Abstract

Lipsticks and eye shadows are cosmetics frequently used by women every day. Some cosmetic products contain heavy metals like lead. This study aims to validate the UV-Visible Spectrophotometer method for analyzing lead in cosmetics and determine the lead content in lipsticks and eye shadows. Sample preparation was conducted using closed wet digestion with reflux. The samples consisted of 5 lipsticks and eye shadows from the King Street Market in Bandar Lampung. Lead content was determined using a UV-Visible spectrophotometer after method validation. Validation parameters included selectivity, linearity (correlation coefficient = 0.998), accuracy (% recovery = 99-100%), precision (%RSD = < 2% for all experiments), detection limit 0.044 µg/mL, and quantitation limit 0.148 µg/mL, all of which met validity criteria. Lead levels in lipsticks ranged from ±6,000 to 11,000 µg/gram, while in eye shadows, they ranged from ±5,000 to 10,000 µg/gram. The results indicate that lead levels in lipsticks and eye shadows exceed the BPOM RI limit of 20 µg/gram.

Abstract

Cassava (Manihot esculenta Crantz) is widely utilized across Indonesia as both a staple food and a traditional medicinal resource, and its leaves are known to contain bioactive secondary metabolites such as flavonoids. This study aimed to quantify the total flavonoid content in several cassava leaf varieties and to isolate a flavonoid compound from the variety with the highest flavonoid content. Total flavonoid content was determined using UV-Visible spectrophotometry. The isolation of flavonoid compounds was carried out using vacuum liquid chromatography (VLC) and column chromatography (CC), with thin-layer chromatography (TLC) employed to monitor the separation results. The total flavonoid content of the Gajah, UJ-3 (Thailand), UJ-5 (Katsesart), Cimanggu dan Kaspro varieties were found to be 1.029 ± 0.003 mgQE/g, 0.746 ± 0.006 mgQE/g, 0.970 ± 0.009 mgQE/g, 0.936 ± 0.004 mgQE/g and 1.220 ± 0.003 mgQE/g, respectively. The Kaspro variety exhibited the highest flavonoid content. Isolation and characterization of the flavonoid from Kaspro leaves yielded a yellowish powder with absorption maxima at 269 nm (Band II) and 334 nm (Band I). Further analysis using shift reagents identified the isolated compound as apigenin, a member of the flavone group.

Abstract

Traditional medicine, derived from medicinal plants, has been used for generations as a form of treatment. The safety, quality, and efficacy of plants used for medicinal purposes are important considerations. Mirabilis jalapa L., also known as the four o'clock flower, is a traditional medicine with known benefits such as diuretic, anti-inflammatory, and antibacterial properties. However, its safety level remains uncertain. To address this, an acute toxicity test was conducted on the ethanol extract of Mirabilis jalapa L. flowers to determine its safety. The extraction process utilized a reflux method with 96% alcohol as a solvent, resulting in a standardized extract. Subsequent toxicity tests were performed on 12 female white mice, using doses of 300 and 2,000 mg/kg body weight. The results revealed an LD50 value exceeding 2,000 mg/kg body weight for the ethanol extract of Mirabilis jalapa L. flowers, indicating its classification as "Practically Not Toxic."

Abstract

Ascorbic acid and its derivatives with nicotinamide are often used as skin lightening, anti-aging, and anti-pigmentation agents. Combining these two active substances may increase the risk of instability due to degradation. This research was conducted to determine the effect of nicotinamide concentration on the degradation process of ascorbic acid and its derivatives, and to determine the degradation products formed using liquid chromatography-mass spectrometry (LC-MS). The study was conducted by combining nicotinamide concentrations ranging from 5 to 15 micrograms per milliliter with incubation times of 1, 2, 3, and 24 hours to react with ascorbic acid and its derivatives. The spectral shape and maximum wavelength were analyzed using a UV-Vis spectrophotometer. Samples that showed changes in chemical structure were subsequently analyzed by LC-MS to identify degradation products. The results showed that only the combination of ascorbic acid and nicotinamide shifted the maximum wavelength and generated a new peak at 310 nm after 24 hours of incubation. Nicotinamide can accelerate the degradation process of ascorbic acid. The LC-MS analysis results show that the degradation products formed from the combination of ascorbic acid and nicotinamide samples are dehydroascorbic acid and 3-hydroxy-2-pyrone. These findings highlight the importance of incorporating stabilizing agents into cosmetic formulations containing unstable active ingredients, such as ascorbic acid, to ensure product stability during storage and use.

Abstract

Background: Channa striata is a protein-rich freshwater fish with therapeutic potential, yet systematic optimization of extraction methods and stable formulation development remain limited. Objective: To compare protein extraction efficiency using three solvents (1% HCl, 0.9% NaCl, and distilled water) and develop optimized capsule formulations. Methods: Protein was extracted at 1:4 (w/v) ratio and quantified using the Lowry method at 595 nm. The method was verified for linearity (R = 0.998), accuracy (98-102%), and precision (RSD <2%). Extracts were formulated into capsules at 0, 300, 400, and 500 mg (F0-F3) and evaluated for quality parameters and 28-day stability at -4°C, 24°C, and 40°C. Data were analyzed using one-way ANOVA with Duncan's post-hoc test. Results: The 1% HCl yielded highest protein content (4.947 ± 0.071 mg/mL) and extraction efficiency (21.390%) compared to 0.9% NaCl and distilled water (p <0.05). All formulations met pharmaceutical standards. Notably, F3 maintained protein retention >95% across all storage conditions (97.1-99.3%), while F0-F2 failed this criterion. Conclusion: The 1% HCl demonstrated optimal extraction efficiency. F3 (500 mg) exhibited superior stability and dissolution characteristics, representing a promising candidate for C. striata-based nutritional supplements.
