Abstract

The aims of this study were (1) to analyze the phenotypic performance of Ettawa Grade (EG) goat; (2)to estimate the heritability of birth weight (BW), weaning weight (WW), yearling weight (YW), and geneticcorrelation between two body weights on the third different period; and (3) to analyze the variance andcovariance component of body weight. The material used were the exiting records of 437 EG goats in BalaiPembibitan Ternak Unggul dan Hijauan Pakan Ternak Pelaihari, South Kalimantan. These goats originatedfrom the crossing between 19 males and 216 females from periods of 2009 - 2012. Nested Design methodwas used to etimate the phenotypic correlation, heritability and genetic correlation. Variance componentswere determined from heritability estimation, while covariance components were determined from geneticcerrelation estimation. Phenotypic correlation between BW and WW, between BW and YW, and betweenWW and YW were 0.19 (low); 0.31 (medium); 0.65 (high); respectively. Heritability of BW, WW, and YW were0.43±0.23 (high); WW 0.27±0.19 (medium); and YW 1.01±0.38 (excludeof the h2 value), respectively.Genetic correlation between BW and WW, between BW and YW, and between WW and YW were -0.04(negative low); 0.49 (positive medium); and -0.41 (negative medium), respectively. Variance components ofbuck, ewes, and kid for BW were 10.76%; 37.16%; and 52.09%, respectively, for WW were 6.67%; 38.52%;and 54.81%, respectively, and for YW were 25.15%; 58.37%; and 16.43%, respectively. Covariancecomponents of buck, ewes, and kid between BW and WW were -3.91%; 66.45%; and 37.46%, respectively,between BW and YW were 65.68%; 16.50%; and 17.82, and between WW and YW were -5.14%; 83.87%; and21.28%, respectively. In conclusions variance component of ewes and kid were high in body weight at birthand weaning time. Therefore, selection should be conducted for body weight at birth and weaning time.

Abstract

Rabies was a new emerging disease in Bali. After the first case of rabies in 2008 there were morethan one hundred of human victims in Bali. The aim of this study was to report the anatomical locationof human body bitten by rabies infected dogs in Bali, and also the day of victims death after the bitten. Aretrospective cross-sectional review of rabies incidences from September 2008 to the end of 2011 was usedin this study. A total of 122 rabies human victims data were used as a population sample. The data on theprofiles of individuals victim were colected , such as the age, gender, living place, anatomical bite site onhuman body, bite date, and type of biting animal. The obtained data were analyzed using descriptiveanalysis. The result showed that the anatomical location of human body that bitten by rabies infected dogwas occurred on leg (52%), hand (32%), body (6%), and head (4%). Of 122 victims, 61.5% were men and38.5% were woman. The death time of the human rabies victims in average occurred at day 95th after thebite date, and time of the victims death depended on the anatomical location of the bite. Bite that occurredon the head caused death in average on day 19th, on the body on day 83th, on the hand on day 122nd, and onleg on day 166th. In conclusion, the most victims of human rabies in Bali was men and the death was foundoccurred on day 95th after the bite date. The bite at the head caused death more quickly than the other partof human body.

Abstract

Avian influenza virus subtype H5N1 (AIV H5N1) is highly pathogenic and fatal in poultry. The virusis still endemic with low virulence rate, although it may play a critical role in causing high morbidity andmortality rates in poultry in Indonesia. In general, diagnostic approach for AIV H5N1 is based onconventional serological and viral isolation methods that have the potential to produce consumings oftime and relatively expensive cost within the laboratory without compromising test utility. Thus, amolecular approach of multiplex reverse transcription-polymerase chain reaction (mRT-PCR) was developedand applied for the detection of matrix gene type A influenza viruses, AIV subtype subtype H5hemagglutinin gene with simultaneous detection of N1 nucleoprotein gene. Thirty sera specimens fromthe diseased commercial chickens that were specifically amplified positive-RT-PCR for AIV H5N1 wereselected for mRT-PCR. The mRT-PCR products were visualized by agarose gel electrophoresis and consistedof DNA fragments of AIV of 245 bp, 545 bp and 343 bp for M, H5 and N1 genes, respectively. Thus, themRT-PCR that can rapidly differentiate simultaneously between these genes is very important for thecontrol and even eradication of AIV transmission in poultry in Indonesia.

