Abstract

Isolation and identification of secondary metabolite stem bark of Polygonum pulchrum and its antioxidant activity were done. Extraction was performed by maceration using methanol solvent. The separation and purification obtained by using vacuum liquid chromatography (VLC), radial chromatography (RC) and monitored by thin layer chromatography (TLC). Identification of isolated compounds determined by spectroscopy method i.e. FT-IR, 1D-NMR (1H, 13C-NMR and DEPT) and 2-D NMR (HMQC and HMBC). White solid compound was isolated and characterized as 5(E)-eicosenoic acid. Antioxidant was analyzed towards DPPH (1,1-diphenyl-picrilhydrazil). The compound showed that has antioxidant potency with IC50 value at 348.10 µM.Keywords: Polygonum pulchrum, 5(E)-eicosenoic acid, antimicrobial, antioxidant

Abstract

Fern is one of the groups of primitive plants rich in secondary metabolites that are commonly used to treat various diseases, including antioxidant, anti-hyaluronidase, anti-inflammation, and respiratory disease but less investigated. Flavonoid is one of the secondary metabolites abundantly present in ferns. This study aims to isolate major compounds found in Blechnum orientale act as 15-lipoxygenase (15-LOX) inhibitors. Inhibition of lipoxygenase decrease the production of leukotriene that induces bronchoconstriction in asthma. Isoquercitrin (Quercetin-3-O-β-glucopyranoside) and trifolin (kaempferol-3-O-β-D-galactoside) have been successfully isolated from Blechnum orientale. Further in silico study was performed to explain the binding mode between flavonoid pyranoside or galactoside and flavonoid aglycone in the 15-LOX cavity and their amino acid residues interaction. Isoquercitrin binds with Ile663, Ile400, Leu408, Leu597, Ala404, and Arg403 in the 15-LOX cavity as a lipoxygenase inhibitor. Trifolin binds the same amino acids as isoquercetin with addition His366, Gln596, and Phe175. Both isoquercitrin and trifolin act as competitive inhibitors against lipoxygenase enzymes.

Abstract

Soft coral Nepthea sp. grows in the seas of South-East Sulawesi, Indonesia. However, information on the chemical and pharmaceutical aspects of this genus is still limited. Therefore, this research aims to explore the chemical contents and biological activities of Nepthea sp. The sample was collected from the waters of Saponda Island by SCUBA diving. It was extracted by ethyl acetate and fractionated using vacuum liquid chromatography. The chemical content was analyzed by phytochemical screening, LC-MS/MS analysis, Total Phenolics Content and Total Flavonoids Contents. Antioxidant potency was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radicals and ABTS (2,2’-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid). Cytotoxicity property was analyzed by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays. The result showed that the fractionation of Nepthea ethylacetate extracts produced six fractions (A-F). Fractions A and B contain non-polar compounds. Based on LC-MS/MS data, the non-polar compounds in Fraction A and B include achillin, atractylenolide II, buthyl isobuthyl phthalate, rengyolester, 2a-acetoxycostic acid, ocotillol acetate, petasitolone and some unidentified compounds that are C33H58O4, C15H21NO, C21H33NO, and C16H20O4. In general, the antioxidant and cytotoxic properties of all samples are in the weak category, however, when examined for each sample, the antioxidant properties of fraction B is slightly better than fraction A based on the IC50 value of DPPH and ABTS. Cytotoxicity of Fraction A is better than Fraction B against Breast Cancer cell lines MCF-7. The non-polar fraction of Nepthea sp. can be developed as raw material for the discovery of new compounds, antioxidant and anticancer agents, especially breast cancer.

Abstract

Reactive oxidative stress (ROS) can lead to cell damage, and one of them is the kidney's cell. Etlingera elatior (Jack) R.M. Smith can be utilized as an agent that can protect the cell from ROS. This study aimed to investigate the protective effect of E. elatior fruit on the kidney's cell. We used experimental animals which were treated with Na-CMC (Group I), Na-CMC (Group II), the extracts of E. elatior fruit 200, 300, 400 mg/kg BW for Group III, IV, and V, respectively, for seven days. The blood was collected after treatment. At day 8, group I, III, IV, and V were induced by CCl4. At day 9, blood was collected and the kidneys were harvested for histopathology analysis. Blood collected were measured for albumin, total protein, urea and creatinine levels. After treatment, albumin and total protein showed no increased levels; urea decreased at doses of 200, 300, and 400 mg/kg BW, respectively, and creatinine levels only decreased at the dose of 400 mg/kg BW (p<0.05). The dose of 200 and 300 mg/kg BW showed protecting effects in the tubular cells of renal. Therefore, the ethanol extract of E. elatior showed a nephroprotective effect by normalizing the urea and creatinine levels of rats and protecting tubular cells of renal.

