Abstract

Mortality and feeding inhibition effects of several plant extracts collected from Merauke Distric, Papua against Crocidolomia pavonana (F.) larvae (Lepidoptera: Crambidae). The objective of this research was to study mortality and feeding inhibition effects of several plants extract species collected from Merauke District Papua as botanical insecticides against Crocidolomia pavonana larvae. Seven plants used in the research were Areca catechu seeds (Arecaceae), Eucalyptus pellita tree barks (Myrtaceae), Myrmecodia pendans tubers (Rubiaceae), Piper aduncum fruits (Piperaceae), Piper betle leaves (Piperaceae), Pandanus conoideus fruits (Pandanaceae), and Zingiber officinale rhizomes (Zingiberaceae). Each plant extract was tested to 2nd instar C. pavonana larvae. The insecticidal activities were assessed including mortality and feeding inhibition activities. Extract concentrations for mortality tests using topical application method were 0,1%, 0,5%, 1%, 2%, and control, while extract concentrations for feeding inhibition tests using choice and no choice methods were 0,25%, 0,5%, 1%, 2%, and control. Each treatment was introduced to ten C. pavonana larvae and replicated five times. The results showed that P. aduncum and P. conoideus fruits extracts gave 100% mortality at 2% rate. Extract of P. betle leaf at 2% rate totally inhibited feeding activity of larvae (100%) using choice method, while extract of Z. officinale rhizome at 2% rate resulted in moderate feeding inhibition effect (75%) using no choice method.

Abstract

The White-Bellied Planthopper (Hemiptera: Delphacidae) Infesting Corn Plants in South Lampung, Indonesia. Corn plants in South Lampung were infested by newly-found delphacid planthoppers. The planthopper specimens were collected from heavily-infested corn fields in Natar area, South Lampung. We identified the specimens as the white-bellied planthopper Stenocranus pacificus Kirkaldy (Hemiptera: Delphacidae), and reported their field population abundance.

Abstract

Detection of seed-borne pathogenic bacteria of northern black wattle (Acacia auriculiformis A. Cunn. ex  enth.). Intensive research of seed-borne pathogen of A. mangium and A. crassicarpa which have been established in industrial timber estate (HTI) was undertaken in Indonesia, while plantings development of northern black wattle have recently been established in the 1990s. Very limited information available on northern black wattle diseases especially seed-borne diseases. The objectives of this study were to identify seed-borne pathogenic bacteria of northern black wattle and the effects on seed germination. Methods for the isolation of bacteria were by seed soaking, seed griding, blotter test, growing-on test on paper and soil. Identification of bacteria by PCR used 63F/1387R primer. The results showed that seed-borne bacteria of northern black wattle were Paenochrobactrum sp., Ralstonia sp., Burkholderia cepacia complex, Pseudomonas stutzeri, Acinetobacter sp., Alcaligenes faecalis, Salmonella bongori, Escherichia hermannii while pathogenic bacteria cause seedling leafspot were Micrococcus luteus and Burkholderia cepacia complex. Burkholderia cepacia complex, A. faecalis, Acinetobater sp., P. stutzeri, S. bongori and Ralstonia sp. reduced seed germination and increased rotten seed, suggested that they were the pathogenic bacteria of northern black wattle seed. Ralstonia sp. significantly increased the percentage of rotten seed and decreased shoot length and root length. P. stutzeri and S. bongori significantly inhibited the root growth. Paenochrobactrum sp. and E. hermannii were assumed as pathogen with weak virulence due to seed germination, the percentage of rotten seed and vigour index were relatively similar to untreated seed.

