Abstract

Diabetes melitus merupakan penyakit tidak menular yang jumlah penderitanya terus meningkat dan dapat menimbulkan berbagai komplikasi apabila tidak dicegah sejak dini. Salah satu tantangan dalam pengendalian penyakit ini adalah masih terbatasnya pengetahuan masyarakat mengenai faktor risiko dan pentingnya deteksi awal. Kegiatan pengabdian kepada masyarakat ini bertujuan meningkatkan pemahaman masyarakat tentang faktor risiko dan upaya pencegahan diabetes melitus melalui program promosi kesehatan di RW 008 Kelurahan Cibodas, Kota Tangerang. Kegiatan dilaksanakan dengan pendekatan edukatif dan partisipatif yang mencakup identifikasi permasalahan, skrining kesehatan sederhana, penyuluhan kesehatan, sesi tanya jawab, serta penilaian pengetahuan menggunakan pre-test dan post-test. Skrining meliputi pemeriksaan tekanan darah, kadar gula darah, dan kadar asam urat. Hasil evaluasi menunjukkan bahwa rata-rata nilai pengetahuan peserta meningkat dari 86,67% sebelum penyuluhan menjadi 93,33% setelah kegiatan. Selain itu, hasil skrining menemukan beberapa peserta dengan tekanan darah dan kadar gula darah di atas batas normal sehingga memerlukan tindak lanjut dan pemantauan kesehatan. Keterlibatan aktif peserta selama penyuluhan dan sesi tanya jawab menunjukkan bahwa pendekatan partisipatif dapat mendukung penyampaian informasi kesehatan kepada masyarakat. Dengan demikian, promosi kesehatan yang dipadukan dengan skrining kesehatan sederhana berpotensi menjadi salah satu intervensi berbasis komunitas untuk mendukung pencegahan dan deteksi dini diabetes melitus.

Abstract

The skin is the body’s primary barrier against free radicals, which can induce oxidative stress and premature ageing. Antioxidants neutralize these radicals. This study aims to formulate and evaluate a cream containing ethanol extract of mung bean (Vigna radiata L.) seed coat. Four formulations were prepared with extract concentrations of 10%, 15%, 20%, and 25%. Physical evaluations included organoleptic tests, homogeneity, pH, viscosity, cream type, spreadability, and adhesiveness. Antioxidant activity was measured using the DPPH method. All formulations met physical standards, and formula F4 (25%) showed the highest antioxidant activity with an IC₅₀ value of 79.49 ppm. These findings indicate the potential of mung bean seed coat extract as an active antioxidant agent in topical preparations.

Abstract

Hypertension remains a major cardiovascular risk factor, and plant-derived compounds may provide leads for adjunctive therapeutic development. This study evaluated bioactive compounds from six Zingiberaceae species using ADMET screening, network pharmacology, and molecular docking. Thirty-two compounds were screened with DeepPK using predicted human oral bioavailability (≥20%) and selected toxicity endpoints. Six compounds met the screening criteria: zingerone, isoborneol, terpinyl acetate, 1,8-cineole, linalool, and α-terpineol. SwissTargetPrediction, GeneCards, STRING, and Cytoscape analyses identified TNF as the highest-ranked hub by both degree and bottleneck metrics. Three independent native-ligand redocking runs against human TNF-α (PDB ID: 6OP0) yielded RMSD values below 2 Å. Pentoxifylline showed a Rerank Score of -43.846, whereas zingerone had the most favorable score among the selected plant compounds (-36.2546) and was predicted non-toxic for the screened endpoints. These computational findings prioritize zingerone for further experimental validation as a potential antihypertensive adjunct candidate.

Abstract

BACKGROUND: Chronic inflammation contributes to diabetes-associated fibrosis, through persistent hyperglycemia, leading to increased interleukin-6 (IL-6), transforming growth factor-β (TGF-β), and collagen levels. While metformin effectively controls blood glucose, it may not adequately suppress these pathogenic pathways. Asiatic acid has demonstrated anti-inflammatory and antifibrotic properties, although its effects within a direct-contact macrophage–fibroblast microenvironment such as in co-culture models remain unclear. This study was conducted to evaluate the effects of asiatic acid on Il6, Tgfb1, and soluble collagen levels in a RAW 264.7–NIH 3T3 co-culture under high-glucose and lipopolysaccharide (HG–LPS) stimulation.METHODS: RAW 264.7 and NIH 3T3 cells were co-cultured at ratios of 1:1–1:6, and the selected co-culture was treated with asiatic acid (2.5, 5.0, and 10.0 µg/mL) for 24 h while HG–LPS stimulation was maintained. Unstimulated NIH 3T3 monocultures and untreated HG–LPS co-cultures served as controls. The inflammatory and profibrotic markers such as Il6 and Tgfb1 mRNA expression were quantified by polymerase chain reaction (PCR), and soluble collagen was measured using a modified Sirius Red assay.RESULTS: Among the tested ratios, the 1:1 co-culture exhibited the highest Tgfb1 mRNA expression (0.35±0.02) and soluble collagen level (1.30±0.00) and was selected for subsequent experiments, whereas Il6 mRNA expression did not differ significantly among co-culture ratios. Under HG–LPS stimulation, Il6 and Tgfb1 mRNA expression increased 3.5-fold and 10.3-fold, respectively, compared with unstimulated NIH 3T3 monocultures. Asiatic acid significantly reduced Il6 mRNA expression at 5.0 and 10.0 µg/mL, Tgfb1 mRNA expression at 2.5 µg/mL, and soluble collagen levels at all tested concentrations, indicating a non-monotonic concentration–response pattern.CONCLUSION: The 1:1 RAW 264.7–NIH 3T3 co-culture was the most responsive under HG–LPS stimulation and could be in vitro platform for investigating macrophage–fibroblast interactions under hyperglycemic inflammatory conditions. Asiatic acid modulated Il6 and Tgfb1 mRNA expression and soluble collagen levels in RAW 264.7–NIH 3T3 co-culture, which suggest that Asiatic acid may be potential for the management of fibrotic inflammation.KEYWORDS: co-culture, collagen, fibroblast dysfunction, NIH 3T3 fibroblast, RAW 264.7 macrophage
