Abstract

Lele masamo (Clarias sp.) merupakan lele varian baru yang memiliki banyak keunggulan dibandingkan dengan lele biasa. Salah satu keunggulannya yaitu pertumbuhan yang cepat sehingga lele masamo banyak diminati oleh pembudidaya. Penggunaan dasar kolam buatan pada budidaya lele masamo dijadikan sebagai salah satu alternatif untuk meningkatkan produksi. Namun, pemberian dasar kolam buatan dalam budidaya mengindikasi ikan mengalami stres. Salah satu cara untuk menanggulangi permasalahan tersebut yaitu dengan pemberian imunostimulan seperti yang terkandung dalam probiotik. Penelitian ini bertujuan untuk mempelajari pengaruh pemberian probiotik terhadap imunitas non-spesifik lele masamoyang dibudidayakan dengan menggunakan dasar kolam buatan. Rancangan penelitian menggunakan 2 perlakuan yaitu perlakuan pemberian probiotik dan tanpa pemberian probiotik dengan tiga kali ulangan. Penelitian berlangsung selama 45 hari dan diamati pada hari ke-0, hari ke-15, hari ke-30, dan hari ke-45. Hasil penelitian menunjukkan bahwa pemberian probiotik berpengaruh nyata terhadap total leukosit, persentase limfosit  dan sintasan lele masamo (P<0,05), namun tidak berbeda nyata terhadap persentase monosit, persentase neutofil dan kadar hematokrit (P>0,05). Pemberian probiotik dalam pakan dan air budidaya mampu meningkatkan total leukosit dari 66.856 sel/mm3 menjadi 103.739 sel/mm3,persentase limfosit dari 68,7% menjadi 75%.

Abstract

Penerapan biosekuritas pada budidaya udang vaname (Litopenaeus vannamei) dan hubungannya secara deskriptif dengan infeksi penyakit virus dan parasit dan performa budidaya  dipelajari untuk keberlanjutan budidaya udang di Kabupaten Pesawaran. Penelitian dilakukan dengan kuisioner standar biosekuritas, diagnosis penyakit virus, identifikasi jenis parasit dan performa budidaya udang. Penelitian menggunakan empat desa yang merupakan sentra budidaya udang di Kabupaten Pesawaran antara lain Hanura, Sidodadi, Gebang, Seribu dengan mengambil 16 petak tambak sebagai sampel. Hasil penelitian menunjukkan terdapat modifikasi penyediaan sarana, prasarana dan aplikasi biosekuritas pada empat desa dibandingkan biosekuritas standar. Modifikasi biosekuritas yang buruk di Desa Hanura menyebabkan infeksi penyakit virus dan budidaya udang tidak berlanjut. Modifikasi biosekuritas di Desa Sidodadi, Gebang dan Seribu yang berbeda mempengaruhi performa budidaya udang. Penerapan standar biosekuritas yang konsisten sesuai aturan lokal, nasional dan internasional diperlukan agar keberlanjutan industri udang vaname di Kabupaten Pesawaran dapat dipertahankan.

Abstract

Lack of distribution, inbreeding depression and environment factors were several negative impacts to tilapia fry quality. It is needed to enhance fry quality by increasing reproduction performance of broodstocks with nutrient and hormonal by using Egg stimulant® and Oodev®. This research purposed to evaluate maturation process with Egg stimulant and Oodev application and effective doses to increase reproduction performance of female tilapia broodstocks. This research conducted within two months at Unit Pelaksana Teknis Dinas Perikanan Budidaya Air Tawar (UPTD PBAT) Wilayah Barat Tanggamus. The research design used was a complete randomized designs two factors and two levels with three replications. Control treatment (K) (Egg stimulant feed 0 g/kg and Oodev injection 0 ml/kg), P1 (Egg stimulant feed 3 g/kg), P2 (Oodev injection 0,5 ml/kg) and P3 (Egg stimulant feed 3 g/kg and Oodev injection 0,5 ml/kg) The results showed that P1, P2 and P3 treatments were not significantly different to eggs diameter, gonadal maturation level, gonadal maturation index, relative fecundity, and fertilization rate. However, Egg stimulant and Oodev were functionally enhance reproduction performances of female tilapia broodstock.  It is showed by treatments has better result than control.  The effective doses of Egg stimulant, Oodev or combination need more longer period of research to applying in tilapia husbandry

