Abstract

Bakteri Pantoea ananatis merupakan salah satu patogen penting penyebab penyakit hawar daun bakteri pada tanaman bawang merah. Salah satu alternatif pengendalian bakteri P. ananatis yang ramah lingkungan yaitu memanfaatkan aktinobakteria filosfer indigenos. Penelitian ini bertujuan untuk memperoleh isolat aktinobakteria filosfer indigenos bawang merah yang mampu menekan pertumbuhan bakteri P. ananatis secara in vitro dengan menghasilkan senyawa biokontrol, dan yang mampu meningkatkan pertumbuhan tanaman, serta produksi bawang merah dengan menghasilkan senyawa biofertilizer. Penelitian dilakukan secara eksperimen terdiri atas 20 isolat aktinobakteria filosfer indigenos yang diulang 4 kali, disusun menggunakan Rancangan Acak Lengkap, Percobaan dilaksanakan di Laboratorium Mikrobiologi, Departemen Proteksi Tanaman dan kebun percobaan, Fakultas Pertanian, Universitas Andalas, Padang pada Januari-April 2025. Percobaan terdiri atas dua tahap yaitu: 1) pengujian kemampuan aktinobakteria filosfer indigenos dalam meningkatkan pertumbuhan tanaman dan produksi bawang merah, dan 2) pengujian kemampuan aktinobakteria filosfer indigenos terseleksi sebagai agens biokontrol dan biofertilizer. Variabel yang diamati yaitu hari muncul tunas, tinggi tanaman, jumlah daun, bobot segar, dan bobot kering umbi, serta produksi senyawa biokontrol dan biofertilizer. Hasil penelitian menunjukkan bahwa sebelas isolat aktinobakteria filosfer indigenos mampu meningkatkan pertumbuhan tanaman dan produksi bawang merah. Terdapat sebelas isolat aktinobakteria filosfer indigenos mampu menghambat pertumbuhan bakteri P. ananatis secara in vitro, delapan isolat menghasilkan enzim protease, sepuluh isolat menghasilkan enzim amilase, satu isolat menghasilkan hidrogen sianida, dan delapan isolat menghasilkan siderofor. Aktinobakteria filosfer indigenos sebagai biofertilizer, sebanyak 18 isolat mampu memfiksasi nitrogen dan menghasilkan IAA, dan lima isolat mampu melarutkan fosfat. Isolat yang berpotensi sebagai agens biokontrol dan biofertilizer adalah NLLP2H, NBSP3A, NLLA3F, NBSP2H, dan NBSP3F.

Abstract

This community service program was implemented in Koto Malintang, West Sumatra, to empower farmer groups through the development of an Aren sugar (Arenga pinnata) production center. The activities consisted of pre-surveys, focus group discussions (FGDs), socialization workshops, participatory planning, and initial demonstrations of intercropping practices. A key component of the program was the financial feasibility analysis, designed to provide farmers and stakeholders with evidence-based tools for decision-making. The analysis was conducted using production costs and revenues from the research location, with a 15-year horizon and a 10% discount rate. Two cultivation scenarios were evaluated: (1) Aren intercropped with seasonal crops and (2) Aren intercropped with robusta coffee. Each scenario considered two product alternatives: molded sugar (gula cetak) and palm sugar crystals (gula semut). The results revealed that molded sugar offered only marginal feasibility, with Net Present Values (NPV) of Rp 28–35 million/ha, Internal Rates of Return (IRR) of 12–15%, Benefit-Cost (B/C) ratios around 1.1, and payback periods of 7–8 years. In contrast, palm sugar crystals showed strong financial viability, with NPVs of Rp 298–330 million/ha, IRRs of 38–45%, B/C ratios of 2.4–2.6, and payback periods of only 4–5 years. Sensitivity analysis confirmed robustness across discount rates of 8–12% and price variations between Rp 40,000–60,000/kg. These findings suggest that palm sugar crystals should be prioritized as the flagship product for Koto Malintang’s Aren sugar industry, supported by institutional strengthening, diversification, and modern marketing to ensure sustainability.

Abstract

The brown spot disease (BSD) caused by Helminthosporium oryzae in rice can lead to yield losses of up to 45 %. Using endophytic bacterial consortia as a biological agent is one of the most potential control methods for resisting the disease. This study aimed to obtain an endophytic bacterial consortium with the best efficacy in controlling BSD and its ability to enhance rice plant growth. Statistical analysis in this study was made using Analysis of Variance (ANOVA) with Duncan’s Multiple Range Test (DMRT) in a randomized experimental design comprising five treatments with five replications. The treatments included: A (Serratia marcescens ULG1E4 + S. marcescens JB1E3), B (S. marcescens ULG1E4 + S. marcescens JB1E3 + Serratia marcescens JB1E2), C (Bacillus sp. SJI + Serratia marcescens ULG1E4), D (positive control), and E (negative control). Testing was conducted in planta, with the introduction of the bacterial consortium (10⁸ cells/mL) via seed and root soaking methods. Subsequently, the leaves of 30-day-old rice plants were inoculated with H. oryzae by spraying a conidial suspension of 10⁶ conidia/ml. Parameters observed included disease progression (incubation period, incidence, and severity), seedling growth metrics (height, leaf number, root length, fresh and dry biomass), and overall plant development (plant height, leaf number, and tiller count). The results demonstrated that the consortium of Serratia marcescens ULG1E4, Serratia marcescens JB1E3, and Serratia marcescens JB1E2 effectively controlled BSD, achieving a 53.69% reduction in disease severity and promoted rice plant growth, evidenced by a 30.63% increase in leaf number

