Abstract

Information on the estimation of genetic distances and differentiation among sheep breeds are needed in crossing and conservation programs. This research aims to study of utilizing behaviour characteristic variables to differentiate and estimate genetic distance between the sheep breeds.  The study was conducted at Cilebut and Bogor Animal House of Indonesian Research Institute for Animal Production.  Five sheep breeds used were Barbados Black Belly Cross (BC), Garut Composite (KG), Garut Local (LG), Sumatera Composite (KS) and St. Croix Cross (SC), with total sample of 50 heads.  A total of 10 variables of behavior traits were observed in this study. Analysis of variances and significance tests were applied to compare between sheep breeds and performed for all of behavior traits using PROC GLM of SAS Program ver. 9.0. PROC CANDISC was used for canonical discriminant analyses, the hierarchical clustering was performed using the PROC CLUSTER by Average Linkage method (Unweighted Pair-Group Method Using Arithmetic Averages, UPGMA), and the dendogram for the five sheep breeds was described using PROC TREE.  The differentiator variables for the behavior traits were standing and feeding duration.  The canonical plotting based on behavioral characteristics could differentiate BC, KS and LG (with KG and SC) sheeps. Estimation of genetic distance based on the behavior traits is less accurate for grouping of sheep breeds. Key Words: Behaviour Characteristic, Differentiation, Genetic Distance

Abstract

Buffalo milk is a source of various lactic acid bacteria (LAB) which is potential as culture starter as well as the probiotic. This study was conducted to isolate and identify LAB from indigenous North Sumatra river buffalo milk. Lactic acid bacteria was isolated and grown in medium De Man Rogosa Sharpe Agar (MRSA). The isolation was conducted to obtain pure isolate. The identification of  LAB was studied in terms of morphology, physiology, biochemistry and survival on low pH. Morphology tests were conducted by Gram staining and cell forming; physiology tests were conducted for growing viability at pH 4.5 and temperature at 45oC; whereas biochemistry tests were conducted for CO2, dextran and NH3 productions. Determination of LAB species was conducted using Analytical Profile Index (API) test CHL 50. Results of identification showed that 41 isolates were identified as LAB with Gram-positive, catalase-negative, rod and round shaped characteristics. Resistance test done to low pH (pH 2) for the lactic acid bacteria showed decrease of bacteria viability up to1.24±0.68 log cfu/ml. The resistant isolates at low pH were L12, L16, L17, L19, L20, M10, P8, S3, S19 and S20. Identification with API test CHL 50 for 10 isolates showed that four isolates were identified as Lactobacillus plantarum, L. brevis, L. pentosus and Lactococuslactis. Abstract

HSD17β13 (17beta 13-Hydroxysteroid dehydrogenases) are important enzymes in steroid metabolism. This study investigated the polymorphisms and expression of the HSD17β13 gene in lamb quality traits in Indonesian sheep. A total of 200 of rams of seven breeds administered in the study were Javanese fat-tailed (JFT), Javanese thin-tailed (JTT), Garut sheep (GS), Jonggol sheep (JS), Garut composite sheep (GCS), Compass agrinac sheep (CAS), Barbados cross sheep (BCS) aged 10-12 months and weighed 20-30 kg. The Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) and MspI enzyme restriction were performed to identify the polymorphism of the HSD17β13 gene. The lamb quality traits were measured for physical quality, flavor, odor, and fatty acid composition. The results showed that the HSD17β13 gene was polymorphic and had three genotypes i.e., CC, CT, and TT. The highest genotype frequency was the CC when compared to the other genotypes. The allele frequency of HSD17β13 gene was not in Hardy Weinberg Equilibrium. The polymorphism of HSD17β13 gene was significantly (P<0.05) associated with the fatty acid composition and Saturated Fatty Acid (SFA), i.e., tridecanoic acid (C13:0), arachidic acid (C20:0), henecosanoic acid (C21:0), and tricosanoic acid (C23:0). The polymorphism was also significantly associated with the Monounsaturated Fatty Acid (MUFA) i.e. ginkgoleic acid (C17:1) and nervonoic acid (C24:1), and Polyunsaturated Fatty Acid (PUFA) i.e. eicosedienoic acid (C20:2). The level of expression of the HSD17β13 gene based on quantitative real time-PCR analysis results was not significantly different (P>0.05) among genotypes for lamb quality traits. It can be concluded that the polymorphism of HSD17β13 gene was associated with the fatty acid composition of lamb in Indonesian sheep.

Abstract

Tujuan utama dalam pembuatan model kurva pertumbuhan adalah untuk deskripsi dan prediksi. Tujuan deskripsi merupakan upaya untuk bisa mempermudah interpretasi dari proses pertumbuhan ternak menjadi hanya beberapa parameter, sedangkan tujuan prediksi lebih fokus bagaimana metode untuk memprediksi dari beberapa parameter. Adapun tujuan penelitian ini adalah untuk membuat model kurva pertumbuhan sapi perah Fries Hollands dari lahir sampai siap kawin yang sesuai dengan situasi dan kondisi skala industri peternakan sapi perah. Data bobot badan sapi betina yang digunakan dalam penelitian ini merupakan hasil penimbangan bobot badan dari sapi perah Fries Hollands yang dipelihara oleh PT Taurus Dairy Farm Sukabumi dari tahun 2001 sampai 2011, sejumlah 1221 ekor, individu yang mempunyai data lengkap dari lahir sampai umur kawin pertama sebanyak 373 ekor. Sedangkan untuk data BPPTU Baturraden 214 ekor data kelahiran sampai ternak siap untuk dikawinkan pertama kali dari 2010 sampai 2011. Data dianalisis menggunakan program SAS 9.2 dengan prosedur NLIN (Non Linear). Kurva yang dihasilkan dapat dipakai sebagai standar kurva pertumbuhan sapi perah Fries Hollands di Indonesia dari lahir hingga siap kawin. Model matematik Logistic dapat dipakai untuk menduga kurva pertumbuhan karena mempunyai tingkat akurasi yang tinggi dengan nilai koefisien determinasi (R2) lebih dari 90%.

