Abstract

Research of prediction parameter genetics in cacao plants at the nursery fase important asinformation on cacao plants that is resistant to attack Phytoptora palmivora and cancharacterized of morphology of that plants. The goal of this research is to know the value ofthought of the parameters of genetics on growth and resistance characters to black poddisease for some of Aceh’s origin genotypes. The research is done in two packages, namelythe growth and resistance plants packages. Design used are thought of random complete(ral)with ten genotypes as treatments and repeated as many as three times. Variables enduranceare qualitative variable (the form of stem of plant, a leaf shape, form the base of a leaf, theform of leaf tips, and flush color) quantitative variable (high plants, long leaves, number ofleaves per stem, wide leaves, and the angle between the leaves with stem), and resistancevariable (the incubation, the percentage of disease, and severity of illness). The result showthat: genotype categorized somewhat endurance element is AT7 and SR45.

Abstract

This article is a dynamics synthesis of milk production data of dairy goat fed diet containingconcentrate supplemented with PUFA (polyunsaturated fatty acid), yeast and curcuma. Twenty (20) dairy goats of Ettawa crossbred in late lactation phage were grouped into fiveof four goats to receive five treatments in in Randomized Complete Block design. Thetreatments were no supplement (PD0), 3 tablets of Asifit (PDA), 5g/d yeast (PDY), 20g/dcurcuma (PDC), and a mix of 5g/d yeast and 20g/d curcuma (PDM). The concentratecontained of PUFA sources (roasted ground corn, roasted soy bean meal, and corn oil),soybean by-product; roughage provided was King Grass. Milk yield was recorded daily, pre,treatment, and post then calculated all the way from pre to post treatment. Milk yield werehigher significantly (P<0.05) in PDA (538.7 g/d) and PDY (522.3g/d) compared to PD0(336.3g/d), PDC (322.3g/d) and PDM (392.0g/d). Average temperature and relativehumidity in the farm were 26.94 ± 2.280 C and 78.83 ± 7.34 %. Yeast supplementation eithersolely or in combination with curcuma is considered better in milk production dynamics inpre, treatment, post.

Abstract

Banana is a climacteric fruit, it produces enough ethylene bringing about speedy changes inphysico-chemical characteristics, including colour, texture, aroma, chemical composition,respiration rate and senescence. Ethylene, a colourless gas, is the main regulator of ripeningin climacteric fruits which leads to the short marketable life and increase postharvest losses.These losses can be reduced by adopting various postharvest management practices that arecurrently in practice all over the world. One of the most effective ethylene removals ispotassium permanganate. However, the usage of non-commercial charcoal from agriculturalwastes that capable to remove or absorb ethylene such as palm kernel shell (PKS), sugarcanebagasse (SB) and coconut shell (CS) is scarce. The treatments were i) control (withoutethylene absorbent), ii) 25g PKS, iii) 25g CS, and iv) 50g SB charcoal with 3 replications. Thepostharvest parameters assessment were internal ethylene production, fruit colour indices,firmness, titratable acidity (TA), soluble solids concentration (SSC), weight loss and starchpattern index (SPI). Internal ethylene production was assessed on daily basis. Meanwhile,other quality parameters were assessed on every four days interval. PKS had the ability todelay the climacteric peak of the ethylene production and prolong the shelf life of Beranganbanana, without significant reduction in its postharvest quality. In addition, PKS charcoalwere light, easy to obtain and cheaper ethylene adsorbent. In contrast, SB charcoal was lesseffective in delaying the ripening of Berangan banana. The shelf life of Berangan banana canbe extended up to 6 days with no apparent changes in colour and weight loss.

Abstract

Citatah karst area is the oldest kars in West Java that located in District Cipatat West Bandung Regency. Karst area in Cipatat District Gunung Masigit Village has 1,053 Ha area, one of the areas that have a potential of cultural heritage and geological reserves. Mining activities conducted in the kars area Citatah Gunung Masigit Village, in the form of active mining and post mining, resulting in decreased quality of physical, chemical, and biological soil fertility. To solve problems in post-mining areas, governments should be able to replace community livelihoods (such as limestone mining) to agriculture, farming, tourism and the creative economy. Through the efforts of plant revegetation. Several options plan that will be developed based on environmental, social, and economic conditions. Selection of plants for revegetation purposes, will be analyzed using Analytical Hierarcy Process (AHP), while to know the public response to revegetation and soil fertility assessment, will be analyzed descriptively. Based on the results obtained from the community response level, a positive response from the AHP analysis of the agrotourism revegetation program has a relative value of 0.834 states agreement for revegetation program with each positive response given that the most positive response is a conative response of 0.395, continued by cognitive response 0.277, affective response 0.199, while the relative value of the lowest response were shown in the negative response of 0.13. On the type of election plant revegetation, has acquired the plant revegetation well developed based on the relative value of the highest to the lowest of which is; 1). Corn (0.338 the highest crop species to be a revegetation commodity in both of social, economic and environmental conditions, 2). Guava (0,216), 3). Albasiah (0.109), 4). Cassava (0.084), 5). Soybean (0,043), 6) Cucumbers, Bananas, yam each have a relative value (0.042). Albasiah and nuts, can be used as pioneer plants to improve the fertility of the land before being developed in fruit and vegetable crops.

