Abstract

Clerodendrum minahassae (Lamiaceae) merupakan tumbuhan yang berasal dari Sulawesi Utara dan dikenal dengan nama leilem. Pada umumnya genus Clerodendrum mengandung senyawa antioksidan seperti senyawa-senyawa fenol dan flavonoid. Penelitian ini bertujuan untuk menentukan aktivitas antioksidan dan toksisitas serta kandungan senyawa fenol dan flavonoid total dari daun leilem. Ekstrak daun leilem dibuat secara maserasi dalam metanol yang selanjutnya difraksinasi dengan n-heksana dan etil asetat. Aktivitas antioksidan diuji dengan metode penangkapan radikal bebas DPPH (1,1-difenil-2-pikrilhidrazil) dan toksistas diuji terhadap larva Artemia salina dengan metode BSLT (Brain Shrimp Lethality Test). Kandungan fenol dan flavonoid dilakukan dengan metode kolorimetri. Hasil penelitian memperlihatkan ekstrak metanol dan fraksi etil asetat daun leilem memiliki aktivitas antioksidan lemah dengan nilai IC50 300,57 µg/ml dan 189,62 μg/ml, sementara itu toksisitas terhadap larva udang Artemia salina diperlihatkan oleh ekstrak metanol dan fraksi etil asetat daun leilem dengan nilai LC50 berturut turut 190,58 μg/ml dan 147,36 μg/ml. Penentuan kadar fenol total dan flavonoid total dilakukan pada fraksi etil asetat dengan menggunakan standar asam galat untuk fenol total dan kuersetin untuk flavonoid total. Hasil penentuan menunjukkan bahwa fraksi etil asetat daun leilem mengandung fenol total 164,38 mg/g ekstrak dan flavonoid total 270,8 mg/g ekstrak.

Abstract

Petai mistletoe (Dendrophthoe praelonga (Blume) Miq) is semi-parasitic plant. Parasite plant has been used traditionally in Indonesia to prevent or treat degenerative diseases, such as cancer. The purpose of this study was to indentify anticancer compound of petai mistletoe leaves extract and to examine its antioxidant activity (based on DPPH method) and cytotoxic activity on breast cancer (MCF-7) cell lines in vitro using Alamar blue method. This research aims to isolating, characterize, determine antioxidant activity and cytotoxic activity. Separation was conducting column chromathography and characterize was conducted using TLC, FT-IR, LC-MS and NMR-1D. From the result obtained, the isolated compound is flavonol quercetine (3,3,7,3’,4’ pethahydroxyflavone) which active antioxidant with IC50 value of 9.4 μg/mL and the value of cytotoxic activity with IC50 750.62 μg/mL

Abstract

Ebony (Diospyros celebica. Bakh) is a native plant of Indonesia that is used as a traditional medicine for diabetes mellitus. Previous studies state that ethanol extract from ebony leaves significantly reduced blood glucose levels of diabetic rats induced by alloxan. This ability is thought to be caused by secondary metabolites which is phenolic and flavonoid compound. However, research related to structure compounds in extract and composition of solvent variations in the extraction process to get the most optimal activity and also their correlation with total phenolic and flavonoid compounds has not been carried out. This study aimed to evaluate the composition of the optimum solvent in the extraction process and also to know the structure compounds contained in the extract. In the present study, we extracted the ebony leaves with three solvents of different polarity (ethanol-water), i.e. ethanol 50% (EtOH), EtOH 70%, and EtOH 96%. Quantitative estimation of total phenolic and flavonoid contents. These extracts were future evaluated for their activity and the content of compounds in the extract was identified using LCMS/MS. The results showed that phenolic compounds that have been identified in ebony leaf extract are methyl gallate and (-)-Epiafzelechin 3-gallate and we found there were significant differences in all test parameters between the three extracts which is EtOH 96% extract gave the best.

Abstract

Chlorella sp. microalgae is a potential source of natural antioxidants and other bioactive compounds used in the food and pharmaceutical industries. Ultrasound pretreatments followed by enzymolysis are widely used to improve the functional properties of biological materials. This work aimed to investigate the effects of ultrasound pretreatments followed by bromelain hydrolysis on the proteins, phenolic content, and antioxidant activity of Chlorella sp. crude extract. The experimental design for the study involved two factors: ultrasound pretreatment time (0, 5, and 10 min) and bromelain hydrolysis time (0, 2, 4, and 6 hours). The protein was measured using Lowry assay while the total phenolic content was measured using a Folin–Ciocalteu assay. The highest protein content (7.09 ± 0.44 mg/mL) and phenolic concentrations (0.28 ± 0.01 ìg/mL) were obtained at 5 min ultrasound pretreatment and 6 hours enzymolysis. For antioxidant activity, 5 min ultrasound pretreatment followed by 4 hours of enzymolysis resulted in the highest DPPH (1,1-diphenyl-2-picrylhydrazyl radical) inhibitory activity (81.74%). Meanwhile, ultrasound pretreatment for 10 min followed by 6 hours of enzymolysis resulted in the highest ABTS (2,20 azinobis-(3-ethylbenzthiazoline-6-sulfonic acid) inhibitory activity (94.10%). This study showed that variation in ultrasound pretreatments followed by bromelain hydrolysis could be used to improve the protein content, phenolic content, and antioxidant activity of Chlorella sp. crude extract.

