Abstract

 Inhibition of the α-glucosidase can be used as antidiabetic therapy because it can reduce postprandial blood sugar levels. The stem bark of mahogany (Swietenia macrophylla King.) have properties as inhibitors of enzyme α-glucosidase. The purpose of this study was to determine the active compound of mahogany stem bark against α-glucosidase enzyme in vitro and in silico. 96% ethanol extract of mahogany stem bark was partitioned with n-hexane, ethyl acetate and water. Testing of inhibition of α-glucosidase enzyme work done with substrate p-nitrophenyl-α-D-glucopyranoside. The water fraction was separated on silica gel column chromatography (SiO2 : dichloromethane-methanol = 10 : 1 ~ 1 : 1 and methanol). Molecular docking using AutoDock 4.2.6 was performed to predict the binding modes of α-glucosidase enzyme with chemical compound from water subfraction of stem bark mahogany. The results showed that water fraction of stem bark mahogany have antidiabetic activity with IC50 value of 1.51 µg/mL. Fraction Fr-1 gave an inhibition of 83.61 ± 1.11 %. Fr-1 contains chemical compound catechin, evodionol, swietenitin E, 11-deoxyswienialide D, swietenitin J and H. In silico study showed that catechin and evodionol free energy value (ΔG) -6.98 and -6.25 Kcal/mol and inhibition constanta (Ki) 7.61 and 26.18 µM.

Abstract

Melanin is the main pigment thfunctions to protect the skin from exposure to ultraviolet light, excessive melanin synthesis can cause skin hyperpigmentation. A. bilimbi and B. macrophylla fruits have bioactive compounds such as phenolics, flavonoids that function as antioxidants and tyrosinase inhibitors. This study aims to formulate a gel preparation of a combination of fruit extracts of A. bilimbi and B. macrophylla, test antioxidant activity using the DPPH method, and test tyrosinase inhibition enzymes. Three formulas were made, namely F0 (Basis), FI (50x extract), and FII (100x extract). Physical evaluation includes organoleptic, homogeneity, pH, adhesion, spreadability, viscosity, viscosity and irritation. Test of antioxidant activity and tyrosinase enzyme inhibition. Then the data were analyzed descriptively and statistically. The results of the evaluation of homogeneous preparations, dispersion, pH, viscosity, mandatory, meet the standards. Adhesion below standard, irritation test (mild irritation). The results of IC50 antioxidants were F0 (2,352 ppm), FI (1,448 ppm), FII (419,74 ppm) and gel brand x (77,429 ppm). The results of the tyrosinase inhibition test were the highest concentration of 10% F0 (7.828%), a concentration of 10% FI (18.245%), a concentration of 10% FII (25.814) kojic acid gel (brand x) as a comparison with a concentration of 10% (51.211%). Abstract

Hypercholesterolemia is closely related to cardiovascular disease, which can increase the risk of atherosclerosis. Oxidative stress is related to the development of atherosclerosis because it is related to cellular responses triggered by an imbalance between oxidants and reductants in various layers of blood vessel tissue. K. anthotheca has pharmacological benefits as an antileishmaniasis and antimalarial. The purpose of this study was to determine the antioxidant and anticholesterol activity of K. anthotheca bark based on the type of solvent and to determine the phenolic and flavonoid content of the most active extract. The results indicated that ethyl acetate extract had the best antioxidant activity compared to n-hexane and ethanol extracts, with an IC50 value of 47.82 ppm, followed by ethanol extract at 48.46 ppm and n-hexane extract at 101.89. The value of antioxidant activity is directly proportional to the anticholesterol activity, where the ethyl acetate extract has an EC50 value of 20.42 ppm, the ethanol extract 445.58 ppm, and the n-hexane extract 1,073 ppm. The ethyl acetate extract has a phenolic content of 229.765 mgGAE/g extract and a flavonoid content of 7.63 mgQE/g.

