Abstract

Effect of the ginger extract (Zingiber officinale Rosc.) on the growth of Pythium sp., the causal agent of cucumber damping off disease. The Pythium sp. often damaged cucumber seedling in seedbed. The objective of this study was to evaluate the effectiveness of ginger extract on the growth and spore production of Pythium sp. The study was conducted in 2005 at the laboratory of plant pathology, University of Lampung. The treatments were arranged in a completely randomized design with four replicates. The treatments were consisted of five level of concentrations of ginger rhizome extract i.e, 0; 2.5; 5.0; 7.5 and 10 %. The results show that all concentrations of the rhizome ginger extract inhibited mycelial growth and spore production of Pythium sp. There was a significant linear relationship between the rhizome ginger extract and both of fungal growth and spore production.

Abstract

Insecticide Preparation of Calophyllum soulattri: Insecticidal and Residual Activity Against Crocidolomia pavonana and Its Savety on Crops. Bioassays were done using leaf-residual method with Crocidolomia pavonana larvae as test insect. The number of dead larvae was recorded daily and was analyzed using probit method. The results showed that methanol extract, and dichloromethane fraction of C. soulattri bark possessed strong lethal effect against C. pavonana larvae with LC50 of 0.15%, and 0.05%, respectively. Dichloromethane fraction of 21 EC and 21 WP sprayed in suspension consentration of 1% on potted broccoli plant had residual activity around 16%-26,7% with range of half-lives of 5.9-7.7 days. Dichloromethane fraction at concentration of 0.2% and 0.3% formulated as EC did not cause phytotoxic to broccoli, tomato, chilli, soybean, red bean, onion, cucumber, and maize. Concerning with this potency, further studies are needed to identify insecticidal compounds.

Abstract

Mechanism of parasitism of Trichoderma harzianum on Fusarium oxysporum on Acacia mangium seedlings. Fusarium oxysporum is one of the most important soil-borne fungi the causal agent of damping-off disease. Detailed information it needed to know how the pathogen can be inhibited by Trichoderma harzianum. The objective of this research was to investigate the inhibition mechanism of T. harzianum on F. oxysporum in vitro and in planta. Green Flourescent Protein (GFP) T. harzianum was used as biocontrol agent of F. oxysporum. An in vitro inhibition test of T. harzianum was performed using dual culture method. In the in planta inhibition tests, seedlings of A. mangium were applied with GFP T. harzianum two days before inoculation of F. oxysporum; GFP T. harzianum was simultaneously applied with F. oxysporum and GFP T. harzianum was applied two days after inoculation of F. oxysporum. The inhibition effect of T. harzianum GFP was observed at seven days incubation, indicated by attachment of T. harzianum to F. oxysporum hyphae. GFP T. harzianum hyphae covered the colonies of F. oxysporum at 12 days after incubation. The highest life percentage of A. mangium seedlings was found on the treatment of GFP T. harzianum two days before inoculation of F. oxysporum (82.22%), whereas the lowest life percentage was found on seedling applied with GFP T. harzianum two days after inoculation of F. oxysporum (64.44%).

Abstract

Sublethal dose effect of imidacloprid on the survival of brown planthoppers on resistant and susceptible rice varieties. Imidacloprid insecticides and resistant rice varieties are commonly used to control the the rice brown planthopper (BPH) Nilaparvata lugens in the field. The aim of this research was to evaluate the resistance of N. lugens to imidacloprid and the influence of sublethal dose of the insecticide applied to a certain resistant rice varieties on the survival of BPH. The experiment was conducted in a greenhouse trial at Indonesian Center for Rice Research, Sukamandi, West Java. Each pair of adult BPH were infested on three rice varieties Ciherang, Inpari 13, and Pelita, and then followed by spraying five doses of imidacloprid. The the effective dose rate of imidacloprid on BPH was determined by the value of LD95 compared with the recommended dose (20 g a.i. ha-1). The nymphal survival was concidered as a result of the applied sublethal doses. The numbers of nymphal progenies were observed until two peaks populations of both first and second generations. The insecticide then was applied at LD50 dose equivalent on adult BPH of the second generation. The number of laid eggs and the abnormal egg shapes were counted. Imidacloprid doses that were unefffective against BPH showed by the values of LD95 were 1224.3, 2799.0, dan 4692.8 g a.i. ha-1 at Ciherang, Inpari 13 dan Pelita rice varieties, respectively. Seven days after application of imidacoprid, about 70-90% of adult BPH still survived. Sublethal dosages of imidacloprid tended to increase the survival of BPH on a resistant rice variety of Inpari 13, as reflected by the number of nymphal progeny of 156-169 BPH/plant at the first population peak. Imidacloprid treatment increased the number of laid egg, but had no effect on the egg sterility.

Abstract

Response of various soybean genotipes against cowpea mild mottle virus (CPMMV) infection. The green house experiment was conducted to investigate the effect of CPMMV infection on growth and yield of various soybean varieties. Treatments were arranged in completely randomized block design in split plot experiment. Mainplots were virus-inoculated and uninoculated plants; subplots were eight soybean varieties: Wilis, Orba, MLG 2521, Taichung, B 3570, Dieng, Tidar, and Pangrango. The results of this experiment showed that CPMMV infection decreased vegetative growth and yield of soybean plant. The decrease of dry weigth of soybean plant proved that CPMMV infection inhibited the vegetative growth. Taichung variety susceptible to CPMMV was indicated by significant reduction of the growth and yield. B3570 line tolerance to CPMMV infection was proved by lower reduction of the yield.

