Abstract

Zika virus caused of the emerging infections characterized by fever, Guillain-Barré syndrome (GBS) for adults. In the current work, we aimed to study the binding orientation of 1,5-benzothiazepine compounds as new potential agent against Zika virus inhibitor through molecular docking and molecular dynamic simulation. Since, 1-5-Benzothiazepines are particular interest for drug discovery and they also has some biological activities. However, their antiviral activities and in silico studies of the binding to their biological targets have not been extensively investigated. Molecular docking study of 1,5-benzothiazepine chalcone derivatives compounds with protein target 5GJB (PDB ID) and this protein was taken from the crystallographic structure. In this study, twelve 1,5-benzothiazepine chalcone derivative compounds were docked to the protein with the grid box along x, y and z radius of 26.85, 28.17 and 24.43 Å, respectively. Suramin was used as positive control. Thus, it can be used as a reference for design new inhibitors for Zika virus helicase. Based on the docking results, it is observed that compounds MA3 and MA8 are estimated to have activity as inhibitors for Zika virus helicase with binding free energy values of -4.6490 and -4.9291 kcal/mol, respectively. MA3 and MA8 were also stable during the MD simulations with the hydrogen bonding are still maintained before and after MD simulation. Furthermore, both of these compounds can be used an early stage for drug design and drug delivery process.

Abstract

The sweet orange (Citrus sinensis (L.) Osbeck) is a popular fruit. The part of the sweet orange that is consumed is only the fruit flesh, while the orange peel is only thrown away and not utilized. The sweet orange peel contains several secondary metabolites, namely alkaloids, flavonoids, tannins, phenols, saponins, and steroids, which have antimicrobial properties. This study aims to determine the effect of the concentration of the ethyl acetate fraction of sweet orange peel on antibacterial and antifungal activity in Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Salmonella typhi, and Candida albicans fungi using the disc diffusion method. The ethyl acetate fraction was prepared in several concentration series, namely 50%, 25%, 12.5%, 6.25%, 3.125%, and 1.5625% w/v. The results obtained showed that all concentrations of the ethyl acetate fraction series showed activity in inhibiting the growth of Salmonella typhi bacteria. Series concentrations of 3.125% to 50% have activity in inhibiting the growth of Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, and Candida albicans fungi. The results of the data analysis using statistical one-way ANOVA showed that the antibacterial and antifungal activities between the concentration series were significantly different (p<0.05) from the negative control.

Abstract

The Bark of Marpuyan Tree (Rhodamnia cinerea Jack) is a plant that has been empirically used by communities in Indonesia for the treatment of infectious diseases caused by bacteria and fungi. The bark of Marpuyan contains flavonoids, phenolics, and saponins. This study aimed to evaluate the antibacterial and antifungal activities of the ethanol extract of the bark of Marpuyan using the disc diffusion method at various extract concentrations, namely 30%, 25%, 15%, 10%, and 5%. The results of the antibacterial activity test against Gram-positive bacteria (Staphylococcus aureus, Staphylococcus epidermidis, and Bacillus subtilis) showed inhibitory zones with respective diameters of 11.67±0.21 mm, 12.29±0.43 mm, and 12.78±0.14 mm. These results indicate weak antibacterial activity. In the antibacterial activity test against Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa, and Salmonella typhi), inhibitory zones were found with respective diameters of 13.05±0.05 mm, 12.58±1.78 mm, and 10.98±0.21 mm. These results also indicate weak antibacterial activity. However, in the antifungal activity test, no activity was found against Candida albicans and Malassezia furfur. Nevertheless, the ethanol extract of Marpuyan bark showed moderate antifungal activity against Trichophyton mentagrophytes, with an inhibitory zone diameter of 15.44±2.02 mm.

Abstract

Green glutinous rice tape is a fermented food made from glutinous rice, katuk leaves (Sauropus androgynus), and yeast, originating from Tembilahan, Riau Province. This product can serve as a source of lactic acid bacteria (LAB) capable of producing antimicrobial compounds to inhibit pathogenic microorganisms, including Aspergillus flavus, a well-known spoilage fungus in food products. However, to date, no reports have explored the ability of LAB from green glutinous rice tape to inhibit the growth of Aspergillus flavus. This study aimed to isolate LAB from green glutinous rice tape, conduct macroscopic, microscopic, and biochemical characterizations, and evaluate the antifungal activity of the LAB isolates against Aspergillus flavus using the good diffusion method. The isolation process yielded four LAB isolates from Tembilahan green tape (TKHT-2, TKHT-3, TKHT-5, and TKHT-7), which were identified as members of the genus Lactobacillus sp., with antifungal activities of 25.50 ± 5.78 mm, 24.33 ± 0.62 mm, 22.16 ± 6.56 mm, and 18.66 ± 4.28 mm, respectively. The corresponding cell-free supernatants (CFS) from these isolates (TKHT-S2, TKHT-S3, TKHT-S5, and TKHT-S7) also demonstrated antifungal activity with inhibition zones of 19.83 ± 3.47 mm, 19.83 ± 4.24 mm, and 14.50 ± 3.26 mm, respectively. LAB cells and their cell-free supernatants partially inhibited Aspergillus flavus, indicating a fungistatic effect. These findings suggest the potential application of LAB from green glutinous rice tape as a natural preservative or antifungal agent in food products.
