Abstract

This study was designed to evaluate the hepatoprotective effect of the powder Eurycomalongifolia Jack. From the activity level of SGPT-SGOT on CCl4-induced in male rats. Twentyfive male rats (150-250 g) divide in to 5 groups. Group I treated with aquadest was kept asnormal, group II treated with a single dose of CCl4 (1 ml/ kg BW i.p), group III and IV weretreated with Eurycoma longifolia Jack. (100 mg/kg BW and 200 mg/kg BW p.o) respectivelyand CCl4 (1 ml/kg BW i.p), group V treated with a single dose of curcumin (100 mg/kg BWp.o) and CCl4 (1 ml/kg BW i.p). Blood was collected from vena porta for determination ofSGPT-SGOT. The study showed the activity level of SGPT from the rats was treated byEurycoma longifolia Jack. 100 mg/kg BW and 200 mg/kg BW, Curcumin, and control groupsare 150.0±5.099 U/L; 113.6±5.508 U/L; 60.5±2.887 U/L; and 129.0±6.055 U/L respectively. Mean while the activity level of SGOT from the rats was treated by Eurycoma longifolia Jack.100 mg/ kg BW and 200 mg/ kg BW, Curcumin, and control groups are 369.4±11.165;263.0±1.803; 194.5±7.448; and 451.5±16.759 U/L respectively. The Eurycoma longifoliaJack. powder and Curcumin significantly (p < 0.05) declines two enzymes (SGPT and SGOT)than control group. The results concluded that Eurycoma longifolia Jack. powder hashepatoprotective effect.

Abstract

(Andrographis paniculata ( Burm.f. ) Ness ) is containing andrografolid, neoandrografolid,and flavonoids known as apigenin-7,4’-di-O-methyl ether, 5-hydroxy7,8,2’,3’tetrametoxyflavon,dihydroxy-dimetoxyflavon. Flavonoids as antioxidants, have potential as an anti-bacterial activity,anti-inflammatory, anti-allergic and anti-thrombosis. The chemical structure of the drug may explainthe nature of the drug and linkage units and structures of drug molecules with biological activity.Compounds with the same functional groups can provide the same biological response due to work onthe same receptors. This present study is determine the total flavonoid content of the ethanol extract ofA. paniculata use Chang method, identification using spectrophotometric method. The measurementresults showed that total flavonoid content of the ethanol extract of A. paniculata amounted to 46.322g/kg.

Abstract

Schizoprenia is one of psychiatric disorders that caused by brain damage. One of schizoprenia handling is giving antipsychotics to prevent the symptoms, that antipsychotics was effective therapy to treat it. This study aims to determine the treatment pattern of schizoprenia patient behind reconciliation program at Puskesmas Mungkid. The method used is analytical survey with retrospective data collection. This research were conducted on 42 medical records of patients in period January-June 2014 which receive antipsychotics medication and were analyzed with descriptive analysis. Obtained data of this research, showed that the most common single therapy of for antipsychotics is haloperidol (54,93%) and the most common therapy was haloperidol and chlopromazine (61,91%). The most common treatment category of schizoprenia is the typical antipsychotic treatmemt (92,96%), while for appropriate dose and the use of antipsychotic drugs were used 97,18% and 2,82% were not appropriate to the standar.

Abstract

Metabolic syndrome causes an imbalance of the immune system and increased levels of HbA1c and IL-17A expression. Black cumin seed oil (BCSO) is known to have antioxidant and immunomodulatory  properties. This study reports the effects of dosage variation in BCSO (1.5 and 3 ml per day for 20 days) on HbA1c levels and IL-17A expression in patients at risk of metabolic syndrome at Jetis 1 Public Health Center in Bantul, Yogyakarta. It employed a crossover design in which a total of 66 patients at risk of metabolic syndrome were divided into two groups receiving a sequence of different treatments. Group 1 (N=33) received treatment A first, which was BCSO at a dose of 1.5 ml/day for 20 days. Then, after a washout period of 7 days, it received treatment B, 3 ml of BCSO per day for 20 days. Group 2 followed the same procedure only vice versa, treatment B, then A. The HbA1c levels were measured by the mean plasma glucose (MPG) method, while the IL-17A expression was detected by flow cytometry. The average HbA1c level and IL-17A expression of the treatment groups were statistically analyzed with 95% confidence level. In response to the treatment regime, the HbA1c level of group 1 was 7.34 ± 2.51% (a decrease), and that of group 2 was 7.72 ± 2.44% (an increase). The IL-17 expression in group 1 was 3.74 ± 3.52% (a decrease), and the one in group 2 was 4.07 ± 3.65% (an increase). The effects of administering 1.5 ml and 3 ml BCSO per day for 20 consecutive days on Hb1c level (p=0.17) and IL-17A expression (p=0.67) were not significantly different. Most patients experience a decrease in HbA1c levels and IL-17A expression after the treatment. Insignificant differences between the two groups mean that at the doses of 1.5 ml/day and 3 ml/day for 20 days, BCSO exhibits the same effects in patients at risk of metabolic syndrome registered at Jetis 1 Public Health Center in Bantul, Yogyakarta.

