Abstract

b-Xylosidase encoding gene from G. thermoleovorans IT-08 had been expressed in the pHIS1525/ B. megaterium MS941 system. The b-xylosidase gene (xyl) was inserted into plasmid pHIS1525 and propagated in E. coli DH10b. The recombinant plasmid was transformed into B. megaterium MS941 by protoplast transformation. Transformants were selected by growing the recombinant cells on solid LB medium containing tetracycline (10 µg/ ml). The expression of the b-xylosidase gene was assayed by overlaid the recombinant B. megaterium MS941 cell with agar medium containing 0.2% ethylumbelliferyl-b-D-xyloside (MUX). This research showed that the b-xylosidase gene was succesfully sub-cloned in pHIS1525 system and expressed by the recombinant B. megaterium MS941. Theaddition of 0.5% xylose into the culture medium could increase the activity of recombinantactivity of recombinant of recombinantb-xylosidase by 2.74 fold. The recombinant B. megaterium MS941 secreted 75.56% of the expressed b-xylosidase into culture medium. The crude extract b-xylosidase showed the optimum activity at 50° C and pH 6. The recombinant b-xylosidase was purified from culture supernatant by affinity chromatographic method using agarose containing Ni-NTA (Nickel-Nitrilotriacetic acid). The pure b-xylosidase showed a specific activity of 10.06 Unit/mg protein and relative molecular weight ± 58 kDa.

Abstract

This research aimed to explore lipase enzyme from soil through metagenomic approach. The DNA was extracted directly from the topsoil and river sediment by SDS-based lysis method. The soil genomic DNA was sheared into approximately 40-kb fragments The construction of genomic library was carried out by cloning the fragment DNA into  fosmid pCC2FOS vector and host E. coli EPI300-T1R accordance with the protocols of CopyControl™ Fosmid production library kit. Screening of lipase-producing clones was performed based on its lipolytic activity using Luria-Bertani agar plate 12.5 µg/ ml chloramphenicol, 10 µg/ml Rhodamine B and 1 % olive oil as a substrate. The assay plates were incubated at 550 C. The lipolytic activity was identified as an orange around colonies. There were 5 (five) clones of 32 screened-clones that expressed lipolytic activity. Abstract

Penelitian ini dengan tujuan untuk mengetahui kebutuhan informasi staf pengajar dalam melaksanakan kegiatan penelitian, perilaku pencarian informasi staf pengajar dalam melaksanakan kegiatan penelitian serta hambatan-hambatan yang dihadapi oleh staf pengajar dalam melaksanakan kegiatan penelitian. Metode penelitian ini adalah metode kualitatif deskriptif. Analisis yang digunakan dalam penelitian ini adalah analisis isi menyajikan hasil akhir dalam bentuk narasi deskriptif. Hasil dari penelitian ini adalah a) Subjek penelitian sesuai dengan bidang kompetensi, b) Sumber informasi yang dibutuhkan buku, jurnal, surat kabar, internet, dan wawancara dengan narasumber, c) Tahun publikasi yaitu terbitan terbaru lima tahun terakhir, akan tetapi tahun publikasi lama juga diperlukan. d) Lokasi sumber informasi diperoleh di luar perpustakaan Polines maupun lembaga terkait dengan bidang yang diteliti, e) Pola pencarian informasi mengikuti pola David Ellis kemudian ditambah dengan menggunakan pencarian informasi melalui akses internet, f) hambatan yang dihadapi paling dominan adalah hambatan dari faktor lingkungan.Kata Kunci : Kebutuhan Informasi, Perilaku Pencarian Informasi, Hambatan dalam Pencarian Informasi, Informasi

Abstract

Recent development in fermentations for bioethanol production were focused three factors, i.e. abundance and cheap substrates,superior yeast fermenting the substrates, and simultaneous saccharification and fermentation (SSF) technology.Nowadays national and world bioethanol production still depend on sugar cane and starchy materials.This research aims to determinate the optimum simultaneous saccharification and fermentation (SSF) conditions to identify the performance of local strain Saccharomyces cerevisiae recombinant [GLO1] in the producing bioethanol from starch.The optimum conditions for SSF process are in a media composition containing glucose 2% (w/v), starch 5% and at aeration rate 50 rpm.At these optimum conditions Saccharomyces cerevisiae recombinant [GLO1] produce 25.36% (v/v) bioethanol at day-20 of the fermentation process design.

