Abstract

Polymerase chain reaction (PCR) is a technique of DNA amplification which its success depends on the wider range of several factors, e.g., the annealing temperatures and MgCl2 concentrations. The aim of this study was to determine the effect of annealing temperatures and MgCl2 concentrations on the amplicon specificity of CSSM018 primer. Annealing temperatures of 50oC to 60oC were used in this study while MgCl2 concentrations were in the range of 1.0mM to 2.5 mM. DNA genome of Garut sheep was used as DNA template. PCR products were visualized by a method of 8% ND PAGE. The findings showed that the annealing temperature of 60oC presented the most specific band of the CSSM018 primer at the right size of 116-134bp. Annealing temperatures that were less than 60oC showed the weaker and non-specific bands. A concentration of 1.25 mM MgCl2 showed the best band of the CSSM018 primer with size of 116-134bp. While concentrations of 1.5; 1.75; 2.0 and 2.5 mM MgCl2 showed sharper with some bands seemed noisy, those indicated non-specific products. This study suggests that annealing temperature of 60oC and MgCl2 concentration of 1.25 mM resulted specific amplicon for CSSM018 primer.

Abstract

An investigation involving large number of animals is often resulting incomplete or in accurate information such as animal parentage, or misidentify on sex due to unlabeled sex samples. A PCR method by applying Y chromosome markers (UTY and SRY) facilitates in determination of unknown sex problem. This study was intended to determine sex from unlabelled sex of blood samples by applying PCR method using a pooled-DNA template. Twenty five of unknown sex blood samples from Nusa Penida, Bali were used in this study. The samples were plotted into 5 pooled-DNA whith each pool DNA consisted of 5 individuals DNA. Two pairs of sex primers, UTY (58oC) and SRY (60oC) with 35 cycles were applied to amplify the samples. The result showed there was only one pooled-DNA (P4) amplified by UTY (484bp). Whereas re-PCR of the positive pooled-DNA (P4) using SRY primer, only one out of 25 samples determined as male Bali cattle (325bp). This finding suggests that UTY and SRY primers are suitable for sex determination and the pooled-DNA could be used as an efficient PCR method both in consumables and PCR process for sex determination. Abstract

The efficiency of expression of recombinant proteins in host cells depends on achieving proper protein folding, solubility, and biological activity. Critical environmental factors, including temperature, pH, nutrient availability, and inducer concentration, significantly influence protein yield, stability, and functionality. Optimizing these conditions is essential to minimize protein misfolding and aggregation, which can adversely affect expression efficiency and reduce bioactivity. This study aims to measure the effect of temperature on the growth of Escherichia coli BL21 harboring the recombinant JSU protein, derived from the ENV-SU gene of the Jembrana virus. A controlled laboratory experiment was conducted in which bacterial cultures were incubated at 25°C and 37°C under identical conditions, except for temperature. The results indicate a statistically significant difference in bacterial growth between the two temperature conditions (p = 0.08), suggesting that E. coli BL21 exhibits enhanced growth at 37°C compared to 25°C. The mean bacterial growth rate at 37°C was 1.756, whereas at 25°C, the average growth was 1.425, demonstrating that higher temperatures facilitate bacterial proliferation. However, due to the relatively small sample size, further validation with a larger dataset is required to confirm these findings. These findings highlight the critical role of temperature optimization in enhancing bacterial growth and improving recombinant protein production efficiency, emphasizing the need for further investigation with a larger dataset to validate the results. Abstract

