Abstract

Silver pompano (Trachinotus blochii) is a mariculture commodity with fast growth characteristics and easy adaptation to the environment. However, the production of its seeds is hindered by the maturation of the parent's gonads out-side the spawning season. Hormonal approaches are needed for the maturation of the parent's gonads. The aim of this study is to investigate the level of steroid hormones, gonadosomatic index, and gonad histology in female silver pompano (Trachinotus blochii) broodstock after dietary supplementation with 17-estradiol. This research is crucial for the advancement of effective and efficient techniques for rearing this fish. The study used a completely randomized design (CRD) consisting of three different 17estradiol concentrations (0 mg/kg (E1), 30 mg/kg (E2), and 60 mg/kg (E3) fed to female silver pompano broodstock for 30 days, beginning at initial feeding, each with eight individual replicates. Absolute body weight growth, steroid hormone levels, gonadal maturation index, gonadal histology, fecundity, and egg diameter of female silver pompano broodstock were all examined in this study. The incorporation of 17-estradiol hormone into the sustenance amplifies the reproductive capabilities of the female silver pompano broodstock. The inclusion of 17-estradiol hormone in the diet renders the most elevated values regarding fecundity and egg diameter in the female silver pompano broodstock. Histological examinations expose that the development of the gonads in the female broodstock treated with 17-estradiol hormone progresses towards maturation. The inclusion of 17- estradiol hormone in the diet at doses of 20 and 60 mg/kg feed yields the highest values in terms of fecundity and egg diameter in female silver pompano broodstock.Keywords:17-estradiolGonadal histologyGonadosomatic indexSteroid hormoneSilver pompano

Abstract

Lack of distribution, inbreeding depression, and environmental factors were several negative impacts on tilapia fry quality. It is needed to enhance fry quality by increasing the reproduction performance of broodstocks with nutrients and hormones by using Egg stimulant® and Oodev®. This research purposed to evaluate the maturation process with Egg stimulant and Oodev application and effective doses to increase reproduction performance of female tilapia broodstocks. This research conducted within two months at Unit Pelaksana Teknis Dinas Perikanan Budidaya Air Tawar (UPTD PBAT) Wilayah Barat Tanggamus. The research design used was a complete randomized design two factors and two levels with three replications. Control treatment (K) (Egg stimulant feed 0 g/kg and Oodev injection 0 mL/kg), P1 (Egg stimulant feed 3 g/kg), P2 (Oodev injection 0,5 mL/kg), and P3 (Egg stimulant feed 3 g/kg and Oodev injection 0,5 mL/kg) The results showed that P1, P2, and P3 treatments were not significantly different to eggs diameter, gonadal maturation level, gonadal maturation index, relative fecundity, and fertilization rate. However, Egg stimulants and Oodev have functionally enhanced reproduction performances of female tilapia broodstock. It is showed by treatments have a better result than control. The effective doses of Egg stimulant, Oodev, or combination need a longer period of research to applying in tilapia.

Abstract

Betta (Betta sp.) is an anabantidae fish with sexual development that can be directed at the embryonic and larval stages. One way to direct the sex development of these fish is by using hormone therapy using natural ingredients. Coconut water is one of the natural ingredients that can be used because it contains high potassium. This potassium can convert cholesterol in fish body tissue to pregnenolone which will affect the increase in testosterone production. This study aims to determine the best dose and age group in directing the sex of Betta fish experimentally using a randomized block design (RBD) with 4 treatments and 3 replications. The immersion treatment of larvae in the age group of 3 days, 5 days, and 7 days for 15 hours with a dose of 0%, 10%, 20%, and 30% coconut water was carried out in a 3 L jar containing 30 larvae. The results showed that coconut water was able to influence the increase in the percentage of male betta fish (P <0.05) with the highest value at a dose of 30% as much as 86.26 ± 8.34% and the 3 day larvae age group of 72.78 ± 16.03, and have a different effect on other treatments. In addition, the survival calculated after immersion shows a value of 100%, meaning that coconut water does not have a harmful effect on fish. Based on the results of this study, the best treatment for directing the sex of betta fish is a dose of 30% coconut water with the 3 day age group.

Abstract

Aplikasi penggunaan bakteri probiotik pada organisme budi daya memiliki salah satu fungsi penting yaitu menyeimbangkan mikroorganisme dalam pencernaan agar tingkat penyerapan zat-zat nutrisi menjadi lebih baik. Penggunaan bakteri probiotik pada pakan telah banyak dilakukan oleh pembudidaya, namun Pokdakan Maju Bersama Mesuji tidak menggunakan probiotik dalam aplikasi budidaya nya. Oleh karena itu dalam pengabdian ini, kami menyarankan Pokdakan untuk menambahkan probiotik dalam pakannya. Probiotik yang digunakan dalam pengabdian ini merupakan probiotik produk dari IRP2I Depok Gut Bioaero. Komposisi probiotik ini yaitu Bacillus cereus, molase dan salin. Bakteri Bacillus cereus yang kemampuan menghasilkan enzim protease, lipase dan amilase untuk meningkatkan kecernaan pakan pada ikan. Bacillus subtillis merupakan bakteri probiotik yang mampu menekan pertumbuhan Aeromonas hydrophila. Aplikasi pemberian probiotik pada media budidaya dan pakan ikan gabus mampu mencegah dan mengobati penyakit pada ikan gabus sehingga mampu meningkatkan produksi ikan gabus. Kendala yang dihadapi dalam budidaya ikan gabus di Pokdakan Maju Bersama Mesuji adalah kurangnya sarana dan prasarana budidaya. Dari hasil kegiatan pengabdian menunjukkan bahwa 10 responden (100%) menyatakan bahwa metode yang digunakan sesuai dengan keadaan sasaran.

