Behavioral, physiological, and blood biochemistry of Friesian Holstein dairy cattle at different altitudes in West Java, Indonesia. Biodiversitas 23: 533-539. For dairy cows, the study of physiological aspects and those related to it is very important based on altitude. The data of this study are the main considerations to determine the ability of homeostasis, prediction of production and appropriate feed management. This study aims to examine the behavior, physiological abilities, and blood plasma biochemistry of sixty dairy cows. It was conducted using three locations in West Java Province based on topography, namely: (i) location with topography 350-500 meters above sea level (masl): Sukabumi Regency; (ii) location with topography 550-750 masl: Sumedang Regency; and (iii) locations with topography > 800 masl: Bandung Regency. Furthermore, 5 mL of blood samples were taken accurately, using a syringe and a 5 mL tube containing EDTA, respectively. Blood sampling was taken carefully from the jugular vein of dairy cows at the beginning of every month for six sampling periods. After each collection, the whole blood was separated from the plasma directly using a centrifuge, with a speed of 4500 ppm for 7 minutes. The plasma obtained was used to measure the value of blood biochemistry related. The plasma analysis was conducted using a Kenza 240TX model spectrophotometer. The procedure for analyzing blood samples followed the instructions listed in the randox and biolabo kits. Furthermore, behavioral and thermoregulation measurements were performed every week during this research. The results showed an increase in dairy cows' time spent lying and drinking at low altitudes compared to dairy cows at high altitudes. In contrast, rumination and feeding activities were decreased, as shown in the blood's physiological response and biochemical profile. Dairy cows appear to be more challenging to adjust physiologically at lower altitudes.

The development of adaptive wheat (Triticum aestivum L.) mutant lines in tropical climate is a long-term solution. The advanced yield trial is an important stage in this development, which is carried out using a selection index and multivariate analysis. This study aims to determine the selection effectiveness of tropical wheat mutant lines by using a selection index and multivariate analysis. the study was conducted in three locations, which included two locations for the follow-up yield test in Kelara Village and Ujung Bulu Village (South Sulawesi, Indonesia), and the third location for the selection validation in Padang Magek Village (West Sumatra, Indonesia). This study was designed by a nested Randomized Complete Block Design (RCBD), with replications nested in locations. The samples included 16 wheat genotypes (13 mutant lines and three comparison varieties) that were replicated three times. The result showed that the selection of tropical wheat mutant lines using selection index and multivariate analysis were considered effectively with high determination value of 0.57 (57%). The results of the selection index show that seven lines were recommended for multi-location trials since they had a better selection index potential than Dewata variety.

Short Communication: The potency of lytic bacteriophage isolated from various environments to control the growth of Citrobacter braakii causing urinary tract infection. Biodiversitas 22: 5550-5554. Citrobacter braakii is one of the pathogenic bacteria causing urinary tract infection (UTI) in humans. Bacteriophages that are specifically infecting C. braakii could be the alternatives to combat antibiotics resistance cases of this bacterium. This study aimed to isolate lytic phages from various environmental samples (tofu factory wastewater, sewage water, rice field water, fishpond water, cattle farm wastewater, and goat farm wastewater), and to analyze their effectivity to reduce UTI-causing C. braakii population in vitro. Results exhibited that phages targeting this bacterium were found in goat and cattle farm wastewater and not present in other samples. Two phages, namely FC1 and FC2, had different plaque morphology characteristics. The number of phages in cattle farm wastewater was 2.8 × 105 PFU/mL and 1.32 × 104 PFU/mL in goat farm wastewater. It was observed that the phages found in these environments also indicate the presence of their host. After 8 h incubation, FC1 and FC2 phage reduced UTI-causing C. braaakii population in vitro from 0.23×108 CFU/mL to 0.03×108 CFU/mL and 0.02×108 CFU/mL, respectively. The phages isolated from these two samples can be further developed for the treatment of UTI caused by C. braakii and goat and cattle farm wastewater treatment to prevent contaminating other areas.

Multispecies pelagic fisheries have contributed as the main fishery production for the Bali Strait fisheries. To maintain the sustainability of the fish stock, sets of harvest control rules (HCRs) have been introduced in terms of fishery management. Conventional single-species surplus production models have been used extensively to determine the future annual allowable biological catch (ABC). However, they may not technically apply to multispecies fishery management. The feedback harvest control rule HCR has been validated and applied to maintain the catch variability of the multispecies fishery. This study aims to technically compare the use of the surplus production model and the feedback HCR in the ABC estimation. The catch and effort data series of the three dominant species from 2010-2019, collected from Pengambengan and Muncar fishing port, were analyzed by two HCRs, the Schaefer surplus production model and the feedback HCR to obtain the ABC. As a result, the Schaefer model showed a higher estimation of ABC than the feedback HCR because the feedback HCR may reflect the recent historical catch consistent with precautionary principles. The feedback HCR showed an initial step toward sustainably managing multispecies fisheries while dealing with unavailability in species-specific data conditions.