Abstract

Ethanol extract of Gratophyllum pictum L (G. pictum L). leaves contain phytochemical compounds such as alkaloids, tannins and flavonoids, which have antibacterial activity against Staphylococcus aureus. This study aims to prove the effectiveness of  ethanol extract of G. pictum L. to reduce the level of TNF-α and NO in Swiss mice infected with S. aureus.The study treated 30 Swiss mice divided into 5 groups, consist of: K (-) normal mice ; K (+) mice were infected with S. aureus ; P1, P2, P3 were infected with S. aureus and given the ethanol extract of G. pictum leaves,75,150, and 300 mg/kgBW/day for 7 days . Mean levels of TNF-α of group K(-), K (+), P1, P2, P3 were 300.29; 550.29; 390.34; 517.67; 327.55 pg / mL respectively. One Way ANOVA test showed that there were significant differences between all treatment groups (p = 0.001), LSD Post Hoc test revealed P1 and P3 groups different significantly with K (+) with p= 0.013 and p = 0.001. Mean levels of NO of group K (-), K (+), P1, P2, P3 were 1.32; 3.56; 2.50; 2.64; and 1.56 μM/mL respectively. Kruskal WallisTest showed that the levels of NO not significantly different, although NO levels at 300 mg/kgBW/day similar with to the levels of NO group K(-). Spearman correlation test showed that there were correlations between TNF-α and NO with the variation results on each treatment groups. Ethanol extract of G. pictum leaves at 300 mg/kgBW/day for 7 days is regarded as the effective dose to reduce TNF-α and NO levels in Swiss mice infected by S. aureus.

Abstract

The development of drug delivery system needs a usefull determination method ofdrug in many kind of samples. This research was purposed to develop a determinationmethod of losartan, an angiotension receptor antagonist II, from the sample of in vitrotransdermal transport. Three methods were studied, i.e. normal spectrophotometric, 1stderivative spectrophotometric, and HPLC. The spectrophotometric method wasconducted using Spectrophotometer Shimadzu tipe UV 1700 controlled by UV Probesoftware (Shimadzu), while HPLC method was performanced by Shimadzu HPLCcontrolled by LC Solution software (Shimadzu). Lichrospher RP 18 250-4 (5 µm) wasused as stationary phase and acetonitril-acetic buffer 0,01 M pH 4 (60:40) was used asmobile phase. Chromatogram was recorded using UV 223 nm and 254 nm as adetector. Some parameters of determination method performance were calculated, i.e.LOD, LOQ, recovery, systemic error, and random error. The results shown thatspectrofotometric methods did not have an enough performance parameters to use intransdermal transport of losartan, while HPLC method had LOD and LOQ 27,329 and91,098 ng/ml using UV 223 nm as detector and 36,178 and 120,590 ng/ml using UV 254nm as detector. Detector UV 223 nm was selected. This HPLC method had recovery106,405% and random error 3,71%, and could be used to determine the losartanconcentration in sample from transdermal transport in vitro.

