Abstract

Diabetes mellitus is a metabolic disease characterized by hyperglycemia, or blood sugar levels higher than normal. This condition is one of the major risk factors for the development of C. albicans, a fungal pathogen commonly found in tropical countries. Infections caused by C. albicans in diabetes mellitus patients can lead to serious complications, including severe sepsis, if left untreated. This study aims to identify the presence of C. albicans in the urine of diabetes mellitus patients at Griya Sehat Link Care, Bulukumba Regency. A descriptive study design was employed, with random sampling of 15 respondents. Analysis was performed using the Sabouraud Dextrose Agar (SDA) method, and results were presented in tabular distribution format. The findings revealed that five respondents (33.3%) tested positive for C. albicans in their urine samples, while ten respondents (66.7%) tested negative. These results emphasize the importance of early detection of C. albicans infection in diabetes mellitus patients to prevent further complications. Additionally, this study provides a reference for developing prevention and management protocols for fungal infections in diabetic patients, particularly in tropical regions like Indonesia. Abstract

Over the past three decades, there have been thousands of women undergoing treatment with tamoxifen. But there is a decrease or even no response to the breast cancer chemotherapy treatment. This phenomenon can occur due to resistance. sinularin has potential as a cancer drug.This study aims to evaluate the interaction of sinularin compounds and its derivatives with breast cancer receptors such as estrogen receptor alpha with PDB code: 5W9C, progesterone receptor with PDB code: 1A28, and human epidermal growth factor receptor 2 using proteins with PDB code: 3PP0 using molecular docking methods to determine anticancer potential.This research method is in-silico research. The research was conducted by screening test compounds such as prediction of compound potential using PASS Online Server, prediction of physicochemical properties using SwissADME, and prediction of compound toxicity using ProTox 3.0. Then validation of the method using AutoDock Vina, Next molecular docking using AutoDock Vina, and visualization of molecular docking results using Biovia Discovery Studio Visualizer. The results showed that sinularin compounds and their derivatives showed anticancer potential through the PASS test, low toxicity, and compliance with the Lipinski Rule of Five. Method validation showed RMSD values ​​below 2.0 Å. In molecular docking, sinularin showed a binding affinity value with a value of -8.027 kcal/mol which was better than tamoxifen with a value of -7.61 kcal/mol, although still below the native ligand of estrogen receptor alpha with a value of -8.815 kcal/mol. Sinularin and its derivatives have potential as anticancer agents through interaction with ERα and HER-2. Keywords: Breast Cancer; Sinularin; Molecular Docking.

Abstract

This study aims to assess the completeness of cough medicine prescriptions based on administrative, pharmaceutical, and clinical aspects at the Mitra Sehat Banjarbaru Pharmacy from July to December 2023. The method used was a descriptive non-experimental, with a retrospective approach to 138 prescriptions, using a total sampling technique. Data analysis used Microsoft Excel 2021 software, namely by entering the obtained data into Microsoft Excel with the final percentage results and displaying them in graphical form. The results of the analysis of the administrative aspects showed that the majority of prescriptions included administrative completeness in accordance with standards, such as the prescription date, patient data, drug name, signature, doctor's SIP number, and the doctor's signature or initials. However, incomplete patient data was found at 100%, namely patient name 0%, patient address 94%, patient date of birth 95%, medical record number 40%, patient age 79%, patient weight 96%, patient gender 99%. In terms of pharmaceutical aspects, the results showed a fairly high level of conformity in dosage form and drug compatibility, each reaching 100%. However, there was a discrepancy in dosage strength in 4% (5 prescriptions). The clinical aspects of this study showed that the majority of prescriptions listed the appropriate route of administration, reaching 99%. The most commonly prescribed drug by doctors was Guiafenesin, at 56% (77 prescriptions). This study concluded that of the 138 prescriptions analyzed, the majority of prescriptions fulfilled most administrative aspects, such as the patient's name, the doctor's Practice License (SIP) number, and the doctor's signature or initials. In terms of pharmaceutical aspects, the results showed a fairly good conformity, in terms of dosage form and drug compatibility, reaching 100%. The clinical aspects of this study showed a completeness result reaching 99%. Nevertheless, it is recommended that pharmacies strengthen prescription review protocols and increase pharmacist supervision to prevent medication errors and improve patient safety in pharmaceutical services. Abstract

