Abstract

This research was conducted in March-April 2017 at Marine Microbiology Laboratory, Faculty of Fisheries and Marine University of Riau. The purpose of this study is to determine the different concentration of detergent (0 ml/L, 1.5 ml/L, 3 ml/L, 4.5 ml/L, 6 ml/L) in different observation time (0, 5, 10, 15, 20) on the growth of heterotrophic bacteria in sea water. Completely randomized design was used in this experimental method. The results showed that bacterial growth of all treated samples decreased on the 5th day of incubation. However, the population began to increase on the 10th day of incubation. The count of maximum bacterial growth was 1.46 x 109 found in the 4.5 ml/L treated detergent, and the lowest growth was 3.73 x 107 in the 1.5 ml/L treated detergent. Statistical analysis (ANOVA) showed that the concentrations in different observation times on the growth of heterotrophic bacteria showed significant effect and the value was (P <0.005).Keywords: heterotrophic bacteria, population growth, rinso, sea water.

Abstract

ABSTRACT Pseudomonas sp. is a hydrocarbonoclasticbacteria that is capable of degrading various types of hydrocarbons. Pseudomonas sp.including bacteria that have the potential to become pathogens. This research was conducted in April to May 2018. This study aims to calculate the density of Pseudomonas sp.bacteriaand heterotrophic bacteria in Dumai sea waters, comparison of the desity of Pseudomonas sp. bacteria to heterotrophic bacteria at each station and the resistance of Pseudomonas sp. to antibiotics. The result showed that densityPseudomonas sp. ranged from 2.25 x 105- 6.08 x 105 CFU/ml. The highest number of Pseudomonas sp.at station 1 and lowest at station 5. The total number of heterotrophic bacteria ranged from 7.37 x 106 - 10.38 x 106 CFU/ml. The highest number of heterotrophic bacteria at station 4 and the lowest at station 5.The highest percentage rasioof density of Pseudomonas sp. bacteri to heterotrophic bacteria was in station 1 (6.26%) and the lowest was in station 4 (2.86%). The results of isolation of bacteria obtained 25 pure isolates and tested for bacterial resistance to antobiotics. Results of isolation bacteria obtained 25 pure isolates and tested the resistance ofPseudomonassp. bacteriato antibiotics.Chloramphenicolantibiotic test results showed 2 isolates classified as sensitive with inhibition zone of 21,3 – 21,9 mm, 1 isolate including resistance with 9,7 mm inhibition zone, with a 22 isolates classified as intermediate with a inhibition zone of 9.7 – 19.6 mm. Two other antibiotics Penicillin and Isoniazid, all bacterial isolates were resistant with the inhibition zone range from 1.2 – 4.6 mm and 1 – 2.8 mm respectively Keyword: Density, Pseuomonas sp.,Heterotrophic Bacteria, Antibiotics

Abstract

                                  AbtrakAzolla microphyllais a water fern that floats and rich of protein, essential amino acids, vitamins and minerals. The ferncan be found in ponds, ditches,warm temperature fields and tropical regions throughout the world. Research on microscopic analysis the growth of Azolla microphylla in brackish water was carried out from April to June 2018. A. microphylla was grown in containers with a volume capacity of 20 liters. The container was filled with planting media, then filled with brackish water to a volume of 10-15 liters and this ecosystem is set to 0 ‰, 1 ‰, 2 ‰ and 3 ‰. The growth of A. microphylla was observed by measuring biomass growth, colony growth, and double time growth, microscopic observations of leaves, rhizomes and rootsA. microphylla. From the results of the study it was found that A. microphylla can grew in brackish water (salinity 1‰, 2‰ and 3‰), however growth tends to be slower than in fresh water (salinity 0 ‰), and also can be seen based on the cross section of leaves, roots and rhizomes that grew optimally at salinity 0 ‰, and experience changes in shape and color in the cross section and do not grew optimally at salinity 1‰, 2‰ and 3‰.Keywords: Azolla microphylla growth, microscopic analysis, brackish water

Abstract

The use of herbal supplements is an alternative to increase food security, because it can stimulate fish appetite and increase fish immunity against disease. The aims of this research to know the total leukocytes, leukocyte differentiation and survival rate of striped catfish fed with dosage fermented herbal supplements, and to obtain the best dosage. The implementation of research was from June until August 2019 at the Parasites and Fish  Diseases Laboratory, Faculty of Fisheries and Marine, Riau University. The method used is a completely randomized design (CRD), with four treatments and three replications, namely P0 (without the addition of herbal supplements), P1 (herbal supplement dose 100 mL / kg of feed), P2 (200 mL / kg of feed), and P3 (300 mL / kg of feed). Maintenance of fish is carried out for 60 days, with a stocking density of 50 fish / m. The results showed that the herbal supplementation at a dose of 200 mL / kg of feed (P2) was the optimal dosage in stimulating total leukocytes and differentiation of catfish leukocytes. P2 treatment resulted in total leukocytes of 11.26 x 104 cells/mm3, further differentiation of leukocytes such as lymphocytes, monocytes, neutrophils and platelets of 81.67%, 6.33%, 5.67% and 6.33%. Abstract

