Abstract

The research was conducted from February 2016 to April 2017 and was aimed to determine the effect of adding different concentrations of detergent on the growth of bacterial populations in the ocean water column and to see the ratio of bacterial persisted capability in different lengths of dissection. This research used experimental method with Completely Randomized Design. Seawater samples were analyzed at Marine Microbiology Laboratory, Fisheries and Marine Faculty of Riau University. Based on the results of the study, from day 0 untill day 10 after given detergent with different concentrations, bacteria experienced a decreasing trend of growth, in day 15 untill day 20, the trend of bacterial growth contaminated by detergent with different concentration relatively increased to stagnant. Based on the result of this study the addition of different detergent concentration in each treatment affect the growth of heterotrophic bacteria population in the sea water, where the higher concentration of detergent added the lower the ability of bacteria to grow and the ability of heterotrophic bacteria to survive at different concentrations of detergent, it can be concluded that the ability of heterotrophic bacteria to grow from days 0 to 20 has decreased in each of its concentrations.   Keywords: heterotrophic bacteria, Surf detergent.

Abstract

 ABSTRACT                Microalgae are very potential to be developed as biofuel feedstocks when compared to food crops because they have several advantages such as rapid growth, high productivity, enabling the use of freshwater and seawater, and production costs are not too high. Spirulinaplatensis has a high nutrient content so it is used as a health food ingredient. Once the importance of the role of nutritional value of S. platensis for humans and some marine organisms, then the right culture medium to get the maximum nutritional content needs to be studied more deeply. This study aims to determine the effect of different doses of nutrition on the density of biomass cultivation of S. platensis microalgae with indoor and semi outdoor system. This research has been conducted from April to May 2018. The method by used the experimental method. The data obtained were analyzed statistically using simple linear regression equation. The results showed that the influence in the dose of nutris media guillard, S.platensis increased with the dose of nutrients as much as 0.60 ml either indoor or semi outdoor system. The relationship experienced between biomass with different doses of nutrients is a positive medium with both indoor and semi-outdoor systems, this can be seen from the results of simple linear regression test, namely r = 0.3443 for indoor system and r = 0.4492 for semi-outdoor system. As for the amount of biomass density S. platensis in the semi-outdoor system the amount of biomass density is higher than the indoor system.Key words: Microalga, Spirulina platensis, Sediment, Media Guillard

Abstract

ABSTRACT               Azolla michrophylla is beneficial for Anabaena azollae because it can act as a host through the supply of oxygen and nitrogen that can be used. Nitrogen fixation carried out by A. Azollae is atmospheric nitrogen gas (N2) converted into ammonia (NH3). This research was conducted at the Laboratory of Marine Microbiology and Marine Chemistry, Department of Mariene Science, Faculty Of Fisheries And Marine Science, University of Riau in April to June 2018. The purpose of this research was to determine the abundance of A. azollae in the growth of A. microphylla plants and determine the levels of nitrate contained in the growth of A. microphylla in brackish water culture. Factorial randomized group design (RAK) was applied in this experimental. Temperature, salinity, and pH were water quality parameters measured. The results showed that the best growth of A. microphylla was obtained at a salinity of 0 ppt with a starter of         A. microphylla 3 gram with a value of 50.42 gram. The highest abundance of A.  azollae is found in salinity 3 ppt with a 3 gram level with the abundance of 5643 cells / ml. A. microphylla and A. azollae had sufficient nitrate levels for their growth in brackish water cultured at 0, 1, 2, and 3 ppt salinitis of in combination with  A. microphylla 1, 1.5 and 3 gram as a starters. The highest nitrate level was obtained on the 14th and 21st day with a value of 1.2625 mg / L and 1.4500 mg / L.   Keywords: Azolla microphylla, Anabaena azollae, Growth, Abundance, Nitrate.

Abstract

This research was conducted on April 2017. Sampling was conducted in estuary of Dumai River, Dumai City, Riau Province. A study aims to determine the relationship between concentration of surfactant and population of heterotrophic bacteria in the estuary waters of Dumai River Dumai City Riau Province. Sampling was done at 6 sampling points (stations). The analysis of surfactant concentration was conducted at the Chemical Laboratory of the Sea and the total analysis of heterotrophic bacteria at the Marine Microbiology Laboratory of Department of Marine Science Faculty of Fisheries and Marine University of Riau, Pekanbaru. The results showed that the highest concentration of surfactant was found at Station 5 (0.8538 mg / l) and the lowest at Station 1 (0.2025 mg / l). Then, the logarithm of the highest average total colony was found at Station 1 (6.18 cells / ml) while the lowest at Station 5 (5.66 cells / ml). The correlation concentration models of surfactant with the total colony using regression was negative correlation Y = 6.3745 - 0.7882 (X), R2 = 0.8622, r = 0.9285 showed that the value of surfactant concentration (X) increased by 1 unit will decrease the total of colony (Y) was 0,7882 cells/ml, with the effect of surfactant to bacteria equal 85% and the level of closeness of the relationship was 93%.Kata Kunci : Surfactant, Heterotrophic bacteria, Estuary of Dumai River

