), maternal knowledge level (p=0.824), or parental income (p=0.699). These findings indicate that these factors are not the sole determinants in this region, and there is a possibility of influence from other external factors not examined in this study, such as environmental sanitation, access to clean water, or the frequency of infections among toddlers. The internal and socioeconomic factors tested were not found to have a direct effect on stunting at the Oesapa Community Health Center. A more comprehensive approach is needed through nutrition education, improved child-rearing practices, interventions targeting environmental factors, and the strengthening of cross-sectoral health services.