Abstract

ABSTRAK Penelitian bertujuan mengkaji pengaruh pemberian limbah pabrik pakan (pakan ceceran) yang difermentasi dengan starter fungsional (terdiri dari ekstrak limbah sayur fermentasi, cairan rumen, pollard dan akuades disebut juga starfungs) terhadap profil kolesterol pada ayam broiler.Materi yang digunakan adalah 105 ekor day old chick (DOC) ayam broiler, pakan limbah pabrik pakan, pakan komersial BR11 dan ‘starfungs’. Penelitian dilakukan menggunakan Rancangan Acak Lengkap (RAL) yaitu 3 perlakuan dengan 7 ulangan.Perlakuan  yang digunakan yaitu T0 = pakan komersil, T1 = pakan tanpa fermentasi dan T2 = pakan fermentasi. Parameter yang diamati meliputi: total kolesterol, low density lipoprotein (LDL) dan high density lipoprotein (HDL) darah ayam broiler. Analisis data yang digunakan adalah analisis ragam dan uji jarak berganda Duncan (UJBD).Hasil penelitian menunjukkan bahwa tidak ada pengaruh nyata pemberian pakan fermentasi limbah pabrik pakan terhadap kadar kolesterol, LDL dan HDL darah. Berdasarkan hasil penelitian dapat disimpulkan bahwa pemberian limbah pabrik pakan yang difermentasi dengan ‘starfungs’ tidak memberikan pengaruh terhadap profil kolesterol darah ayam broiler dilihat dari kadar kolesterol, LDL dan HDL.Kata Kunci: limbah pabrik pakan fermentasi (pakan ceceran); ayam broiler; kolesterol; LDL dan HDL ABSTRACT The research was aimed to study the effect of waste poultry feeds (scattered feed) that was fermented with fungsional starter (that contain extarct of fermented vegetable waste, rumen fluid, pollard and aquades also called starfungs) on cholesterol profile in broiler chickens. The material used was 105 day old broiler chick (DOC), waste poultry feeds, commercial feed BR11 and 'starfungs'. A completely randomized design (CRD) was applied with 3 treatments and 7 replications of each treatment. The treatments measured were commercial feed (T0), feed unfermented (T1) and fermented feed (T2). The parameters observed were: total cholesterol, low density lipoprotein (LDL) and high density lipoprotein (HDL) on broiler blood. Analysis of variance and Duncan multiple range test (DMRT) were used to analysis data. The results of the research showed that there was no significant effect of feeding fermented waste poultry feeds on blood cholesterol levels, LDL and HDL. The conclusion was that waste poultry feeds fermented with starfungs had no effect on blood cholesterol profile of broiler that could be seen on the level cholesterol, LDL and HDL.Keywords: waste poultry feeds (scattered feed); broiler chicken; cholesterol; LDL and HDL

Abstract

The objectives of the research was to determine and assess the effect variation of level and different time periode fermentation cattle feed based agricultural waste on in vitro level of VFA and NH3. Statistic analysis used Completely Randomized Design (CRD) with a 4x4 factorial pattern repeated 3 times to assess the influence of the levels of A. niger (0, 2, 4 and 6% of dry matter feed) and time periode cattle feed quality (0, 7, 14 and 21 days). The results showed different level starter and time periode fermentation did not show interaction (P<0,05), but each of treatments affect on VFA and NH3 content. Each adding A. niger and different time periode affect the increased VFA and NH3. Each of adding A. niger was increased the levels of VFA and NH3 highest in T4 treatment (A. niger cedar 4%). Each curing time was increase levels of VFA and NH3 highest in treatment B2 (time periode was 14 days).Keywords : agricultural by-product and A. nigerABSTRAKTujuan penelitian adalah mengetahui dan mengkaji pengaruh aras dan lama pemeraman fermentasi limbah dan hasil samping pertanian dengan A. niger terhadap kadar volatile fatty acid (VFA) dan NH3 secara in vitro. Rancangan yang digunakan adalah rancangan acak lengkap (RAL) dengan pola faktorial 4 x 4 dengan 3 ulangan. Faktor pertama adalah aras A. niger (T0, T1, T2, T3) masing – masing (0, 2, 4, 6% dari BK (Bahan kering)). Faktor kedua adalah lama pemeraman (B0, B1, B2, B3) masing – masing (0, 7, 14, dan 21 hari). Parameter yang diukur adalah kadar VFA dan NH3. Hasil penelitian menunjukkan bahwa tidak adanya interaksi (p>0,05) antara kombinasi perlakuan aras A. niger dan lama pemeraman pada fermentasi pakan berbasis limbah dan hasil samping pertanian terhadap VFA dan NH3. Penambahan aras A. niger dan perbedaan lama peram berpengaruh terhadap peningkatan VFA dan NH3. Masing – masing penambahan aras A. niger dapat meningkatkan kadar VFA dan NH3 tertinggi pada perlakuan T4 (aras A. niger 4%). Masing – masing peningkatan lama peram dapat meningkatkan kadar VFA dan NH3 tertinggi pada perlakuan B2 (Lama pemeraman 14 hari).Kata kunci: limbah pertanian dan A. niger