Abstract

Kegiatan tentang pemanfaatan teknologi pengelolaan organisme penganggu tanaman (OPT) pada tanaman sayuran berbahan baku ramah lingkungan telah dilakukan di Kelompok Tani Mitra di Nagari Lasi Kecamatan Canduang Kabupaten Agam pada bulan September hingga November 2016. Kegiatan ini bertujuan untuk meningkatkan pengetahuan petani tentang serangan OPT sayuran dan bagaimana mengelola OPT menggunakan biopestisida dan pestisida botani. Kegiatan ini meliputi: pemantauan tingkat serangan OPT di lahan mitra, penyuluhan, pelatihan, dan evaluasi hasil kegiatan. Hasil pemantauan tingkat serangan OPT pada tanaman sayuran mitra berkisar antara 18 - 27% untuk penyakit dan 15 - 20% untuk hama. Dari kegiatan dapat dikesimpulkaan yaitu: (1) tingkat OPT pada tanaman sayuran mitra cukup tinggi, (2) Petani belum memahami OPT yang menyerang tanaman sayuran mereka, (3) Petani tidak memahami metode pengelolaan OPT menggunakan biopestisida dan pestisida botani, dan (4) penyuluhan dan praktek lapangan, telah meningkatkan pengetahuan petani tentang pengelolaan OPT sayuran dengan menggunakan biopestisida dan pestisida botani.

Isolation and selection of maize plants rhizobacteria with the potential of entomopathogens against Spodoptera litura F (Lepidoptera: Noctuidae). Biodiversitas 21: 753-758.  Spodoptera litura is a polyphagous pest that can be controlled with entomopathogen. Furthermore, Rhizobacteria derived from the rhizosphere of corn plants are expected to control this pest. This research, therefore, was conducted to obtain the entomopathogenic rhizobacteria to control S. litura derived from the rhizosphere of corn plants. Data sampling was carried out at corn plantations in West Pasaman Regency, while Isolation, selection, and testing of S. litura were conducted at the Biological Control Laboratory of the Plant Protection Department, Faculty of Agriculture, Andalas University. The method uses serial glowing techniques cultured on Nutrient Agar (NA) media, with the selected isolates tested for their ability as an entomopathogen against S. litura. The feed dip method given to S. litura larvae was used to test for rhizobacteria virulence determination with entomopathogen, used to observe the larvae mortality rate, percentage of pupae, and adult formed. Furthermore, observation of morphological and physiological characters was conducted on entomopathogenic rhizobacteria, with hypersensitive testing conducted on Mirabilis jalapa plants. The results of rhizobacterial isolation from corn rhizosphere obtained 12 entomopathogenic isolates. Deadly larvae of S. litura 51-91.11%, 0.00-8.89% pupae, and 0.00- 6.67% adult were formed. The gram test results obtained approximately 8;4 positive (+) and four negative isolates (-), respectively. Hypersensitivity test results found that one of 12 isolates caused necrotic reactions in Mirabilis jalapa. Therefore, a total of 11 rhizobacteria isolates are entomopathogenic and safe to be used as S. litura biological control agents.

The potential of beneficial microbes to suppress the development of bacterial leaf blight in rice plants caused by Xanthomonas oryzae pv. oryzae. Biodiversitas 24: 4209-4217. Xanthomonas oryzae pv. oryzae (Xoo) is caused bacterial leaf blight in rice plants. One technique for controlling the bacterial leaf blight that currently being developed is using beneficial microbes. The purpose of this study was to isolate and identified beneficial microbes that can suppress bacterial leaf blight development in rice plants. In the present study fifteen isolates of beneficial microbes were investigated for their potential as an antagonist against Xoo, the causal agent of bacterial leaf blight of rice plants. Four isolates, namely Act-SK2, Act-Mn2, Act-Hr21, and Act-Pha4, showed the ability to reduce the area under progress curve (AUDPC) values by 51.33, 51.00, 84.00, and 82.33, respectively. The effectiveness of AUDPC suppression ranged between 80%-88%. Two out of four isolates showed potential in reducing the severity of BLB disease by 3.65% (Act-Sk2) and 3.88% (Act-Mn2) as compared to control. The effectiveness of both isolates in reducing disease severity was 86.34% (Act-Sk2) and 85.48% (Act-Mn2). Furthermore, identification based on 16S rRNA gene sequence analysis showed that three isolates, namely Act-Sk2, Act-Hr21, and Act-Pha4, belong to Actinobacteria. Each isolate showed 99.68% similarities with Streptomyces sp. strain KS02 (Acc. No. AB373961, 99.84% with Streptomyces sp. strains Al-Dhabi-119 (Acc. No.MK675528), and 99.64% with Streptomyces griseus strain K 2 (Acc. No. MK811436). Isolate Act-Mn2 showed 100% similarity with Penicillium janthinellum strain CMV006C1 (Acc. No. MK450697). The results of this study indicate that the four microorganisms tested have potential to be developed as biological agents.
