Abstract

Minimally processed fruits provide convenience and freshness characters. Weight loss is a critical factor affecting the shelf life of minimally processed apples. These apples were limited by brown coloration that develops on surface. The objective of this study is to evaluate the effectiveness of edible coat and film made from aren starch against weight loss and browning on minimally processed apples. Edible coat and film solutions were prepared from 3% (w/v) aren starch, 6% (w/w) palmitic acid, and 1% (w/v) sorbitol. Minimally processed apples were either dipped in the coating solution or wrapping by edible film and then stored at ambient temperature for 8 hours. Browning appearance was determined every 4 hours and weight loss of apples was measured every hour.  Edible film was much effective on weight loss control than coating during storage. Color analysis showed that pati aren edible  coating and film wrapping did not significantly delayed browning reaction on  minimally processed apples.

Abstract

Karakterisasi molekuler anjing Kintamani berperan penting dalam penentuan status trah dan mencegah penurunan kemurnian akibat perkawinan silang dengan trah anjing lain. Anjing Kintamani sangat popular di Bali. Anjing Kintamani dipercaya berasal dari Kintamani, Bangli, tetapi asal usulnya sampai kini belum jelas. Mikrosatelit merupakan runutan nukleotida terulang yang didistribusikan secara acak dalam gen vertebrata. Lokus mikrosatelit ini telah diketahui sangat polimorfik, karena keragaman jumlah nukleotida yang terulang. Mikrosatelit merupakan petanda allel utama yang dapat digunakan untuk pemetaan gen, genetika populasi dan identifikasi individu serta alat untuk mengungkap karakter genetik hewan. Pada penelitian ini diungkap karakteristik genetik anjing Kintamani menggunakan mikrosatelit. Jumlah dan ukuran allel pada 425 anjing Kintamani dianalisis menggunakan 116 macam primer mikrosatelit. DNA diisolasi dari sel hasil swab pipi. Amplifikasi 116 lokus mikrosatelit menggunakan PCR dalam 12 multiplek. Produk PCR dipisahkan dengan gel bis-akrilamid 6% dalam automated DNA sequencer. Flurosesnsi yang dihasilkan dideteksi dengan program Genescan 3.1, dan program Strand versi 2.2.39 digunakan untuk menghitung jumlah allel. Hasil penelitian menunjukkan bahwa jumlah allel yang didapat adalah 1128. Jumlah allel perlokus berkisar antara 3 (AHT136) sampai 41 (FH2138). Rataan PIC adalah 0,68 dan semua lokus bersifat polimorfik.

Abstract

Hard or stony coral is one important component to structure the coral reef ecosystem and play a significant role for its marine life and its surrounding. Therefore, the condition of reef ecosystem needs to be maintained. Research on stony coral condition and distribution in waters of Wori Village, North Sulawesi, was conducted in April 2008. Line Intercept Transect was applied to assess the coral condition. A 50 m line transect was set on the 5 - 7 m depth horizontally coastal line with assumption that in this depth, coral growth well. The results of this study show that the condition of coral was between 1,00 and 53,44% indicating the condition ranged from bad to good category. Stony coral distribution was found up to 20 m depth, with the highest diversity and evennes indices were 1,06 at station 3, and 0,56 at station 4, respectively. Numbers of stony coral were 99 species belonging to 40 genus and 14 families.

Abstract

Food poisoning cases need to be examinated intensively. There have been five poisoning cases reported between 2003-2004 in Sleman area. Three of them were caused by contaminated tuna fish. The objective of this research was to determine bacteriological quality of salted tuna fish from several markets in Yogyakarta. Salted tuna fish sample were collected from five markets, namely Beringharjo, Condongcatur, Demangan, Kranggan and Sentul. In each market, four vendors were selected. Bacteriological test were done including total microbe cells utilizing PCA medium, Coliform test utilizing VRBA medium. Identification and isolation of Vibrio parahaemolyticus use pre-enrichment selective medium of Alkaline Petone Water and selective Chromogenic medium agar for Vibrio. Salmonella test utilizing Chromogenic agar continued by utilizing TSI and LIA medium. The result showed that bacteriological quality of salted tuna fish from markets in Yogyakarta did not fulfill the requirements based on SNI 01-2717-1992. It was shown that total microbial count were high, i.e. 2.4 x 105 – 5.27 x 107 CFU/ g. Coliform count ranged from < 2.5 x 101 - > 6.5 x 106 CFU/g. From 20 samples that were brought to analyzed, 60% was positively presumed contaminated by V. parahaemolyticus and 50% was positively presumed contaminated by Salmonella.