Abstract

Alginat Sargassum telah terbukti mampu meningkatkan imunitas udang vannamei. Kegiatan Pengabdian kepada Masyarakat (PkM) ini  bertujuan melakukan diseminasi hasil riset suplementasi alginat untuk meningkatkan produksi udang vannamei di tambak masyarakat di Desa Berundung, Ketapang, Lampung Selatan. Metode PkM dilakukan dengan survei lokasi, pemetaan masalah, ceramah, diskusi, pelatihan, dan diseminasi alginat di tambak udang vannamei. Peserta PkM adalah para teknisi tambak, anak kolam, dan pemilik tambak di sekitar kecamatan Ketapang, Lampung Selatan dan sebagian berasal dari Kecamatan Pasir Sakti dan Kecamatan Labuhan Maringgai, Lampung Timur. Hasil kegiatan PkM menunjukkan adanya peningkatan pemahaman dan ketrampilan peserta terkait pembuatan alginat dari Sargassum, pemanfaatan alginat pada budidaya udnag vananmei dan manajemen kesehatan dalam budidaya udang di tambak. Hasil diseminasi menunjukkan adanya peningkatan laju sintasan udang vannamei dan dapat dilakukan panen parsial sebanyak dua kali pada hari budidaya (day of culture, DOC) 60 dan 67. Hasil sangat dirasakan manfaatnya oleh mitra setelah dua siklus budidaya sebelumnya selalu mengalami kegaglan panen. Kegiatan ini telah memberikan dampak positif bagi masyarakat khususnya mitra, baik secara ekonomi maupun sosial sehingga diharapkan tetap berlanjut dan memberikan dapak positif bagi pembudidaya udang vannamei lainnya

Abstract

Industri perikanan budidaya laut menghadapi tantangan besar dalam pengendalian penyakit yang disebabkan oleh bakteri patogen. Salah satu patogen yang sering menyerang ikan budidaya adalah Vibrio parahaemolyticus, yang dapat menyebabkan vibriosis pada ikan laut. Penelitian ini bertujuan untuk mengetahui karakter bakteri patogen Vibrio parahaemolyticus yang ditemukan pada ikan Bawal Bintang (Trachinotus blochii) di perairan Teluk Hurun, Lampung, serta menguji potensi antibakterinya. Karakterisasi bakteri V. parahaemolyticus meliputi uji media selektif CHROMagar Vibrio (CV), konfirmasi gen toxR dengan metode PCR, aktivitas hemolitik pada medium blood agar plate (BAP)  uji biokimia dengan metode API 20E, uji patogenisitas secara in vio pada bawal bintang, dan uji sensitivitas terhadap antibiotik dan bahan antimikrobia. Hasil penelitian menunjukkan bakteri V. parahaemolyticus tumbuh pada media CAV dengan warna biru kemerahan, teramplifikasi gen toxR, hemolitik positif, morfologi koloni berwarna krem dengan tepi bergelombang, mampu memfermentasi beberapa karbohidrat, termasuk glukosa, mannitol, dan sukrosa. Uji patogenisitas in vivo pada ikan bawal bintang menunjukkan gejala klinis berupa lesu, peradangan pada sirip, dan perdarahan, yang menandakan bahwa Vibrio parahaemolyticus merupakan patogen penyebab vibriosis pada ikan. Uji antibakteri menunjukkan bahwa enrofloxacin adalah antibiotik yang paling efektif, diikuti oleh tetrasiklin dan oksitetrasiklin. Ekstrak jintan hitam menunjukkan hasil yang lebih bervariasi, meskipun tetap menunjukkan potensi antibakteri alami. Penelitian ini diharapkan dapat memberikan dasar yang lebih kuat untuk pengendalian vibriosis di perikanan budidaya serta mendorong pengembangan alternatif antibakteri alami.

Abstract

Padina, one of the brown algae, grows in many tropical waters including in Lampung. Padina contains fucoidan, a sulfated polysaccharide, which has been shown to have a wide range of bioactivity. This study aims to determine the effectiveness of fucoidan Padina sp. from Lampung waters to enhance the non-specific immune response in Pacific white shrimp. Padina sp. was collected from Ketapang Beach, Pesawaran, Lampung then the Padina’s fucoidan was acidly extracted by previously method. Fucoidan then supplemented to 10 vananmei shrimp (size 10 g) each container (45 L) for 14 days. The study was conducted with a complete randomized design trial (CRD) consisting of 3 treatments and 4 repeats including fucoidan supplementation at a dose of 0 mg/kg (control); 750 mg/kg; and 1500 mg/kg of feed. Every day, shrimp were fed as much as 3% of biomass with a frequency of 4 times. Four nonspecific immune response of shrimp including total hemocyte count (THC), phagocytosis activity (PA), phagocytosis index (PI), and total plasma protein (TPP) were mesured on day 0; 7; and 14 after fucoidan supplementation. The results showed the administration of fucoidan extract Padina sp. gives significantly different results to THC and TPP enhancements. Supplementation treatment of 750 mg/kg fucoidan Padina is the best treatment to increase nonspecific immune response in Pacific white shrimp.