Abstract

High level of blood glucose is an indicator for diabetes mellitus (DM) condition. The condition iscaused by low level of insulin secretion or impairement of insulin receptor. The number of DM patientincreases every year. The World Health Organization reported that the number of DM patient in Indonesiawas the 4th highest in the world, after following China, India, and the United States of America, respectively.This study was conducted to analyze the effect of sea cucumber (Holothuria scabra J) on the profile ofantioxidant copper, zinc-superoxide dismutase (Cu, Zn-SOD) in the pancreatic tissues of diabetic rats. Atotal of 25 male white rats (Sprague Dawley) were used in this study. They were divided into five groups;(1) negative control (KN), (2) positive control, diabetic rats (KP), (3) diabetic rats treated with hydrolyzatedprotein of sea cucumber (HDL), (4) diabetic rats treated with concentrated protein of sea cucumber (KST),and (5) diabetic rats treated with isolated protein of sea cucumber (ISL), respectively. Diabetic conditionwas obtained by alloxan injection 110 mg/kg bw. The treatments were done for 28 days. At the end oftreatment period, the rats were sacrificed and pancreatic tissues were collected and fixed in Bouin solution and then processed to paraffin embedding standard method. The tissues were then stained withimmunohistochemical staining techniques using monoclonal antibody of Cu, Zn-SOD. The results showedthat treatment of HDL, KST, and ISL of sea cucumber (Holothuria scabra J) increased the content ofantioxidant Cu, Zn-SOD either in Langerhans islets and acinar cells of pancreatic tissues-diabetic rats.The HDL of sea cucumber treatment gave the best effect in increasing the antioxidant content of Cu, Zn-SOD in pancreatic tissue of diabetic rats.

Abstract

Calcitriol supplementation in ovariohisterectomized rat was known to decreased calcium retention.The objective of the research was to study the response of femur bone to calcitriol supplementation for 30weeks in ovariohisterectomized rats consuming casein. Twenty female Wistar rats at 8 weeks of age wererandomly divided into four groups (unovariohisterectomy without calcitriol supplementation (N),unovariohisterectomy with calcitriol supplementation (ND), ovariohisterectomy without calcitriolsupplementation (O) and ovariohisterectomy with calcitriol supplementation (OD) of five each. Thirtyweeks after surgery, femur was taken for histopathological and immunohistochemistry examination.Immunohistochemistry of distal femur metaphysis in group O and OD were revealed decreasing tartrateresistant alkaline phosphatase 5b (TRAP5b) in trabecular bone, which was located in bone marrow space,and also in trabecular speculum surface. Histopathological analysis of distal femur metaphysis in groupN and ND were showed normal structure, meanwhile, distal femur metaphysis in group O and OD wereshown some abonormalities, such as increased of bone marrow space, domination of adipocytes in the bonemarrow, and decreased of trabecular bone speculum in metaphysis. Based on the results, it was concludedthat femur bone of ovariohisterectomized rats fed casein with and without calcitriol 8ng/daysupplementation for 30 weeks were showed unbalanced between resorption and formation of bone whichwas domination by bone resorption.

Abstract

The growth curve is a reflection of the ability of an individual or population to actualize themselves aswell as the size will be the development of the parts of the body until it reaches the maximum size (adult)on the existing environmental conditions. The main goal of this study is to determine the growth curvemodel of Friesian Holstein cattle from birth until ready to mate based on birth rates with Logistic model,Gompertz, and von Bertalanffy. The data was used in this study is the result of the weighing of dairy cowsbody weight of female Friesian Holstein as many as 335 head number collected by PT Taurus Dairy FarmSukabumi from 2001to 2011, which have complete data from birth until first mating with a birth rate thatis different from the birth of the one to five. The data used in the analysis of growth curves using three nonlineargrowth curve model i.e. Logistic models, Gompertz, and Von Bertalanffy. The results obtainedindicate that the growth curve model shave different levels of accuracy, depending on the environment andage or duration of observation, the longer observed to mature weight (A), the age of puberty, and pubertygreater weight. The Conclusion of this study is a model of Logistic close to field conditions. Morover thehigher the birth rate will be heavier birth weight and adult weight. These three non-linear mathematicalmodel used in this study has a high degree of accuracy.

