Abstract

One way to ensure that efficacy and quality are guaranteed is that a product/medicinal ingredient quality standard needs to be met by standardizing the extract. Ceremai fruit (Phyllantus acidus) is thought to have elastase enzyme inhibitor activity because it contains vitamin C and flavonoids. The elastase enzyme functions in skin elasticity. This research aims to determine the standardization of ceremai fruit extract (P. acidus) and the inhibitory activity of the elastase enzyme. The sample was extracted using the maceration method using 70% solvent. Parameters in determining extract standardization are specific and non-specific. The inhibitory activity of the elastase enzyme was analyzed using the ELISA method. The results of determining the standardization of extract-specific parameters, the water-soluble essence content was 65.57%, and the ethanol-soluble essence content was 27.875. Non-specific parameters for determining water content 9.17%, determining total ash content 5.68%, determining acid insoluble ash content 1.5%, determining residual solvent 0.01%, determining microbial contamination ALT and AKK 10-1, heavy metal examination with undetectable results. Ceremai fruit extract (EBC) is positive for containing flavonoids, tannins, saponins, alkaloids, steroids, and phenolic compounds. The elastase enzyme inhibitory activity of ceremai fruit extract (EBC) resulted in an average IC50 value of 281.09 ± 11,3µg/mL and retinol with an average IC50 value of 23.81 ± 0,4 µg/mL. This research concludes that based on standardization testing of specific and non-specific parameters, ceremai fruit extract meets raw material quality standards and ceremai fruit extract (EBC) has weak category elastase enzyme inhibitory activity.

Abstract

Hypercholesterolemia is closely related to cardiovascular disease, which can increase the risk of atherosclerosis. Oxidative stress is related to the development of atherosclerosis because it is related to cellular responses triggered by an imbalance between oxidants and reductants in various layers of blood vessel tissue. K. anthotheca has pharmacological benefits as an antileishmaniasis and antimalarial. The purpose of this study was to determine the antioxidant and anticholesterol activity of K. anthotheca bark based on the type of solvent and to determine the phenolic and flavonoid content of the most active extract. The results indicated that ethyl acetate extract had the best antioxidant activity compared to n-hexane and ethanol extracts, with an IC50 value of 47.82 ppm, followed by ethanol extract at 48.46 ppm and n-hexane extract at 101.89. The value of antioxidant activity is directly proportional to the anticholesterol activity, where the ethyl acetate extract has an EC50 value of 20.42 ppm, the ethanol extract 445.58 ppm, and the n-hexane extract 1,073 ppm. The ethyl acetate extract has a phenolic content of 229.765 mgGAE/g extract and a flavonoid content of 7.63 mgQE/g.

Abstract

Erectile dysfunction (ED) is a common condition marked by difficulty maintaining a penile erection, often linked to reduced nitric oxide (NO) production. This study investigates the aphrodisiac properties of ethanolic white ginger extract (Z. officinale Rosc. var. officinarum) in male Wistar rats. Sixteen male and sixteen female rats were divided into four groups: two groups received 25% and 50% ginger extract, one served as a negative control, and the other as a positive control using sildenafil. Ginger rhizomes were dried, powdered, and extracted with ethanol. Phytochemical analysis revealed alkaloids, flavonoids, tannins, saponins, and terpenoids. Behavioral parameters, including introduction (approach to females), climbing, and coitus, were observed to evaluate sexual activity. The group treated with 50% ginger extract showed the highest frequency of introduction and climbing, while the sildenafil group exhibited the highest coitus frequency. Statistical analysis using the Kruskal-Wallis test indicated significant differences in sexual behavior across the groups. The study suggests that white ginger extract, especially at higher concentrations, may have aphrodisiac effects by enhancing sexual behavior in male rats. This could be due to its bioactive compounds affecting hormone levels or the nervous system. The findings propose white ginger as a potential natural treatment for ED, with further research required to explore its implications in humans.