Abstract

The objective of the present study was to evaluate the effect of heat-moisturetreatment (HMT) and sodium tripolyphosphate (STPP) addition on the quality ofinstant noodles from sweet potato starch. The diversification of raw material used innoodle production often necessitates processing modifications. Heat-moisturetreatment was applied to native sweet potato starch (HMTSPS) in three conditions(1100C; 10, 20 and 30 min) and STPP was added in three level (0,60; 0,75 and 0,90%).HMTSPS, STPP, water, lecitin and CMC were mixed. The mixture was pelleted,steamed and extruded. The extruded noodles were dried using a microwave oven. Thenoodles were evaluated both when dry and after cooking. The HMT method andSTPP addition improved noodle quality. Swelling index, solid loss and cooking loss ofthe noodles was decreased, while the texture and the elastisity of the noodles washigher. But noodles made with added of STTP more than 0,75% had low elastisity.The best instant noodle prepared by HMT method for 20 min and added of STTP0,75%.

Abstract

Aktivitas musiman enzim pyruvate kinase (PK), dan citrate synthase (CS) pada otot kaki siput, Littorina littorea (Gastropoda, Littorinidae) telah diteliti. Enzim-enzim ini berperan sebagai enzim kunci pada rangkaian metabolisme (metabolic pathways). Perbedaan yang signifikan terlihat pada aktivitas adalah CS dan PK sepanjang musim. Aktivitas spesifik dari CS bervariasi antara 0,04 dan 0,4 μg berat basah dan PK antara 0,1 dan 1,7 μg berat basah. Aktivitas enzim tersebut lebih tinggi pada musim dingin dibandingkan dengan musim panas. Hubungan negatif antara suhu dan aktivitas enzim terdeteksi dalam penelitian ini. Terdapat perbedaan yang nyata antara suhu air dan aktivitas enzim CS, dan suhu air dan udara pada enzim PK (P<0.05). Teramati tendensi yang sama untuk prosentasi perubahan pada aktivitas enzim PK dan CS pada bulan pertama pengamatan dan bulan berikutnya. Hasil penelitian setuju dengan hipotesis yang mengatakan bahwa terdapat kemungkinan perubahan aktivitas enzim CS dan PK selama siklus musiman. Fenomena ini dibahas dalam pembahasan tulisan ini.

Abstract

Curcumin which is isolated from Curcuma longa L. rhizome, has been used as a food additive, medicinal agent, cosmetic and fabric dye, whithout known its side effects. This research is to study the effects of curcumin on the structure of forelimb finger. Twenty virgin female rats, 2 month old, 150 g body weight were mated with fertile male rats. The present of vaginal plug or spermatozoa in the vagina were determined as the first day of pregnancy. Pregnant animals were devided into 4 groups of 5. Treatment were started from 10th to 17th day of pregnancy. The first 2 groups for control and vehicle only. Curcumin was dissolved in aquous 5% Tween 80. The second 2 groups were given curcumin at the dosage of 100 and 150 mg kg -1 bw. Rats were sacrificed at 18th day of pregnancy. Foetuses were dissected out, fixed in 10% formalin. Morphology of the foetuses were observed, forelimb were processed by paraffin method, and plantar paraffin section were made and used for histological observations and cell development of fingers. The results showed that curcumin at the dosage of 100 mg kg-1 body weight caused some foetuses dead, resorption of the embryo, hemorrhagi, unilateral phocomelia. Microscopical observations showed the treatment caused induced of nail formation, inhibition of indensity interdigity, and the increased of new blood vessel formation. It was concluded that the curcumin has no effect on the growth of foetuses. However it was affected the development of forelimb finger which is non dosage dependent.