Abstract

Tourism has proven itself to be able to support the economic growth through opportunity for investation, occupation, work, business which accumulated in the prosperity of the people. Since re-open, the effect of the development of Mount Kelud tourism post-eruption aren’t clearly identifiable. Given that reason, the goals of this research are to: 1). Identify the locals farmers’ perception about the development of Mount Kelud tourism post-eruption which include its process and development. 2). Identify the effects of post-eruption Mount Kelud tourism development to the local farmers which include occupation-changes, working hours, per-family income, land price, price of the daily needs, local product values. The research conclude that; 1) local farmers’ perception in Sugihwaras village to the development of the posteruption Mount Kelud tourism are generally positive and they mostly agree with the tourism site’s existence. 2) from the ecenomic aspect, the post-eruption Mount Kelud tourism site has significant effects to the rise of land selling price and type of jobs/occupation and the income of the local farmers who work on the tourism site. But it doesn’t significantly affected the working hours, the price of daily needs and other local products.

Abstract

Sucrose transporter (SUT) is a protein that functions to translocate sucrose from source to sink. Protein activity is determined by the accumulation of sucrose in sugarcane stem. The level of sucrose accumulation is also influenced by the activity of  sucrose phosphat synthase (SPS) and neutral invertase (NI) in conjunction with the synthesis and hydrolysis of sucrose. This aim of this study is to determine the sucrose transporter protein expression in Genetically Modified Product (GMP) of sugarcane.Genetically Modified Product GMP sugarcane events 1, 2, 3, 4, 5, 18, 20 and non GMP were used as materials. The analysis was performed by Western Blot method to observe the presence of SUT protein expression, stem sucrose content by resorcinol method to observe the translocation of sucrose, the SPS enzyme activity and NI that play a role in determining the accumulation of sucrose.The results showed that (1) an increase in SUT protein content in GMP plants events 2 and 18 with a protein band that is thicker than the non GMP plants, (2) SPS and invertase activity at all events GMP crop plants to increase compared to non GMP. Sucrose content of the stem at all events GMP plant to increase with age segment. The older age segments, the higher the sucrose  content. Abstract

This research aimed to explore lipase enzyme from soil through metagenomic approach. The DNA was extracted directly from the topsoil and river sediment by SDS-based lysis method. The soil genomic DNA was sheared into approximately 40-kb fragments The construction of genomic library was carried out by cloning the fragment DNA into  fosmid pCC2FOS vector and host E. coli EPI300-T1R accordance with the protocols of CopyControl™ Fosmid production library kit. Screening of lipase-producing clones was performed based on its lipolytic activity using Luria-Bertani agar plate 12.5 µg/ ml chloramphenicol, 10 µg/ml Rhodamine B and 1 % olive oil as a substrate. The assay plates were incubated at 550 C. The lipolytic activity was identified as an orange around colonies. There were 5 (five) clones of 32 screened-clones that expressed lipolytic activity. Abstract

Surface sterilization is a very important key for reaching a successful explants introduction to the in vitro environment. However, to get the best protocol of explants surface sterilization is not easy. Each explants has its characteristics and the contaminants (which usually are bacteria and fungus) are vary depending on its source. In order to know the best sterilization protocol of Phaleria macrocarpa leaves, some sterilizing agents were tested. This experiment suggests that ethylene bis dithiocarbamate may eradicate fungal infection by metalloenzymes inhibition, which is showed by 91.7% survivals. In the other hand, streptomycin sulfate may eliminate bacterial infection by inhibition of protein synthesis, which is showed by 95.8% to 98.3% survivals depending on the exposure time. Abstract

Sea has benefits to conncet islands in Indonesia, which is one of the natural resources that is very potential to be empowered and can be used as one of the basic capitals of national development. The purpose of this study was to determine the correlation between strategy of management vaname shrimp (Litopenaeus vannamei) and respondents’ ages, to know the difference of strategy of management vaname shrimp (Litopenaeus vannamei) based on respondents’ genders, and to know the difference of strategy of management of vaname shrimp (Litopenaeus vannamei) based on respondents' education levels at Molang Beach Tulungagung. Demographic factors used are ages, genders, and education levels to determine the management strategy of vaname shrimp fishermen at Molang Beach Tulungagung. Molang Beach at Tulungagung was choosen as the research object because it has long beach line and it is managed and used as farming land. The results are the respondents’ ages have a correlation with the strategy that used to manage vaname shrimp, strategy of management vaname shrimp (Litopenaeus vannamei) differs based on genders, and strategy of management vaname shrimp (Litopenaeus vannamei) differs based on levels of education at Molang Beach Tulungagung.

Abstract

Estrogen deficiency can changes the reproductive system especially in menopausal women, such as a decreasing of smooth muscle contraction, a thinning of vaginal structure, epithelial atrophy and a collagen decreasing. The disorders can be overcome by phytoestrogens treatment. Tempeh is a fermented soy product contains isoflavones that has estrogen-like structure with 17β-estradiol so that it can be able to bind estrogen receptors. Ovariectomized animal as animal test model has estrogen deficiency. Therefore, the purpose of this study to determine the effect of tempeh soy flour extract on vagina histological structure of Swiss Webster ovariectomized mice. The method used animals tested by treating 45 female Swiss Webster mice that were divided into five groups: negative control group (normal mice had not ovariectomized was not given tempeh flour extract), positive control (ovariectomized mice was not given tempeh flour extract), the treatment group (ovariectomized mice and was given tempeh flour extract D1 (0.21 g/ml / day), D2 (0.42 g/ml/day), and D3 (0,63 g/ml/day). Soy tempeh flour extract was given for 10, 20, and 30 days by gavage. The histological parameter were observed include tunica mucosa epithelium and tunica muscularis thickness of vagina. The result showed that tempeh flour extract increase epithelium thickness and tunica muscularis of the vagina ovariectomized mice. Treatment by taking longer time and giving higher dosage of tempeh flour extract could increase the ephitelium thickness of tunica mucosa and tunica muscularis vagina. The treatment of 0.63 g/ml/day dosage during 30 days showed the highest thickness average values of epithelium and tunica muscularis. The ephitelium thickness was 33.90 μm and the tunica muscularis dense was 90.20 μm.
