Abstract

ABSTRACTA study of optimization of red cabbage (Brassica oleracea.L. var. capitata f. rubra) gel extract formula has been performed by using Simplex Lattice Design (SLD) method and antioxidant activity of the formula gel was also evaluated by using in vitro method.The red cabbage was extracted by soxhletation by using ethanol 96% followed by optimization of red cabbage extract in antioxidant gel preparation used SLD method by Design-Expert® software version 7 (DX7) and determination of its IC50 used UV-Spectrophotometry. The stability of optimum gel formula is seen through comparison of physycal properties at the beginning and after four weeks storage used ANOVA, with a 95% significant level. Optimum gel formula of red cabbage extract obtained in the proportion of Metolose 3,883%, propilen glikol 13.5%, Tween 80 1.117%. The evaluation results of optimum gel formula of red cabbage extract is the surface area of gel dispersive of 38.99 ± 3.27cm2; viscosity gel dPa.s of 295.56 ± 1.93 and viscosity change 3.89 ± 0.96%. From the results of statistical analysis of one t-test sample was concluded that there was no difference between the prediction price of software with the observation result (p> 0.05). The IC50 test result of the optimum formula of red cabbage extract gel was 257.25 ± 0.35 µg / mL. The testing result of the physical stability of the optimum formula of red cabbage extract gel suffered a pH decrease after 4 weeks of storage (p <0.05).

Abstract

Green paprica (Capsicum annum L.) is one of fruits which have contain–carotene, vitamin E and vitamin C, which have antioxidant activity. The aim of thisresearch is to know an antioxidant capacity of green paprica. An analysis of theantioxidant activity of this research used radical scavenging DPPH(1,1–difenil–2–pikrilhidrazil) method. The sample was preparated with maceration for4 days using methanol as the solvent. Crude extract was evaluated qualitatively using0,4 mM DPPH reagent. This result of qualitative analysis showed that green papricahave an antioxidant activity. Then, absorbance was measured using UV–Visspectrophotometer at wavelength of 517 nm. The result of this analysis obtained EC50value of green paprica was 0,3399 ± 0,01408 mg/ml.

Abstract

Stroke murupakan keadaan gawat darurat yang memerlukan penanganan dengan cepat dan tepat di IGD, namun kenyataannya sering dilaporkan kejadian medication error (ME) di IGD. Keterbatasan waktu, kondisi klinis pasien dan kondisi Instalasi Gawat Darurat (IGD) merupakan faktor yang diduga berkaitan dengan peningkatan kejadian ME di IGD. Belum diketahui dengan pasti berapa kejadian ME pasien stroke di IGD rumah sakit X dengan keterbatasan sarana prasarana. Penelitian ini bertujuan menentukan gambaran kejadian ME di IGD RS daerah dengan keterbatasan sarana dan prasarana di rumah sakit X.Telah dilakukan penelitian observasional deskriptif dengan pengambilan data pasien secara prospektif pada bulan Desember 2014 – April 2015 di IGD Rumah Sakit X di Yogyakarta. Didapatkan sebanyak 106 pasien dengan kriteria inklusi subjek adalah pasien dengan diagnosis stroke non hemoragik maupun stroke hemoragik, baik dengan penyakit penyerta maupun tanpa penyakit penyerta. Dirawat dalam ruangan IGD di Rumah Sakit X di Yogyakarta pada periode penelitian yaitu selama Desember 2014 – April 2015 dan bersedia terlibat sebagai responden dalam penelitian. Data obat dan riwayat pengobatan diambil dari catatan rekam medis. Peneliti melakukan wawancara dengan pasien atau keluarga pasien untuk melengkapi data riwayat pengobatan. Dari data tersebut dilakukan kajian terhadap kemungkinan terjadinya ME pada fase prescribing dan administrasi (cara pemberian) berdasarkan PERDOSSI. Pengolahan data dilakukan dengan analisa monovariat untuk mengetahui gambaran karakteristik umum pasien (gambaran demografi), karakteristik klinik pasien serta gambaran kejadian, distribusi dan fase ME. Data gambaran kejadian, distribusi dan fase ME disajikan secara deskriptif (persentase).Hasil dari penelitian ini adalah jumlah pasien yang berusia lebih dari 60 tahun  sebanyak 67 pasien (63,2%). Sebanyak 85,8% pasien didiagnosis  stroke non hemoragik dan sisanya sebanyak 14,2%  didiagnosis stroke hemoragik. ME terjadi pada 104 pasien (98,1%). Distribusi kejadian ME pada fase prescribing dan administrasi per pasien adalah 28 pasien mengalami satu kejadian ME, 48 pasien mengalami dua kejadian ME, Sebanyak 20 pasien yang mengalami tiga kejadian ME, 8 pasien mengalami empat kejadian ME. Total kejadian medication error yang terjadi sebanyak 216 dari 106 pasien dengan nilai rata-rata 2 kejadian error per pasien. Medication error di fase prescribing sebanyak 98,11% dan medication error di fase administrasi sebanyak 61,32%.Kata kunci : Stroke, geriatri, medication error, prescribing, administrasi.