Abstract

The aim of the study was to determine sequence and genotype diversity of protective antigenic gene ofBacillus anthracis isolated from Central Java and Yogyakarta. Pag-A gene which encodes for antigenicprotein is one toxin component and the virulent factor of B. anthracis. As many as five isolates fromSemarang, Sragen, and Boyolali (Central Java) and Sleman (Yogyakarta) were used. The gene wassequenced and amplified used three set of primers PA1857/PA2436, PA8/PA5, and PA-5F/PA-5R. Theresult showed that the nucleotide sequences of gene from five isolates were identical and only had onenucleotide difference as compared to B. anthracis sterne M22589. All isolates were confirmed as genotypebased on pag-A sequence. It was concluded that all B. anthracis from Central Java and Yogyakarta haveidentical pag-A sequence and belong to genotypt-1. Further studies are needed to investigate B. anthracisisolates from other regions of Indonesia.

Abstract

The aim of this research were to determine antigenicity, sensitivity, and specificity of Toxocara canisprotein used as antigen in indirect-ELISA for the detection antibody against the worm in the infected hostin order to proper diagnose kit. The design used was true experimental, with Post-test Only ControlGroups Design. Mouse was immunized with various worm homogenates used to antigenicity, sensitivityand specificity tests of T. canis protein with indirect-ELISA technique. The independence variable werevarious immunogens (homogenates); the dependence variables were antigenicity, sensitivity and specificityvalues interpreted by optical density (OD) value of mouse sera; and controlled variable were mouse strain,feed and retrieval time of sera. The result showed that OD values of mouse sera immunized with T. canisand T.cati homogenate were signicantly difference (p<0.01) as compared to those immunized withAncylostoma spp., Dipylidium caninum and control sera. Using the diagnosis based on the finding ofToxocara, the sensitivity of OD value by ELISA result from mouse sera immunized with Toxocara spp.homogenate were 100%. Using negative OD value by ELISA from mouse sera immunized with Ancylostomaspp. and D. caninum homogenate, the specificity of the test was 87.5%. In conclusion, protein of T.canishas the same antigenicity against anti-T. canis and anti-T. cati sera, but they had the lower antigenicityagainst anti-Ancylostoma spp. and anti-D.caninum sera. As the sensitivity value of 100% and specificityvalue of 87.5%, in detecting antibody against toxocariasis, the possibility of obtaining false positive was12.5%.

Abstract

Avian Paramyxovirus type 1 (APMV-1) is a virus infecting birds with mortality level varies accordingto its pathotype. Although the virus is normally originated from birds, the recent report showed that it canbe isolated from pig. In order to find out APMV-1 infections on pig in Bali, a serologic study was conductedto detect the antibody against APMV-1 among pigs in Bali. Indirect enzyme-linked immunosorbentassay (ELISA) was used to detect the antibody against APMV-1 in pig sera. The serum samples withoptical density value over the cut off value (COV) was considered as those containing Ab against APMV-1. Of 215 samples collected from traditional farms of eight regency and city in Bali namely Negara,Tabanan, Badung, Denpasar City, Gianyar, Bangli, Karangasem and Buleleng Regency, 174 samples(80,93%) were positive antibody against APMV-1. On the other hand, there were 125 (98,42%) positivesample among 127 sample collected from Pesanggaran abattoir. Furthermore, the 8-9 month age pigsshowed that their antibody against APMV-1 were higher (p<0.01) than those of 3-6 months age. TheELISA positive samples were further confirmed using Western immunoblotting assay. The anti-APMV-1antibody positive sera reacted with the virus specific protein band of 258 kDa and the negative sera did notreact with the viral protein. As pig has never been vaccinated with APMV-1 vaccine, the presence of Abagainst APMV-1 showed that the pigs were naturally infected with APMV-1.

Abstract

Commonly, native chicken is raised in an extensive management system. The chickens were free toscavenge in the backyard, and eat a wide variety of feeds including, rice, corn, insects, or even grit. Grit is anabrasive material that can help to grind feedstuffs and use as a source of mineral for chicken. In anintensive farming system, native chickens are given full feeding program throughout the production period,without grit. The objective of this research was to study the development of gastrointestinal tract andcarcass bone of native chickens supplemented to grit. Eighty chickens were used in the study usingrandomized block design. The four treatments were: R0 = basal feeds without grit ; R1 = R0 + grit 0,25% ofbody weight ; R2 = R0 + grit 0,50% of body weight ; R3 = R0 + grit 0,75% of body weight. The results showedthat administration of grit up to the level of 0.75% body weight significantly increased grit consumption,digestive tract length, gizzard weight and carcass bone weight of native chicken.