Abstract

Sabun terbentuk melalui reaksi antara asam lemak dan basa kuat, dan umumnya digunakan sebagai pembersih untuk menghilangkan kotoran pada kulit. Salah satu jenis sabun yang diminati pada saat ini oleh masyarakat yaitu sabun cair. Hal ini dikarenakan penggunaannya yang lebih praktis dan efisien. SMA Presiden 2 Tanjung Lesung merupakan sekolah umum semi militer yang memiliki 2 bidang ilmu yaitu IPA dan IPS. Kegiatan ini bertujuan untuk memberikan pengetahuan dan keterampilan kepada siswa/i SMA Presiden 2 Tanjung Lesung tentang pengaplikasian ilmu dan tekonologi dalam menghasilkan sabun mandi. Metode kegiatan yang dilakukan berupa pemaparan materi dan pelatihan pembuatan sabun cair dengan campuran ekstrak daun sisik naga (bahan alam) serta memberikan kuisioner berupa pretest dan postest untuk mengetahui pengaruhnya kepada siswa/i. Hasilnya didapatkan bahwa terdapat peningkatan pengetahuan dari kegiatan pengabdian kepada masyarakat (PKM) dari nilai rata-rata prestest 70,23 menjadi 86,28 untuk nilai posttest. Soap is formed through a reaction between fatty acids and a strong base and is commonly used as a cleanser to remove dirt from the skin. One type of soap that is currently popular among the public is liquid soap due to its practical and efficient use. SMA Presiden 2 Tanjung Lesung is a semi-military public school that offers two fields of study: Science and Social Studies. This activity aims to provide knowledge and skills to the students of SMA Presiden 2 Tanjung Lesung on the application of science and technology in producing bath soap. The methods used in this activity include material presentation, hands-on training in making liquid soap with dragon scale leaf extract (a natural ingredient), and pretest and post-test to measure the impact. The results showed an increase in knowledge from this community service activity, with the average pretest score rising from 70.23 to an average post-test score of 86.28.

Abstract

Caffeine is a stimulant substances that can stimulate the central nervous system (CNS) increasing concentration, and reducing drowsiness. The use of caffeine generally comes from coffee plants which can increase gastrin secretion. Long-term use of caffeine can cause side effects such as feelings of anxiety, irritability, insomnia and faster heart rate. An alternative stimulant that has limited side effects is needed. White pepper (Piperis albi fructus) is potential as stimulant. The extract of White pepper contained piperine compound. Piperine can potentially be a stimulant because it can stimulate the formation of energy in the body. The purpose of this study was to determine the in vivo stimulant activity of ethanol extract White pepper in male white mice. The method used in this research is Natatory exhaustion. 25 test animals were divided into 5 groups. The doses used are white pepper extract 0.32; 0.65 and 1.3 mg/20 g BW, caffeine 0.26 mg/20 g BW and 0.5% CMC-Na. Data analysis with SPSS 16 with the Kruskal-Wallis test and the Mann-Withney test. The results of the stimulant activity test of white pepper ethanol extract at a dose of 0.32; 0.65 and 1.3 mg/20 g BW for male white mice with the DDY strain with an average % increase in stamina of 63.92; 120.81 and 179.31%,mean while in the positive control of caffeine by 232.55% and the negative control of CMC-Na by 21.10%. White pepper ethanol extract dose of 1.3 mg/20 g BW has stimulant activity which is not significantly different from caffeine positive control.

Abstract

Mahogany (Swietenia macrophylla King.) is one of the plants used as an antidiabetic agent. 96% ethanol extract of S. macrophylla stem bark can inhibit the α-glucosidase enzyme. Methanol and aqueous extracts of S. macrophylla stem bark can reduce rat blood glucose levels induced by streptozotocin. Molecular docking is one of the most widely used structural-based drug design strategies due to its wide application in the analysis of binding energy and molecular interactions between drug compounds and receptors. This study aims to identify the compounds in the stem bark of S. macrophylla and analyze the molecular anchoring of the compounds produced by the aid. The research was carried out by means of the water fraction of the stem bark of S. macrophylla and was purified by column chromatography. Fraction 2 was identified by LCMS-MS. Molecular docking using AutoDock 4.2.6. The results of Fraction 2 showed the presence of catechins, evodionol, swietemacrophyllanin, swietenitin K and β-sitosterol. The results of an in-silico study showed that swietemacrophyllanin provided a better affinity than acarbose with a binding energy value of -8.57 kcal/mol and an inhibition constant of 0.52 µM. Forms hydrogen bonds with catalytic residues (Glu277 and Asp352) from the active site of the α-glucosidase enzyme. Swietemacrophyllanin has potential as an antidiabetic agent
