Abstract

Noni (Morinda citrifolia L.) is a traditional medicinal plant that is widely used in the treatment of various degenerative diseases. However, scientific data regarding the composition of its bioactive compounds are still limited, especially those analyzed using a high-precision instrument approach. This study aims to identify the phytochemical composition in 70% ethanol extract of noni fruit using Liquid Chromatography-Mass Spectrometry (LC-MS) method. Extraction was performed by Ultrasound-Assisted Extraction (UAE) method and compound analysis using Q-ToF type LC-MS instrument with positive ionization mode. The results showed that noni extract contained 28 compounds, with 13 compounds successfully identified consisting of 7 alkaloids, 3 glycosides, 1 phenolic, 1 amino acid, and 1 aminoglycoside. The major compound identified was Methyl (5R)-2,3,4-tris-O-(methoxymethyl)-5-{2-[(2R,3R,4R,5R,6R)-3,4,5-trihydroxy-6-(hydroxymethyl)-2-piperidinyl]ethyl}-α-D-xylopyranoside, belonging to the glycoside group with a molecular weight of 485.25 g/mol. These results indicate that noni fruit contains diverse secondary metabolite compounds with the dominance of alkaloid and glycoside compounds. This study contributes to providing primary data based on LC-MS technology on the chemical composition of Indonesian natural ingredients and can be a scientific basis for the development of medicinal plant-based pharmaceutical preparations with noni active ingredients.

Abstract

Quercetin are the main marker compounds in extracts of Sonchus arvensis which show nephrolithiasis activity. For quality control of herbal medicines, Densitometric Thin Layer Chromatography (TLC) method has been developed in this study using quercetin as phytochemical markers. Objective: Determination for quercetin in herbal medicines. Determination of quercetin by determining the composition of the mobile phase, the wavelength of the analysis, the saturation time, the elution distance and the volume of the spot. The parameters observed in the optimization method and stability test are area, retardation factor (Rf), peak shape and resolution (Rs). The composition of selected mobile phase which results good separation and symmetrical peak shape is chloroform: methanol: dichloromethane: acetonitrile: formic acid (6: 2: 2: 0.05: 0.05 v/v/v/v/v) with a wavelength of 335 nm, time of saturation is 30 minutes, elution distance is 8 cm, volume of spots is 10 µL and Rf 0.48. In this study, it was shown that there were no factors that had a significant effect so that the method could be applied for the determination of quercetin levels in herbal medicines.The research data showed that the range of levels on quercetin 3.26%.

Abstract

Latar belakang: Pisang kepok (Musa paradisiaca L.) merupakan salah satu buah yang banyak dikonsumsi karena daging buahnya memiliki rasa manis dan enak. Buah pisang kepok mengandung vitamin C yang dapat meningkatkan daya tahan tubuh dari radikal bebas. Vitamin C merupakan sumber elektron yang dapat menyumbangkan sebuah elektron kepada radikal bebas sehingga dapat merendam reaktivitas radikal bebas. Tujuan: mengetahui perbandingan kadar vitamin C pada buah pisang kepok dengan proses pematangan secara alami dan karbid. Metode: sampel yang digunakan dalam penelitian ini yaitu pisang kepok 1 sisir yang sudah tua namun masih berkulit hijau dibagi menjadi dua kelompok, satu kelompok matang alami sedangkan kelompok kedua ditambahkan karbid kemudian dibiarkan selama 2-3 hari. Uji kualitatif menggunakan 3 perlakuan berbeda yaitu KMnO4, NaOH, FeSO4, dan iodin serta pada penetapan kadar vitamin C menggunakan spektrofotometri UV-Vis. Hasil: sampel menunjukkan bahwa pisang kepok matang alami memiliki kulit buah berwarna kuning cerah, rasa manis dan daging buah lembut sedangkan pisang kepok matang karbid memiliki kulit buah berwarna kuning pucat, dan rasa yang hambar. Simpulan: Hasil kadar vitamin C pada buah pisang kepok matang alami sebesar 1,9668 mg/100g dan matang karbid sebesar 1,6742 mg/100g