Abstract

Penelitian mengenai keanekaragaman ikan air tawar di Sungai Opak DIY telah dilakukan dan dipublikasikan, tahun 2006, 2011 dan 2013. Data mengenai keanekaragaman ikan di Sungai Opak perlu dilakukan penambahan data, terutama data menyeluruh dari hulu hingga muara. Penambahan data perlu dilakukan, karena dimungkinkan ditemukan jenis ikan yang belum sempat tertangkap pada penelitian sebelumnya. Data jenis ikan yang tidak tertangkap pada penelitian sebelumnya, dapat menambah keanekaragaman jenis ikan di Sungai Opak. Penelitian ini bertujuan untuk melengkapi dan menambah data keanekaragaman jenis ikan yang terdapat di sepanjang Sungai Opak, dari hulu hingga muara. Pengambilan sampel dari hulu hingga muara menggunakan metode Purposive Random Sampling dengan bantuan jaring besar, kecil dan jala tebar. Sampling secara umum dibagi dalam empat bagian yaitu: hulu, tengah, hilir dan muara. Pada penelitian yang kami lakukan, terdapat beberapa jenis ikan yang tidak dijumpai pada penelitian sebelumnya, yaitu: lima jenis ikan dibagian hulu, di bagian tengah terdapat tiga jenis ikan yang tidak dijumpai pada penelitian sebelumnya, baik sampling hulu maupun hilir. Di bagian hilir dijumpai tujuh jenis ikan saat penelitian ini dan tidak dijumpai pada penelitian sebelumnya. Pada bagian muara, dijumpai lima belas jenis ikan yang tidak dijumpai di hilir pada penelitian sebelumnya. Total terdapat 28 jenis ikan yang dijumpai di tahun 2013 tetapi belum dijumpai di penelitian sebelumnya.

Abstract

In molecular studies, extraction can be time consuming and costly (especially in large scale studies), therefore some studies examine the success rate of amplification using alternative DNA isolation methods (without extraction). This study aims to determine the most effective DNA isolation method in Javan Plover molecular sexing by comparing several methods, such as extraction kit, alkaline lysis and direct PCR.This study used filter paper (5) and EDTA stored blood (8) as samples, and both sample types would be isolated using extraction kit, alkaline lysis, and direct PCR method. Molecular sexing was carried out using 2550F/2718R primer. The results showed that DNA in filter paper and EDTA stored blood, both using kit extraction, alkaline lysis and direct PCR, methods were successfully amplified with 2550F/2718R primer which produced 621 bp sized CHD1-Z and 459 bp sized CHD1-W band. The results suggested  that extraction kit is the most effective DNA isolation method for Javan Plover molecular sexing.  AbstrakDalam studi molekuler, ekstraksi dapat memakan waktu dan biaya saat diterapkan dalam penelitian skala besar, sehingga beberapa studi mengkaji tingkat keberhasilan amplifikasi menggunakan metode isolasi DNA alternatif (tanpa ekstraksi). Penelitian ini bertujuan untuk mengetahui metode isolasi DNA paling efektif untuk molecular sexing Cerek Jawa. Penelitian ini menggunakan 5 sampel darah kertas saring, dan 8 sampel darah EDTA yang diisolasi dengan metode ekstraksi kit, alkalin lisis dan PCR langsung. Molecular sexing dilakukan dengan primer 2550F/2718R. Hasil penelitian menunjukan bahwa DNA pada sampel darah kertas saring dan EDTA, baik metode ektraksi kit, alkalin lisis dan PCR langsung, berhasil diamplifikasi dengan primer 2550F/2718R yang menghasilkan pita CHD1-Z dengan ukuran 621 bp dan pita CHD1-W dengan ukuran 459 bp. Penelitian ini menunjukan bahwa ekstraksi DNA dengan kit menghasilkan DNA template yang paling efektif untuk molecular sexing Cerek Jawa.

Abstract

Kemampuan burung gosong sebagai satwa yang mampu menghasilkan telur dengan ukuran diatas rata-rata dan ditetaskan oleh alam telah menjadi pemicu adanya eksploitasi tidak terkendali oleh manusia. Keadaan tersebut disebabkan adanya perbedaan cara pandang di tengah masyarakat. Penelitian ini bertujuan untuk mengetahui pengetahuan dan sikap masyarakat di Kecamatan Seram Utara Barat terhadap keberadaan burung gosong. Penelitian dilakukan dalam 2 tahap, yaitu Juli 2017 dan Agustus 2018 di dua desa utama (Labuan dan Pasanea) dan disertai observasi lokasi bertelur di beberapa gugusan pulau setempat (Pulau Tujuh). Hasil penelitian menunjukkan para pengumpul telur memiliki pengetahuan yang lebih lengkap tentang karateristik burung gosong dibandingkan masyarakat biasa. Pengetahuan tersebut kemudian dimanifestasikan dalam beberapa sikap konservasi untuk melindungi habitat dan populasi burung gosong, yaitu penggalian telur secara manual, rekonstruksi sarang bertelur, pelepasan anak burung, evaluasi kondisi telur, dan reinkubasi telur bertunas. Sikap tersebut belum mampu mengendalikan tingkat eksploitasi diantara masyarakat sehingga penting untuk dirumuskan program konservasi dan aturan hukum setempat yang sifatnya mengikat.