Diversity, coverage, distribution and ecosystem services of seagrass in three small islands of northern Papua, Indonesia: Liki Island, Meossu Island and Befondi Island. Biodiversitas 22: 5544-5549. Papua waters are one of the hot spot areas with high marine biodiversity in the world. Yet, the less accessible location and varying geographical conditions become the significant constraints of biodiversity study in this region, resulting in limited information about the diversity of marine resources, including the seagrass ecosystem. This research aims to investigate the diversity, coverage and distribution of seagrass in three small islands in the northern waters of Papua, namely Liki Island, Befondi Island, and Meossu Island, as well as to study the ecosystem services provided by the seagrass. This research is part of the Nusa Manggala Expedition, which was carried out in November 2018, consisted of seven observation stations. Data collection was conducted using the transect line method combined with plot quadrat. Information regarding seagrass ecosystem services was gathered through observation and direct interviews to the community. The results showed that there were seven species of seagrass in the studied sites, namely Cymodocea rotundata, Enhalus acoroides, Halodule uninervis, Halodule pinifolia, Halophila ovalis, Thalassia hemprichii, and Syringodium isoetifolium. Liki Island had the highest seagrass cover value of 82.24%, followed by Meossu Island and Befondi Island with seagrass cover of 63.23% and 31.25%, respectively. There were 13 seagrass ecosystem services identified in this research. The people of Liki Island and Meossu Island get the benefits of the existence of the seagrass ecosystem. The direct benefits are the presence of seagrass is associated with biota commonly used as food sources such as Siganus sp and sea cucumbers. There are efforts made by the community to maintain the sustainability of the seagrass ecosystem, which is known as sasi.

Abstract. Agus A, Agussalim, Sahlan M, Sabir A. 2021. Honey sugars profile of stingless bee Tetragonula laeviceps (Hymenoptera: Meliponinae). Biodiversitas 22: 5205-5210. Honey was a functional food to improve human health, but irresponsible people used this circumstance to make fake honey. This study aimed to evaluate the profile of the sugar of stingless bee honey [Tetragonula laeviceps (Smith, 1857)] from different geographical origins in Indonesia. Honey, three samples were directly collected from three other sources for meliponiculture of T. laeviceps in Indonesia: Sleman, Klaten, and Gunungkidul. The honey sugars profile was analyzed: glucose, fructose, sucrose, reducing sugar, the sum of fructose and glucose, glucose to moisture ratio, fructose to glucose ratio, and honey pH. Glucose and fructose were analyzed by HPLC, sucrose by Luff Schoorl, reducing the sugar by Layne-Enyon, and pH by a pH meter. The current findings revealed that the geographical origins had a highly significant effect on glucose, fructose, the sum of fructose and glucose, glucose to moisture ratio, fructose to glucose ratio, and honey pH (P<0.01) and significant effect on reducing sugar (P<0.05), but not on sucrose content. Thus, it can be concluded that the origins were affecting the honey sugars profile and honey from Sleman has the highest sugars content, followed by honey from Klaten and the lower was honey from Gunungkidul.

Detection of marine microalgae (phytoplankton) quality to support seafood health: a case study on the west coast of South Sulawesi, Indonesia. Biodiversitas 22: 5179- 5186. The research aimed to detect marine microalgae quality to support seafood health was carried out from January to November 2020 along the west coast of South Sulawesi, Indonesia. Samples were collected from the coastal waters of Pangkep District, Maros District, and the northern part of Makassar City. Phytoplankton cell counts were obtained using the deposition method developed by Uthermol. Phytoplankton cell abundances were calculated through sweeping (census) using a Sedgwick Rafter Cell (SRC). Two-way analysis of variance (ANOVA) was used to compare the distribution of marine microalgae community abundance between observation stations and periods. Based on the types and relative abundance of phytoplankton present, i.e., harmful algal bloom (HAB) forming or not (non-HAB), the results showed the quality of marine microalgae, specifically, phytoplankton was relatively good. Many more non-HAB (94-98%) than HAB (2-6%) marine microalgae were detected. Thus, the phytoplankton flourishing in these waters is mostly suitable as food for other organisms, including fish and shellfish. This also means that if fishers harvest these fish and shellfish, they should be fit and safe for human consumption.