Self-medication or self-treatment is an attempt by a person to treat themselves by recognizing the symptoms or illnesses they are experiencing and choosing their own medicine. However, in reality, self-medication is not accompanied by good drug waste management. So this study aims to measure students' knowledge about self-medication and drug waste management at Insan Harapan Islamic Boarding School and analyze the relationship between students' self-medication knowledge and drug waste management. The approach used in this study is the Cross-Sectional method. The study population consisted of all high school students at Insan Harapan Islamic Boarding School in Karawang, with a sample of 187 students selected using non-probability sampling. Data were analyzed using the spearman rank test. The results showed that 60% of respondents had a high level of knowledge about self-medication for minor illnesses, while 71% demonstrated good practices in drug waste management. This study also found a significant relationship between students' knowledge of self-medication for minor illnesses and their practices in drug waste management. The correlation test shows a significance p-value test is 0.01, which is less than 0.05, so it can be concluded that there is a relationship between the level of knowledge of self-medication for minor illnesses and the management of drug waste at the Insan Harapan Karawang Islamic Boarding School. However, this relationship is low with a coefficient correlation value of 0.25. These findings highlight the importance of enhancing health education on self-medication and drug waste management, as well as developing policies to regulate drug waste management to prevent adverse environmental impacts. Abstract

Breast cancer remains the leading cause of death among women worldwide. Resistance to chemotherapy and adverse effects of conventional treatments have driven the search for alternative therapeutic agents derived from natural compounds. One of the key molecular targets involved in cancer cell apoptosis is the Bcl-2 protein family. This study aimed to systematically review in silico molecular docking studies of natural compounds from tropical plants and synthetic molecules targeting the Bcl-2 protein as potential therapeutic agents for breast cancer. A total of 21 original research articles published in the last five years were analyzed based on the source of compounds (plant-derived or synthetic), active constituents, protein database (PDB) codes, and binding affinity values. The results indicated that most compounds demonstrated strong inhibitory potential against Bcl-2, with notable examples such as Niaziminin B from Moringa oleifera (−14.96 kcal/mol) showing one of the highest binding affinities. This review highlights the potential of natural compounds from tropical plants and synthetic molecules as promising candidates for breast cancer therapy through Bcl-2 inhibition. Further in vitro and in vivo studies are required to confirm their biological activity and toxicity profiles in the drug development process. Abstract

Antioxidant compounds were known to reduce the reactivity of free radicals, thereby helping to prevent damage to cells and bodily molecules. These compounds were commonly found in various plants and fruits and were considered relatively safer than synthetic antioxidants. In this study, the author utilized white rind extract of watermelon (Citrullus lanatus) as a natural antioxidant source in the formulation of a peel-off  gel face mask intended for dry skin, using different concentrations of carbopol  940 as a gelling agent. An experimental method was employed, involving formulation development and product evaluation. Based on the assessment of three peel-off  mask formulations containing 3.5% watermelon white rind extract, the optimal concentration of carbopol  940 was determined to be 0.5%. This conclusion was drawn from evaluations of homogeneity, organoleptic properties, spreadability, pH, viscosity, drying time, and product stability. The peel-off  gel face mask formulated with 0.5% carbopol  940 and watermelon rind extract appears safe based on pH range and physical tests; however, further safety assessments. Abstract

Cancer is a leading cause of death worldwide, with nearly 10 million fatalities reported in 2020, including 2.3 million new cases of breast cancer. Due to its high prevalence, effective treatment options remain a critical priority. Clitoria ternatea L., a medicinal plant widely found in Indonesia, is traditionally used for various therapeutic purposes and may possess anticancer properties. This study aimed to evaluate the cytotoxic effects of ethanol extracts of Clitoria ternatea L. collected from Ternate Island. Cytotoxicity was assessed using two methods: the Brine Shrimp Lethality Test (BSLT) to determine general toxicity, and the PrestoBlue assay to evaluate antiproliferative effects on MCF-7 human breast cancer cells. In the BSLT, Artemia salina larvae were exposed to various concentrations of the extract (7.81-1000 µg/mL) for 24 hours, and LC50 values were calculated using probit analysis. For the PrestoBlue assay, MCF-7 cells were treated with extract concentrations ranging from 7.81 to 1000 µg/mL for 48 hours, followed by measurement of cell viability via absorbance at 570 nm. Results showed that the LC50 for the BSLT was 105.681 µg/mL, indicating low toxicity of the extract. In contrast, the extract did not demonstrate significant cytotoxicity against MCF-7 cells, with an IC50 value exceeding 1000 µg/mL. In conclusion, the ethanol extract of Clitoria ternatea L. exhibits low general toxicity but lacks significant cytotoxic effects on MCF-7 breast cancer cells under the tested conditions. Further studies are warranted to isolate active compounds and explore their potential anticancer mechanisms. Keywords: BSLT; Clitoria ternatea L.; MCF-7; Prestoblue Assay.

