Abstract

ABSTRACT The objective of this research was to produce an alternative media for aquatic bacteria using fish broth as pepton source and commercial consumption agar as material. This experiment consisted of six treatments; four treatments used fish broth with doses 200 g/L; 400 g/L; 600 g/L and 800 g/L; two treatments as controls which were commercial agar as negative control and tryptic soy agar (TSA) as positive control. The result showed that treatment 200 g/L had performed as good as TSA for bacterial growth. Keywords: solid media, fish broth, aquatic bacteria  ABSTRAK Tujuan dari penelitian ini adalah menciptakan alternatif media padat nonselektif untuk bakteri akuatik dengan memanfaatkan kaldu daging ikan sebagai sumber pepton dan agar-agar konsumsi komersial (AKK) sebagai pemadat. Penelitian ini terdiri atas enam perlakuan yaitu; empat perlakuan menggunakan kaldu ikan yang dibuat dengan dosis 200 g/L; 400 g/L; 600 g/L and 800 g/L; dua perlakuan sebagai kontrol yaitu ART sebagai kontrol negatif dan tryptic soy agar (TSA) sebagai kontrol positif. Hasil penelitian menunjukkan bahwa pemanfaatan kaldu daging ikan pada dosis 200 g/L memiliki performa yang sama baiknya dengan TSA sebagai media tumbuh bakteri. Kata kunci: media padat, kaldu ikan, bakteri akuatik

Abstract

Feed is an important factor in the success of cultivation, feed is also the component that costs the most that is about 70% of the total production cost. Therefore, it is necessary to do techniques that can increase production, one of which is by improving feed nutrition using EM4 probiotics. In addition, it can also be by applying bioflok technology that utilizes bacteria that can process aquaculture waste which is a problem in water quality into a continuously available source of fish feed.  This study aims to find out if the application of bioflok and feed technology with EM4 can increase cultivation production. There are 3 treatments that will be used, namely, P1 (bioflok application), P2 (EM4 application), P3 (without bioflok and EM4). The fish used is pearl catfish seed (Clarias gariepinus) with a size of 6.5±0.5 cm with a dense spread of 150 heads / m3. Maintenance is carried out for 30 days (1 month) with the frequency of feeding