Abstract

Penggunaan  obat tanpa resep dalam upaya swamedikasi telah dilakukan secara luas oleh masyarakat untuk mengobati berbagai kondisi penyakit ringan. Obat-obat yang sering digunakan dalam swamedikasi pada umumnya termasuk ke dalam golongan obat tanpa resep. Perilaku masyarakat daam swamedikasi dipengaruhi beberapa hal salah satunya kemudahan mengakses berbagai informasi mengenai obat, dan juga merupakan salah satu pertimbangan konsumen dalam pemilihan obat. . Perkembangan konsep “Pelayanan Kefarmasian” berarti Apoteker secara langsung bertanggung jawab pada pasien dalam peningkatkan mutu pelayanan sehingga Apoteker memiliki kewajiban dalam pemberian informasi yang benar terkait penggunaan obat-obat tanpa resep. Penelitian ini bertujuan untuk mengetahui pendapat konsumen apotek mengenaikonseling obat tandap resep dan gambaran pelaksanaan swamedikasi di wilayah bantul. Rancangan penelitian deskriptif dengan metode penelitian survei secara langsung menggunakan kuesioner dan lembar observasi. Penentuan sampel apotek dan pasien dilakukan menggunakan teknik purposive sampling. Hasil dari penelitian ini menunjukkan bahwa tiga golongan obat yang paling banyak dibeli dalam pelaksanaan swamedikasi adalah analgesik antipiretik (28,8%), vitamin/suplemen (19,3%), dan obat batuk pilek (15,1%). Sebagian besar konsumen telah mengetahui aturan pemakaian obat (71%0, dan Apoteker merupakan faktor pertimbangan dalam pemilihan obat (34%). Sebanyak 95,7% konsumen mengaku membaca label obat pada saat pertama kali pembelian (95,7%). Pendapat konsumen mengenai konseling yaitu bahwa sebagian besar memerlukan adanya konseling obat tanpa resep (89%), sebanyak 24,8% pernah mendapatkan konseling obat tanpa resep dengan durasi konseling 1-5 menit. Kesimpulan dari penelitian ini adalah Gambaran pelaksanaan swamedikasi obat tanpa resep di Wilayah Bantul menunjukkan bahwa golongan obat tanpa resep yang paling banyak dibeli adalah analgesik antipiretik. Pelaksanaan swamedikasi obat tanpa resep di wilayah Bantul sudah dilakukan dengan baik, ditunjukkan dengan sebagian besar konsumen pernah mendapatkan konseling dengan durasi 1-5 menit. Konsumen juga merasa perlu mendapatkan konseling obat tanpa resep oleh Apoteker.

Abstract

The extract quality can be affected by the components and concentration of chemical compounds containedin the extract.Type and quantities of chemical compounds that are dissolved in the extraction process is stronglyinfluenced by the solvent extraction. The aim of this study is to determine the effect of concentration of ethanol assolvent extraction on naphthoquinone content in Henna leaves extract. The extraction method was done bymaceration for one hours with water, ethanol 50% , 70% , 90% and 95% v/v solvens of the dried Henna leavespowder. Each macerate was made 5 % w/v concentration and than was spotted 5 µL on plate of silica gel F 254 andeluted with mixture of chloroform - methanol (17:3 v/v) as mobile phase. The spots on chromathogram werescanning and measured the area under curve with TLC densitometer at maximum wave lenght. The results showedthat spot at Rf 0.31 had similar spectra with naphthoquinone standard and same maximum wavelength at 279 nm.The result of solvent optimization indicated that the best solvent extraction for Henna leaves was ethanol 50% v/vwith the highest naphthoquinone contentat 1.43 ± 0.1942% w/v.

Abstract

Elephantopus scaber Linn had been used as traditional medicine. It contain some lacton sesquiterpene with antiproliferative activity. The study aim was to explore the effect of chloroform fraction of ethanolic extract of E.scaber on the expression of p53 on T47D cell line. The ethanol extract was found by maceration method. The extract was then fractionated using petroleum ether and chloroform respectively to get the chloroform fraction. The expression of p53 was observed by immunocytochemistry method with antip53 antibody. The p53 cell line was cultured and treated with fraction with concentration of 7.06 and 3.53 µg/ml. The expression of p53 was showed by brown colour in the nucleus of the cell. The result showed that chloroform fraction treatment with concentration of 7.06 µg/ml induced apoptotic cell death in the end stage (necrosis). While the treatment with lower concentration 3.53 µg/ml could increase the p53 expression.

Abstract

Cancer is the formation of new tissue which is abnormal and malignant. A group of cells suddenly become disorganized and reduplicate themselves rigorously (hyperproliferation). Nigella sativa L. is one of the herbs which have an anticancer effect. This research aims to assess the cytotoxic and antiproliferative effect of Nigella sativa L. ethanol extract of Myeloma cells. Ethanolic extract was produced from Nigella sativa L. powder with maseration method. The cytotoxicity test was done by incubating Myeloma cells with the treatment concentration group of N. sativa L. ethyl acetic fraction of ethanolic extract 2000; 1000; 500; 250; and 62.5 µg/ml, respectively. The test was done with an MTT method and then with a calculation of its death percentage. The LC50 is calculated using a probit analysis method. The test was then continued with the antiproliferative test to assess the doubling time at treatment concentration 125; 62.5 µg/ml and cellular control at hours 24, 48, and 72. The results showed that Nigella sativa L. ethanolic extract had cytotoxic activity towards the Mieloma cells with an LC50 value 177.01 µg/ml. The antiproliferative test showed that there was a growth inhibition, even cell death at the extract treatments. The doubling time was 253 hours at 62.5 µg/ml concentration, 298.4 hours at 125 ug/ml, while the cell control had 54.52 hours