Abstract

The effect of Aglaia odorata Lour. twig extract on development of a parasitic wasp Eriborus argenteopilosus (Cameron) in its host Crocidolomia binotalis Zeller. This laboratory study was conducted to evaluate the effect of ethyl acetate soluble fraction of methanolic twig extract of Aglaia odorata Lour. (Meliaceae) on the development of a parasitic wasp Eriborus argenteopilosus (Cameron) (Hymenoptera: Ichneumonidae) in its host larvae Crocidolomia binotalis Zeller (Lepidoptera: Pyralidae). Extract treatment was given through the host. C. binotalis larvae were fed broccoli leaves treated with A. odorata extract at LC25, LC50, and LC70, then were exposed to the female adults E. argenteopilosus. Mortality of the parasitized host larvae, survival of immature stages of the parasitoid in the host larvae, and longevity and fecundity of the emerging female parasitoids were recorded. The results showed that combinations of extract treatments (at LC25, LC50, and LC70) and parasitization increased the mortality of the parasitized host larvae (38,4 - 42,7%) more than twice that of the parasitized control larvae. Those extract treatments, however, did not exert any negative effects on developmental time of the parasitoid immature stages nor on some morphological characters, longevity and fecundity of the emerging female parasitoids. Thus, A. odorata extract showed promise to be integrated with the parasitoid for the control of C. binotalis.

Abstract

The potential of fungi from bamboo rhizosphic soils as endophytic and biocontrol agents of clubroot disease (Plasmodiophora brassicae) on Brocoli. Clubroot, caused by Plasmodiophora brassicae is a serious soilborne disease of plants worldwide, capable of severe infection on broccoli, cabbage and cauliflower. Clubroot is the most destructive diseases on cruciferae in Indonesia. The existing control measures, including biological control do not provide satisfactory result. The objective of the study was to explore fungi from bamboo rhizospheric soils as endophytic that can suppress clubroot disease in broccoli. There were two steps in this study: (1) exploration and identification of fungi from bamboo rhizospheric soils. (2) Effectiveness test of the fungi in suppressing clubroot disease. There were four species of fungi as endophytes in brocoli in this study, e.g Aspergillus sp., Mortierella sp., Paecilomyces sp., and Chaetomium globosum. They had colonized broccoli root endophytically and suppressing clubroot disease. Paecilomyces sp. of endophytic fungi can suppress clubroot diseases incidence for 18.75%.

Abstract

Detection using PCR (Polymerase Chain Reaction) Candidatus Liberibacter asiaticus, Huanglongbing causal Organism on Siem Mandarin with different types of symptoms. Huanglongbing (HLB) or Citrus Vein Phloem Degeration (CVPD) is one of major diseases on Siem mandarin in Indonesia. HLB is caused by bacteria Candidatus liberibacter asiatus (LAS). The bacteria only live in the phloem cells of host tree and only recently it was reported to be successfully cultured on agar medium. Early detection method of LAS is needed to support healthy Siem mandarin cultivation program. This research was conducted to detect LAS in different types of HLB leaf symptoms based on Polymerase Chain Reaction (PCR) method with specific primer forward MHO 353 and reverse MHO 354. The results suggested that 8 types of HLB leaf symptoms were found on the samples used in this experiment. LAS was detected at 60% on the leaves without any symptom followed by the leaves with completely chlorosis symptom at 66%. The leaves with unevenly yellow showing higher percentage of LAS detection ranged from 80-86%. PCR technique successfully amplified DNA of LAS with the size target of 600 bp.

Abstract

This predator is efective to control population of aphids, so the aim of research was to study the field population abundance, predator preference to this prey, and fitness character of M. sexmaculatus as predator. The population fluctuation was observed at conventional and organic farming. The preference test was conducted by choice and no choice test for some types of prey. Fitness test was performed by studying its prey, the number of eggs produced and number of eggs hatched. The results showed that M. sexmaculatus population abundance was fluctuated, the abundance higher in chili cultivated conventionaly than cultivated in organic farming. But in conventional farming decreased with increasing age of chili. Preference test showed that almost all species of aphids preferred by M. sexmaculatus. Some prey species found were aphids, thrips and Neotoxoptera. Predator fitness levels showed by the longevity of females ranged 9-10 days. Oviposition period was on average five days, while the post-oviposition was two days. The number of eggs produced ranged 100-200 eggs with an average of 135.3 eggs.

Abstract

Effect of infestation of Verticillium lecanii on the severity of coffee leaf rust on plants and the incidence of its colony on coffee leaves. V. lecanii often antagonizes H. vastatrix on lesions caused by the pathogen on coffee leaves in the field. The objectives of this study were to evaluate the efficacy of V. lecanii to control disease severity on coffee plants in a green house and to compare the incidence of V. lecanii colonies on coffee leaves naturally infested by V. lecanii in the field and that in the leaves with additional infestation in the laboratory. For efficacy test and additional infestation in the laboratory, conidia were harvested from YMPD medium. Suspension with 107 conidia ml-1 was sprayed into coffee foliage until both surfaces of the leaves were saturated with the suspension. After the leaves were air dried, they were sprayed with urediospores (104 ml-1). The results show that disease severity in plants treated with V. lecanii and control plants was not significantly different (P < 0.05), except that obtained at the fourth week when disease severity on the treated plants was significantly lower that that in control plants. The number of V. lecanii colonies on coffee leaves naturally infested with the biocontrol agent was not increased by additional infestation with V. lecanii conidia in the laboratory.