Abstract

Etawah grade goat is a result of grading up line between Kacang and Etawah goats, which well adapted to Indonesia’s humid tropical climate. This goat is a dual-purpose goat breed for milk and meat production. The objectives of this study were to evaluate the polymorphism of fecundity genes through the investigation of four candidate genes for prolificacy, i.e., BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1), and KISS1 (intron 1) and their associations with the litter size of Etawah-Grade does. The diversity of BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1), and KISS1 (introns 1) genes were analyzed by the PCR-RFLP technique. A total of 106 of DNA samples were collected from Etawah-Grade does at BPTU-HPT Pelaihari in Pelaihari, South Kalimantan (51 heads) and Cordero Farm in Bogor, West Java (55 heads). Genetic polymorphisms between the two Etawah-Grade groups were calculated as well as their allele and genotype frequencies and the Polymorphic Informative Content (PIC). The Kruskal-Wallis one-factor ranked analysis of variance was used to determine whether there was any effect of genotype of the fecundity genes on litter size. The polymorphic information content (PIC) estimated at BPTU-HPT Pelaihari was 0.313, whereas at Cordero farm was 0.174, and overall PIC was 0.244. There was no association between genotypes of BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1), and KISS1 (intron 1) genes and litter size. Results indicated that BMP15 (exon 1) and KISS1 (exon 1) genes were monomorphic. However, the other two loci showed polymorphisms. The BMPR1B (exon 1) locus had two genotypes with the frequency for CC (96.23%) and for GG (3.77%), and KISS1 (intron 1) locus had three genotypes with the frequency for TT (80.19%), AT (17.92%), and AA (1.89%).

Abstract

Sound analysis has been carried out in various activities including identification and differentiation of species as well as the preparation of the taxonomy of some animals’ species because of several advantages, including no need to capture or too close to the subject observed. Analysis of voice used to differentiate and to estimate of breeds’ sheep genetic distance has not been reported. This research was conducted to study the character of a few breeds’ sheep sound and likely to be used as a predictor of genetic distance between breeds of sheep. The study was conducted in the Animal House at Indonesian Research Institute for Animal Production, Bogor.  A total of 20 head adult of five sheeps (St. Croix cross / SC, Barbados Black Belly cross / BC, Local Garut/LG, Composite Garut / KG and Composites Sumatra / KS) used in this study. Call sound recorded using a digital voice recorder. Sound analysis performed by Raven Software Pro 1.3 for Windows to count as many as 24 variables sound. Analysis of variance of each variable sound was performed using PROC GLM of SAS software Ver. 9.0. It used PROC CANDISC for canonical discriminant analysis and then PROC TREE to build a dendogram. The results showed that there were variations in amplitude, energy, power and frequency variables among the five breeds of sheep. By plotting canonical, LG, KS and BC sheep were from a different group. It was concluded that the sound characteristics variables which can be used as a differentiator breeds of sheep were the third quartile frequency, center frequency, maximum frequency and the first quartile time. Dendogram showed that KG sheep was in the less accurate group. Genetic distance estimation method using voice characteristic data may be applied on sheep. Abstract

Bali cattle (Bos javanicus) are common species distributed throughout Indonesia to survive in tropical hot, and humid climates. Fluctuating asymmetry (FA) is a random deviation from perfect bilateral symmetry. FA is often used to measure developmental stability in individuals and can be used as a marker for the impact of environmental and genetic stress. This study aims to investigate the pattern of developmental instability in Bali cattle as caused by environmental stress at various altitudes using the FA index. FA indexes in this study were used to compare asymmetrical physical traits of Bali cattle in Sembalun high altitude and Serading low altitude. Sixty-five Bali cattle were used in this study reared at different altitudes, namely Serading, Sumbawa Island (50 m above sea level), and Sembalun, Lombok Island (1,186 m above sea level). The physical traits of Bali cattle measured were horn circumference (horn), a distance of hip to spine bone (pelvic), metatarsal circumference (metatarsal), and metacarpal circumference (metacarpal). The FA1 and FA5 indexes showed significant metacarpal differences between Bali cattle reared at Sembalun and Serading population (P<0.05). FA11 multiple trait index as a composite of all traits revealed a significant difference as well. (P<001). It can be concluded that various heat stress by altitude impacts the asymmetry of Bali cattle.

Abstract

The study of behavior has a very important role in wildlife management process both for cultivationand for preservation in nature. The purpose of this study was to observe and document behaviors and dailyactivities of weris. Observation of hatching process and behavior of juvenile weris used direct observationmethod, while the behavior of adult weris in captivity used scanning sampling method. The data wereanalyzed descriptively. Weris had six stages of hatching processes starting from the breakdown of the eggshell until the chick moved out from the shell. The placement of the eggs in hatching machine was byputting the dull part on upside. Weris is a diurnal bird that active in the morning until late afternoon.Young and adult weris birds had different activities, i.e., adult birds did bathing and flying activities,while the young birds just rest and sleep. Observation on behavior of adult weris (n = 10) showed that thehighest activity for 12 hours was moving (314,6 minutes), then followed by resting (283,1 minutes), eating(51,8 minutes), drinking (29 minutes), bathing (22,9 minutes), and the lowest was preening (18,6 minutes).Inherited behavior pattern, such as bathing, did not change, eventhough the environment changed.
