Abstract

Medical masks play an important role in minimizing infection risk; however, they may also act as reservoirs for bacterial contamination. Mask wear duration is a key factor influencing microbial accumulation. While previous studies have focused on healthcare, industrial, or community settings, limited attention has been given to campus environments, where prolonged mask use is common among students and staff. This study aimed to evaluate bacterial contamination on disposable medical masks across different wear durations in a campus setting. This study involved 10 student who wore disposable medical masks for 0 (control), 3, and 6 hours under campus conditions. Bacterial contamination was assessed using the Plate Count Agar (PCA) method following serial dilution, and results were expressed as CFU/cm². Data were analyzed using using the Kruskal–Wallis test. The results showed a progressive increase in bacterial contamination with longer mask wear duration, from 0 hours (0 ± 0 CFU/cm²), to 3 hours (34.8 ± 28.04 CFU/cm²), and sharply at 6 hours (7297.59 ± 9552.08 CFU/cm²). Significant differences were observed between wear durations. In conclusion, extended mask wear duration may increase the risk of bacterial accumulation; therefore, appropriate mask replacement practices are recommended to support personal hygiene and health protection

Abstract

This study investigated the temporal dynamics of sweet potato rhizosphere microbial communities across different growth phases to understand their functional metabolic shifts, which is critical for optimizing sustainable soil management. Rhizosphere soil samples were collected monthly (1 to 5 months) during the crop growth period. Community-Level Physiological Profiling (CLPP) was conducted using the Biolog EcoPlate approach combined with quantitative spectrophotometry analysis, with data evaluated through Average Well Color Development (AWCD), Shannon diversity (H’), and Evenness (E) indices. The results revealed a distict shift in microbial metabolic activity, which peaked at 2 months and gradually declined toward the harvesting phase at 5 months. Ecologically, the substrate utilization pattens shifted dynamically over time: early-stage microbial communities (1-2 months) predominantly utilized carbohydrates and polymers, reflecting the high energy demand and typical root exudates of young plants. Conversely, late-stage communities (3-5 months) transitioned toward amines and polymers, indicating an adaptation to more complex organic matter as the plant matured. This study demonstrates that sweet potato growth stages strongly drive rhizosphere microbial functional diversity. These insights offer an agricultural blueprint for the precise timing of biofertilizer applications aligned with the plant's natural metabolic windows.

Abstract

This study aims to morphologically distinguish the two closely related loaches, N. chrysolaimos and N. fasciatus using an integrated approach that combines allometrically-adjusted morphometrics measurements, meristic counts, and observable morphological features. Loaches samples were collected from six river systems representing the Progo, Serayu, and Bengawan Solo watersheds in Central Java and the Special Region of Yogyakarta. Morphometric measurements were adjusted using Elliott formulation to reduce allometric bias prior to Principal Component Analysis (PCA). The value of PC1 and PC2 accounting for 42.31%. The results shows that. N. chrysolaimos showed greater caudal peduncle depth and dorsal fin length, whereas N. fasciatus was more strongly associated with dorsal fin height and caudal fin length. Meristic characters provided additional evidence, with the examined specimens of N. chrysolaimos showing higher than N. fasciatus for lateral-line scales (198 vs 156), transverse scale rows (14 vs 10), preorsal scales (57 vs 39), and circumpeduncular scales (27 vs 18), as well as dorsal (10 vs 9) and pecotral (9 vs 7) soft raysi.e. body color pattern, mouth shape and barbel orientation  also distinguished the species. These findings demonstrate that combined morphometric, meristic, and morphological characters provide reliable diagnostic evidence for distinguishing between the two species in Java.

Abstract

This study aims to sequence and characterize the complete mitochondrial genome of the Javan ferret-badger and to contruct its phylogenetic position within the family Mustelidae. The gDNA from the femoral muscle was extracted and subjected to Illumina whole-genome sequencing. The resulting sequence achieved a mean genome sequencing depth of approximately 1,545×. The mitochondrial genome was assembled using SPAdes v3.15.5 and annotated using MitoZ v3.4 and MITOS2. Phylogenetic relationships were analyzed against nine mustelid mitogenomes using Maximum Likelihood analysis under the GTR model with 1,000 bootstrap replicates. The circular mitogenome lenght of M. orientalis is 16,274 bp consisted of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a control region. The results showed that the mitogenome is compact with short intergenic spacers, conserved gene overlaps, and an overall GC content of 45.0%. Relative Synonymous Codon Usage analysis indicated an uneven codon distribution, with a general preference for A/T-ending codons. Mitogenomic data were used to examine the phylogenetic position of M. orientalis within Mustelidae. The analysis reveals the phylogeny of M. orientalis within Melogale supports the monophyly of the genus. These findings provide a valuable genomic data for further systematic, phylogeographic, population genetic, and conservation studies.

Abstract

Plastic waste remains a major environmental concern worldwide. Approximately 418,262 tons of plastic waste, accounting for 20% of the total waste accumulation, are deposited annually at the Pasir Bajing Landfill (TPA) in Garut, Indonesia. Under these conditions, plastic serves both as a selective substrate for certain microorganisms and as a potential carbon source. This study aimed to identify bacteria associated with plastic waste at the Pasir Bajing Landfill using molecular techniques. Polystyrene waste samples were collected from three different sampling sites within the landfill area. Phylogenetic analysis based on 16S rRNA gene sequences revealed three bacterial isolates: isolate 2 was identified as Acinetobacter baumannii, isolate 5 as Atlantibacter hermannii, and isolate 6 as a member of the family Enterobacteriaceae. These isolates belong to the phylum Proteobacteria and may possess the potential to degrade plastic materials. Further studies are required to evaluate their plastic-degrading capabilities and to elucidate the metabolic pathways involved in the biodegradation process.

Abstract

Local rice cultivars are traditionally developed and maintained through farmer-based agricultural practices. Identifying traits associated with drought tolerance in local rice germplasm is essential for the development of high-yielding, drought-resistant varieties. Therefore, this study aimed to evaluate the drought tolerance characteristics of local rice cultivars from West Java, Indonesia. Fifteen local rice cultivars were collected from three locations: Tanjungsari District (Sumedang Regency), Ciletuh District (Sukabumi Regency), and Pandeglang Regency. The experiment was arranged in a randomized block design with two treatment factors and three replications. The ten local rice cultivars evaluated exhibited different responses to variations in soil water content relative to field capacity. Based on the Drought Sensitivity Index (DSI), five cultivars were classified as sensitive, four as moderately tolerant, and one as tolerant to drought stress. Under optimal water availability, the Ampera, NN Banten, and Rancung cultivars demonstrated high grain yield potential. However, the Rancung cultivar was found to be susceptible to drought stress. In contrast, the Huma Hideung Ciletuh cultivar was classified as drought-tolerant because it showed only minor reductions in grain weight and grain number under drought conditions compared with optimal water availability.