Peningkatan keamanan pangan menggunakan bahan alami, seperti kunyit, kencur, dan temulawak sebagai suplemen pada pakan disebabkan karena bahan tersebut memiliki sifat yang ramah lingkungan, biodegradable, dan mudah ditemukan di alam. Penelitian ini bertujuan untuk mendapatkan dosis suplemen herbal dan padat tebar yang efektif dalam meningkatkan status kesehatan ikan ditinjau dari profil hematologis. Rancangan yang digunakan adalah Rancangan Acak Lengkap pola Faktorial. Perlakuan yang diberikan adalah dosis suplemen herbal (D1= 100 mL/kg pakan, D2= 200 mL/kg pakan, dan D3 = 300 mL/kg pakan) dan padat tebar ikan P1 = 50 ekor/ m3 dan P2 = 75 ekor/ m3. Ikan patin (Pangasius hypophthalmus) yang digunakan berukuran 8-12 cm, pemeliharaan dilakukan selama 60 hari di keramba jaring apung berukuran 1,5x1x1m. Pengambilan sampel darah dilakukan sebanyak tiga kali, yakni sebelum perlakuan, kedua pada hari ke-30 dan hari ke-60 pemeliharaan. Hasil penelitian menunjukkan bahwa penambahan dosis suplemen herbal 200 mL/kg pakan dan padat tebar 75 ekor/m3 yang dipelihara selama 60 hari memberikan hasil tertinggi terhadap profil hematologis, yaitu total eritrosit sebesar 2,81 x 106 sel/mm3, nilai hematokrit 39,00%, kadar hemoglobin 10,73 g/dL, total leukosit 11,41 x 104 sel/mm3 dan kelulusanhidup 100%. Hasil penelitian dapat disimpulkan bahwa kombinasi padat tebar 75 ekor/m3 dan dosis suplemen herbal 200 mL/kg pakan adalah yang terbaik untuk peningkatan kesehatan ikan patin.

Abstract

One of the interesting and environmentally friendly microbiology strategies and approaches to control the impact of microplastics is to approach bioremediation technology by harnessing the potential of microbes or indigenous bacteria (local bacteria).  Dumai sea waters currently show a high enough of microplastic pollution which allows the potential of indigenous bacteria to adapt to a plastic environment. The purpose of this study is to isolate and identify indigenous bacteria to degrade plastics from the sea waters of Dumai and to know whether or not there is a difference in the number of bacteria found between stations in this study. This research was conducted in October-December 2020 with experimental methods at the Marine Microbiology Laboratory Faculty of Fisheries and Marine Science Universitas Riau. Based on the results of the study, 12 bacterial isolates were isolated from the research stations.  Isolates of these bacteria have diamaters ranging from 0.2-1.1 cm. Microplastic degradation test results by bacteria found that ISL 10 is an isolate that shows the highest PET degradation activity, which is 17.27% and the diameter of biofilm formation 0.8 cm. Based on biochemical and morphological tests, similar results were obtained that ISL 10 bacteria are a bacterium of the genus Bacillus. The most bacterial colonies were seen in statiun IV (TPI) with an average number of bacteria of 214.9 x 104 CFU/ml.

Abstract

The mangrove forest ecosystem in Sungai Kayu Ara Village, Sungai Apit Subdistrict, Siak Regency, Riau Province has a unique or distinctive appeal, which has the potential to be developed as a mangrove forest ecotourism destination. The research objective was to analyze the potential for developing mangrove forest ecotourism in Sungai Kayu Ara Village in realizing sustainable mangrove forest conservation. The method used in this research is the survey method, which was carried out in May-July 2020. Mangrove density data was taken using the line transect plot technique, which consists of 3 observation transects. Each transect has 3 plots, each plot measuring 10 x 10 m2. The identification results found 6 types of mangroves that grow in the mangrove forest area of Sungai Kayu Ara Village, namely Rhizophora apiculata, Avicennia alba, Bruguiera gymnorrihiza, Sonneratia alba, Nypa fruticans, and Xylocarpus granatum. The mangrove density value is 1234 ind / ha. The average value of the Tourism Suitability Index (IKW) is 76.75%. (S2 category), which is suitable for development as a mangrove forest ecotourism area. Mangrove forest ecotourism development strategies based on SWOT analysis include 1) Increasing stakeholder cooperation for mangrove forest conservation; 2) Developing facilities and infrastructure for the development of mangrove forest ecotourism; 3) Provision of information and promotion of manrove ecotourism through print and electronic media; and 4) Increased quality of skilled and skilled human resources for the development of mangrove forest ecotourism.