Abstract

Gastrointestinal surgery is a surgery that has a high risk of the incidence of surgical wound infection (ILO).Surgical wound infection is the result of bacterial contamination when the operation happens. The purpose of thisstudy was to determine the pattern of post operative antibiotics usage in patients undergone gastrointestinalsurgery at PKU Muhammadiyah Hospital in Bantul. The result of this study was expected to be useful as a sourceof information on patterns of post operative antibiotics that used, as well as for a base for next studies. This studywas a descriptive non-experimental research that prospectively held at PKU Muhammadiyah Bantul YogyakartaHospital. Subjects for this study were all patients who had undergone gastrointestinal surgery, which hospitalizedat PKU Muhammadiyah Bantul Yogyakarta Hospital and met the inclusion criteria. As for the inclusion criteria inthis study i.e., patients who have undergone gastrointestinal surgery, patients undergo hospitalization, as well aspatients that use antibiotics for ILO's prevention. The data analyzed was data of patterns on antibiotics usage thatused to prevent ILO incident. Based on this study showed that the usage of the most widely used post operativeantibiotics is a combination of Ceftriaxon and Cefadroxil as many as 28 patients (47,46%). Intravenous antibioticsthat mostly used is Ceftriaxon as many as 43 patients (6,29%), whereas for the most of oral antibiotics isCefradoxil as many as 45 patients (76,27%).

Abstract

Food product forgery begins to become a new problem in Indonesia, the main problem is there is a concern that food product is mixed with forbidden meat, one of may is rat meat in meatball.  This study is expected to become one basic in order to meet meatball authenticity guarantee.Study about the utilization of infrared spectrophotometry Fourier Transform (FTIR) which was combined with chemometric was carried out to identified rat meat in meatball quantitative and qualitatively. Concentration variation of beef and rat meat in producing meatball is 0%, 25%, 35%, 65%, 75% and 100% rat meat in meatball dough and 5 samples of market meatball. Beef and rat fat which were contained in meatball was extracted by Soxhlet in temperature ≤ 70oC for 7 hours with n-hexane solvent.Qualitative and quantitative analysis was carried out in wave range 750-1600 cm-1 by using partial least square (PLS) multivariate analysis technique and principal component analysis (PCA). The result of calibration with y = 0,9720x + 1,580, coefficient of determination (R2) = 0,9941, and root mean square error of calibration (RMSEC) value was 1.63%. Validation model with, mean root mean square error of cross validation (RMSECV) value was 1.79%, and root mean square error of prediction (RMSEP) value was 2.60%. Using Principal Component Analysis (PCA), grouping of cow fat and mouse fat in the meatballs successfully.

Abstract

Essential oil of sere leaf has been proven as repellant against mosquitoes. This research aims to know about the influence of concentration sere essential oil (Cymbopogon citratus (DC) Stapf) in a vanishing cream bases to physical properties and repellant testing against female Aedes aegypti mosquitoes. Essential oil of sere (Cymbopogon citratus (DC) Stapf) was isolated with steam water distillation and was evaluated the organoleptic appearance and refraction index. Furthermore, the essential oils were formulated in a vanishing cream base with the different formulas, contain formula I: 20% v/w; formula II: 30% v/ w and formula III: 40% v / w. Then this three formulas were evaluated for physical properties including adhesive, dispersive and the repellent activity testing against female Aedes aegypti mosquitoes amount of twenty mosquitoes per cage while seven female volunteers as the probandus. The data have been analyzed using Kolmogorov-Smirnov test and t-test (Main-Whitney), with 95% signifancy level. The results were obtained that the adhesion of formula I: 0.90 ± 0.029 seconds, formula II: 0.60 ± 0.017 seconds, formula III: 0.46 ± 0.040 seconds, while the dispersion of formula I is 64.89 ± 4.559 cm2 , the formula II is 64.71 ± 4.600 cm2 and the formula III is 54.96 ± 14.783 cm2 . The longest time repellent was formula III showed 3014.2 ± 20.163 seconds or 50.23 minutes. It was concluded that the increase of concentration essential oils of sere leaf followed by the increase of repellant activity.