Abstract

ABSTRACT                Heterotrophic bacteria are decomposing bacteria that have an important role in maintaining the continuity of the life cycle of biota in the sea and as a standard of fertility in the waters. Heterotrophic bacteria obtain sources of food, oxygen and energy derived from the results of the decomposition process of the rest of other organisms, waste, or substances contained in the bodies of other organisms. This study aims to determine the effect of adding different concentration and duration of surf detergent to the growth of heterotrophic bacteria in sea water. This research was conducted from April to May 2018 using the experimental method. The results showed that the bacterial population in each treatment decreased with increasing concentration of detergent. The results showed that the highest number of heterotrophic bacteria was found at a concentration of 0.60% at station 1 exposure on the 10th day at 6.60 x 107 (CFU / ml) and 0.90% at the station on the 10th day of the day exposure the 10th was 6.72 x 107 (CFU / ml) and the lowest was found at a concentration of 0.90% on day 5 exposure at station 1 at 3.83 x 107 (CFU / ml). The value of the correlation coefficient (r) obtained is 0.177. Significant treatment of detergent concentration is different, which is equal to 0.00 <(0.550) and duration of exposure which is equal to 0.00 <α (0.45). Based on the results of this study, there is a negative effect of the addition of surf detergent with different concentration and duration of exposure to the growth of heterotrophic bacteria in seawater. Keywords: heterotrophic bacteria, Surf detergent.

Abstract

Three candidates of probiotic bacteria isolated from kakap putih (Lates calcarifer) had been examined for their antagonism against Vibrio alginolyticus. However, the ability to suppress the pathogen in co-culture system has not been tested. This research aimed to test the ability of probiotic candidates to suppress the growth of V. alginolyticus in co-culture trial. Three probiotics candidates (KP1, KP2, and KP3) were used in this experiment method. Four treatments consisted of P1 (broth culture of KP1 + V. alginolyticus), P2 (KP2 + V. alginolyticus ), P3 (KP3 + V. alginolyticus), P4 (V. alginolyticus) as control were prepared in triplicate. Total counts of each probiotic candidate and V. alginolyticus was calculated after 48 hours culture incubation. KP1 isolate showed highest ability suppress the growth of V. alginolyticus, followed by KP3 and KP2 isolates.Keywords: Probiotics, Kakap Putih, Growth, V. alginolyticus, Co-culture

Abstract

This research aims to determine the effect of “Attack” detergent addition at different concentration and exposure time on heterotrophic bacterial growth in sea water. The research was conducted from February to April 2017 used experimental method. The results showed that the highest number of heterotrophic bacteria was observed on the A5 day 1st exposure treatment of 6,69 x 107 (CFU/ml) and the lowest was in the 16th day of exposure treatment of 0,098 x 107 (CFU/ml). At day 1st, the value of the correlation coefficient (r) obtained by 0.277 showed the relationship between two variables with positive linearity, while day 6th to day 21th, the correlation coefficient (r) are 0,982, 0,9426, 0,9778 and 0,986, showed the relationship between moderate two was very strong variables with negative linearity. The regression relationship between exposure time and bacterial growth showed a strong correlation with negative linearity, where the coefficient value (r) was 0,87. Significant treatment of detergent concentration that was equal to 0,00 and exposure time that was equal to 0,00 < α (0,05). The bacterial growth on treatment A1 (control) was significantly different with treatment A2, A3, A4 and A5. Bacterial growth at day 1st was significantly different from 6th, 11th, 16th and day 21th. The result of this research showed that there were a effect of “Attack” detergent addition with different concentration and exposure time to the growth of heterotrophic bacteria in seawater.Keyword: Detergent, Concentration, Exposure, Growth, Heterotrophic Bacteria

Abstract

The presence of antibiotic-producing bacteria from microhabitat in mangrove ecosystems can be useful to produce new antibiotics for industrial and environmental conservation. Mangrove ecosystems in Dumai have the potential to possess antibiotic-producing bacteria that have not been much studied. The objective of this research was to obtain antibiotic-producing bacteria from extreme microhabitats in the mangrove ecosystem in suppressing the growth of pathogenic bacterial pathogens (Vibrio alginolyticus). Samples were taken from several microhabitat mangrove areas in Dumai (Kelurahan Purnama Dumai), Riau Province. The survey method is used for identification, antibacterial test against pathogenic bacteria (V. alginolyticus). Bacterial isolation was carried out using the pour plate method and purification was carried out using the quadrant streak method. Identification is carried out through observations of macroscopic, microscopic morphology, biochemical tests, antagonism tests (agar diffusion method on agar nutrient media) and DNA analysis. Sequencing of 16S rDNA from the best antibiotic-producing bacterial isolates indicated that molecularly identified of Bacillus amyloliquefaciens, Bacillus cereus, Enterobacter hormaechei, Klebsiella pneumonia, and Enterococcus gallinarum. All bacteria were able to inhibit the growth of V. alginolyticus