Abstract

ABSTRACT The research was conducted from July to September 2017 in Estuary of Siak River. The purpose of this research is to examine thepopulation of heterotrophic bacteria in Estuary of Siak River, and heterotrophic bacteria antagonism against pathogenic bacteria (Vibrio sp, Aeromonas sp and Pseudomonas sp). And than identified by phenotype and genotype. This research use survey method. The samples analisis were conducted at the Marine Microbiology Laboratory of Marine Science Department of Fisheries and Marine Faculty and DNA analisis conducted at the Genetic Laboratory of Biology Department Mathematic and Natural Sciences. The result of antagonism test from 25 isolate heterotrophic bacterial was found 10 isolates which have ability to inhibit the growth of pathogenic bacteria (Vibrio sp, Aeromonas sp and Pseudomonas sp). From Vibrio sp bacteria the mean of clear zone is 1P11 isolate of 20,3 mm and the smallest is 1P19 isolate of 5 mm. Aeromonas sp bacteria the largest clear zone is 2P20 of 9,3 mm and the smallest is 1P12 isolate of 3,3 mm. Pseudomonas sp bacteria the largest clear zone is 1P12 isolate of 17 mm and the smallest is 1P11 and 2P22 isolate of 8,3 mm. the result of molecular identification using PCR 16S rRNA method and BLAST analisis showed 10 isolates of heterotrophic bacteria having kinship with bacteria of the genus Bacillus that is Bacillus cereus (1P11, 1P12, 1P14, 1P15, 1P19, 2P20, 2P21, 2P23), Bacillus toyonensis (2P22) and Virgibacillus salarius (1P18). Keyword : Heterotrophic Bacteria, Isolation, Antagonism test, Identification

Abstract

ABSTRACT Lactic acid bacteria (BAL) have the ability to inhibit the growth of pathogenic bacteria. BAL is bacterium that has an important role in the processof fermentation of organic materials. BAL can be found in traditional fishery products such as cincaluk (fermented shrimp). This study was conducted from February to April 2018. Test of isolate activity of BAL and protease enzyme was carried out at Marine Microbiology Laboratory, Faculty of Fisheries and Marine Scince, University of Riau.The study aimed was to examine the BAL activity of cincaluk against pathogenic bacteria and tested the protease enzyme produced by BAL from the cincaluk produced the community in Bengkalis and Tembilahan, Riau. This research used survey method where fresh cincaluk sample was used as abservation object in isolating the BAL and testing antagonism against three different pathogenic bacteria (Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus). The results of antagonism test showed that all bacterial isolated have ability to inhibit the growth of the three pathogenic bacteria. The highest inhibition zone P.aeruginosawas demonstrated by a GT isolate (11.0 mm), toward the highest inhibition zone against E. coliwas indicated by PB isolate (8.7 mm), and toward S. aureuswasdemonstrated by MB isolate (6.6 mm). All bacterial isolates produced protease enzyme indicated by clear zone around the colonies on skim milk medium. Keywords: BAL, Antagonism, Inhibition zone, Pathogens, Protease

Abstract

Lactic acid bacteria (LAB) has been applied as probiotic mainly to prevent pathogens. This group of bacteria can be found in marine fish products including fermented, shrimp product, cincaluk. The objective of this research was to isolate LAB from cincaluk, and examined the activity against pathogen V. alginolyticus and A. hydrophila. The research was conducted on March-Juny, total of 12 isolates were obtained from cincaluk survey method. The result of research showed that all isolates round, creamy white, smooth-edged, raised surface, and the size is 1.0 to 3.0 mm. The physiological and biochemical test showed that get all isolates had a Gram-positive, catalase positive and are indole negative, 10 isolates were motile, as many as five isolates have citric positive, 10 isolates is Methyl Red (+), all isolates ferment sucrose and lactose, while only 9 isolates ferment glucose, and as much as 6 isolates that produce sulfide. All isolates were found to have inhibitory effects against bacteria V. alginolyticus and A. hydrophila. The highest antimicrobial activity is indicated by B isolates with an average diameter of 12.3 mm and inhibition zone antivibrio activity of 6.3 units. Isolates B with an average diameter of inhibition zone of 9.7 mm and 3.7 units antiaeromonas activity.Keywords: Isolation, Lactic Acid Bacteria, Cincaluk, V. alginolyticus, A. hydrophila