Abstract

Nepthea sp., a soft coral In Indonesia's South-East Sulawesi, it thrives. The chemical and pharmacological properties of this genus have not been studied in this location. As a result, the goal of this page is to offer a broad overview of both properties of Nepthea sp. The sample was taken from Hoga Island. Methanol was used to extract the material, which was then fractionated with hexane and ethyl acetate to produce n-hexane, ethyl acetate, and methanol fractions. Vacuum liquid chromatography was also used to fractionate the ethyl acetate fraction. Phytochemical screening, LC-MS/MS, Total Phenolics Content (TPC), and Total Flavonoids Content (TFC) were used to determine the chemical contents. DPPH radicals and ABTS were used to assess antioxidant activity and MTT assays for cytotoxicity. Six subfractions are produced by fractioning the ethyl acetate fraction (160 g), with the most polar subfraction (F) weighing 15.4 g. The phenolics and flavonoids constituents in Subfraction F have IC50 values of 68.77±4.8 mg GAE/g extract and 0.22±0.1 mgQE/g extract, respectively. The presence of phenolics, flavonoids, and alkaloids in the subfraction F is supported by the LC-MS/MS data, Subfraction F has 3-ter-butyl-4-methoxyphenol (terpenoids or phenolics compound), isosalsoline (alkaloids), valine (amin acid), and several unidentified chemicals with molecular formulas C15H23NO3, C33H47NO4, C29H49NO3 (alkaloids or amino acid) and C8H18O3 (phenolic compound).The IC50 values (mg/L) of 101.1 ± 8.8 (DPPH) and 89.76 ± 7.5 (ABTS) indicate that F has antioxidant capacity, and it is also cytotoxic against breast cancer cell lines (MCF-7) with an IC50 of 170.72±6.5mg/mL.

Abstract

Etlingera elatior (Wualae) termasuk dalam tumbuhan famili zingiberaceae yang memiliki aktivitas antiinflamasi. Namun, bioavailabilitas dan permeabilitas zat aktif yang rendah sehingga diperlukan suatu sistem penghantaran dalam komplek lipid-bahan alam. Sistem yang dikenal dengan fitofosfolipid ini merupakan penghantaran fitokonstituen dalam matriks fosfolipid yang dapat meningkatkan permeabilitas dan bioavailabilitas zat aktif melalui karakteristik ampifilik dan aksi emulsifikasi oleh fosfolipid. Penelitian ini bertujuan untuk melakukan formulasi dan karakterisasi sistem vesikel sehingga diperoleh sistem yang optimum antara ekstrak etanol buah wualae dan fosfatidilkolin yang memiliki karakteristik vesikel terbaik. Preparasi dilakukan    menggunakan    teknik    hidrasi    lapis   tipis dengan adanya variasi komposisi ekstrak dan fosfatidilkolin, yakni 0,5:0,5 (Formula 1); 0,5:1 (Formula 2); 1:0,5 (Formula 3); 1:1 (Formula 4); 1:2 (Formula 5); dan 2:1 (Formula 6). Hasil yang diperoleh adalah komposisi formula optimum antara ekstrak dan fosfatidilkolin yaitu pada F4 dengan komposisi 1:1 dengan karakterisasi meliputi pengamatan morfologi berbentuk bulat (sferis) menggunakan mikroskop optic, efisiensi penjerapan vesikel diperoleh 99,20%, ukuran partikel 898,2 nm dan indeks polidispersitas 0,573 dengan kategori vesikel jenis LUV (Large Unilamellar Vesicle) menggunakan Particel Size Analyzer.

Abstract

Sappan wood extract (Caesalpinia sappan) provides better antioxidant properties than ascorbic acid. Empirically, Sappan wood is often used as traditional medicine. To support the utilization of Sappan wood, the isolation and identification of active antioxidant compounds from the methanol extract of Sappan wood had been conducted from this study. Extraction was performed with maceration and evaporation methods. The spots of active antioxidant compounds in TLC-DPPH observations were isolated using liquid vacuum chromatography to obtain two pure compounds. The results of the LC-MS/MS identification of the two compounds, respectively, exhibited peak fragmentation of molecular ions at m/z 271.09, 257.15, 167.03, 151.06, 123.12, and 104.06 at the retention time of 6.35 minutes which were translated as Alpinetine and peak fragmentation of molecular ion at m/z 287.09, 177.06, 164.05, 123.04 and 110.04 at a retention time of 5.75. minutes which were translated as 3-Deoxysappanone B. The antioxidant activity quantitatively showed that Alpinetine and 3-Deoxysappanone B were capable of scavenging DPPH radicals with IC50 values ​​of 20.11 M and 15.28 M, respectively.

