Abstract

Background: Quercetin, luteolin, sinensetin and stigmasterol each is the main marker compound in extracts of Sonchus arvensis, Plantago major, Orthosiphon stamineus, and Strobilanthes crispus, respectively. These extracts show nephrolithiasis activity. For quality control of herbal medicines, a high performance liquid chromatography (HPLC) method has been developed in this study using quercetin, luteolin, sinensetin and stigmasterol as phytochemical markers. Objective: to show optimal conditions of analysis and evaluate the stability of quercetin, luteolin, sinensetin and stigmasterol. Methods: The optimal conditions for analysis were carried out by determining the composition of the mobile phase, the flow rate, and the detector's wavelength. Zorbax Eclipse Plus C18 150 x 4.6 mm, 5 μm was used as the column. The stability test was done by analyzing the standard and samples stored at 4oC for 0, 3, 6 and 24 hours. Results: The best separation of the extract was achieved under isocratic conditions using a mixture of water: methanol: phosphoric acid: acetic acid : acetonitrile (50: 30: 0.05: 0.05: 20 v/v/v/ v/v) as mobile phase with detector wavelength of 352 nm, a mobile phase flow rate of 1 mL/min, and a sample injection volume of 10 μL. Conclusion: In this study, the optimal condition for analysis of quercetin, luteolin, sinensetin and stigmasterol. Quercetin, luteolin, sinensetin and stigmasterol were not stable during 6 hours storage, therefore, standard solutions and samples should be made fresh to maintain the stability.

Abstract

Quercetin are the main marker compounds in extracts of Sonchus arvensis which show nephrolithiasis activity. For quality control of herbal medicines, Densitometric Thin Layer Chromatography (TLC) method has been developed in this study using quercetin as phytochemical markers. Objective: Determination for quercetin in herbal medicines. Determination of quercetin by determining the composition of the mobile phase, the wavelength of the analysis, the saturation time, the elution distance and the volume of the spot. The parameters observed in the optimization method and stability test are area, retardation factor (Rf), peak shape and resolution (Rs). The composition of selected mobile phase which results good separation and symmetrical peak shape is chloroform: methanol: dichloromethane: acetonitrile: formic acid (6: 2: 2: 0.05: 0.05 v/v/v/v/v) with a wavelength of 335 nm, time of saturation is 30 minutes, elution distance is 8 cm, volume of spots is 10 µL and Rf 0.48. In this study, it was shown that there were no factors that had a significant effect so that the method could be applied for the determination of quercetin levels in herbal medicines.The research data showed that the range of levels on quercetin 3.26%.

Abstract

Daun patikan kebo merupakan tanaman yang mengandung senyawa metabolit sekunder golongan flavonoid, terpenoid, tanin dan fenolik. Penelitian ini bertujuan untuk mengetahui uji perbandingan pelarut metanol dan etanol 96% terhadap hasil kromatografi lapis tipis flavonoid pada tanaman daun patikan kebo (Euphorbia hirta L.). Metode daun patikan kebo diekstraksi dengan maserasi dengan pelarut yang digunakan yaitu metanol dan etanol 96%. Skrining dilakukan menggunakan reagen Pb Asetat 10% dan NaOH 20%. Kromatografi lapis tipis (KLT) menggunakan fase gerak n-butanol : asam asetat glasial : air (4:1:5) dan fase diam plat silika gel G60 F254. Hasil skrining fitokimia yaitu negatif pada penambahan reagen Pb Asetat 10% dan hasil positif pada penambahan reagen NaOH 20%. Pada hasil penelitian dianalisis terjadi perubahan warna pada plat KLT dari biru menjadi fluoresensi nyala biru muda. Kesimpulan didapatkan nilai Rf ekstrak metanol empat bercak flavonoid dengan nilai Rf 033, 0,47, 0,61 dan 0,88, sedangkan ekstrak etanol 96% terdapat tiga bercak flavonoid dengan nilai Rf 0,29, 0,46 dan 0,59. Abstract

Dengue Hemorrhagic Fever is an infectious disease caused by the Dengue virus through the Aedes aegypti mosquito vector, which is controlled both at the larval and adult stages. The use of chemicals that are difficult to degrade in nature can pollute the environment. The Paitan plant (Tithonia diversifolia) contains secondary metabolite compounds that have the potential to be natural larvicides. This study aims to determine the natural larvicidal activity of Paitan leaf infusion. This study is an experimental study. The results of this study indicate that (1) Paitan leaf infusion contains alkaloid, flavonoid, terpenoid, saponin, phenolic, and tannin compounds. At concentrations of 1.8%, 3.2% and 5.6%, the mortality percentage results were 41.30%, 50.60% and 60% respectively; (2) The administration of Paitan leaf infusion has an effect on morphological damage Aedes aegypti larvae in the thorax, digestive tract and siphon. The content of different metabolite compounds causes different levels of morphological damage at each concentration

Abstract

Pluchea indica L. is a herbaceous plant of the Asteraceae family that has been known to the public as fresh vegetables and traditional medicine since ancient times. This study aims to determine the antioxidant activity of beluntas leaves obtained from the extraction using 70% ethanol and ethyl acetate. The identification of phytochemical screening of beluntas leaves revealed the presence of flavonoids, tanninsand alkaloids. Abstract