Abstract

Pests of star fruit in Blitar District, East Java. Pest attack causes the production of star fruit decreases. This study aim was to identify the pests of star fruit in Blitar District which could provide an information about pest species, damage symptoms and damage intensity. The research was conducted during August – September 2014 in Gogodeso, Karangsonoand Pojok Villages at Blitar District, East Java. Observations of the pests ware done by determining the four locations of plantation on each villages randomly. Three plants sample were choosen in each plantation location diagonally. Each plant was divided into four quadrant according to wind directions. One branch in the bottom was choosen in each quadrant. Nine species were recorded as pests of starfruit in Blitar district. Pest was identified on the leaf was Pteroma plagiophleps, on the branch was Zeuzera coffeae, on the flower were Toxoptera aurantii and Maconellicoccus hirsutus, whereas those on the fruit were Toxoptera aurantii, Maconellicoccus hirsutus, Thrips javanicus, Helopeltis bradyi, Cryptophlebia leucotreta, Bactrocera dorsalis, and Bactrocera carambolae. The population density of M. hirsutus was higher in Karangsono than in other villages. The highest damage intensity was caused by Bactrocera spp., C. leucotreta and H. bradyi (occured in Gogodeso), whereas the highest damage intensity of T. javanicus occurred in Pojok.

Abstract

Efficacy of clove oil and neem extract control rust (Phakopsora rapachyrhizi) disease on soybean (Glycine max). Rust is the major disease on soybean. It was widely spread in almost all soybean production areas in the world. Yield losses can reach 85%. One of the control measured was sprayed with a mixture of clove oil and neem extract. The objective of the research was found the information of the clove oil and neem extract effectivity control soybean rust. The research consisted of two phases as followed: 1) the efficacy of clove oil and neem extract dilution control soybean rust disease. 2) the efficacy of clove oil, neem extract and soap berry (Sapindus rarak) extract control soybean rust disease. The research was conducted at Kendalpayak Experimental Station and the green house of Indonesian Legumes and Tuber Crops Research Institute, Malang District, in dry season of 2012 and 2013 respectively. The research was arranged in randomized block design. Treatments were various comparison between clove oil, neem extract and soap berry. Parameters observed were rust disease intensity and yield components. Results showed that a mixture of clove oil and neem extract with ratio of 60:40 (v:v) can inhibit rust disease intensity by 45% compared with control, increase the number of pods by 60% and preventing the yieldloss of 20%. Furthermore, a mixture of clove oil, neem extract and berry soap (Sapindus rarak) with a 50:30:20 ratio (v:v:v) can inhibit rust disease intensity up to 28% and 24% in the field and green house respectively and prevent yield loss by 12%.

Abstract

Effectiveness of the biopesticide of Bacillus subtilis BNt 8 and botanical pesticide in controlling banded leaf and sheath blight disease on maize. Banded leaf and sheath blight disease (BLSB) caused by the fungus Rhizoctonia solani is difficult to control because it pertained soil borne fungus that can survive in a long time in the soil. Control the disease with synthetic pesticide causing contamination to the environment, so that an environmentally friendly alternative control is needed. This study aimed to obtain a Bacillus subtilis formulation as biological agents and selected botanical pesticides that effective to control BLSB in the field. The study was conducted at the Plant Pathology Laboratory of Indonesia Cereals Research Institutein Maros and at the Bajeng Experimental Farm in Gowa, held from February to August 2015. The reatments consists of several botanical pesticides, B. subtilis formulation, a synthetic fungicide, positive and negative controls. In vitro test was inhibition test between botanical pesticide with R. solani and antagonistic test between the B. subtilis and botanical pesticides, each of them consists of 6 treatments and 3 replications, while the field activity consists of test of effectiveness of single treatment and combination between B. subtilis formulation and botanical pesticides. The results showed that combination of formulated B. subtilis with botanical pesticide of cloves leaves, betel leaves, and turmeric were not significantly different from single treatment of formulated B. subtilis and botanical pesticides. Formulated B. subtilis suppressed the severity of BLSB as much as 39.1% and yield reached 8.4 t/ha.

Abstract

Cassava mealybug, Phenacoccus manihoti Matile-Ferrero (Hemiptera: Pseudococcidae): Geographic distribution in Java and initiation of biological control. Cassava mealybug, Phenacoccus manihoti Matile-Ferrero (Hemiptera: Psedococcidae), is a recently introduced pest in Indonesia. Parasitoid Anagyrus lopezi (De Santis) (Hymenoptera: Encyrtidae) was imported to control the pest. Studies were conducted to determine geographic distribution of P. manihoti throughout Java, and to evaluate the potential of the parasitoid as a biological control agent. Geographic distribution of the pest was determined through field survey, whereas evaluation of the parasitoid was studied through cage experiment and field release. Our survey revealed that the cassava mealybug has spread throughout Java. Heavy infestations caused shortened and distorted stems, complete defoliation, and stunted growth. In cages containing only mealybugs, all cassava plants (100%) died after two months. Whereas in cages containing both mealybugs and three pairs of parasitoid, rate of parasitization was 25% and plant mortality 20%. Parasitoids released in the field were able to survive, reproduce, and establish under Bogor climatic condition. These might indicate that parasitoid A. lopezi is a potential natural enemy to be used in biological control program of the cassava mealybug.