Abstract

Minyak cengkeh telah banyak digunakan oleh pembudidaya sebagai anestesi pada transportasi ikan. Akan tetapi penggunaannya pada beberapa jenis ikan dengan kepadatan yang berbeda menunjukkan kebutuhan dosis penggunaan yang berbeda. Tujuan penelitian ini adalah untuk menentukan dosis minyak cengkeh sebagai bahan pembius ikan tambakan (Helostoma temminckii) dan untuk mengetahui pengaruh pemberian minyak cengkeh pada transportasi ikan tambakan. Penelitian ini membandingkan kondisi transportasi yang menggunakan minyak cengkeh dan tanpa minyak cengkeh (kontrol) dengan kepadatan berbeda. Kepadatan yang digunakan adalah 10, 12, dan 14 ekor L-1. Ikan tambakan yang digunakan berukuran 15 ± 0,5 cm. Penelitian ini dibagi menjadi dua tahap yang terdiri dari penentuan konsentrasi efektif-100 10 menit (EC100 10 min) dan pengaruh minyak cengkeh terhadap kepadatan selama 12 jam transportasi. Hasil penelitian menunjukkan bahwa nilai EC100 10 min adalah 0,026 mL L-1. Sesaat setelah transportasi, tingkat kelangsungan hidup dan tingkat konsumsi oksigen tidak berbeda nyata antara perlakuan dan kontrol pada kepadatan yang berbeda. Akan tetapi kadar glukosa darah pada kepadatan 10 ekor L-1 dan 12 ekor L-1 lebih rendah daripada kontrol. Pemantauan kesehatan ikan setelah 7 hari pascatransportasi menunjukkan bahwa tidak ada perbedaan nyata antara kontrol dan perlakuan baik kelangsungan hidup maupun kadar glukosa darah. Oleh karena itu, pemanfaatan minyak cengkeh pada dosis 0,026 mL L-1 untuk transportasi ikan tambakan dapat diaplikasikan dengan kondisi kepadatan 14 ekor L-1 selama 12 jam. Pemanfaatan minyak cengkeh pada ikan tambakan perlu dilakukan kajian lebih lanjut terkait penambahan kepadatan dan waktu transportasi atau aplikasinya untuk transportasi benih ikan tambakan.Clove oil has been widely used by farmers as an anesthetic agent in fish transportation. However, its use on several fish with different densities indicates the need for different doses. The aim of this study was to determine the dose of clove oil as an anesthetic agent for kissing gourami (Helostoma temminckii) and to determine the effect of administering clove oil on the transportation of kissing gourami. This study compared transportation conditions using clove oil and without clove oil (control) with different densities. The densities used were 10, 12, and 14 L-1. The kissing gourami used measured 15 ± 0.5 cm. This study was divided into two stages consisting of determining the effective concentration-100 10 minutes (EC100 10 min) and the effect of clove oil on density during 12 hours of transportation. The results showed that EC100 10 min value was 0.026 mL L-1. Immediately after transportation, survival rates and oxygen consumption levels were not significantly different between treatments and control at different densities. However, blood glucose levels of 10 fish L-1 and 12 fish L-1 were lower than the control. Fish health status monitoring after 7 days post-transportation showed that there was no significant difference between control and treatment in terms of survival rate or blood glucose levels. Therefore, the use of clove oil at a dose of 0.026 mL L-1 for the transportation of kissing gourami can be applied at a density of 14 fish L-1 for 12 hours. Further studies are required to determine the effects of clove oil as an anesthetic agent applied at denser stocking densities and longer transportation period of kissing gourami seeds.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun nipah sebagai imunostimulan pada ikan nila. Rancangan percobaan menggunakan rancangan acak lengkap (RAL) yang terdiri atas empat perlakuan berupa suplementasi dosis ekstrak daun nipah yang berbeda dalam pakan yaitu tidak diberi ekstrak daun nipah (P0), serta supplementasi ekstrak daun nipah sebanyak 1 g kg-1 (P1), 1,5 g kg-1 (P2), dan 2 g kg-1 (P3) pakan. Ikan nila yang telah diberi pakan perlakuan masing-masing selama 14 hari, diuji tantang dengan bakteri Aeromonas hydrophila melalui perendaman selama 60 menit dengan kepadatan bakteri 106 cfu mL-1 pada hari ke-15. Parameter diamati antara lain gejala klinis, kadar hematokrit (He) sebelum dan sesudah infeksi, prevalensi, pertumbuhan bobot dan panjang mutlak, dan kelangsungan hidup. Data dari semua parameter dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa penambahan ekstrak daun nipah memberikan pengaruh terhadap pertumbuhan, nilai hematokrit, prevalensi, kelangsungan hidup, dan gejala klinis ikan nila yang diinfeksi A. hydrophila. Perlakuan penambahan ekstrak daun nipah sebanyak 2 g kg-1 (P3) menunjukkan hasil yang lebih baik dibandingkan perlakuan lainnya yaitu gejala klinis yang lebih ringan, kadar hematokrit setelah infeksi 41,86-43,47%, kelangsungan hidup setelah infeksi 96,33%, prevalensi 4,16%, pertumbuhan panjang mutlak 1,60 cm, dan pertumbuhan bobot mutlak 4,87 g. Hal ini berhubungan dengan adanya senyawa bioaktif pada daun nipah yang berperan sebagai imunostimulan dan antibakteri. Oleh karena itu, dapat disimpulkan bahwa pemberian ekstrak daun nipah sebanyak 2 g kg-1 efektif berperan sebagai imunostimulan dan dapat diaplikasikan untuk meningkatkan produksi budidaya ikan nila.This study aimed to determine the effect of nipa leaves extract as an immunostimulant in tilapia. The experimental design used a completely randomized design (CRD) consisting of four treatments in the form of supplementation with different doses of nipa leaves extract in the feed, namely no nipa leaves extract (P0), and supplementation of nipa leaves extract as much as 1 g kg-1 (P1), 1.5 g kg-1 (P2), and 2 g kg-1 (P3) feed. Tilapia fish that had been given experimental feed for 14 days were challenged with Aeromonas hydrophila bacteria by soaking for 60 minutes with a bacterial density of 106 cfu mL-1 on day 15. Parameters observed included clinical symptoms, hematocrit levels (He) before and after infection, prevalence, absolute weight and length growth, and survival. Data from all parameters were analyzed descriptively. The results of the study showed that the addition of nipa palm leaves extract had an influence on the growth, hematocrit level, prevalence, survival, and clinical symptoms of tilapia infected with A. hydrophila. Treatment with the addition of 2 g kg-1 nipa leaves extract (P3) showed better results than other treatments, namely lighter clinical symptom, hematocrit levels after infection of 41.86-43.47%, survival after infection of 96.33 %, prevalence of 4.16%, absolute length growth of 1.60 cm, and absolute weight growth of 4.87 g. This is related to the presence of bioactive compounds in nipa palm leaves which act as immunostimulants and antibacterials. Therefore, it can be concluded that giving 2 g kg-1 of nipah leaves extract effectively acts as an immunostimulant and can be applied to increase the production of tilapia farming.