Lip tint merupakan salah satu jenis kosmetik bibir yang memiliki tekstur ringan dan cair. Salah satu zat utama dalam lip tint adalah zat warna. Bunga rosela (Hibiscus sabdariffa L.) dapat dimanfaatkan sebagai bahan baku pewarna alami karena mengandung senyawa antosianin. Tujuan dari penelitian ini untuk mengetahui kandungan total antosianin pada lip tint yang diformulasi dengan ekstrak bunga rosela sebagai pewarna alami serta membandingkan kadar total antosianin pada ekstrak bunga rosela dan lip tint. Metode analisis untuk penetapan kadar total antosianin dilakukan secara spektrofotometri UV-Vis sesuai Farmakope Herbal Indonesia II. Hasil pengujian diperoleh kadar total antosianin pada ekstrak bunga rosela dan lip tint masing-masing 5,9371 mg ⁄ L ± 0,30 dan 7,5568 mg ⁄ L ± 0,15. Perbandingan kadar total antosianin diuji statistik menggunakan uji T berpasangan dan diperoleh nilai sig. 0,038. Kesimpulan penelitian ini yaitu kadar total antosianin dalam lip tint lebih tinggi dibandingkan ekstrak murninya. Lip tint is a type of lip cosmetic with a light and liquid texture. One of the main components in lip tint is a coloring agent. Roselle flower (Hibiscus sabdariffa L.) can be utilized as a natural coloring ingredient due to its anthocyanin content. This study aims to determine the total anthocyanin content in lip tint formulated with roselle extract as a natural colorant and compare the total anthocyanin levels between roselle extract and lip tint. The analysis method for determining total anthocyanin content was conducted using UV-Vis spectrophotometry according to the Indonesian Herbal Pharmacopoeia II. The results showed that the total anthocyanin content in roselle extract and lip tint was 5.9371 mg/L ± 0.30 and 7.5568 mg/L ± 0.15, respectively. The comparison of total anthocyanin levels was statistically analyzed using a paired t-test, resulting in a significance value of 0.038. The conclusion of this study is that the total anthocyanin content in the lip tint increased compared to its pure extract. Abstract

Noni (Morinda citrifolia L.) is a traditional medicinal plant that is widely used in the treatment of various degenerative diseases. However, scientific data regarding the composition of its bioactive compounds are still limited, especially those analyzed using a high-precision instrument approach. This study aims to identify the phytochemical composition in 70% ethanol extract of noni fruit using Liquid Chromatography-Mass Spectrometry (LC-MS) method. Extraction was performed by Ultrasound-Assisted Extraction (UAE) method and compound analysis using Q-ToF type LC-MS instrument with positive ionization mode. The results showed that noni extract contained 28 compounds, with 13 compounds successfully identified consisting of 7 alkaloids, 3 glycosides, 1 phenolic, 1 amino acid, and 1 aminoglycoside. The major compound identified was Methyl (5R)-2,3,4-tris-O-(methoxymethyl)-5-{2-[(2R,3R,4R,5R,6R)-3,4,5-trihydroxy-6-(hydroxymethyl)-2-piperidinyl]ethyl}-α-D-xylopyranoside, belonging to the glycoside group with a molecular weight of 485.25 g/mol. These results indicate that noni fruit contains diverse secondary metabolite compounds with the dominance of alkaloid and glycoside compounds. This study contributes to providing primary data based on LC-MS technology on the chemical composition of Indonesian natural ingredients and can be a scientific basis for the development of medicinal plant-based pharmaceutical preparations with noni active ingredients.

Abstract

Latar belakang: Pisang kepok (Musa paradisiaca L.) merupakan salah satu buah yang banyak dikonsumsi karena daging buahnya memiliki rasa manis dan enak. Buah pisang kepok mengandung vitamin C yang dapat meningkatkan daya tahan tubuh dari radikal bebas. Vitamin C merupakan sumber elektron yang dapat menyumbangkan sebuah elektron kepada radikal bebas sehingga dapat merendam reaktivitas radikal bebas. Tujuan: mengetahui perbandingan kadar vitamin C pada buah pisang kepok dengan proses pematangan secara alami dan karbid. Metode: sampel yang digunakan dalam penelitian ini yaitu pisang kepok 1 sisir yang sudah tua namun masih berkulit hijau dibagi menjadi dua kelompok, satu kelompok matang alami sedangkan kelompok kedua ditambahkan karbid kemudian dibiarkan selama 2-3 hari. Uji kualitatif menggunakan 3 perlakuan berbeda yaitu KMnO4, NaOH, FeSO4, dan iodin serta pada penetapan kadar vitamin C menggunakan spektrofotometri UV-Vis. Hasil: sampel menunjukkan bahwa pisang kepok matang alami memiliki kulit buah berwarna kuning cerah, rasa manis dan daging buah lembut sedangkan pisang kepok matang karbid memiliki kulit buah berwarna kuning pucat, dan rasa yang hambar. Simpulan: Hasil kadar vitamin C pada buah pisang kepok matang alami sebesar 1,9668 mg/100g dan matang karbid sebesar 1,6742 mg/100g