Abstract

Exploration and identification of endophytic Trichoderma in banana. Endophytic fungi Trichoderma is an organism that can used as biocontrol agent. This study aims to isolate and identify endophytic Trichoderma in roots of healthy banana plants from three districts in Yogyakarta, which will be used as biological control agent against the pathogen Fusarium oxysporum f.sp. cubense. Isolation was conducted using TSM (Trichoderma Selective Medium). We obtained six isolates of endophytic Trichoderma spp., i.e., Swn-1, Swn-2, Ksn, Psr-1, Psr-2, and Psr-3. Molecular identification was done by using ITS1 and ITS2 primer pain and sequenced. The sequence of DNA obtained was analysed and compared with NCBI database by using BLAST-N programe. The results showed that all isolates were amplified at 560-bp. Phylogenetic analysis showed that isolates Swn-1, Swn-2 and Psr-1 are homologous to Trichoderma harzianum, isolate Ksn homologous to Trichoderma asperrellum, isolate Psr-2 homologous to Trichoderma gamsii, and isolate Psr-3 homologous to Trichoderma koningiopsis, with the homologous value of 99%.

Abstract

Isolation method of Pyricularia oryzae the causal agent of rice blast disease. Rice blast disease is a disease that reduces rice productivity and threatens global food reserves. The study of diversity and distribution of race fungal causing rice blast disease required a fungal isolates collection from different places and times. One of the challenges in collecting these fungi is the difficulty of isolation process. The purpose of this research was to study the proper isolation method of rice blast pathogen. The most appropriate isolation method of Pyricularia oryzae was to moisten the infected panicle, place on moist filter paper in a petri dish, and incubate plate for 2 days at room temperature under fluorescent lamp. Afterward, conidium was picked using sterile needle and transfered to potato dextrose agar without lactic acid.

Abstract

Feeding rate, aestivation and mobility of golden snails (Pomacea canaliculata) at different depths of water. The research was studied in Malaka District, East Nusa Tenggara Province, during the dry and the rainy season (October 2014 - March 2015). Studies on capacity of damage to the Ciherang rice seedlings were carried out for three weeks. The size of snails used in this experiment was 11-20; 21-30; and 31-40 mm. The numbers of seedlings that were planted were 3, 6, 9, 12, 15, 18 and 21 seeds. The treatments were repeated 5 times. The numbers of seeds which are damaged by the snails then were tabulated and analyzed. Snails with the size of 11-20 mm damaged the seeds in 31.67%, followed by snails with the size of 21-30 mm and 31- 40 mm with potency of damaging were 64.76% and 97.38%, respectively. The size of snails affects the level of damage to rice seedlings when transplanting. Study about aestivation was conducted in five villages, namely Kamanasa, Wehali, Umakatahan, Harekakae and Kletek when the rice fields were fallow. The observation point was observed at the corner of the dike and along the dike, evaluated every 10 meters away. The results showed snails aestivate when the fields were dry. Studies on snail’s mobility conducted on the experiment arena on the mud in buckets, which planted with 20 paddy seedlings. The treatment depth of puddles in the experiment were without puddles, half, the same height and twice the height of snails shell. The treatments were repeated 10 times. Measurement of snail’s mobility was performed routinely every 2 hours, observed from the beginning when the snails were placed for 24 hours in advance. The results showed that the depth of the water significantly affected the mobility of snails. Without puddles, the mobility of snails was severely hampered in the mud. The mobility of the juvenile was significantly higher than the imago. The implication of this study was benefit to that the regulating the level of water in the paddy field which is known as an appropriate effective technology and for controlling the attack of the golden snails.
