Abstract

Abstract

Bogor Botanic Gardens is an ex-situ plant conservation area in Indonesia. Since BBG is 103 years old, many collections are 100 years old or older. These antique collections may sustain damage, such as broken or collapsing, endangering visitors and employees. As a result, monitoring tree health at BBG is a critical task. According to the tree health monitoring data, 73 of 244 trees were further checked using the PiCUS Sonic Tomograph. Trees from the Fabaceae (31%) and Myrtaceae (10%) families were the most frequently checked. Walnuts trees from the Burseraceae family had the most specimens (47,94%). The PST effectively provides an immediate picture of the stem condition by calculating solid and decaying wood percentage values.

Abstract

The IUCN red listed Pinanga javana Blume, also known as Javanese Pinang, as a protected and threatened species that is endemic to Java island, Indonesia. This research focused on study of the germination and growth process of P. javana seedlings using four types of planting media: cocopeat, husks, sand, and sawdust. The results showed that P. javana seed water content slowly decreased from 33.96% to 24.45% during the storage period from day 0 to day 21. The planting medium had a significant influence on the germination rate, with sand exhibiting the highest value (2.34) and husks providing the best seedling growth. Cocopeat had the highest moisture level (90%), while sand generated the lowest moisture level. The pH of all media was within the normal range (6-7), which supports optimal germination. The analysis of various parameters concluded that sand was the optimal planting medium for P. javana in this study. Keywords: germination, planting media, Pinanga javana, viability, water content

Abstract

DNA isolation is a routine procedure in molecular analysis. The method of plant genome DNA extraction has been widely available throughout global laboratories. Several labs made some modifications to obtain optimal results. This research was aimed to analyse two types of DNA extraction methods for ironwood plant (Eusideroxylon zwageri), one tropical rain forest woody plant known for its high strength and durability. The results indicate that isolation DNA kit produced high DNA purity with lower concentrations while CTAB methods generated lower DNA purity with higher concentrations. This may be used as consideration of DNA isolation for downstream molecular analyses.   Keywords: DNA extraction, ironwood, CTAB, genomic DNA

Abstract

The local communities have local knowledge in utilizing and managing landscape units. The threat of modernization has led to the degradation of local knowledge, including the Javanese community on Mt. Slamet. This study aimed to analyze the plant diversity characteristics of landscape units and determine the important value of landscape units and plant species used by the community. This research was conducted in the Ragatunjung, Cipetung, and Pandansari village, Paguyangan District, Brebes Regency, Central Java. Semi-structured interviews collected Ethnoecological data with eight key informants and 83 respondents selected by purposive sampling and snowball sampling. Data on the level of landscape utilization were analyzed using the Local User's Value Index (LUVI). The community recognizes nine types of landscape units, such as perawisan (yard), wanah (production forest), Majegan (gardens), Sabin (rice fields), Kubang buyut (protected forest plan), tea plantations, Telaga Ranjeng Nature Reserve, Tanah Bengkok and tuk (water sources). Wanah was the most important landscape unit in the community in Ragatunjung (31.27), Cipetung (53.55), and Pandansari (28.17). Oryza sativa had the highest importance value in Sabin (22) and wanah (12) of Ragatunjung. In contrast, Syzygium aromaticum had the highest in Majegan (6.68). Brassica oleracea has the highest importance value in the two landscape units of Cipetung, namely, Majegan (4.20) and settlements (3.5), while in the wanah is Zea mays (11.38). Solanum tuberosum had the highest value in each landscape unit in Pandansari, both in wanah (10.33), Majegan (6.80), and Protection Forest Plan (PFP) (4.13). Categorizing landscapes and their utilization by maintaining certain landscapes, such as the Telaga Ranjeng Nature Reserve and Kubang buyut, directly impact the sustainability and integrity of ecosystems and natural resources in and around the area.