Abstract. To’bungan N, Pratiwi R, Widyarini S, Nugroho LH. 2021. Cytotoxicity extract and fraction of knobweed (Hyptis capitata) and its effect on migration and apoptosis of T47D cells. Biodiversitas 23: 572-580. Hyptis capitata Jacq. has an ethnobotanical history. The leaves and stems of H. capitata have been reported to have potential anticancer properties. This study was conducted to determine the anticancer potential of the roots, stems, leaves, and flowers of H. capitata. Organs of H. capitata were extracted in stages using chloroform and continued with methanol to obtain eight extracts. Crude extracts were tested for cytotoxicity on T47D and WiDr cells with the MTT method. Potential inhibition of cell migration by potential fraction was tested with scratch wound healing assay and the content of secondary metabolites was analyzed with gas chromatography-mass spectrometry (GC-MS). An Annexin V-Pl apoptosis test was performed to determine the number of living cells, necrosis, and apoptosis. The results showed that the root chloroform extract showed the highest cytotoxic activity against T47D cells (34.90 ± 4.78µg/mL) and WiDr (44.65 ± 12.07 µg/mL), and it was referred to as a potential extract. Fractionation of the root chloroform extract resulted in four fractions (F1, F2, F3, and F4). Fraction F2 had the highest cytotoxic activity (13.8 ± 0.65 µg/mL) and was selective against T47D cells (selectivity index 3.71). Fraction F2 showed its antimetastatic and apoptotic potential against T47D cells. Gas chromatography-mass spectrometry analyses combined with library showed that the root chloroform extract and fraction F2 contain ferruginol, campesterol, and stigmasterol.

Abstract. Bao TQ, Viet LH, Hai NH, Tuan NT, Cuong LV. 2021. Population dynamics and regeneration of Shorea roxburghii, a threatened timber species in Southern region, Viet Nam. Biodiversitas 22: 5649-5656. Despite its wide distribution in South and Southeast Asia, Shorea roxburghii G. Don is classified as Vulnerable in the IUCN Red List of Threatened Species due to over-exploitation for its valuable timber. This study aims to investigate the population dynamics and regeneration of S. roxburghii in southeastern Vietnam, serving for conservation and sustainable development plan. Six plots of 2,500 m2 each (50 m × 50 m) were established at two forest conditions: i) forest with standing volume > 200 m3/ha (SV>200), and ii) < 200 m3/ha (SV<200); to investigate stems with diameter at breast height (DBH) ? 5 cm and that < 5 cm. Moisture (%), pHH2O of the topsoil and the occurrence of regenerating S. roxburghii were collected from 125 points in the study site. The results indicated that mean stem height, DBH, basal area, and aboveground biomass were significantly higher at the forest stand with SV>200 than that at the forest stand with SV<200 for the pool of all species. Similar higher figures were found in SV>200 for the pool of S. roxburghii compared to that in SV<200, except mean DBH, which was larger in SV<200. There were missing stems with DBH < 5 cm in SV<200, while it was numerous in SV>200 (1,313 stems/ha). DBH/stem distributions in both forest types had an inverted-J shape with a peak at 10-15 cm DBH for the pool of all species, while there was no clear pattern for the pool of S. roxburghii in both forest types. Regression analysis indicated that higher total crown area of stems ? 23 tall leads to higher S. roxburghii stems, while the higher total crown area of stems < 23 tall leads to fewer S. roxburghii stems with DBH < 5 cm. It is concluded that the total crown area of forest stand plays an important role in the sustainable regeneration of S. roxburghii. Silvicultural treatment should be applied to reduce stand density and crown area of stems < 23 m tall to enhance sunlight reaching the forest floor for germination, growth, and survival of S. roxburghii. The moisture of the topsoil ranging from 60 - 80% and the pHH2O ranging from 3.8 - 4.6 are suitable conditions for natural regeneration of S. roxburghii.

Genetic variation of Austropuccinia psidii in some species of Myrtaceae as host plants in Java, Indonesia based on simple sequence repeats (SSR) markers. Biodiversitas 23: 256-263. Austropuccinia psidii is pathogenic rust with a wide host and is considered a biosecurity threat to the Myrtaceae family in many countries. The genetic variation of this rust in Java, Indonesia is poorly reported. Therefore, this study aimed to determine the genetic variation among A. psidii from different Myrtaceae hosts, namely Syzygium polyanthum, S. myrtifolium, and Melaleuca cajuputi from three different locations in Java using seven simple sequence repeats (SSR) markers. Data were collected from 28 A. psidiisamples from three different hosts and locations in Java. The genetic variations of A. psidii were found in six isolates in locus USYD_Pp168, PpSSR161, and PpSSR195*. The results showed that the expected heterozygosity value among 12 isolates of A. psidii is 0.312. The dendrogram illustrates two clusters constructed with cluster I consisting of subcluster IA (S1, S3, S4, S5, K1, K2, K3, K4, and P1) and IB (P2); and cluster II consists of isolates S2 and S6. Principal Coordinate Analysis (PCoA) was used to demonstrate the similarity and dissimilarity among isolates based on microsatellite sites. Axis 1 and 2 explained 87.7 % of the total variations and sep a rated isolate S1, S2, S3, S4, S6 from other isolates, and grouping the rest of isolates together. The dendrogram and PCo analysis demonstrated A. psidii isolates tend to have close genetic similarity based on their host.