Abstract

Betta (Betta sp.) is an anabantidae fish with sexual development that can be directed at the embryonic and larval stages. One way to direct the sex development of these fish is by using hormone therapy using natural ingredients. Coconut water is one of the natural ingredients that can be used because it contains high potassium. This potassium can convert cholesterol in fish body tissue to pregnenolone which will affect the increase in testosterone production. This study aims to determine the best dose and age group in directing the sex of Betta fish experimentally using a randomized block design (RBD) with 4 treatments and 3 replications. The immersion treatment of larvae in the age group of 3 days, 5 days, and 7 days for 15 hours with a dose of 0%, 10%, 20%, and 30% coconut water was carried out in a 3 L jar containing 30 larvae. The results showed that coconut water was able to influence the increase in the percentage of male betta fish (P <0.05) with the highest value at a dose of 30% as much as 86.26 ± 8.34% and the 3 day larvae age group of 72.78 ± 16.03, and have a different effect on other treatments. In addition, the survival calculated after immersion shows a value of 100%, meaning that coconut water does not have a harmful effect on fish. Based on the results of this study, the best treatment for directing the sex of betta fish is a dose of 30% coconut water with the 3 day age group

Abstract

Kerapu cantang (Epinephelus fuscoguttatus x Epinephelus lanceolatus) memiliki nilai strategis dalam marikultur di Indonesia dan menjadi salah satu komoditas unggulan perikanan di Lampung. Namun keberadaan ektoparasit berpotensi menurunkan produksi kerapu Cantang. Penelitian dilakukan untuk mengevaluasi prevalensi, intensitas, dan identifikasi ektoparasit pada kerapu Cantang di perairan Ringgung dan Durian Kabupaten Pesawaran. Metode penelitian dengan pengambilan sampel secara acak di dua lokasi karamba jaring apung. Pengambilan sampel sebanyak tiga kali dengan jumlah ikan sebanyak 30 ekor pada setiap lokasi. Pengamatan ektoparasit dilakukan di Laboratorium Kesehatan Lingkungan Balai Besar Perikanan Budidaya Laut Lampung. Hasil penelitian menunjukan ada 3 jenis ektoparasit yang menginfeksi kerapu cantang di Perairan Ringgung dan Durian yaitu Pseudorhabdosynochus sp., Neobenedenia sp. dan Brooklynella sp.. Ektoparasit Pseudorhabdosynochus sp. memiliki nilai prevalensi 100% pada lokasi satu, dan ektoparasit Brooklynella sp memiliki nilai intensitas 873 ind/ekor pada lokasi dua. Indeks keanekaragaman (H’) pada lokasi satu 0,18 dan lokasi dua 0,10. Indeks keseragaman (e) pada lokasi satu 0,27 dan lokasi dua 0,09. Indeks dominansi (c) pada lokasi satu 0,91 dan lokasi dua 0,96. Berdasarkan hasil tersebut nilai prevalensi tertinggi yaitu pada Pseudorhabdosynocus sp. dan nilai intensitas tertinggi pada Brooklynella sp.. Hasil juga menunjukkan adanya co-infeksi sebesar 30% pada sampel kerapu cantang berasal dari perairan Ringgung. Organ insang dominan terinfeksi oleh Pseudorhabdosynocus sp., sedangkan lendir didominanasi terinfeksi oleh Neobenedenia sp. dan Brooklynella sp.. Dibutuhkan pendekatan lain untuk mengontrol kelimpahan ektoparasit pada budidaya kerapu cantang di Teluk Lampung disamping menggunakan perendaman dengan air tawar yang selama ini dilakukan pembudidaya.