Abstract

Coffee husk fermentation (CHF) might be utilized as an alternative plant protein source. This research evaluated dietary CHF supplementation's effects on growth for striped catfish. This study consisted of five grade levels of CHF as a treatment. The treatments were P0 (without the addition of CHF), P1 (addition of 6% CHF), P2 (addition of 12% CHF), P3 (addition of 18% CHF), and P4 (addition of 24% CHF). The results determined that dietary CHF inclusion had no significantly different effect on specific growth rate (SGR) and feed efficiency (P > 0.05). However, the P1 group (6% CHF) produced the best protein retention (PR), which was significantly different from the control (P0) and other CHF treatments (P < 0.05). No significant differences were observed in survival rates among the treatments. The inclusion of CHF up to 24% in the diet was found acceptable, as it did not negatively impact the striped catfish’s overall health or growth performance. Although growth rate, feed efficiency, and survival did not significantly differ (P > 0.05).

Abstract

Bilimbi (Averrhoa bilimbi) is plant medicinal used to control fish diseases both prevention and treatment. This study aims to determine the protection period of bilimbi-fed which different duration in catfish to enhance catfish (Clariassp.) immunity in order to protect fish fromAeromonas hydrophilainfection. The Randomized Block Design Factorial (RBDF) with 2 factors was used in this experimental. The first factor (A) was the period of feeding containing bilimbi juice consisting of 3 treatments, namely 0 days (P0), 7 days (P1), and 14 days (P2). The second factor (B) was the protection period after the administration of bilimbi juice to the infection ofA. hydrophilabacteria consisting of 3 treatment, 5 days (T5), 10 days (T10), and (T15). This research was conducted at the Laboratory of Aquaculture Experiment Pond, Faculty of Agriculture, Sriwijaya University in October-December 2018. The parameters observed included prevalence of infection, growth, survival, feed efficiency and water quality. The result revealed that by giving 300 mL.kg-1 of bilimbi juice with different periods significantly affected the period of fish protection forA. hydrophilainfection.The best prevalence value was 3,33% (P2T5),and the best survival of the fish before infection was 94,17% (P2), while the best fish survival post infection was 100% (P2T5), the best feed efficiency value was 56,47% (P2), water quality under experiment were 26-29,70C for temperature, pH 6.8-7.7, dissolved oxygen 4.35-5.37 mg/L and ammonia 0.09-0.21 mg/L.Keywords:Averrhoa bilimbiAeromonas hydrophilaEthnomedicinal plantFish immunityMotile Aeromonas Septicemia