Introduction: The ongoing SARS-CoV-2 pandemic has profoundly emphasized the pressing need for accurate and reliable diagnostic procedures. Given the potential health risks associated with nasopharyngeal swabs, there has been growing interest in seeking alternative diagnostic mediums. In this context, our study delved into evaluating saliva as a potential diagnostic tool, simultaneously assessing its efficiency in relation to patient demographics and their exhibited clinical symptoms. Methods: Spanning from May to December 2020, we conducted a comprehensive cross-sectional analysis. We meticulously examined medical records to gather insights on patient characteristics, existing health conditions, onset of symptoms, clinical manifestations, and compared the results obtained from both salivary and nasopharyngeal RT-PCR tests for SARS-CoV-2. Results: Among the individuals suspected of SARS-CoV-2 infection, the mean age stood at 52.4 years, with males representing 60.3% of this group. Interestingly, a significant 76.9% reported underlying health conditions, predominantly hypertension and diabetes. The most commonly reported symptoms encompassed respiratory challenges, notably coughing and shortness of breath, succeeded by symptoms like nausea, fever, and a general sense of fatigue. The performance of saliva tests, in terms of accuracy, appeared to be significantly influenced by the timing of symptom emergence. Conclusion: The RT-PCR tests utilizing saliva samples demonstrated considerable promise, especially during the early stages of symptom manifestation, providing a reliable alternative to traditional nasopharyngeal swabs. The findings suggest a superior diagnostic sensitivity when utilizing saliva during the initial phases of a SARS-CoV-2 infection.

Abstract

Analisis produk unggulan pada sektor pertanian pada kantor Dinas Pertanian Kabupaten Maros belum adanya aplikasi tersendiri dalam melakukan analisis produk unggulan atau masih diterapkannya proses manual. Tujuan dari penelitian ini adalah untuk menghasilkan analisis, pengujian dan menerapkan sistem di setiap wilayah Kabupaten Maros, dan mengembangkannya menggunakan analisis Location Quotient dan Shift Share. Metode dari penelitian ini adalah menggunakan RAD (Rapid Aplication Development). Sumber data penelitian berupa data PDRB sektor pertanian. hasil pengujian Black Box yang dilakukan untuk admin dapat menginput dan mengubah data pada menu Kecamatan, Kabupaten, komoditas serta pendapatan pada sistem, halaman user dapat mengetahui perkembangan sektor pertanian dengan metode Location Quotient dan Shift Share. hasil dari Kecamatan dijadikan wilayah studi dan daerah Kabupaten Maros dijadikan pembanding atau wilayah referensi yaitu dari perhitungan LQ Kec Simbang tahun 2020 produk padi diperoleh hasil 1,253, LQ>1 = Sektor Basis. Kesimpulan adalah dengan adanya aplikasi ini dapat mempermudah instansi di Dinas Pertanian mengolah data dan masyarakat Kabupaten Maros melihat informasi produk unggulan sektor pertanian.

Abstract

Indonesia merupakan salah satu negara dengan konsumsi mi instan terbesar. Permintaan impor gandum meningkat seiring dengan meningkatnya konsumsi mi berbahan tepung terigu. Tepung sagu direkomendasikan untuk digunakan sebagai bahan baku mi. Kandungan gizi ikan dapat meningkatkan nilai gizi produk mi. Penelitian ini bertujuan untuk menentukan konsentrasi tepung ikan cakalang yang optimal untuk mi kering berbahan tepung sagu berdasarkan karakteristik kecerahan dan kesukaan konsumen. Mi kering berbahan dasar sagu terdiri dari lima perlakuan, yaitu kontrol, tepung ikan cakalang 2, 4, 6, dan 8%. Parameter yang dianalisis meliputi lightness dan organoleptik. Perlakuan terbaik kemudian dianalisis kadar protein, abu, dan mikrostruktur. Penambahan tepung ikan cakalang memberikan pengaruh yang nyata (p<0,05) terhadap parameter lightness, aroma, dan keseluruhan. Lightness, aroma, dan nilai keseluruhan mi tepung ikan cakalang 8% lebih baik dibandingkan dengan kontrol dan perlakuan lainnya. Mi kering dengan penambahan tepung ikan cakalang 8% memiliki aroma harum, warna agak kurang bening, rasa mi enak, dan tekstur mendekati kenyal sehingga masih dapat diterima panelis. Perlakuan mi dengan penambahan tepung ikan cakalang 8% menghasilkan kadar protein tinggi, yaitu 5,56% (bk) dan kadar abu 1,12% (bk). Mi dengan penambahan tepung ikan cakalang 8% memiliki butiran elips agak terpotong, sedangkan mi gandum memiliki butiran elips.