Abstract

Padina, one of the brown algae, grows in many tropical waters including in Lampung. Padina contains fucoidan, a sulfated polysaccharide, which has been shown to have a wide range of bioactivity. This study aims to determine the effectiveness of fucoidan Padina sp. from Lampung waters to enhance the non-specific immune response in Pacific white shrimp. Padina sp. was collected from Ketapang Beach, Pesawaran, Lampung then the Padina’s fucoidan was acidly extracted by previously method. Fucoidan then supplemented to 10 vananmei shrimp (size 10 g) each container (45 L) for 14 days. The study was conducted with a complete randomized design trial (CRD) consisting of 3 treatments and 4 repeats including fucoidan supplementation at a dose of 0 mg/kg (control); 750 mg/kg; and 1500 mg/kg of feed. Every day, shrimp were fed as much as 3% of biomass with a frequency of 4 times. Four nonspecific immune response of shrimp including total hemocyte count (THC), phagocytosis activity (PA), phagocytosis index (PI), and total plasma protein (TPP) were mesured on day 0; 7; and 14 after fucoidan supplementation. The results showed the administration of fucoidan extract Padina sp. gives significantly different results to THC and TPP enhancements. Supplementation treatment of 750 mg/kg fucoidan Padina is the best treatment to increase nonspecific immune response in Pacific white shrimp.

Abstract

This study evaluates the hematological profile of potential broodstock silver pompano (Trachinotus blochii) through hormonal induction and phyto-biotic supplementation via feed. An experimental trial was conducted for 30 days to assess the hematological profile of potential broodstock Trachinotus blochii. This study employed a completely randomized design (CRD) in two research experiments. The first experiment utilized hormonal induction with estradiol-17β hormone on female silver pompano broodstock, consisting of 3 treatments: E1 (0 mg/kg), E2 (20 mg/kg), and E3 (60 mg/kg), whereas the second experiment employed phyto-biotic induction using Tribulus terrestris extract (ETT) and gonadotropin hormone, consisting of 5 treatments: T1: ETT 0 mg/kg feed, T2: ETT 50 mg/kg feed, T3: ETT 250 mg/kg feed, T4: ETT 50 mg/kg feed + gonadotropin 1000 IU/kg body weight, and T5: ETT 250 mg/kg feed + gonadotropin 500 IU/kg body weight, both experiments used 8 individuals as replicate per treatment. The results of the study indicated that all major hematological indices such as total erythrocytes, total leukocytes, and hematocrit levels were higher in male silver pompano broodstock supplemented with Tribulus terrestris extract compared to female silver pompano broodstock induced hormonally with estradiol-17β hormone.

Abstract

Ikan kerapu (subfamily Epinephelinae) merupakan salah satu kelompok ikan terbesar di dunia. Identifikasi kerapu, khususnya genus Epinephelus, dilakukan berdasarkan karakteristik morfologi seperti warna, bentuk tubuh, dan ukurannya. Namun, proses identifikasinya kadang sulit untuk dibedakan secara morfologi karena memiliki karakteristik yang sangat mirip. Salah satu metode yang dapat diaplikasikan adalah penggunaan DNA barcoding. Penelitian ini bertujuan untuk membandingkan ikan kerapu dari Arab Saudi dan Indonesia. Hasil identifikasi morfologi dan molekuler menunjukkan bahwa kerapu dari penelitian ini (dari Yanbu, Arab Saudi dan Lamongan, Indonesia) adalah Epinephelus areolatus (kerapu sirip putih). Secara morfologi, sampel kerapu ekor putih dari Yanbu Arab (Saudi) adalah sebagai berikut: sirip dorsal X-XI/12-15; sirpi anal II-III/8-9; sirip pectoral 13-15; sirip pelvic I-5; sisik linea lateralis 48-53; vertebrae 24. Sementara hasil meristik dari kerapu dari Lamongan (Indonesia) adalah sebagai berikut: sirip dorsal X-XI/15-17; sirip anal II-III/8; sirip pectoral 16-19; sirip pelvic I-5; sisik linea lateralis 48-53; vertebrae 24. Perbedaan E. areolatus dari Arab Saudi dan Indonesia secara morfologi adalah dari bintik dan ekor caudal-nya. Hasil molekuler pada E. areolatus menunjukkan perbedaan clade. Sampel dari Arab Saudi termasuk clade Western Indian Ocean sementara Indonesia termasuk Western Pacific. Hal ini menunjukkan bahwa terdapat perbedaan morfologi dan molekuler antara E. areolatus dari Yanbu (Saudi Arabia) dan Lamongan (Indonesia). Sekuens gen COI dari DNA ikan kerapu ekor putih pada penelitian ini telah didaftarkan ke NCBI (PP388919.1 untuk Lamongan dan PP388920.1 untuk Arab Saudi). Data penelitian ini dapat dijadikan sebagai referensi untuk konservasi.