Abstract

ABSTRAKTujuan penelitian yaitu mengkaji keberadaan bakteri Escherichia coli dan Salmonella sp pada limbah pabrik pakan unggas (pakan ceceran) yang difermentasi dengan starter fumgsional (yang terdiri dari ekstrak limbah sayur fermentasi, cairan rumen, pollard dan akuades disebut juga ‘starfungs’). Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 4 ulangan. Perlakuan yang diamati adalah T0 : pakan ceceran + 0% ‘starfungs’, T1 : : pakan ceceran + 1% ‘starfungs’, T2 : : pakan ceceran + 2% ‘starfungs’, T3 : : pakan ceceran + 3% ‘starfungs’. Parameter yang diamati adalah keberadaan bakteri Escherichia coli dan Salmonella sp pada limbah pabrik pakan (pakan ceceran). Hasil penelitian menunjukkan penambahan ‘starfungs’ mulai level 1% (T1) mampu menekan keberadaan Escherichia coli. Penambahan ‘starfungs’ tidak berpengaruh terhadap keberadaan Salmonella sp. Abstract

Agricultural intensification program in Indonesia which is carried out by using high yield variety, high chemicals use and soil disturbances tends to trigger land exploitation. Land exploitation, performed without considering the lands capability can generate degradations on the land itself. Various methods have been used to determine land exploitation level, including evaluation of soil microbe resources as on soil component. This research is aimed to assess land exploitation level, based on the amount of microbial population. The result of this research is expected to add the soil quality standard criteria. In the case study performed in Dieng plateau, representative soil sampling method was used. The amount of microbial population can be enumerated using plating and MPN method. Based on nutrient availability to indicate the soil biological characteristics, the soil under the trees, shrub, and Colocasia were classified as “below normal”, and the soil under the grass, tobacco, cabbage and potato were classified as normal. It shows that the land exploitation at the agricultural soils were still in the range of its land capability.

Abstract

Pseudomonas fluorescensandP. putida are Plant Growth Promoting Rhizobacteria (PGPR) that can produce growth hormone. The objective of this study is to know the effects of those two combined species of PGPR on seedling root growth of Jatrophacurcas. The condition of the seedling root determines the success of dry land cultivation. The root which has wider coverage, is larger in number, and is bigger in diameter makes seedling more resistant to stress in dry land environment. In the experiment, two kinds of plant materials are used for seedling, the Jatropha seed and stem material, which are treated in a mixed culture of PGPR. For the Jatropha seed, this mixed culture of PGPR is given at the same time of cultivating the sprout on the seedling medium. For the stem cutting, the PGPR is poured in together during the first watering of the seedling cultivation medium. In the fourthweek, the observed growth parameters are root length, root diameter, primary and secondary lateral root numbers, Root Length Density (RLD), Frequency of Lateral Root (FLR), and Specific Root Length (SRL). These data are analyzed using analysis of variant with DMRT test at 0.05 level of significance. The result of this study shows that PGPR tend to reduce FLR values on the seedling root made from seeds. On the seedling root made from stem cutting, PGPR increase the root length, primary and secondary lateral root numbers, root diameter, FLR and SRL values as well. Abstract

Natto and Soy Yogurt are fermented food products that are very beneficial for improving health. This community service activity aims to socialize the benefits and make them known to the wider community. However, community service activities during the Covid-19 pandemic cannot be carried out face-to-face and have activities with the community in large numbers. Therefore, this community service activity was carried out through webinar and practical activities. Online seminars was chosen so that the coverage was wider and more people could participate in the pandemic. Face-to-face and community activities are carried out with a limited number of participants, according to health protocols. The level of success, benefit and acceptance of the community from this community service activity is known from the responses of the participants during the activities. Based on the results of the assessment/ response of participants who were present virtually and those present at the location, In general it can be concluded that the PKM activity entitled "socializing the benefits and making natto and soy yogurt through webinars and practices" is going well, but better preparation is still needed.The material presented by the resource person is suitable for the current pandemic conditions, easy to understand, easy to practice and can be developed for home businesses. The Zoom Meeting application can be used as a medium for community service activities in pandemic conditions even though it still cannot reach the wider community.  Keywords: Socialization, natto, soy yogurt, webinar, practical