Abstract

Cervical cancer is one of the highest causes of death in Indonesian women caused by mutations of normal cervical cells turning into abnormal cells. Treatment of cervical cancer causes significant side effects with a relatively long treatment period, so many researchers are exploring various natural ingredients from marine ecosystems as candidates for anticancer drugs. One of them is the sea sponge Petrosia sp. containing various secondary metabolites, one of which is an alkaloid that has a cytotoxic effect on cancer cell lines. This study aims to determine the cytotoxic activity of the ethanol extract of Petrosia sp. in HeLa cervical cancer cells. The ethanol extract of Petrosia sp. obtained by maceration using 96% ethanol and obtained extract as much as 55.9 g and yield value of 2.94%. Cytotoxic activity test was carried out in vitro using the Presto Blue method with varying concentrations of ethanol extract 7.81 ppm; 15.62 ppm; 31.25 ppm; 62.5 ppm; 125 ppm; 250 ppm; 500 ppm; and 1000 ppm. The cytotoxic parameter used was IC50 which was determined using the GraphPad Prism software version 5. The IC50 value of the marine sponge extract Petrosia sp. of 97.20 ppm or 97.20 g/ml with an active category as cervical anticancer. Abstract

Buah walay (Meistera chinensis) mengandung senyawa fenolat, flavonoid, steroid, terpenoid, alkaloid, dan saponin. Beberapa metabolit sekunder, termasuk triterpenoid dan flavonoid, memiliki potensi sebagai antioksidan, antibakteri dan toksisitas. Berdasarkan penelitian sebelumnya dengan menggunakan metode Brine Shrimp Lethality Test ekstrak buah Meistera chinensis memberikan efek sangat toksik dengan IC50 dari 5,02 ± 1,11 mg/mL. Penelitian ini bertujuan untuk mengetahui efek toksik yang ditimbulkan dari ekstrak buah walay ditinjau dari penentuan nilai LD50. Jenis penelitian ini adalah penelitian eksperimental. Penentuan kategori toksisitas berdasarkan Generally Recognized As Safe/GRAS dan perhitungan LD50 menggunakan Reed-Muench Penelitian menggunakan ekor mencit yang dibagi ke dalam 5 kelompok. Kelompok kontrol diberi Na-CMC 0,5 % dan kelompok perlakuan diberi ekstrak buah dengan dosis 50 mg/KgBB; 500 mg/KgBB; 5000 mg/KgBB dan 15.000 mg/KgB Rute pemberian yang digunakan adalah secara oral. Parameter yang diamati dalam penelitian ini adalah gejala toksik yang timbul dan nilai LD50 yang diperoleh berdasarkan perhitungan jumlah hewan uji yang mati. Hasil penelitian menunjukkan bahwa ekstrak buah walay  memberikan gejala toksik berupa penurunan aktivitas gerak, laju nafas cepat serta tremor dan nilai LD50 adalah 865,765 mg/KgBB (Toksisitas ringan). Berdasarkan hasil penelitian dapat disimpulkan bahwa ekstrak buah walay (Meistera chinensis) memiliki efek toksik ringan.

Abstract

Etlingera alba (EA) is one of the endemic plants grown in Sulawesi. We determined the cytotoxic and anti-metastatic activity of isolates obtained from EA rhizome extract. IC50 value for 1,7-diphenyl-6-heptene-3-one, sitostenone, sinapyl alcohol diacetate, and sinapyl alcohol acetate were 134.59; 170 ; 128.11; and 161.96 μg/mL, respectively. The compounds also had anti-migration activity. 200-μg/mL Sitostenon exhibited the lowest migration rate (3.32%). The free bond energies of sitostenon, 1,7-diphenyl-6-heptene-3-one, sinapyl alcohol acetate, and sinapyl alcohol diacetate were -11.81; -8.25; -6.64; and -6.28 kcal/mol, respectively. All compounds were more effective in stabilizing the Matrix metalloproteinase 1 complex. Isolation from EA rhizomes have the potential to be developed as anti-metastatic for Triple-Negative Breast Cancer.