Abstract

Streptococcus mutans is one of the bacteria that causes teeth disorders. This bacterium is a typical gram-positive, round-shaped bacteria often found in pairs or chains. This study aims to determine the performance of the combination of kepok banana peel extract and clove oil as an herbal mouthwash formulation that inhibits the growth of Streptococcus mutans bacteria. The method carried out in this study is an experimental method using the disc diffusion method. The components of the three formulations designed, formulation I, use 1 gram of kepok banana peel extract and 5 ml of clove oil. In formulation II, 2 grams of kepok banana peel extract and 7.5 ml of clove oil were used, while in formulation III, 3 grams of kepok banana peel extract and 10 ml of clove oil were used. The evaluation test of mouthwash preparations is carried out several times, including organoleptic tests. The study included antibacterial activity tests. In this study, the results of the antibacterial activity test of herbal mouthwash formulations were obtained to have inhibitory power in Streptococcus mutans. The inhibition zone produced by formulation I was 13.38 mm (strong), formulation II was 11.64 mm (strong), and formulation III was 10.55 mm (strong). Based on research, it can be concluded that this herbal mouthwash formulation has the potential as a natural alternative in controlling Streptococcus mutans and preventing oral diseases. Abstract

Parijoto fruit extract has antioxidant activity with an IC50 value of 19.73 ppm. Parijoto extract needs to be formulated into a lip balm to maximise its potential. Lip balm preparations require a stiffening agent used to maintain shape, texture, and stability. The stiffening agent used in this study is cera alba. The aim of this research is to determine the effect of varying concentrations of cera alba as a stiffening agent on the physical properties of lip balm preparations made from parijoto fruit extract (Medinila speciosa Blume). This research is an experimental study. The parijoto fruit extract was prepared by maceration extraction using 96% ethanol. The preparations were made with 3 formulas with different concentrations of cera alba as a stiffening agent: FI (10%), FII (15%), and FIII (20%). Subsequently, physical tests of the lip balm preparations were conducted, including organoleptic tests, pH tests, homogeneity tests, adhesion tests, spreadability tests, viscosity tests, moisture content tests, and irritation tests. The results of the physical property analysis of the lip balm preparations were analysed descriptively and inferentially using statistical software with one-way ANOVA and post hoc Tukey tests. Based on the results, the variation in cera alba concentration as a stiffening agent influences the physical properties of the lip balm extract from parijoto fruit (Medinila speciosa Blume), including pH, viscosity, adhesion, and spreadability, but does not significantly affect moisturising ability. In this study, FI with 10% cera alba concentration was established as the best formula based on the fulfilment of all physical property tests (pH, viscosity, adhesion, spreadability) and the statistical test results. Abstract

Determination of drug concentration in biological specimens, including blood, plasma, serum, urine, tissue, and feces is a critical aspect of bioanalysis. The increasing interest in research on drug molecules, both existing drugs and their metabolites, and new drug candidates shows that a new development approach is needed for their bioanalytical methods. Sample preparation is an important step to eliminate the influence of the matrix and improve the analytical performance of a method. Molecularly imprinted polymer (MIP) as a separation method with unique features that are selective to target molecules has been applied to various applications and benefits, especially when used as an adsorbent in preparation methods for the analysis of complex biological matrices. The purpose of this study was to provide an overview of the use of MIP in the bioanalysis of drug compounds. The method used in this study uses a review of articles related to the concept of bioanalysis and the application of MIP for the analysis of drug components in biological matrices. Based on the results of the study, there has been an increase in the number of studies on the development of MIP over the last decade. Investigations show satisfactory results in MIP-based bioanalysis methods. The results of the study show the superiority of MIP as a preparation method that has high accuracy, reproducibility, sensitivity, speed, and cost-effectiveness, which makes it suitable for clinical use. This study can be a preliminary for further research on the development of MIP for bioanalysis of drug molecules.