Abstract

Dexamethasone as glucocorticoids has been widely used to reduce inflammation and tissue damage in a variety of conditions. It was commonly prescribed in combination with the other compounds such as chlorpheniramine maleate for the relief of asthma. It was important to observe the quality and safety of the combination dosage form, called pulveres or puyer in order to serve the patient oriented medication. This research aimed to develop analytical method validation and determine the content uniformity of the divided powder containing dexamethasone compounded by pharmacy in Yogyakarta and hospital in Central Java, Indonesia. A validated high performance liquid chromatography (HPLC) method was chosen to determine dexamethasone in the samples. The column of ACE 5 C18 (250 x 4.6 mm) was used in this research and the methanol:water (65:35) was used as the mobile phase at the flow rate of 1 mL/min. The UV detection was performed at 239 nm. Analytical method validation was performed on the parameters of system suitability, limit of detection, limit of quantification, accuracy, precision, and linearity. The results showed that dexamethasone contains in divided powder compounded by pharmacy in Yogyakarta and hospital in Central Java were met the acceptance requirement.

Abstract

The purpose of this research is to determine the total phenolic content of Hibiscussabdariffa calyx in variations of growing area. Red Rosell calyxs were collected fromGlagah, Kediri, and Samigaluh. Phenolic compounds of Hibiscus sabdariffa calyx wereextracted using maceration method with methanol. Total phenolics content weredetermined using visible spectrophotometry method with Folin Ciocalteau reagent. Theprinciple of this method is the formation of blue complex compound fromphospomolybdate-phosphotungstate reduced by phenolic compound in the basiccondition, which can be measured by spectrophotometry. Galic acid was used ascomperator in this research. Total phenolic content in red calyx Glagah, Kediri andSamigaluh were respectively 1.40 g GAE/100 g extract, SD 0.06 (n=12), 1.41 gGAE/100 g extract, SD 0.07 (n=12) dan 2.12 g GAE/100 g extract, SD 0.05 (n=15).Based on this results it could be concluded that growing area affected total phenoliccontent in the methanol extract of red calyx Rosell.

Abstract

Ganyong rhizome (Canna edulis Ker.) empirically use for treating gastritis (peptic ulcers). Ganyong rhizome contains carbohydrates and flavonoid that can reduce inflammation in the gastric tissues and againsts free radicals that are contributing to the pathogenesis of peptic ulcers. This research aims to know the gastroprotective effect of ganyong rhizome on white female rats, Wistar strain, which were given alcohol (ethanol 96%) by dose 1 ml/200gBB and measuring the total flavonoids content in ganyong rhizome. This research is Post Test Only Control Group Design, using 30 rats, weight 150-200 g, age 6-8 weeks, health condition. It is divided into six groups. Group I (normal) were fed and watered only, Group II (control) were given a CMC-Na 0,5%, Group III-V were given a suspension of ganyong rhizome starch (SGRS) with each doses 250, 500, and 1000 mg/kgBB, then Group VI were given a sucralfate dose 360 mg/kgBB. Rats were given oral treatment for 14 days. An hour after the treatment on 14th day, the whole groups unless the group I were given ethanol 96% 1 ml/200gBB orally. Twenty four hours later the rats were sacrificed, dissected and taken its gastric for further analyzed the number of ulcers as well as the description of histopathology of the gastric. The results showed that ganyong rhizome starch can decrease the index ulcer and increase the protection ratio value to the control group with results of doses SGRS 250, 500, 1000 mg/kgBB sequentially i.e., 12,5; 50; and 75%. The description of histopathology also showed significant improvement of gastric tissue at a dose of 1000 mg/kgBB. In addition, on TLC-Densitometri test also obtained the total flavonoid compound in ganyong rhizome is 112,49 ± 7,97 ppm. Conclusions of this research are that the ganyong rhizome positive contains flavonoid and it has gastroprotective effect by decreasing the number of index ulcer, increasing the protection ratio value, and improving the histopathology of gastric tissue which had given alcohol.

Abstract

High Performance Liquid Chromatography (HPLC) was one of analytical methodscommonly used to determine the concentration of epigallocatechin gallate (EGCG) on green teaextract. The method must be validated in order to fit to its purpose. The aim of this research was toprove that the used method has selectifity, liniearity, precise, accurate and know limit of detection(LOD) and limit of quantification (LOQ) is acceptable. The selectivity of analytical method wasdetermined by calculating the resolution value between two peak. Data from 10 μg/mL and100 μg/mL with 5 replicates would give precition and accuration. Precition was known from CV value and accuration was known from recovery value in each concentration. Liniearity was knownfrom regression linear between concentration and wide area of peak. From regresion linear couldcalculate LOD and LOQ. Research show that method of analyse have selectificity withRs= 2.27>1.5; liniearity with r= 0.99; precision with CV 8.74% at concentration 200 µg/mL and3.74% at concentration 500 µg/mL; accuration with recovery 99.76% at concentration 200 µg/mLand 100.52% at concentration 500 µg/mL and the value of LOD is 33.28 μg/mL and LOQ is110.93 μg/mL.