Abstract

In order to create an antisense construct of the gene encoding Ornithine Decarboxylase (ODC) from tobacco (Nicoticum tabacum L.) cv. Temanggung, the target gene must be isolated. In this paper. we present the PCR amplification of a fragment from putative gene encoding ODC from tobacco cv. Temanggung. Leaf genomic DNA was isolated and used as the template for PCR. PCR optimization was done by adjusting the annealing temperature and the cycle number. Verification of the fragment obtained was also done using the second primer pairs.

Abstract

Asparagine synthetase (AS) is one of the enzyme involves in ammonium metabolism in plant, it catalyzes the transfer of the amino group of glutamine to aspartate giving asparagine. Asparagine serves as a major nitrogen transport and storage molecule in many higher plants. This research used legume plant Coronilla rostrata callus culture for primary and secondary metabolism study. On primary metabolism, the culture were used to study the phenomenon of asparagine accumulation. Because of the responsiveness of the culture, we investigated the use of this culture for model system to conduct molecular biology study of the nitrogen metabolism. The aim of this study was to isolate cDNAs enconding asparagine synthetase from C. rostrata because of the asparagine accumulate in this culture. Several PCR based approaches were conducted such as RT-PCR, LM-PCR, and RACE. Using these methods, two AS cDNAs was isolated from C. rostrata AS1 (GenBank no AY081945) which a complete cDNA sequence and AS2 which a partial cDNA sequence. (GenBank no AF488726). These two cDNAs had high homology with the legume AS.

Abstract

Calophyllum bicolor (Clusiacea) is a big tree from Indonesian rain forest in Palangkaraya, Central Kalimantan. Calophyllum or bintagor is one of many sources of natural bioactive compounds that can be used in the fields of health and pharmaceuticals. The aim of this research was to explore the antiplasmodial activity of methanol extract of Calophyllum bicolor P.F. Steven against Plasmodium falciparum. The methanol extract was purified by colomn chromatography system, hexane – ethyl acetate was used as solvent with increasing polarity. One pure compound was obtained and was elucidated based on the 1H-&13C-NMR and 2D-NMR, [COSY, HMBC and HMQC] data and the isolated compound was identified as xanthone. Methanol extract showed antiplasmodial activity growth inhibition against P. falciparum with IC50 5.2 ppm and the new 5-methoxy trapezifolixanthone compound have maximum inhibition at concentration 0.11 nMol.

Abstract

Currently marine bacteria are considered as important source of natural products for drug discovery. The objective of this study is to conduct an in vitro bioactivities (antidiabetic, antioxidant and antibacterial) screening of 9 Indonesian marine bacteria isolated from sponges that belongs to the Research Center for Oceanography, Indonesian Institute of Sciences collections. The marine bacteria were cultured for 2 days in liquid medium containing yeast, peptone and sea salt under shaking condition and extracted with ethyl acetate. Antidiabetic was measured using inhibition of -glucosidase inhibitory activity method; antioxidant was measured using DPPH free radical scavenging activity method; antibacterial was tested using disc diffusion method. Screening results showed that at sample concentration of 200 μg/mL, there was significant -glucosidase inhibitory activity detected in the extracts of strain Sp 7.9 (84 % inhibition) and Sp 8.10 (75 % inhibition), however the antioxidant activities of these two strains were low only around 30 % inhibition, antioxidant activities of other strains were very low. Screening for antibacterial activities using 10 μL samples show that extract of strain Sp 8.5 was best for Staphylococcus aureus (14 mm inhibition); Sp 7.9, and Sp 8.5 for Bacillus subtilis (18 mm inhibition); Sp 8.10 for Escherichia coli (10 mm inhibition); Sp 8.9 and Sp 8.10 (10 mm inhibition) for Pseudomonas aeuriginosa. Based on these results marine bacteria strain Sp 7.9 and Sp 8.10 were selected to be used for further studies in the isolation of bioactive that has potential as antidiabetic and antibacterial. Results of molecular identification conducted by InaCC show that identity of both strains based on BLAST Homology using NCBI database were Bacillus thuringiensis.
